Variation of Anthocyanins and Isoflavones between Yellow-Cotyledon and Green-Cotyledon Seeds of Black Soybean

  • Analysis of black soybeans [Glycine max (L.) Merr.; 59 Korean varieties] revealed that 100-seed weights of green cotyledon seeds (33.5 g, n=31) were higher than those of yellow ones (28.9 g, n=28). Contents of delphinidin-3-glucoside (D3G), cyanidin-3-glucoside (C3G), petunidin-3-glucoside (P3G), and total anthocyanins in seed coats of black soybeans were 0.03-4.15, 0.74-18.36, 0.02-1.60, and 0.87-23.52 mg/g, respectively, among which most prominent anthocyanin was C3G (80.9% of total content), followed by D3G (13.6%) and P3G (5.5%). No significant differences were observed in color parameters $a^*$ and $b^*$ between black soybeans with yellow cotyledon (BYC) and green cotyledon (BGC). Total isoflavone content of BGC was higher than that of BYC, and negative correlation was found between total anthocyanin and isoflavone contents.

멜론(Cucumis melo L.) 유묘 절편으로부터 고빈도의 체세포배발생과 식물체 재분화 (High Frequency Somatic Embryogenesis and Plant Regeneration in Seedling Explant Cultures of Melon (Cucumis melo L.))

  • 멜론의 배발생 캘러스의 유도와 식물체 재분화 시스템을 확립하기 위해서 자엽과 배축의 절편을 여러 농도의 2,4-D 와 BA가 첨가된 MS 기본배지에 치상하여 관찰한 결과, 초기배양에서는 4 mg/L 2,4-D와 1 mg/L BA 그리고 2 mg/L 2,4-D+O.1 mg/L BA가 첨가된 배지에서 연한 노란색의 배발생 캘러스가 형성되었고, 위의 조건에서 형성된 캘러스로부터 배발생 캘러스는 0.5와 1 mg/L 2,4-D가 첨가된 동일배지에서 약 75%가 형성되었다. 그리고 생장조절제를 첨가하지 않은 기본배지에서 대부분의 체세포배는 식물체로 재분화되었으며, 두개의 자엽을 갖춘 정상적인 체세포배보다 비정상적인 나팔형의 자엽을 가진 것이 3배 높은 빈도로 나타났다. 이러한 체세포배들은 MS 기본배지에서 식물체로 발아되었으며 생육상에서 생장하여 견실한 열매를 맺었다.

모/폴리에틸렌테레프탈레이트 복합막의 제조와 염색성 (Preparation of Wool/Poly(ethylene terephthalate) Composite Membrane and It's Dyeablities)

  • Composite membranes having different mixing ratio of Wool(SCMK) and poly(ethylene terephthalate) (PET) were prepared by dissolving wool/PET in hexafluoro-2-propanol(HFIP), casting the obtained solution on a glass plate and evaporation the solvent in the presence and absence of an electric field. The internal structure of the prepared membrane was investigated using polarise microscope dyeing and dye permeation method. In the composite membrane prepared under electric field, both components were micro mixing, while in the membranes prepared under nonelectric field, the two components formed a random sea/island structure according to different mixing ratio. Such characteristic membrane structure was influenced the permeation behavior of C.I. Acid Red 118 through the membranes from an aqueous solution.

Novel strategy for isolating suppressors of meiosis-deficient mutants and its application for isolating the bcy1 suppressor

  • A novel strategy was developed for isolating suppressors from sporulation-deficient mutants. The mutation in the BCY1 gene, which codes for the regulatory subunit of cAMP-dependent protein kinase, when homozygous, results in diploids being meiosis and sporulation deficient. Two plasmids, YCp-MAT.alpha. and YEp-SPOT7-lacZ, were introduced into MAT.alpha. BCY1$\^$+/ or MAT.alpha. bcy1 haploid cells. The transformant of the BCY1$\^$+/ haploid cell produced .betha.-galactosidase under nutrient starvation, but the bcy1 transformant did not. Using this system, the mutagenesis experiment performed on the bcy1 transformant strain resulted in a number of sporulation mutants that produced .betha.-galactosidase under nutrient starvation. One complementation group, sob1, was identified from the isoalted suppressor mutants and characterized as a single recessive mutation by tetrad analysis. Genetic analysis revealed that the sob1 mutation suppressed the sporulation deficiency, the failure to arrest at the G1 phase of the cell cecle, and the sensitivity to heat or nitrogen starvation caused by the bcy1 mutation. However, the sob1 mutation did not suppress the sporulation deficiency of ime1 and of ime2 diploids. These results suggest that the sob1 mutation affects a gene which functions as a downstream regulator in both meiosis and cell cycle regulation.

국내 양말 소비자의 브랜드 인지도 및 디자인 선호도 분석 (Analysis of Korean Consumer Brand Awareness and Preferred Types of Sock Design)

  • The purpose of this study was to develop a large representative data base for socks marketing strategy. The study was to determine socks brand awareness in ralation to segmented distribution regions and such demographic variables as sex and age. The authors also analyzed preferred design. A total of 650 questionnaires were distributed and 611 reliable ones were used for statistical analysis. A SAS statistical package including frequency tables and chi square test was used. The results are as follows: Brand awareness involves "brand recall" based on asking a person to name the brand he or she recalls first, and also "brand recognition" based on asking subjects to identify brand names from 30 given brands. 'SOCKSTOP' was found to be a dominant brand as a result of the brand recall test, and 'BYC' was found to be a dominant brand as a result of the brand recognition test. Brand recognition was significantly different in the segmented distribution regions, three age groups and different sex groups. People considered design first in purchasing sports/casual socks, while they considered color first in purchasing dress socks. The most favored type of sock style was the common crew sock type. The results of a chi square test showed that preferred type of sock design was related with the sex and age variables.
