Purification and Characterization of Xylanase from Fomitopsis palustris in Rice Straw Culture (볏짚분해과정 중에 생산하는 Fomitopsis palustris 균체 외 Xylanase의 분리정제 및 효소특성)

  • An extracellular xylanase from the brown-rot fungus Fomitopsis palustris grown on rice straw culture was purified to a single protein band. On SDS-PAGE, the molecular mass of purified enzyme was estimated to be about 43 kDa. The amino acid sequence of the proteolytic fragments showed high homology with fungal glycoside hydrolase family 10 xylanases. The $K_m$, $K_{cat}$ and $V_{max}$ for birch xylan were $31mg/m{\ell}$, $2.3{\times}10^4/min$ and 252.3 U/mg, respectively. The optimal activity of the purified xylanase from F palustris was observed at pH 4.0~5.0 and $70^{\circ}C$.

  • The development of Korean glaze originated from the development of ash glazes. Ash not only has strong solubility but also can shows the glaze a variety of different colors according to what the glaze has in it as the main component. In addition, it gives a feeling of lucidness and softness. For these reasons, there are a lot of needs for ash and many potters want to take advantage of ashes as glaze. But natural ashes have not been widely used as glaze primarily because it is relatively hard to find or manufacture. Considering the difficulty of finding or manufacturing natural ashes, this study aims to formulate synthetic ash which not only is available to the potters in general but also has the sam ε characteristics as the natural ashes. To achieve this aim, this study examines the characteristics of the pine tree ash, the main component of the glaze of celadon porcelains, and the red pepper stem ash, the main component of the brown glaze, both of which have been used by the Korean traditional potters. In this study, the alkaline component of the glaze. A important ingredient when the ashes are synthesized, was supplied by mixing of $Na_2CO_3$, Chungju limestone and rice straw ash. Furthermore, the synthetic ash, when it shows no change of pH in its composition rate of 6:2:2, was found to be usable as the most stable material. In conclusion, the formula which frits some raw $materials-Na_2CO_3$, Chungju limestone and rice straw ash-can duplicate the synthetic ash which is simila to the natural ash.

Studies on the Heavy Metals in Paddy Rice and Soils in Jang-hang Smelter (제연소인근지역(製鍊所隣近地域)의 토양(土壤) 및 수도체중(水稻體中) 중금속함량(重金屬含量)에 관(關)한 조사연구(調査硏究))

  • Soils and rice plants subject to smoke from the Jang-hang Smelter were samples at two depths to assess the nature and extend of Cd, Cu, Pb, and Zn contamination, particularly with respect to distance from pollution source. Soils at east site within 1km of the center of the smelter were highly contaminated with Cd, Cu, Pb, and Zn. Metal levels in the smoke-polluted area were higher in surface soil than in subsurface soil, and decreased rapidly with distance reaching nearly background levels at 5km from pollution source. A position correlation was found between contents of total and 0.1 N HCl-extractable, or 1.0N $CH_3COONH_4$- extractable heavy metals in surface soils of pollution. Contents of heavy metals in soils were positively correlated with soil organic matter and cation exchange capacity. The contents of heavy metals in brown rice collected from the paddy field around the Jang-hang Smelter were in the range of 0.23-1.33 ppm for Cd, 2.39-6.25 for Cu, 0.95-8.32 ppm for Pb and 14.60-27.31 for Zn. The contents of Cd, Cu, and Zn in rice straw were positively correlated with those in the brown rice. Negative correlation was found between the contents of Cd, Cu, Pb, and Zn in brown rice and the distance from the source.

Heavy Metals in Paddy Rice and Soils in Mangyeong River Area (만경강(萬頃江) 유역(流域)의 토양(土壤) 및 수도체중(水稻體中) 중금속(重金屬) 함량(含量))

  • Soils and rice plants in wastewater irrigated area of the Mangyeong River receiving waster from the Jeonju Industrial Complex and municipal sewage were sampled at two depths to assess the nature and content of Cd, Cu, Pb and Zn, particularly with respect to distance from wastewater source. For metal levels of soils in these area, no difference in the heavy metal contents between the surface and the subsurface soils was found. Total contents of Cu, Pb and Zn in soils were negatively correlated with distance from the source. A positive correlation was found between contents of total and 0.1N-HCl extractable or $1N-CH_3COONH_4$ extractable heavy metals in surface soils of these area. Total contents of heavy metals in soils were positively correlated with clay, soil organic matter and cation exchange capacity. Heavy metal contents of brown rice sampled at the Jeon-ju Industrial Complex area ranged from 0.15 to 0.91 ppm for Cd, from 1.13 to 5.68 ppm for Cu, from 0.22 to 7.16 ppm for Pb and from 11.74 to 38.66 ppm for Zn. Negative correlation was found between the contents of Cd, Cu, Pb, and Zn in the brown rice and the distance from the source. The contents of Cd, Cu and Zn in rice straw were positively correlated with those in the brown rice.

Effect of Cold Water Irrigation on the Growth, Yield and Grain Quality in rice (냉수관개가 벼 생육, 수량 및 미질에 미치는 영향)

  • This experiment was carried out at Chuncheon substation, Crop Experiment Station to investigate the effect of cold water irrigation on the growth and grain yield of rice in 1990. Irrigation of cold water (17$^{\circ}C$) delayed 10 to 19 days of heading, and reduced about 17 to 31% of plant height in terms of culm and panicle length. Number of panicle per plant was decreased and / or increased by cold water irrigation. Cold water irrigation reduced spikelet number per panicle and percent of filled grain. Grain yield was increased but, straw weight was decreased according to water temperature gradient from inlet to outlet. There was varietal differences in head rice recovery by cold water treatment. In general the dark brown and / or black colored kernel increased at cold water inlet but decreased percent of green kernel. Alkali digestible value increased and amylose content decreased at cold water inlet.

  • A study has been conducted to investigate the behavior of carbofuran in a miniature paddy agrosystem simulated for paddy field. Carbofuran applied onto the paddy water was rapidly absorbed and translocated into rice plants. Carbofuran concentration in rice plant reached its maximum level between 1 to 3 days after treatment and gradually decreased thereafter. Half life of carbofuran concentration in paddy water was 4 days in both application rates of 0.12 and 0.24Kg a.i./10a. Carbofuran residue in paddy soil was gradually dissipated with the half life of 8 and 12 days in 0.12 and 0.24㎏ a.i./l0a respectively. Range of carbofuran residue in brown rice and rice straw harvested from the paddy agrosystem was 0.01∼0.02 ppm and 0.37∼0.57 ppm irrespective of the two application rates respectively.

Effect of Slaked Lime and Borax Application upon Yield and Yield Components of Paddy Rice (소석회 및 붕사의 시용이 수도의 수량 및 수량구성요소에 미치는 영향)

  • This study was conducted in order to investigate the effects of slaked lime and borax on the Akiochi paddy field. The pot trial(slaked lime was treated 0, 6, 10, 15g and borax 0, 0.75, 1.5g per pot) and the field trial(slaked lime was treated 0, 100, 200kg and borax 0, 10, 30, 50kg per 10are) respectively. The results are summarized as fo11ows. I. Pot Trial: 1. In a heavy treated of borax, appeared chlorosis on whole leaves(bottom) in early growing stage, However, in about 25 days after transplanting, these chlorosis were disappeared gradually. And in proportion to close by maturity after the ear-heading, changed to dark brown color and rolled at the end of upper leaves, finally withered. At the same time the part of the end of grains was also colored to dark brown. 2. Slaked lime effected to increase the ti1lering, hence increased the number of ear, on the other hand, borax showed the tendency of repression the tillering, conversely. 3. Borax effected to hurried up to heading date of rice plant, to which borax had been the plot of B$_1$ to 3 days and B$_2$ to 5 days, hurried up than control respectively, regardless of the quantities of slaked lime application. 4. According to the application of borax, the length of culm were shortened, otherwise the length of panicle were longished, then no recognized the. independent effect of slaked lime. 5. The number of grains per panicle were seemed like increased by treatment of slaked lime, but the grain were decreased in the heavy treatment of slaked lime and borax. These phenomena were showed in ratio of maturity likely tendency as above. 6. In the 1, 000 grains weight of brown rice, in the case of slaked lime was almost no significant, otherwise by increase the application of borax, increased the weight of 1, 000 grains and the size of the grains was greater. 7. The weight of rough rice and straw were showed the same tendency as the number of grains and maturity ratio. particularly, showed the phenomenon of the reduction by the heavy application of borax. II. Field trial: 1. In a heavy treated of borax, appeared chlorosis in a early growing stage, according to close by maturiting date after the ear-heading changed to dark brown the end of upper leaves, finally dried. 2. Slaked lime application accelerated the growth and increased the number of ear, but borax showed the tendency of repression the tillering. 3. Borax effected to hurried up to heading date of rice plant, according to heavy treatment of borax quickened heading date for 4 to 5 days. 4. As a whole, culm length was repressed by borax treatment. 5. Borax application 10kg per 10are advanced progress the maturity, but in the case of above 30kg per 10are of borax showed the tendency obstruction the maturity. 6. The yield of rough rice and brown rice recognized the independent effect of slaked lime and borax, respectively. the yield was decreased by the abundant of borax.

Study on the Technological System of the Cooperative Cultivation of Paddy Rice in Korea (수도집단재배의 기술체계에 관한 연구)

  • For the purpose of establishing the systematized technical scheme of the cooperative rice cultivation which has most significant impact to improve rice productivity and the farm management, the author have studied the cultivation practices, and the variation of rice growth and yield between the cooperative rice cultivation and the individual rice cultivation at random selected 18 paddy fields. The author also have investigated through comparative method on the cultivation practices, management, organization and operation scheme of the two different rice cultivation methods at 460 paddy fields. The economic feasibility has been ana lysed and added in this report. The results obtained from this study are summarized as follows; 1. In the nursery, the average amount of fertilizer application, especially, phosphate and potassium, and the frequency of chemicals spray for the disease, insect and pest control at the cooperative rice cultivation are significantly higher than those of the individual rice cultivation. 2. The cultivation techniques of the cooperative rice farming after the transplanting can be characterized by a) the earlier transplanting of rice, b) the denser hills per unit area and the lesser number of seedlings per hill, c) the application of larger quantities of fertilizer including nitrogen, phosphate and potassium, d) more divided application of fertilizers, split doses of the nitrogen and potassium, e) the increased frequencies of the chemicals spray for the prevention of disease, insect and pest damages. 3. The rate of lodging in the cooperative rice cultivation was slightly higher than that of the individual rice cultivation, however, the losses of rice yield owing to the occurrence of rice stem borer and grass leaf roller in the cooperative rice cultivation were lower than that of the individual rice cultivation. 4. The culm length, panicle length, straw weight and grain-straw ratio are respectively higher at the cooperative rice cultivation, moreover, the higher variation of the above factors due to different localities of the paddy fields found at the individual rice cultivation. 5. The number of panicles, number of flowers per panicle and the weight of 1, 000 grains, those contributing components to the rice yield were significantly greater in the cooperative rice cultivation, however, not clear difference in the maturing rate was observed. The variation coefficient of the yield component in the cooperative cultivation showed lower than that or the individual rice cultivation. 6. The average yield of brown rice per 10 are in the cooperative rice cultivation obtained 459.0 kilograms while that of the individual rice cultivation brought 374.8 kilograms. The yield of brown rice in the cooperative rice cultivation increased 84.2 kilogram per 10 are over the individual rice cultivation. With lower variation coefficient of the brown rice yield in the cooperative rice cultivation, it can be said that uniformed higher yield could be obtained through the cooperative rice cultivation. 7. Highly significant positive correlations shown between the seeding date and the number of flowers per panicle, the chemical spray and the number of flowers per panicle, the transplanting date and the number of flowers per panicle, phosphate application and yield, potassium application and maturing rate, the split application of fertilizers and yield. Whilst the significant negative correlation was shown between the transplanting date and the maturing rate 8. The results of investigation from 480 paddy fields obtained through comparative method on the following items are identical in general with those obtained at 18 paddy fields: Application of fertilizers, chemical spray for the control of disease, insects and pests both in the nursery and the paddy field, transplanting date, transplanting density, split application of fertilizers and yield n the paddy fields. a) The number of rice varieties used in the cooperative rice cultivation were 13 varieties while the individual rice cultivation used 47 varieties. b) The cooperative rice cultivation has more successfully adopted improved cultivation techniques such as the practice of seed disinfection, adoption of recommended seeding amount, fall ploughing, application of red soil, introduction of power tillers, the rectangular-type transplanting, midsummer drainage and the periodical irrigation. 9. The following results were also obtained from the same investigation and they are: a) In the cooperative rice cultivation, the greater part of the important practices have been carried out through cooperative operation including seed disinfection, ploughing, application of red soil and compost, the control of disease, insects and pests, harvest, threshing and transportation of the products. b) The labor input to the nursery bed and water control in the cooperative rice cultivation was less than that of the individual rice cultivation while the higher rate of labor input was resulted in the red soil and compost application. 10. From the investigation on the organization and operation scheme of the cooperative rice cultivation, the following results were obtained: a) The size of cooperative rice cultivation farm was varied from. 3 ha to 7 ha and 5 ha farm. occupied 55.9 percent of the total farms. And a single cooperative farm was consisted of 10 to 20 plots of paddies. b) The educational back ground of the staff members involved in the cooperative rice cultivation was superior than that of the individual rice cultivation. c) All of the farmers who participated to the questionaires have responded that the cooperative rice cultivation could promise the increased rice yield mainly through the introduction of the improved method of fertilizer application and the effective control of diseases, insects and pests damages. And the majority of farmers were also in the opinion that preparation of the materials and labor input can be timely carried out and the labor requirement for the rice cultivation possibly be saved through the cooperative rice cultivation. d) The farmers who have expressed their wishes to continue and to make further development of the cooperative rice cultivation was 74.5 percent of total farmers participated to the questionaires. 11. From the analysis of economical feasibility on the two different methods of cultivation, the following results were obtained: a) The value of operation cost for the compost, chemical fertilizers, agricultural chemicals and labor input in the cooperative rice cultivation was respectively higher by 335 won, 199 won, 288 won and 303 won over the individual rice cultivation. However, the other production costs showed no distinct differences between the two cultivation methods. b) Although the total value of expenses for the fertilizers, agricultural chemicals, labor input and etc. in the cooperative rice cultivation were approximately doubled to the amount of the individual rice cultivation, the net income, substracted operation costs from the gross income, was obtained 24, 302 won in the cooperative rice cultivation and 20, 168 won was obtained from the individual rice cultivation. Thereby, it can be said that net income from the cooperative rice cultivation increased 4, 134 won over the individual rice cultivation. It was revealed in this study that the cooperative rice cultivation has not only contributed to increment of the farm income through higher yield but also showed as an effective means to introduce highly improved cultivation techniques to the farmers. It may also be concluded, therefore, the cooperative rice cultivation shall continuously renovate the rice production process of the farmers.

  • For the increase of protein content in the straw without detrimental effect on grain yield by top dressing with various nitrogen amount IR667 (Suwon 214) and Jinheung were grown under field condition and the value as feed was investigated by chemical analysis on the harvested straw. 1. In straw IR667 was higher in crude protein, crude fiber and ash, and lower in total carbohydrate than Jinheung, and crude fat content was not much different. 2. The contents of crude protein and total carbohydrate increased with nitrogen level but crude fiber, crude fat and ash decreased, and those phenomena were clear in IR667. 3. The content of phosphorus in straw decreased with increasing nitrogen level more clearly in Jinheung. 4. Protein content was greatly increased in brown rice and calcium content tended to decrease with the increase of nitrogen application amount. 5. Country mean of crude protein in straw was 4.06% much lower than that of mixed wild grasses and forages (6.71) but crude protein could be increased above that of wild sources, moreover with grain yield increase by the improvement of nitrogen fertilizer application.

  • The objectives of the present study were to find out the differences in growth and weed competition of rice under the foggy and non-foggy condition, and finally, the fundamental data for the establishment of the paddy weed control system under the locational foggy regions. The research was carried out on tin trays ($0.12m^2$) in greenhouse equipped with an Auto Foggy System(SAE KI RTN Co.). The results are summarized as follows: Exp. I. Difference in rice growth under the foggy and non-foggy condition. 1. While the plant height was not affected, the number of tillers was decreased by the foggy condition. The ratio of the number of effective tiller, however, became higher under the foggy condition. 2. Due to the fog present, the heading rate was decreased and heading time was delayed. 3. The foggy condition did not affect the dry weight of rice straw whether they were grown under the foggy or non-foggy condition. However, yield components such as the number of ears, the number of grains per ear, thousand kernel weight and percentage of ripeness were reduced. Particularly, weights of perfect brown and unpolished rice were also decreased. Exp. II. Effect of the duration of competition between weed and rice grown under the foggy condition on the growth of rice plant. 1. There was no difference in the height of rice grown under a different duration of competition. There was a clear tendency that the number of tillers of rice grown under the foggy condition was decreased as the duration of competition was decreased. 2. When the duration of competition was longer than 60 days, the heading rate was decreased and the initiation of heading was also delayed by 2-4 days. 3. Under the foggy condition, the duration of competition for more than 40 days affected dry weight of rice straw and the difference in yield was greatest in the non-competition and competition for more than 40 days.
