We isolated the Cucumber green mottle mosaic virus(CGMMV) particles from watermelon leaves and designated as CGMMV-HY1 as a watermelon isolate and attempted to characterize the pathogenic isolate responsible for such an epidemic in watermelon and also to monitor dominant viral isolates in greenhouse. The watermelon plants infected with CGMMV generally showed mottle mosaic, mosaic, growth stunting, necrosis and deformed fruit. The reactions of indicator plants to CGMMV-HY1 were the local lesions on Nicotiana tabacum cv. White Burley, Nicotiana tabacum cv. Samsun, and Chenopodium amaranticola, and the mosaic symptoms only on Cucumis sativus, but the CGMMV-HY1 did not infect Nicotiana sylvesytis, Datura stramonium, Chenopodium quinoa, and Petunia hybrida. Purified virus particles were rod-shaped and about 300 nm long. The coat protein (CP) of purified CGMMV-HY1 was single band with molecular weight of about 16.5 kDa which was confirmed by western blot analysis probed with monoclonal antibody of CGMMV-HY1. The genomic and subgenomic RNAs of 6.4 kb and 0.75 kb were revealed by the electrophoresis on 1.2% formaldehydedenatured agarose gel. Viral and complementary CGMMV-specific primer sets were designed for spanning the genome using previously reported CGMMV sequences. A 464bp of CP gene of CGMMV-HY1 was amplified by RT-PCR and cloned into PGEM-T easy vector. The nucleotide sequence of CP gene of CGMMV-HY1 shared 98%, 99%, and 100% identities with that of CGMMV strains W, KOM, and KW respectively. Based on these results, we identified CGMMV-HY1 as a CGMMV isolate of watermelon, a member of Tobamovirus.
This study was conducted to investigate the effects of using barley seeder attached to rice harvest combine that could be used in rice harvesting and barley seeding simultaneously on the growth characteristics of barley and labor-saving efficiency. In developed seeding system, burley seeding was earlier about 5 days than in the conventional system, because of conducting rice Harvesting and barley seeding simultaneously. The germination of barley seed after seeding was protected from drought damage by the rice straw covering. Among the growth characteristics of barley using developed seeding system, the number of spikes per m$^2$ was lower than that of conventional system, but others showed longer culm length, hove kernel numbers per spike and heavier 1,000 kernel weight than those of conventional system. Developed seeding system resulted in about 57% labor-saving efficiency compared with conventional system. The cost of whole works from seeding to harvest of barley was saved about 8% compared with conventional system. A total of income increased about 16% than that of conventional system.
Varietal response to cool temperature(18$^{\circ}C$) and short (8hr.)-day treatment were investigated under controlled conditions of Phytotron in the Suwon Agronomy Experiment Station. Five flue-cured and one burley tobacco varieties (Nicotiana tabacum L.) were used, and the length of treatment ranging from 8 to 32 days at 3-day intervals and also includes continuous treatment up to the plants were flowered. The days to flower decreased significantly than the no treatment when the length of treatment was 11 to 14 days for the varieties NC82, Br.21 and NC22NF, and 17 to 26 days for SPG-28, Mc.944 and TC499, respectively. And also the number of leaves decreased significantly than the no treatment when the length of treatment was 8 days for NC22NF, 11days for NC82 and Br.21, 14 days for Mc.944, and 17 days for SPG-28 and TC499, respectively. The maximum decreasing ratio of the leaf number by the cool temperature and short-day treatment were 47.7 to 58.5% for NC82, Br.21 and NC22NF, and 38.9% for Mc.944, 33.4% for SPG-28 and 29.0% for TC499, respectively.
Studies with the Tobacco Mosaic Viruses; W. S Kim, and So, I Y., (Dept. of biology Sung Kyun Kwan Univer. Seoul, Korea.). Using the common strain of tobacco mosaic virus (TMV) which was sent from the Dept. of Plant Pathology, University of Wisconsin, U.S.A. as control virus, a possible new strain of tobacco mosaic virus (SMV) was isolated from tobacco leaves collected from Tobacco Experiment Station farms as well as from various blends of manufactured Korean cigaretts. SMV was isolated by single lesion isolation method and by inoculating the virus through various species of host plants. The two viruses, TMV and SMV were indentified by the difference in symptoms, host range, serological reaction, and electron micrograpy. As the results of the above experiment the author believes the virus isolate SMV is a different strain of TMV. The experimental evidences that SMV belongs to the TMV group are as follows; 1. Both viruses produced local necrotic lesions on Nicotiana glutimosa L. 2. Both showed a dilution end point of $10^8$. 3. Aphid transmission was failed with the viruses. 4. Both had an isoelectric point around pH 3.3. 5. Two viruses were serological reactive. 6. The size of the virus particles was around 270-300mu as they were observed under the electron microscope. The virus SMV, however, is different from the common strain of TMV and the experimental evidences are as follows; 1. SMV produced quite different symptoms from TMV on various host plants like tobacoo(Nicotiana tabacum L., White Burley), Nicotiana rustica L., Chenopodium Koreanse Nakai. Bata vulgaris L., and Datura tatula L., SMV produced distinct local lesions on these host plants whereas TMV incited largely mosaic diseases. 2. The serological titers obtained from the heterologous combinations were lower than those from homologous combinations of antigens and antiser.
Six cigarette brands produced in Korea and U.S.A. were studied to find the effects of relative humidity, moisture sorption time, temperature, and the amounts of water extracts and total sugar in the brands on the equilibrium moisture contents. The equilibrium contents were higher in the domestic brands than foreign ones and the sorption rates of moisture were faster in the domestic cigarettes. The Korean brands with high contents of water extracts and total sugar had higher equilibrium moisture contents in the range of RH 40-80% at $20^{\circ}C$ for 72hours. These are the reasons that Korean brands contain higher ratio of flue-cured tobacco and American brands are blended with higher ratio of Burley tobacco.
The aim of this study is to develop a simple and rapid field method for the determination of chloride content in tobacco green or cured leaves. To set up a practical process, several factors such as the proper sampling position of each leaf, the size of leaf lamina pieces for blending, the substitution of deionized water fur the extracting solution, 0.1N $HNO_3$, the submerging durations before blending, and the standing times before measuring were studied using flue-cured and burley tobacco with a stable and reproducible Chloride Meter, KRK CL-2Z. From the results, chloride contents of tobacco leaves could be analyzed with the Chloride Meter(CL-2Z) using the suspension of 14 mm diameter leaf disks after soaking in distilled water for 30 min., blending with a mini-blender for 30 seconds. The chloride contents measured in this way showed highly significant correlations with ones by the conventional potentiometric method using an ion meter-chloride electrode and $AgNO_3$ titration. In our method, the procedures such as drying, powdering and weighing samples, and using chemical reagents for extraction (0.1N $HNO_3$) and titration(0.028N $AgNO_3$) could be eliminated. These results suggest that the newly constructed method using the Chloride Meter is more efficient to determine the chloride content of tobacco leave in comparison with the one by the Ion $Meter:AgNO_3$ titration.
It would be clear that the constituents of the leaf surface lipid is ye비 important as an evaluation index of tobacco leaf quality since the quality of tobacco specific aroma with leaf species depends on the contents of the lipid and the strength of the aroma is determined by the amounts of the lipid secreted. For the reason, a rapid and peproducible method to quantify DVT, which is a kind of lipid, has been studied. The biosynthesis procedure of DVT in leaf growing processes, and the volatile or decompositional characters of DVT in leaf drying processes were also discussed. In consequence, it might be possible to get the data available to the cultivation of better tobacco leaf and the manufacture of cigarettes with better aroma and taste. The results obtained from this study are as follows. 1. Chloroform/dichloromethane solvent was better than chloroform alone for DVT extraction. The extraction yields of the leaf surface lipid were about 5% 2. The extractives with dichloromethane were treated by silylation with BSTPa and the quantitative analysis of DVT was carried out using SE -54 fused silica capillary column. It was found that rapid and reproducible data could be obtained from these methods. 3. In flue - cured tobacco species, DVT contents were $30.3\mu\textrm{g}/cm^2$ in the beginning stage of leaf drying processes and $12.1\mu\textrm{g}/cm^2$ corresponded to 30% levels of the beginning stage, in the end stage. 4. DVT contents in Burley mere 2 times as large as those in fluecured tobacco. DVT in the upper stalk position of leaf was 3 times larger than that in the lower stalk position. 5. DVT of tobacco leaves was decomposed by $SO_2$ gas or the sun light. The decomposition rate was largest in the sample used methanol as a extraction solvent.
This study was conducted to develop a resistant tobarro against Potato virus Y (PVY) by transformation of the plants with genetically engineered viral genes. The complimentary DNAs (cDNAS) of potato virus Y-necrosis strain (PVY-Vn) replicase mutant genes (3'-deleted, 5'-deleted and ADD-mutant Nlbs) were synthesized through RT-PCR by using purified PVY-VN RNA and synthesized primers, and cloned in the sense orientation into a plant expression vector (pMBPI), The cDNAS of the genes were transferred into Agrobacterium tumefaciens LBA 4404, and then transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Regenerated plants were tested for PVY resistance by inoculation test; 13 transgenic plants including 7 for 3'-deleted Nlb, 3 for 5'-deleted Nlb, and 3 for ADD-mutant Nlb appeared to be resistant at 4 weeks after inoculation with PVY-VN. Among the 13 transgenic tobacco plants, 8 plants had no symptom up to 14 weeks after inoculation. The progenies ($T_1$) from self-fertilization of the transgenic lines varied 0.0% to 81.2% in their resistance (% of resistant plants). The analysis of Nlb-31deleted, -5'deleted and -ADD mutant in the $T_1$ plants by polymerase chain reaction (PCR) showed that Nlb-3'deleted, -5'deleted and -ADD mutants were detected in all of the resistant plants. These results suggest that the PVY resistance was inherited in the $T_1$ generation.
This Study was conducted to estimate the degree of dominance and gene frequency of some sueful characters in tobacco. The eight parents and a set of 28 crosses of F'1s was F'2s were used as materials, and planted on oriental's and burley cultivated system as randomized block designs, respectively. The observed characters were six agronomic characters which were plant height, number of leaves per plant, leaf length, leaf width, days to flowering and yield, and the data obtained from the experiment were analyzed from methods by Hayman's and Jinks. The results obtained are summarized as follows: 1. In Vr-Wr graphical analysis, number of leaves per plant, leaf length, days to flowering and yield were found to be inherited in partial dominance, and plant height was over dominance to be similar to complete dominance, but leaf width was inherited with partial dominance close to complete dominance. 2. In the gene frequency, two varieties Xanthi-Basma and KA 102, for days to flowering and yield had larger number of dominant genes as those were situated near the point of origen. 3. Additive effects of genes(D) were greater than dominance effects of Genes(H) for six agronomic characters except plant height, and mean degree of dominance over all loci was lower than 1 for days to flowering yield, leaf length and number of leaves per plant.
The study was conducted to obtain the genetic informations for some useful characters in tobacco breeding. The eight parents and a set of 28 crosses of F1's and F2's were used as materials, and planted on two different cultivated systems, i. e., oriental's and burley systems, during 1986-1988 at taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. The results obtaining are summarized as follows: 1. The higher heritabilities were found for days to flowering yield and nicotine in both generations, but values for number of leaves per plant, leaf length, total nitrogen and protein nitrogen were lower than other characters. 2. Genotypic correlation coefficients among all pairs of characters were slightly higher than the corresponding phenotypic correlations in F1's and F2's on two different cultivated systems. The relationship between leaf length and leaf width was the positive correlation, but that between number of leaves per plant and leaf width was negative. 3. From the genotypic correlations between yield and other characters, a conclusion that the yield was highly correlated with plant height, leaf length, leaf width and days to flowering was given. 4. Quality was positively correlated with number of leaves per plant and nicotine, but negatively with the other agronomic characters and chemical components.
