This study was performed to evaluate the cytotoxicity induced by ischemia-like condition (ILC) in cultured neuroglial cells (C6 glioma cells). The protective effect of kaempferol (KAE), flavonoid against the cytotoxicity induced by ILC induction was assessed. In addition, antioxidative effects of KAE were done by colorimetric assays. Cell viability and the antioxidative effects such as DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lipid peroxidation (LP) were analyzed. ILC induction decreased cell viability in a dose-dependent manner, and the $XTT_{90}$ value (low cytotoxicity value) and $XTT_{50}$ value (high cytotoxicity value) were determined during ILC induction for 15 and 40 minutes, respectively. The butylated hydroxytoluene (BHT) antioxidant significantly increased cell viability damaged by the ILC-induced cytotoxicity. In the protective effect of KAE on ILC-induced cytotoxicity, KAE protected the ILC-induced cytotoxicity by the significant increase of cell viability, and also it showed DPPH-radical scavenging ability, SOD-like ability and inhibitory ability of LP. From these results, it is suggested that ILC induction showed cytotoxicity in these cultures and the oxidative stress is involved in the ILC-induced cytotoxicity. While, KAE prevented ILC-induced cytotoxicity by antioxidative effects. In conclusion, natural products like KAE may be a putative therapeutic agent for the treatment of disease associated with oxidative stress such as ischemia.

In our search for bioactive phenolics from plants, the culms of Phyllostachys bambusoides has been selected for investigation of anti-cariogenic and 1,1-diphenylpicrylhydrazyl (DPPH) radical scavenging agents based on the initial screening results. Fractionation process of n-hexane and $CHCl_3$ extracts afforded four phenolic constituents, ferulic acid (1), vanillin (2), coniferaldehyde (3), and coniferyl alcohol (4) as guided by their DPPH free radical scavenging activities. Additionally, activity-guided fractionation of EtOAc extract with anti-cariogenic activity has resulted in the isolation of coniferaldehyde (3), 2,6-dimethoxy-p-benzoquinone (5), p-methoxycinnamic acid (6), (${\pm}$)-balanophonin (7), and 6-methoxychromanone (8). The structures of 1 - 8 were determined by spectroscopic data interpretation, and also by comparison of their data with the published values. Phenolic compounds 1 - 4 exhibited similar DPPH radical scavenging activities compared with the synthetic antioxidant, butylated hydroxytoluene (BHT), and compounds 3 and 5 - 8 showed significant antibacterial activity against cariogenic oral streptococci, Streptococcus mutans and S. sobrinus.
The purpose of this study is to analyze antioxidant activity and multidrug resistance reversing activity in several Korean colored soybean (Glycine max Merr.) cultivars. Antioxidant activity of methanol extracts from colored soybean cultivars was evaluated by TBARS (thiobarbituric acid reactive substances), DPPH (1, 1-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) methods. By means of TBARS, cultivar "Jeonnam #1" showed the highest activity until 7 days, and followed by "Black #1", "Jinyul" and "Black #3", showing lower activity than that of BHT (butylated hydroxytoluene). Methanol extracts of all cultivars proved that DPPH radical scavenging activity is dose-dependent. Methanol extract from cultivar "Jeonnam #1" showed highest DPPH radical scavenging activity, and followed by cultivars "Black #1". MDR (multi-drug resistance reversing) activity, however, showed the highest effect in "Black #3" and the lowest "Black #1" cultivar. These results suggest that seed colors of soybean may play an important role in antioxidant activity and MDR activity.
Crude extracts of thirty seaweeds collected in Korea were obtained using 50% ethanol, and total phenolic contents and antioxidant activities were compared. Two brown algae, Ecklonia cava (E. cava) and Sargassum siliquastrum (S. siliquastrum), showing high antioxidant activity based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide anion radical scavenging activity were further investigated for their inhibitory effects on tyrosinase activity. The E. cava extract had the highest total phenolic content among the seaweeds extracts. Total phenolic contents were strongly correlated with antioxidant activity in the thirty seaweed extracts ($R^2\;=\;0.9169$). The E. cava and S. siliquastrum extracts exhibited higher inhibition to tyrosinase activity than butylated hydroxytoluene (BHT) and epigallocatechin gallate (EGCG).
The present GC-MS analysis elucidated the composition of the essential oil obtained from the herb of Elsholtzia ciliata(Lamiaceae). Overall, the content of monoterpenes was higher than that of sesquiterpenes. Monoterpenes rich in this oil were carvone (peak area, 26.180%), camphor (2.304%), borneol (9.974%), dihydrocarveol (3.296%), ${\alpha}$-citral (=geranial, 4.025%), geranic acid (2.961%), while sesquiterpenes occupying relatively higher percentage were ${\alpha}$-humulene (0.918%), (-)-spathulenol (0.974%), ${\alpha}$-caryophyllene oxide (2.014%), globulol (1.362%), ${\beta}$-caryophyllene oxide (0.750%). The components characterizing this oil were 1-octen-3-ol, acetophenone, and butylated hydroxytoluene. The $IC_{50}$ of this oil on acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) were $42.37{\mu}g/ml$ and $121.34{\mu}g/ml$, respectively, suggesting that the essential oil of E. ciliata may be active on the memory loss of patients suffering from Alzheimer's disease.
The antioxidative activity of Alpinia oxyphylla was investigated through measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and inhibitory activity for linoleic acid peroxidation. Two antioxidative diarylheptanoids, yakuchinone A (1) and oxyphyllacinol (2), were isolated from the fruits of A. oxyphylla using thin layer chromatography (TLC) autographic assays. The DPPH scavenging activities of the compounds ($IC_{50}=1$, $57{\pm}2.1\;{\mu}M$; 2, $89{\pm}3.1\;{\mu}M$) were lower than vitamin C ($IC_{50}=51{\pm}1.1\;{\mu}M$), but higher than butylated hydroxytoluene (BHT, $IC_{50}=99{\pm}2.2\;{\mu}M$). Also, inhibitory activities for linoleic acid peroxidation of the compounds ($IC_{50}=1$, $0.19{\pm}0.011\;mM$; 2, $0.31{\pm}0.009\;mM$) were higher than those of vitamin C ($IC_{50}=0.59{\pm}0.017\;mM$) and BHT ($IC_{50}=0.52{\pm}0.014\;mM$). In addition the $^{13}C-NMR$ data of oxyphyllacinol (2) have been first reported in this paper.
This study investigated the effects of bovine plasma protein (PP) hydrolysates on the antioxidant and quality properties of pork patties during storage. Pork patties were divided into 4 groups: without butylated hydroxytoluene (BHT) and PP hydrolysates (control), 0.02% BHT (T1), 1% PP hydrolysates (T2), and 2% PP hydrolysates (T3). Pork patty supplemented with PP hydrolysates had higher pH values and lower weight loss during cooking than the control patties. Results showed that lightness and hardness both decreased upon the addition of PP hydrolysates. All samples containing BHT and PP hydrolysates had reduced TBARS and peroxide values during storage. In particular, 2% PP hydrolysates were more effective in delaying lipid oxidation than were the other treatments. It was concluded that treatment with 2% PP hydrolysates can enhance the acceptance of pork patty.
This study was conducted to investigate the extraction characteristics including total polyphenol compound content (TPC) and the antioxidative effect of Rumex crispus root extract on commercial corn oil. Extraction yield was increased with extraction temperature but decreased with extraction ratio. No significant differences in aromatics content were found among the extracts prepared with various extraction conditions; extraction ratio, extraction temperature, extraction pH and composition of extractant. Total flavonoid content of the extract was increased with extraction temperature and extraction pH, and highest value of it was found when extractant composition of ethanol in water was 75%. Total polyphenol compound content (TPC) of the extract with 10 fold extraction ratio showed the highest value, but no significant difference in TPC was found with extraction temperature. Reducing power and DPPH (${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging ability (RSA) of the extract in spectrophotometric absorbance were decreased with extraction ratio but increased with extraction temperature and showed 63.1%~98.4% and 67.6%~86.7% of those of ascorbic acid in reducing power and DPPH RSA, respectively. The antioxidative effects of the extract on corn oil were 84.8~93.0% of that of commercial butylated hydroxytoluene, the antioxidant index value was highest when the ethanol composition to water in extractant was equal ratio.
Several wild plant species are known to contain biologically active substances that are allelopathic to weed species as well as antioxidant to foods. Plant extracts or residues from leaves of 4 species, Achyranthes japonica (speedwell), Cucumis sativus (Cucumber), Trifolium repens (white clover), and Vicia angustifolia (narrowleaf vetch) were bioassayed against Medicago sativa (alfalfa) or Echinochloa crus-galli (barnyard grass) to determine their allelopathic effects, and used for measurement of antioxidant activities. The aqueous extracts applied on filter paper significantly inhibited root growth of alfalfa. Aqueous extracts or residues from V. angustifolia showed the most inhibitory effect on alfalfa or barnyard grass seedling growth and followed by A. japonica and T. repens. Oxidative stability by Rancimat method, antioxidant activity by TBA (2-thiobarbituric acid) method and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity for the ground samples or methanol extracts were the greatest in V. angustifolia, although were less than those of commonly used antioxidants, BHT (butylated hydroxytoluene) and ascorbic acid. These results suggest that the wild plant species had potent allelopathic and antioxidant activities, and that their activities differed depending on plant species.
Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts ($319.2\;mg\;kg^{-1}$) as the greatest component, and followed by A. lappa ($96.3\;mg\;kg^{-1}$) and Y. sonchifolia ($22.9\;mg\;kg^{-1}$). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg $100\;g^{-1}$) and followed by Y. sonchifolia (11.2 mg $100\;g^{-1}$) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg $100\;g^{-1}$ exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity ($r^2$ = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.
