Inhibition of VLA-4/VCAM-1-mediated Cell Adhesion by Triterpenoid Saponins from Bupleurum falcatum L

  • Discovery and isolation of compounds capable of blocking the interactions between VCAM-1 and VLA-4, a major pair of adhesion molecules contributing to the different steps of leukocyte migration across the endothelium in inflammatory responses, has been a major goal of this lab. Through bioactivity-guided fractionation, five saikosaponins were subsequently isolated from the methanol extracts of the roots of Bupleurum falcatum L. Their structures were elucidated by spectroscopic analysis ($^1H-$, $^{13}C$-NMR and 2D-NMR), as follows, saikosaponins: A (1); D (2); C (3); B3 (4); B4 (5). Compounds 1 and 2 inhibited interaction of sVCAM-1 and VLA-4 of THP-1 cells with respective $IC_{50}$ values of 7.8 and 1.7 ${\mu}M$. The aglycone structure of 2 also showed cell adhesion inhibitory activity with an $IC_{50}$ value of 21.1 ${\mu}M$. With these results, we suspect these two saikosaponins from the Bupleurum falcatum L. roots to be prime candidates for therapeutic strategies towards inflammation.

Effect of Plant Growth Regulators on the Adventitious Root Formation from Bupleurum falcatum Callus (생장조절물질(生長調節物質)이 시호(柴胡) 캘러스의 부정근(不定根) 형성(形成)에 미치는 영향(影響))

  • Calli induced from the leaf segment of Bupleurum falcatum were cultured on Mu-rashige and Skoog's(MS) medium supplemented with 2, 4-D, IBA, IAA and NAA of 0.1 mg/l , The induction of adventitious roots from callus was the best in MS medium supplemented with 0.1 mg/l 2, 4-D and the lateral root was the same. The pretreatment of 0.1 mg/l 2, 4-D for 120 hours was most effective for the formation and grwoth of adventitious roots. The number of adventitious roots per micro callus pre-treated with 0.1 mg/l 2, 4- D was 5. 3 which was the highest level. The callus subcultured for 4 weeks were best for the adventitious root formation. The callus subcultured for more than 4 weeks decreased the adventitious root formation and turned to brown in color.

  • Karyotype analysis was carried out in four lines of Bupleurum falcatum L. cultivated in Korea and SDS-PAGE was applied to determine the seed protein profiles among the lines. Chromosomes were classified into two groups, large and small ones. Two kinds of karyotype, 2n=20 and 2n=26, were identified. Chromosome 1 of 2n=20 were all submedian, while that of 2n=26 were median. Chromosomes 2, 3 and 5 of 2n=20 showed polymorphism in size and arm-ratio. Chromosome 2 was submedian, while others were median in the line of 2n=26. Karyotypcs of cultivars native of Korea were similiar each other, while those introduced from Japan showed different patterns. In SDS PAGE gels, qualitative difference s in high molecular weight proteins, more than 45KD, were detected among the lines. The numbers of specific band were three in lines of 2n=20 and two in 2n=26.

Saikosaponin Content in Adventitious Root Formed from Callus of Bupleurum falcatum L. (시호(Bupleurum falcatum L)의 캘러스로부터 형성된 부정근의 Saikosaponin 함량)

  • This study was carried out to analyse saikosaponin contents in adventitious root formed from callus of Bupleurum falcatum L. The induction of adventitious root from callus of 4 weeks of culture was most favorable to MS medium supplemented with 0.1 mg/L 2,4-D. The adventitious root formation ratio per callus was highest when the size of callus was 900 to 1,000 $\mu$M The content of saikosaponin a and d was 2089$\pm$124 $\mu$g and 4778 $\pm$ 214 $\mu$g at 150 days of culture respectively on the basis of g dry root wt, whereas that of saikosaponin c was 3492 $\pm$ 123 $\mu$g at 60 days of culture.

Effects of Gelling Agents on the Saikosaponin Content of Bupleurum falcatum Callus (배지응고제가 시호캘러스의 Saikosaponin함량에 미치는 영향)

  • In order to product the saikosaponin which is one of the secondary product from Bupleurum falcatum efficiently through the tissue culture, several levels of agar and gellan gum as the gelling agent, 2,4-D as the growth regulator, and L-phenylalanine as the precursor were used with single or combination treatment on MS basal medium. Gellan gum was more effective than agar as the gelling agent in fresh and dry weight increase of callus induced from Bupleurum falcatum leaf segment. Gellan gum medium supplemented with L-phenylalanine produced 1.6 times of fresh weight more than that of agar. The fresh weight was remarkably high in gellan gum when the calli was treated with the combination of 2,4-D and L-phenylalanine similar to the single treatment of 2,4-D or L-phenylalanine. However, the saikosaponin content in callus was high in gellan gum with the single treatment of L-phenylalanine. Especially, the saikosaponin content in gellan gum supplemented with 1.0mg/L L-phenylalanine was 2 times(2.4 mg/g) higher than that in agar medium supplemented with 0.1 mg/L 2,4-D(0.9 mg/g).

  • Mycoplasma-like organisms (MLO) were identified from three plants. The disease symptoms show stunting, yellowing and witche's broom on Cnidium officinale Makino collected from Ulreung Island, Bupleurum falcatum L. collected from Jangsu, and Plantago asiatica L. collected from Daegwanreung. The particles of MLO were observed in the phloem tissues by a Hitachi Hu-11E electron microscope. C. officinale infected with MLO was frequently observed in Ulreung Island.

Saikosaponin Contents in Somaclones Derived from Different Aged Calli of Bupleurum falcatum L. (배양 기간이 다른 캘러스로부터 재분화된 시호 (Bupleurum falcatum L.) 체세포클론의 사이코사포닌 함량 변이)

  • The contents of saikosaponin a, c and d in the root of somaclones and wild plants of B. falcatum L. were determined. The saikosaponin contents of wild plants collected from Chongson-gun, Kangwon Province and Yongdok-gun, Kyongbuk Province were higher than those of the somaclones derived from 3-month and 2-year aged calli. The mean values of saikosaponin content in 2 year-old somaclones derived from the callus cultured three months were higher than those in 1 year-old somaclones from 2-year aged callus. Variation of saikosaponin content depending on the culture periods was not found. Four plants among 1-year old somaclones showed saikosaponin content twice as much as wild plants had. This result implies the possible use of somaclonal variation for crop improvement.

Studies on Seed Germination Characters and Germination Inhibitors of Bupleurum falcatum L. (자호(紫胡)의 발아특성(發芽特性)과 발아억제물질(發芽抑制物質)에 관(關)한 연구(硏究))

  • This experiment was conducted to investigate the optimum temperature and the effects of sulfuric acid, flowing water and $GA_3$ on the germination of Bupleurum falcatum. At the same time, the effect of germination-related compounds in Bupleurum falcatum seed was tested. Among the tested temperatures $(15,\;20,\;25\;and\;30^{\circ}C)$ the highest germination rate was obtained at $15^{\circ}C$. The optimum soaking time of sulfuric acid was 5 minutes at the concentration of both 0.1 and 1.0%. At the same time, the germination rate was highest when Bupleurum falcatum seed was washed for 2 days in the flowing water before germination test. The effect of $GA_3$ on the germination was highest at 100 ppm out of 10, 50 and 100 ppm. Regardless of treatments, the germination rate of Jeongseon species was higher than that of Mishima species. SEM photographs revealed that most of germination-inhibiting substances in the seed coat were washed away by rinsing in the flowing water for 2 days thus the seed coat became more smooth. The 4, 000 ppm alcohol extract of Bupleurum falcatum seeds inhibited the germination of lettuce seeds as much as 41.7% in Jeongseon species and 58.3% in Mishima species. The higher extract concentration from seed of Bupleurum falcatum, the lower the germination of lettuce seeds was obtained. Out of phenolic acids, the contents of orchinol, pyrogallol, and p-hydroxybenzoic acid were highest in both varieties. Compared with Mishima species, Jeongseon species showed much higher content of salicylic and vanillic acid.
