Isolation and Characterization of Halophilic Kocuria salsicia Strains from Cheese Brine

  • Kocuria salsicia can survive in extreme environments and cause infections, including catheter-related bacteremia, in humans. Here, we investigated and evaluated the characteristics of nine K. salsicia strains (KS1-KS9) isolated from cheese brine from a farmstead cheese-manufacturing plant in Korea from June to December, 2020. Staphylococcus aureus American Type Culture Collection (ATCC) 29213 was used as a positive control in the growth curve analysis and biofilm-formation assays. All K. salsicia isolates showed growth at 15% salt concentration and temperatures of 15℃, 25℃, 30℃, 37℃, and 42℃. KS6 and KS8 showed growth at 5℃, suggesting that they are potential psychrotrophs. In the biofilm-formation analysis via crystal violet staining, KS6 exhibited the highest biofilm-forming ability at various temperatures and media [phosphate buffered saline, nutrient broth (NB), and NB containing 15% sodium chloride]. At 25℃ and 30℃, KS3, KS6, and KS8 showed higher biofilm-forming ability than S. aureus ATCC 29213. The antimicrobial resistance of the isolates was evaluated using the VITEK® 2 system; most isolates were resistant to marbofloxacin and nitrofurantoin (both 9/9, 100%), followed by enrofloxacin (7/9, 77.8%). Five of the nine isolates (5/9, 55.6%) showed multidrug resistance. Our study reports the abilities of K. salsicia to grow in the presence of high salt concentrations and at relatively low temperatures, along with its multidrug resistance and tendency to form biofilms.

이비인후, 안, 치아 및 피부증상의 민간요법에 관한 고찰 (Study on the Herb Remedies of ENT, Eyes, Teeth and Skin Problems)

  • The intention of this study is to apprehend the con. tents and methods of herb remedies that are commonly conducted when there are health-problem cases of ENT, eyes, teeth and skin. Methods of this study are divided into two stages : 1) For a period of six months from December 1994 to June 1995, some 40 persons who are believed to be well versed with herb remedies have been randomly chosen, and we made a survey on herb remedies by symptoms: and 2) we have endeavored to make their grounds evident through the studies on literatures with the focus on the basic data collected. Their results are as follows: 1) When one feels a pain in ears, such herb remedies are employed as pouring the vapor into ears, which is made by steaming Alaska pollack, or as applying or wiping with the juices of radish or the ginkgo, or' alum. Applying the radish juice is effective for sterilization and fever removal: and applying the ginkgo juice is effective for cleanliness. But, plastering alum, sesame oil or castor oil, or pouring the vapor of Alaska pollack into ears are perhaps effective but do not have any pharmacologic grounds. 2) When one bleeds at the nose, such kinds of herb remedies are applied as stimulating nose or head with cold water, pressing nose or ridge of nose, or filling up nares with mashed mugworts. In addition, they have utilized garlic or leeks. Such methods as stimulating with cold water or just pressing nose and ridge of nose is based on reasonable grounds, i.e. vasoconstriction and vascular compression ; and applying mashed garlic on the sole of foot is good for the circulation of Qui ; and the use of mugworts and leeks is based upon the pharmacological function of hemostasis. 3) When one feels a sore throat such kinds of herb remedies are employed as gargling or rinsing throat with brine, drinking hot gruel or water, or drinking the juice of mugwort, radish, ginger or Chinese quince. Gargling with brine or drinking the juice of mugwort, radish or ginger is based upon the pharmacological function of pain alleviation, fever removal, and detoxication. 4) When a boil is formed in mouth, such herb remedies are applied as spreading honey, brine or alum water, and taking gall nut, Chinese matrimony vine, lotus root, etc, for drugs. Spreading honey, brine or water that is made by infusing gallnut, Chinese matrimony vine, lotus root is based upon such functions as hematosis, astriction, antibacterial, and antiphlogistic, Alum, eggplant and licorice are said to be effective, but their pharmacological effects have no grounds. 5) When one has conjunctivitis such herb remedies are commonly applied as irrigation with brine and dropping breast milk in eyes. Moreover, such other drugs are used as plantain. shepherd's purse, and purslane, etc. The use of brine, breast milk, plantain, shepherd's purse and purslane is based upon such functions as sterilization, antiphlogistic, disinfection and pain relieving. Eriocaulon sieboldianum, bean stem, bean pod and narcissus leaves are said to be effective, but their pharmacological action have no basis. When one has a stye, such herb remedies are applied as extracting eyelashes, stimulating by a massage of middle finger, third finger or big toe, as well as sear ing with a heated bamboo comb that is fine-toothed. Other than these, plantain and nightshade's nuts are used as drugs for it. Extracting eyelashes corresponds with exclusing suppurative node and draining the stye of pus ; and the use of plantain is based upon disinfection: and nightshade's nuts are said to be effective, however, their pharmacological action has no grounds. 6) For a treatment of toothache, such herb remedies are commonly employed as rinsing mouth with brine and holding cold water or gasoline in the mouth ; and as the drugs that are believed to be effective have been Welsh onion, ginger and castor-oil, plant, etc. The use of Welsh onion is based upon pain killing, antiinflammatory actions, and the use of ginger is based upon detoxication and disinfection ; and seeds of castor-oil plants are said to be effective, but they have no pharmacological basis. 7) When one has hives, such herb remedies are commonly applied as rubbing burned straw in affected parts, exposing to its smoke, rubbing with salt, sweeping down with a broom, and spreading and drinking boiled water of trifoliate orange. The use of cassia tora seeds, walnut, aloe and radish is said to be effective. The use of cassia tor a seeds has the functions of intestinal order, anti-paralysis, etc. The use of walnut has resulted in an increase of blood by invigorating spirits ; and the use of aloe is based upon disinfection, antibiotic, anti-salt, antihistamine and detoxication action. But, the effects of radish juice and straw's smoke have no pharmacological grounds. 8) When one gets a boil, such herb remedies are commonly used as applying a plaster, paste of flour mixed with yolk, soy sauce or honey, as well as spreading pounded elm tree. Other remedies that have been said to be effective are ; heating with mugwort, brine, wild rocambole, aloe, onion, squid's bone, etc. The use of mugwort is based upon pain killing, astringent antiinflammatory and tranquility. Wild rocambole is based upon the generation and maintenance functions of cell-joining textures ; elm tree upon antiphlogistic ; aloe upon fever removal and antiphlogistic ; onion on pain killing, fever removal, antiphlogistic and tranquility ; squid's bone on astriction: and brine or vinegar on sterilization. Pine resin and gardenia seed are said to be effective, but they have no pharmacological basis. 9) When one cuts his skin, such herb remedies are commonly employed as spreading mugwort's juice or squid's bone powder, or pressing the wounds. In addition, kalopanax, onion and fine soil are employed. The use of mugwort, kalopanax and squid's bone is based upon such functions as hemostasis, sedation, pain killing, antibacterial ; and fine soil is said to be effective, but it has no pharmacological basis. 10) When one suffers from whitlow, such herb remedies are commonly utilized as heating with boiled soy sauce, spreading soybean paste, or dipping into eggs, etc. Other drugs that have been employed are onion root, brine, eggplant, potato, loach, etc. The use of onion is based upon pain killing and antiphlogistic functions ; and that of brine upon antiphlogistic function. The use of soy sauce or soybean paste, fomentation, eggplant, potato and loach is said to be effective, but it has no pharmacblogic ground. 11) For the treatment of frostbite, such herb remedies are commonly used as dipping the affected part into frozen soybean sack, using boiled water of eggplant stem, garlic caulis, onion, hot pepper, caulis. Onion is based upon antiphlogistic and tranquility actions garlic upon disintection, metabolic exacerbation, tonic and aphrodisiac actions and the use of eggplant and hot pepper is based upon help blood circulation, dissolution and excretion of waste matters in vein. 12) For the treatment of burn, such herb remedies or drugs are commonly used as cleansing with Korean gin, spreading eggs, cleansing with cold water and soap water ; and as brine, cactus, moss, soybean paste, oil, etc. The cleansing with Korean gin, cold water, soap water, brine, vinegar is based upon cleaning and sterilizing functions ; and the use of cucumber is based upon nu. trition provision, and strengthening of resisting power by adjustment of metabolism. The use of potato, cactus, moss, oil and eggs is said to be effective, but their phamacological functions are not clarified. In view of the above results, we can realize that the drugs that have been employed in herb remedies are quite diverse. However, in regard to majority of herb remedies that have been employed by symptoms, the pharmacological functions of their drugs have not been clarified, and they are merely known as effective. Furthermore, they have not been recorded in the literature as yet ; and we confirm that there have been many herb remedies that were executed without the proper knowlege of their effects. It is now our view that the results of this survey may be utilized for consulting data in regard to the use of herb remedies.

온배수 및 염배수의 근역혼합특성 분석을 통한 방류시스템의 최적설계 (Optimum Design of Outfall System by Analyzing Mixing Characteristics of Heat and Brine Discharge at Near Field Region)

  • 해양방류시스템을 계획할 때 우선목표는 방류수의 초기희석을 극대화하는 것이다. 이를 효과적으로 달성하기 위해서는 방류수와 주변수의 혼합현상, 특히 초기혼합영역인 근역에 대한 특성분석이 선행되어야 한다. 최근까지 최적의 방류시스템(outfall system) 구현을 위하여 근역(NFR)에서의 온배수 방류에 관한 많은 연구가 이루어지고 있지만 온배수(heat discharge)와 염배수(brine discharge)를 동시에 방류할 경우를 고려한 성과물을 찾기는 어려운 실정이다. 이에 본 연구에서는 온수와 염수를 동시에 고려한 방류시스템의 최적설계를 위한 기초자료를 제공하기 위하여 방류수의 거동양상 특히, 근역에서의 혼합성을 분석하였다. 수치실험은 표면방류, 단일확산관 방류, 다공확산관 방류를 해당 해역의 법적 수질기준에 부합하는지 여부를 주로 평가하였다. 그 결과 표면방류와 단일확산관 방류는 방류형식이 부적절하였고, 다공확산관의 경우도 기하학적인 형상에 따라 결과가 조금 상이하였다.

절임배추 및 김치 양념의 저장기간 중 품질 특성 변화 (Changes in quality characteristics of salted Kimchi cabbage and kimchi paste during storage)

  • 절임배추와 양념으로 구성된 제품에 대한 품질 특성 정보를 제고하고, 버무린 형태로 저장된 김치와 절임배추와 양념을 분리 저장한 후 버무린 김치의 품질 특성의 차이를 확인하기 위해 $4^{\circ}C$에서 저장하면서 1주 간격으로 품질특성을 분석하였다. 절임배추의 산도 및 일반세균수 변화 결과, 염수 침지에 따른 절임배추의 품질유지 효과가 나타났으며, 외관 및 관능적 품질 특성 결과를 토대로 염수 침지하지 않은 절임배추의 저장한계는 1주, 염수침지한 절임배추의 저장한계는 2주 이내임을 알 수 있었다. 특히, 염수량이 많을수록 품질 유지에 더욱 효과적이었으며 관능적 특성에서도 다소 높은 점수를 받았다. 양념은 저장 8주 동안 산도가 증가하는 경향을 보였으나 미생물학적 및 관능적 품질특성에는 큰 변화가 없었다. 0일차에 제조하여 $4^{\circ}C$에서 저장한 김치와 $4^{\circ}C$에서 각각 저장된 절임배추와 양념을 이용하여 매주 제조한 김치의 품질 특성을 분석한 결과, 0일차에 제조하여 저장한 김치보다 저장된 절임배추와 양념으로 매주 제조한 김치의 pH가 높고, 산도가 낮아 발효 지연효과가 있는 것으로 나타났다. 특히 3주차 저장된 절임배추와 양념으로 제조한 김치의 경우 신냄새, 신맛 등의 지표에서 김치형태로 저장된 것보다 유의적으로 낮은 점수를 받았다. 전반적인 기호도에서도 분리 저장 후 제조한 김치에서 높은 점수를 받았으나 유의적인 차이는 확인되지 않았다. 이상의 결과, 절임배추와 양념으로 제품화할 경우 절임배추는 품질유지를 위해 염수에 침지하여 포장하는 방법이 효과적이며, 분리 저장된 절임배추와 김치 양념을 혼합하여 김치로 제조하여도 처음부터 김치 형태로 제조하여 저장한 것과 비교하여 발효가 지연되면서 관능적으로도 문제가 없어 김치의 가식기간을 연장할 수 있는 방법이 될 것으로 생각된다.

  • In previous paper (Lee et al., 1983) processing method of sardine meat "surimi" was described as a part of the wort to develop new types of ready-to-cook food materials with dark fleshed fishes. As the other part of the work, processing of low salt mackerel fillet was investigated, in this paper, in which fresh mackerel was filleted, salted in brine or with dry salt for an adequate time until the expected salt concentration reached, washed, air dried (3 m/sec, 15 to $20^{\circ}C$), and finally packed individually in K-flex film bag by vacuum or $N_2$ gas substitution. Salting time and salt concentration of brine was decided by the salt level penetrated into the fillet. As the final salt level was fixed to 4 to $5\%$, salting for 20 hours with $10\%$ dry salt or in $15\%$ brine at $5^{\circ}C$ was enough to get that level of salt. If the final salt level was set 5 to $6\%$, salting for 20-24 hours with $15\%$ dry salt or in $20\%$ brine was adequate. Salt penetration, however, was not much influenced by salting method and temperature. Changes in VBN and salt soluble protein occurred more rapidly in cases of salting with dry salt at $20^{\circ}C$ than salted in brine at $5^{\circ}C$, although it was not significant in the period of 20 to 24 hours. Oxidation of lipid and histamine formation during salting at $20^{\circ}C$ could not be neglected if it was delayed loger than 25 hours. Insolubilizing the salt soluble proteins during the storage of salted fillet occurred rapidly regardless of storage temperature. Browning and histamine formation, however, was depended on temperature and packing condition. In case of air pack, deterioration by browning and rancid was deeply developed but not the case for the packs by vacuum or $N_2$ gas substitution. The shelf-life of the salted mackerel fillet based on panel scores of brown color and rancidity, appeared 21 days for the air packed, and more than 30 days for vacunm or $N_2$ gas packed fillet at $20^{\circ}C$.

직접 접촉식 막증류 공정에서 담수 투과량 및 열효율 극대화를 위한 수치적 연구 (A Numerical Study for the Maximizing Water Vapor Flux and Thermal Efficiency in Direct Contact Membrane Distillation (DCMD) Process)

  • 본 연구에서는 직접 접촉식 막증류 공정에서 운전인자에 따른 담수 투과량과 열효율을 예측하기 위해 열 및 물질전달 방정식을 이용하여 1차원 해석모델을 개발하였다. 이 해석모델의 타당성을 검증하기 위해 해석모델 결과와 기존 연구자들에 의해 수행된 실험 결과를 비교하였고 만족할 만한 결과를 얻었다. 이를 통해 DCMD 모듈에서 염수와 증류수의 입구온도 및 입구속도가 담수 투과량 및 열효율에 미치는 영향을 분석하였다. 그 결과 염수의 입구온도와 입구속도가 증류수의 입구온도와 입구속도보다 담수 투과량과 열효율 증가에 미치는 영향이 크기 때문에 지배적인 운전특성이라는 것을 알 수 있었다. 염수의 입구온도가 $60^{\circ}C$에서 $95^{\circ}C$로 증가할 때 담수 투과량이 21.22 $kg/m^2h$에서 71.26 $kg/m^2h$로 3.4배 증가하였고 열효율은 0.556에서 0.765로 37.5% 증가하였다. 한편, 염수의 입구속도가 60에서 300 m/h로 증가함에 따라 담수 투과량이 27.91 $kg/m^2h$에서 36.33 $kg/m^2h$로 30% 증가하였고 열효율은 0.6에서 0.646로 7.5% 증가함을 알 수 있었다.

염수 침지식 및 전극 접촉식 저전압 Ohmic 해동 처리가 돈육의 이화학적 특성에 미치는 효과 (Effects of Brine Immersion and Electrode Contact Type Low Voltage Ohmic Thawing on the Physico-chemical Properties of Pork Meat)

  • 본 실험은 전압의 증가에 따른 염수 침지식 및 전극 접촉식 ohmic 해동 처리가 돈육의 이화학적 특성에 미치는 효과를 규명하고자 실시되었다. 염수 침지식 ohmic 해동방법은 전압에 의한 염수의 온도 상승에 의하여 시료가 해동 됨으로써 시료 표면의 과열 현상이 효과적으로 억제되었고, 해동 후 높은 보수력 및 전단력의 저하가 관찰되었지만, 육색이 다소 저하되는 단점을 나타내었다. 반면 전극 접촉식 ohmic 해동 방법은 해동 과정 중 얼음의 상전이 구간을 빠르게 통과함으로써 빠른 해동이 가능하였고, 해동 후 식육의 품질 저하가 최소화되었지만, 시료의 크기 및 전압의 증가에 따른 심한 표면 과열 현상이 발생할 수 있는 단점을 보였다. 따라서 염수 침지식 해동은 보다 높은 전압을 이용하여 식육을 빠르게 해동하고, 전극 접촉식 해동은 낮은 전압을 이용하여 표면 과열을 억제하면 보다 효과적인 해동이 가능할 것으로 사료되었다.

2상 밀폐 서모사이폰을 이용한 빙축열 시스템의 성능 (An Experimental Study on an Ice Storage System by a Two-Phase Closed Thermosyphon)

  • A two-phase closed thermosyphon is applied to an ice storage system. The thermosyphon is used to freeze the water in a storage tank. The experiment has been performed to investigate the effects of the important parameters such as the quantity of the fluid filled with, the ratio of the length of the evaporator to the condenser, and the temperature and the mass flow rate of the brine. It is found that the higher thermal performance of the thermosyphon is obtained as the ratio of the length of the evaporator section to that of the condenser section is decreased and the temperature of the brine is lowered. The increase of the quantity of the working fluid also favors the performance of the system. The experimental data can be utilized for the basic design of ice storage systems with thermosyphons.

  • This paper describes total flow control of an ice slurry system for pump energy saving. Similar turbo machinery has a characteristic that input power ratio is proportional to the three time of revolution speed ratio. To reduce the energy cost of brine pump in ice slurry storage systems, inverter is adapted instead of 3-way valve to control the speed of brine pump motor. One type of cooling load profile was used as driving load of the system, generated by a boiler and warm water storage tank. As results of the laboratory test, energy consumption and cost of the pump were reduced by 11.4%.

Cytotoxicity and antinociceptive activity of Jasminum sambac leaves

  • The ethanolic extract of Jasminum sambac leaves were tested for its cytotoxicity and possible antinociceptive activity in experimental animals. The extract showed potent cytotoxic activity in brine shrimp lethality assay and the LC50 was found only 25 mg/ml. The extract significantly and dose dependently inhibited the acetic acid induced writhing in mice (56.83%, P < 0.001 and 43.17%, P < 0.001 for 500 and 250 mg/kg body weight, respectively). The results supported its traditional uses.
