Effects of Liming and Nitrogen Sources on the Yield and Quality of Burley Tobacco II. Effects of Chemical Constituents of tobacco leaves (석회의 질소원이 버어리종 담배의 수량과 품질에 미치는 영향 II. 잎담배의 화학성분에 미치는 영향)

  • A field experiment was conducted to find out the effects of liming (soil pH) and sources of N on the chemical constituents of soil and leaf lamina of burley tobacco. Treatments consisted of liming (nonliming, liming to soil pH 5.5 and 6.5) as the main plot and N sources[compound fertilizer of containing 3.9% $NH_4-N$ and 6.1% $NH_2-N,\;NaNO_3,\;(NH_2)_2CO\;and\;(NH_4)_2SO_4$]as the sub-plot. The soil pH was high in $NaNO_2$ plot, while low in $(NH_4)_2SO_4$. But the differences of Ca concentration in top soil among N sources were not detected. The $NO_3-N$ concentration in top soil was high in high limed and $NaNO_3$ plot. The $NO_3-N$ content of leaf (lamina) at 75 days after transplanting was high in $NaNO_3$ plot and CaO con-tent of leaf at 45 days after transplanting was high in high limed plot. But neither liming nor N source had effect on the contents of total nitrogen, $P_2-O_{5}\;and\;K_2O$ of leaf during growing season. There was no significant differences in total alkaloid and total nitrogen contents of cured leaf (lamina) to liming and N source. But when the source of N was $NaNO_3$, the content of total alkaloid was increased by adding lime. When the source of N was $(NH_4$)_2SO_4$, the content of $K_2O$ in cured leaf was high while CaO was low. But neither liming nor N source had effect on the contents of $P_2-O_{5}$ and MgO in cured leaf.

  • Interrelationships between yields of Burley 21 variety and meteorological factors such as precipitation, duration of sunshine, and evaporation in Suweon, Chungju, Daegu, and Jeonju were investigated during the growing seasons from 1976 to 1985. Cultivation period from the 41st to the 60th day after transplanting which showed the highest transpiration by the tobacco plant was corresponding to relatively dry season in Korea from the middle of May to early of June. The meteorological factors during the middle stage of the tobacco growth, especially the 41st to the 80th day after transplanting, seemed to have higher correlation with tobacco yield, than any other periods. This result implied that the middle stage of the growth when tobacco plant consume more soil moisture by transpiration proved to be a important growth stage having a great influence upon tobacco productivity.

Effect of Light Quality on Appearance of Photobleaching leaves During the Cure of Burley Tobacco (Burley종 잎담배 건조시 광질이 백화엽 발생에 미치는 영향)

  • This experiment was designed to determine the effect of light quality on the appearance of photobleaching leaves during the cure of Burley Tobacco. The harvested and browned tobacco leaves were exposed to sunlight in pipe houses covered with 8 kinds of color vinyls (white, red, black, yellow, purple, orange, blue, green), and exposed to ultraviolet rays(20W x 3) and infrared rays (150W x 2) in curing chamber (1.2 x 1.2 x 1.2m). Photobleaching occured more at lower position leaf and after the leaves being browned when the curing was done in sunlight under a transparent vinyl. But photobleaching leaves were 5-6% of total cured leaves in sunlight under all kinds of color vinyl houses. It seems that photobleaching mainly induced by ultrabiolet rays of sunlight, and humidity too influenced. Yellow, orange and purple vinyl were durable and effective as shading material of color vinyls. since white and red vinyl tore easily in two monthes in strong sunlight and under black and blue vinyl houses curing period was longer than others.

Effects of Planting Density and Fertilizer Level on the Growth, Yield, Quality and Nitrogenous Compounds of Burley Tobacco (재식밀도 및 시비량이 버어리종 잎담배의 생육, 수량, 품질 및 질소화합물에 미치는 영향)

  • Under the different conditions of planting density and compound fertilizer level, some agronomic and chemical characteristics of burley tobacco were investigated from 1982 to 1984. Leaf area and dry leaf weight per plant, crop growth rate, relative growth rate and net assimilation rate for 40-60 days after transplanting were higher with increasing plant spacing and fertilizer, but leaf area index was lower with increasing plant spacing. At topping stage, the leaf size was increased with increasing plant spacing and amount of fertilizer applied and the stem diameter was increased by increasing plant spacing. Leaf area, leaf weight per plant and weight per unit leaf area of harvested leaf were higher when plant spacing and fertilizer increased. It was estimated that the optimum plant spacing was 105cm x 34cm and level of com-pound fertilizer (N-P$_2$ O$\sub$5/ -K$_2$O =10-10-20) was 263kg/l0a for high yield. There were trends toward increase the contents of total alkaloid and total nitrogen with increasing plant spacing and fertilizer application. There was significant positive correlation between plant spacing and total nitrogen, and between fertilizer application and total nitrogen. The plant spacing of 105cm x 35 to 40cm and 227.5kg/l0a of fertilizer level may be profitable for farm economy and the low nitrogenous leaf.

  • The study was conducted to investigate the relationships between tobacco leaf pro-duction rate by quality grade and the value of climatic factors in tobacco growing season. In flue cured tobacco(NC82), high quality was positively corretated with precipitation in late April and May, with sunshine' hours in early May and late June, with average temperature in middle July, significanly. In Burley tobacco(Br21), high quality was positively correlated with precipitation in late April and May, with sunshine hours in early May and early July, with average temperature in middle July, significantly.

  • A Solid-phase micro extraction(SPME) was evaluated as a tool for headspace sampling of tobacco samples. Several experimental parameters (sampling temperature, pH, and type of SPME fibers) were optimized to improve sampling efficiency in two aspects ; maximum adsorption and selective adsorption of volatile organic acids onto SPME fibers. Among four types of SPME fibers such as PDMS(Polydimethylsiloxane), PA(Polyacrylate), Car/PDMS (Carboxen/Polydimethylsiioxane) and PDMS/DVB(Polydimethylsiioxane/Divinylbenzene) which were investigated to determine the selectivity and adsorption efficiency. A variety of tobacco samples such as flue cured, burley and oriental were used in this study. The effect of these parameters was often dominated by the physical and chemical nature (volatility, polarity) of target compounds. This method allowed us to make important improvements in selectivity and sensitivity. The Car/PDMS fiber was shown to be the most efficient at extracting the 10 selected volatile organic acids. The parameters were optimized: $80^{\circ}C$ adsorption temperature, 30 min of adsorption time, $240^{\circ}C$ desorption temperature, 1 min of adsorption time.

  • Spinaches showing dark green mosaic symptoms were used for identification of broad bean wilt virus. In host reaction test, that virus caused local lesions on the inoculated leaves and mosaic symptoms on upper leaves of Chenopodium amaranticolor, Chenopodium quinoa and Vicia faba, and developed mosaic symptoms on Physalis floridana, Spinacia oleracea, Nicotiana tabacum, (White burley, Bright yellow) Nicotiana glutinusa. In agar gel-diffusion test, the virus showed positive reaction with broad bean wilt virus antiserum. Spherical virus particles with size of 25nm in diameter were observed in electron microscope.

  • New shoot growth of Panax ginseng root was investigated comparing with burley and soybean from l0$^{\circ}C$ to 30$^{\circ}C$ under dark. Shoot growth ceased by 12days at 30$^{\circ}C$ and optimum temperature appeared to be 15$^{\circ}C$/20$^{\circ}C$ (15hrs/9 hrs) , and 15$^{\circ}C$/15$^{\circ}C$ for ginseng. Shoot growth seems to be Poor below l0$^{\circ}C$. Temperature for maximum growth 20$^{\circ}C$/20$^{\circ}C$ for barley and 20$^{\circ}C$ /25$^{\circ}C$ for soybean. Barley did not germinate above 25$^{\circ}C$/25$^{\circ}C$, but grow better than soybean below 15$^{\circ}C$/25$^{\circ}C$. Fresh weight of 2 weeks suggesting cessation of water uptake at higher temporal use. Ginseng showed greater root ply s shoot of ginseng was linearly increased at 15$^{\circ}C$ but did not increased at 25$^{\circ}C$ after occurence of die-back of new shoot or root rot above 25$^{\circ}C$.

  • The study was conducted to investigate the relationships between tobacco leaf yield and changes of leaf dry matters by tobacco growing season. In flue-cured tobacco, leaf yield was high significantly correlated with leaf dry matters from 50 days to 70 days after transplanting. In Burley tobacco, leaf yield was affected differently by the leaf dry matters according to the different growing season. High significant correlation was noted between yield and leaf dry matters at 70 days after transplanting, and having low significant correlation with leaf dry matters at 55, 60, 65 days after transplanting.

Scopoletin Production Related to Induced Resistance of Tobacco Plants Against Tobacco mosaic virus

  • A fluorescent material was accumulated in inoculated leaves showing necrotic local lesions of tobacco plants with N gene, Nicotiana tabacum cvs. Xanthi-nc NN, Samsun NN, Burley 21 and KF 114, and N. glutinosa, and Datura stramonium at the early growth stages by the inoculation of Tobacco mosaic virus (TMV). It was identified as a coumarin phytoalexin, scopoletin. Although the material was most prominently produced in TMV-inoculated tobacco leaves with local necrotic lesions, its accumulation was also noted in uninoculated leaves of TMV-inoculated plants. Its accumulation was somewhat greater in high resistance-induced leaves than low resistance-induced and intact leaves. Scopoletin treatment induced the expression of a pathogenesis-related protein, PR-1, prominently at the concentration of 500 or 1000 ${\mu}$g/ml. This suggests that scopoletin is a phytoalexin abundantly accumulating in N gene-containing resistant plants in response to TMV infection, and may be related to hypersensitive responses (HR) and systemic acquired resistance (SAR) in the resistant tobacco plants.
