Relationship between Volatile Oil Components of Tobacco and Sensory Attributes of Tobacco Smoke (잎담배의 휘발성 정유성분과 담배연기의 관능특성과의 관계)

  • This study was conducted to evaluate the prediction of sensory attributes of tobacco smoke by the use of volatile oil components of tobacco. For analytical and sensory evaluations, twelve aging tobaccos (i.e., 3 crop years; 1998, 1999, 2000, and 4 stalk positions) were prepared in flue-cured and burley tobaccos. 61 volatile oil components and 5 sensory attributes such as irritation, impact, after taste, bitter and green were investigated. Irritation of flue-cured tobacco, and irritation and impact of burley tobacco were significantly increased with the ascending stalk position, whereas after taste of burley tobacco was significantly decreased. Significantly positive correlations among irritation, impact, and bitter were observed in flue-cured tobacco. A significantly positive correlation between irritation and bitter was observed, significantly negative correlations between after taste and irritation and between after taste and impact were observed in burley tobacco. Except for green of burley tobacco, all probabilities of multiple linear regression equations between volatile oil components of tobacco and sensory attributes of tobacco smoke were significant(P$\leq$0.05). This study suggests that the multiple linear regression equations may be useful to predict the sensory attributes of tobacco smoke with a few selected volatile oil components of tobacco.

Varietal Response to Days to Flower and Number of Leaves under Cool Temperature and Short-day in Burley Tobacco (Nicotiana tabacum L.) (연초 버어리종의 저온단일에 따른 품종간 개화일수 및 엽수 반응)

  • This experiment was conducted to investigate varietal responses to cool temperature (18℃) and short photoperiod (8 hours a day) in a controlled room of phytotron. Five burley tobacco varieties (Nicotiana tabacum L.), Awelius (very early flowering type), Burley 21 and KB 103 (medium), Banket Al and MB 1 (slightly late) and TC 681 (late) were compared in number of leaves per plant and day to flower in different treatment periods (10, 15, 20, 25 and 30 days). The days to flower decreased significantly than the control (in the phytoron with 14hr photoperiods and a minimum temperature 20'c) when the treatment periods were over 15 days for the varieties Aurelius, Burley 21 and KB 103, and 20 days for TC 681, and 30 days for Banket Al, respectively. And also the number of leaves decreased significantly than that of the control when the periods of treatment were over 10 days for Burley 21 and Aurelius, 15 days form 103, and 20 days for TC 681 and Banket Al, and 30 days for MB 1, respectively. The decreasing ratio in the number of leaves per plant under cool temperature and short photoperiod treatments was the lowest in Banket Al and MB 1. Relationships of Midrib Ratio to Some Agronomic Characters in Burley Tobacco(N.tabacum L.) (버어리종 잎담배의 주맥비율과 형질 상호간의 관련성)

  • This study was conducted to find out the relationship among agronomic characteristics including midrib ratio in Burley tobacco(N.tabacum L.) Four cultrivars, Burley 21, KB108, KB111 and KB301, were grown at Suwon Experimental Station in 2001. Several agronomic characteristics of these cultivars were measured for the analysis of correlations among agronomic characteristics. Midrib ratio of KB301 was somewhat higher than those of the other cultivars, but there is no significant differences among cultivars at $5\%$ probability level. Among four stalk positions, midrib ratio of cutters was the highest. Midrib ratio was correlated positively with length of leaf at $1\%$ level and negatively with weight of lamina at $5\%$ level, respectively.

Changes in Physico-Chemical Properties of Burley Leaf Tobacco by Expansion Process. (팽화처리에 의한 Burley종의 물리화학적 특성변화)

  • This study was carried out to compare the change physico-chemical properties in the 10 grades of Burley leaf tobacco expanded by $CO_2$. The filling capacity of AB3T was increased from 5.44cc/g to 9.52cc/g with the expansion rate 75.0% and that of CD3W was increased from 5.57cc/g to 10.16cc/g with the expansion rate 82.4%. But the rate of cut tobacco longer than 3.36 mm decreased from 77.2% to 49.9% and from 67.3% to 41.2% in grade B1T and C1W, respectively. The contents of nicotine and total volatile base in the of expanded cut tobacco decreased 11.9% and 10.4% respectively. The contents of onganic acids, except oxalic and palmitic acid, essential oil, amino acids, nicotine, tar and ammonia contents in the cigarette smoke decreased by expansion procecs. These results suggest that expanded tobacco was very useful to make less irritative cigarettes.

  • The uncertainty of measurement in nitrate from burley leaf tobacco by continuous-flow analysis method was evaluated following internationally accepted guidelines. The sources of uncertainty associated with the analysis of nitrate were weight of standard and sample, purity of standard, dilution of standard solution, calibration curve, water content, etc. The calculation of uncertainty based on the GUM(Guide to the Expression of Uncertainty in Measurement) and EURACHEM/CITAC Guide. An expanded uncertainty was obtained by multiplying the combined standard uncertainty with a coverage factor (k) calculated from the effective degree of freedom. The concentration of nitrate from burley leaf tobacco was $2.09\%$ and the expanded uncertainty by multiplying by the coverage factor(k, 2.20) was $0.13\%\;at\;a\;95\%$ confidence level.

Molecular Cloning of Putrescine N-Methyltransferase Gene from Burley 21 Tobacco (Burley 21 담배에서 Putrescine N-Methyltransferase 유전자의 클로닝)

  • Recently, many researches for plant alkaloids, one of the largest groups of natural products, are reported because of their various pharmacological activity. This study was carried out to clone putrescine N-methyltransferase (PMT) gene which is a key enzyme in diverting polyamine metabolism towards the biosynthesis of nicotine and related alkaloids from Burley tobacco. To induce expression of PMT gene in tobacco plant, the floral meristem was removed and then mRNA was purified from root. cDNA encoding PMT gene was isolated by RT PCR and cloned. Three different groups of clones were screened by PCR and restriction enzyme digestion analysis and were characterized. The data of these screening revealed that three types of PMT are present in Burley tobacco. Comparison of the nucleotide sequence of this three genes encoding putative PMT with those of other tobaccos revealed that two types of PMT are newly discovered from Nicotiana tabacum cv. Br21 tobacco and they were same as PMT2, PMT3 of N. tabacum cv. Xanthi.

Inheritance of DVT and Cis-abienol in F$_1$ Generation among Burley and Sun-cured Cultivars of China(N. tabacum L.) (연초(N. tabacum L.) 버어리종과 중국 양건종 교잡 후대의 DVT 및 cis-abienol 유전)

  • To obtain the genetic information about DVT( $\alpha$- and $\beta$-4,8,13- duvatriene-1,3-diols) and cis-abienol contents, two burley(Burley 21, KB 108), three sun-cured cultivars(N. tabacum L. cv. Yonbyun 3, Jahengyon and Jaraehong), TI 1068, and thirty F$_1$ hybrids derived from them were sampled and the diterpenes were analysed using thin layer chromatography (TLC) procedures. DVT exudation from the leaf scerface could Ie detected in all coltivars and F$_1$ hybrids tested. TI 1068 and Yonbyun 3 had cis-abienol exdudates. Burley 21, KB 108, Jaheungyon, and Jaraehong had no spot of cis-abienol. It is considered that cis-abienol exdudation might be controlled by dominant gene(s). The cytoplasmic effect on the cis-abienol exdudation was not detected.

Varietal Difference of Sucker producing Characteristics in Burley Tobacco(Nicotiana tabacum L. var. burley) (버어리종 연초 곁순발생의 품종간 차이)

  • This study was carried out to obtain the basic information about the sucker producing characteristics of certain burley tobacco varieties(Nicotiana tabacum L. var. burley) at Suwon and Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 1999. The number and weight of suckers for 12 varieties or breeding lines were investigated three times, 45 days after transplanting for ground sucker, 10 days after topping, and 10 days after maleic hydrazide(MH) applying for upstalk sucker. KB 103 and KB 9416-1 produced less ground suckers and upstalk suckers than other entries did. KB 9210-8 produced less ground suckers and (Male sterile TC 612 x KB 108)F$_1$ also produced less upstalk suckers than other entries did. There was no differences among entries in sucker production at 10 days after MH treatment. Between the weight of total sucker and days to flower revealed the negative correlation, but not significant. The low sucker producing entries described above could be used as a parental line in the breeding program for the low sucker producing variety.

Mechanical properties of Korea Tobacco (한국산 잎담배의 역학적 특성)

  • The tensile strength (Pm) . elongation percentage ($\varepsilon$) , elastic modulus (I) , and specific volume (Sv) were calculated from the data of tensile characteristics of By 104 .and Burley 21 tobacco leaves, which were measured over a relative humidity from 60 to 80% at $20\pm2^{\circ}C$.The important results obtained were as follows.(1) Tensile strength(Pm) The "Pm"of By 104 was greater than that of Burley at. The "Pm"in both varieties were increased toward the upper stalk position and according to the decrease of relative humidity. The "Pm" of parallel piece to tile vein in a leaf were greater than that of right angle piece to the vein. (2) Elongation percentage ($\varepsilon$). The "s" of By 104 was higher than that of Burley 21. The "s" in both varieties were increased toward the upper stalk position and according to the increase of relative humidity. The "s" of parallel Piece to the vein in a Burley leaf was greater than that of right angle piece to the vein. (3) Elastic modulus (E) The "E" of Burley 21 leaf was higher than that of By 104. The "E" in both varieties were increased toward the lower stalk position and according to decrease of relative humidity. The "E"of parallel piece to the vein in both varieties leaves were greater than that of right angle piece to tile vein. (4) Specific volume (Sv) The "Sv" of Burley 21 leaf was increased than that of By 104. Influences of Nitrogen and Potassium Rates on the Chemical and Physical Properties of Cured Leaf on Burley Tobacco (질소 및 가리 시용량이 버어리종 잎담배의 성분 및 물리성에 미치는 영향)

  • A field experiment was conducted to find out the effects of nitrogen and potassium rates on the chemical constituents and physical properties of burley tobacco (1988; Burley 21. 1989; Burley 21 and KB 101) during successive two years. KB 101 was low in total nitrogen and total alkaloid contents and potassium/total nitrogen ratio of cured leaf. but high in the lightness, red and yellow color and filling power, comparing to Burley 21. The increasing rates of total nitrogen and total alkaloid contents were larger when nitrogen applied to 22.75kg/10a, but they were smaller when nitrogen applied above 22.75 kg/l0a. The potassium content, potassium/total nitrogen ratio, filling power and shatter resistance index of cured leaf were decreased by nitrogen fertilizer addition. The potassium content and shatter resistance index were increased by potassium fertilizer addition. The effect of nitrogen rate on the potassium/total nitrogen ratio of cured leaf was larger than the effect of potassium rate. When the nitrogen fertilizer being applied above 22.75 kg/l0a, there were no advantageous effects on the chemical and physical properties. It is considered to be sufficient that the potassium application is about 35.0 kg/l0a.
