Gonadosomatic index (GSI) of the crucian carp (Carassius auratus) was investigated to clarify annual reproductive cycle from February in 1992 to October in 1994. The values of GSI were high with individual variation from April to July which period was coincided with the breeding season of fish. The GSI was very low in August and September, when follicular atresia developed in the ovaries. GSI value began to increase in October and reached a peak around the following March, which indicated that ovarian follicles may grow during this period. Human chorionic gonadotropin (HCG 10 IU), $17\alpha$, 20\beta-dihydroxyprogesterone\;(1-100{\mu}g/ml)$ and phorbol 12-myristate-13-acetate (TPA, protein kinase C activator, 0.1-10${\mu}M$) induced germinal vesicle breakdown (GVBD), but $4\alpha-phorbol$ 12, 13- didicanoate ($4\alpha-PDD,\;phorbol\; ester\;analogue,\;25{\mu}M$) did not induce germinal vesicle breakdown in the follicular oocytes. Prostaglandin $F_{2\alpha}$$(0.1-10 {\mu}g/ml)$ and TPA $(0.1-10 {\mu}M$ induced ovulation of the oocytes, but $4\alpha-PDD$$(25{\mu}M)$ did not induce ovulation of the follicles. $17\alpha-hydroxyprogesterone$ production was examined from the isolated follicles to investigate the steroid production ability in the crucian carp ovaries. HCG (1 lU, 10 lU) and forskolin (adenylate cyclase activator, 0.1-10 ${\mu}M$) stimulated $17\alpha-hydroxyprogesterone$ production. The time course of HCG (10 lU) and forskolin $(10\;{\mu}M)$ stimulated $17\alpha-hydroxyprogesterone$ production within 3 hours, the elevated levels were maintained during the rest of the culture period. The data indicates that cyclic AMP and protein kinase C may play important roles in the oocyte maturation and ovulation in crucian carp.

Soybean yield has been low and unstable in Japan and other areas in East Asia, despite long history of cultivation. This is contrasting with consistent increase of yield in North and South America. This presentation tries to describe perspective of breaking stagnation of soybean yield in East Asia, considering the factors of the different yields between regions. Large amount of rainfall with occasional dry-spell in the summer is a nature of monsoon climate and as frequently stated excess water is the factor of low and unstable soybean yield. For example, there exists a great deal of field-to-field variation in yield of 'Tanbaguro' soybean, which is reputed for high market value and thus cultivated intensively and this results in low average yield. According to our field survey, a major portion of yield variation occurs in early growth period. Soybean production on drained paddy fields is also vulnerable to drought stress after flowering. An analysis at the above study site demonstrated a substantial field-to-field variation of canopy transpiration activity in the mid-summer, but the variation of pod-set was not as large as that of early growth. As frequently mentioned by the contest winners of good practice farming, avoidance of excess water problem in the early growth period is of greatest importance. A series of technological development took place in Japan in crop management for stable crop establishment and growth, that includes seed-bed preparation with ridge and/or chisel ploughing, adjustment of seed moisture content, seed treatment with mancozeb+metalaxyl and the water table control system, FOEAS. A unique success is seen in the tidal swamp area in South Sumatra with the Saturated Soil Culture (SSC), which is for managing acidity problem of pyrite soils. In 2016, an average yield of $2.4tha^{-1}$ was recorded for a 450 ha area with SSC (Ghulamahdi 2017, personal communication). This is a sort of raised bed culture and thus the moisture condition is kept markedly stable during growth period. For genetic control, too, many attempts are on-going for better emergence and plant growth after emergence under excess water. There seems to exist two aspects of excess water resistance, one related to phytophthora resistance and the other with better growth under excess water. The improvement for the latter is particularly challenging and genomic approach is expected to be effectively utilized. The crop model simulation would estimate/evaluate the impact of environmental and genetic factors. But comprehensive crop models for soybean are mainly for cultivations on upland fields and crop response to excess water is not fully accounted for. A soybean model for production on drained paddy fields under monsoon climate is demanded to coordinate technological development under changing climate. We recently recognized that the yield potential of recent US cultivars is greater than that of Japanese cultivars and this also may be responsible for different yield trends. Cultivar comparisons proved that higher yields are associated with greater biomass production specifically during early seed filling, in which high and well sustained activity of leaf gas exchange is related. In fact, the leaf stomatal conductance is considered to have been improved during last a couple of decades in the USA through selections for high yield in several crop species. It is suspected that priority to product quality of soybean as food crop, especially large seed size in Japan, did not allow efficient improvement of productivity. We also recently found a substantial variation of yielding performance under an environment of Indonesia among divergent cultivars from tropical and temperate regions through in a part biomass productivity. Gas exchange activity again seems to be involved. Unlike in North America where transpiration adjustment is considered necessary to avoid terminal drought, under the monsoon climate with wet summer plants with higher activity of gas exchange than current level might be advantageous. In order to explore higher or better-adjusted canopy function, the methodological development is demanded for canopy-level evaluation of transpiration activity. The stagnation of soybean yield would be broken through controlling variable water environment and breeding efforts to improve the quality-oriented cultivars for stable and high yield.

This work was carried out to investigate the performance of large-type Korean native ducks at middle laying phase (30 to 50 weeks). A total of one hundred and sixty eight laying ducks were divided into A and B strains (6 replications/strain, 14 birds/replication and fed practical diet, for 20 weeks). There was no significant difference in body weight, daily feed intake and average egg weight. With increasing rearing period, mean body weight tended to be decreased and showed the lowest value (2,649 g) at 42 weeks of age (P<0.05). Similar trend was observed for weekly feed intake and average egg weight which decreased with passing weeks and was lowest (190.5 g and 83.8 g, respectively) at the age of 42~46 weeks (P<0.05). Daily egg production of B strain was higher than that of A strain at the age of 46~50 weeks (P<0.05). Weekly egg production of A and B strains commenced to decrease from the age of 42 weeks, however, B strain recovered at 46~50 weeks of age (P<0.05). At the age of 30~38 weeks, the egg production in B strain was higher than A strain (P<0.05), whereas, there was no significant difference between both the strains during 38~50 week. Feed conversion ratio of A strain was higher than that of B strain at 46~50 weeks of age (P<0.05). Weekly feed conversion was highest at the age of 42~46 weeks (P<0.05). In conclusion, the present results provides the basic information of the performance record of large-type Korean native ducks for the middle laying phase.
This study was conducted to establish the efficient screening system for resistant radish to Fusarium oxysporum f. sp. raphani. Five radish cultivars ('Myoungsan', 'Chungdu', 'Jangsaeng', 'Hannongyeorm', and 'Chungsukungjung') showing different degree of resistance to the fungus were selected. And the development of Fusarium wilt of the cultivars according to several conditions such as root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease was tested. In distinguishing the resistance degree of the radish cultivars to the disease, non-cut roots were more effective than cut roots. And occurrence of Fusarium wilt of the radish plants increased in the proportion to increase of root-dipping period and spore concentration of the fungus. Thus, optimum conditions to differentiate susceptible and resistant cultivars to the disease were root-dipping period of 0.5 hour and spore concentration of $1{\times}10^7\;conidia{\cdot}mL^{-1}$. Disease severity of Fusarium wilt on the cultivars was changed with incubation temperature and the radish seedlings incubated at $25^{\circ}C$ represented the most difference of resistance and susceptibility to Fusarium wilt. From the above results, we suggest that the efficient screening method for resistant radish to Fusarium oxysporum f. sp. raphani would be to dip non-cut roots of fourteen-day-old radish seedlings in spore suspension of $1{\times}10^7\;conidia{\cdot}mL^{-1}$ for 0.5 hour and to transplant the inoculated plants to plastic pots with fertilized soil, and then to incubate the radish plants at a temperature of $25^{\circ}C$ for development of Fusarium wilt.
Sensitivities of PLBs of four Phalaenopsis cultivars, P. 'Taisuco Windian', P. 'Nancy Amour', P. 'Pink Twilight' and P. 'Taipei Gold' to kanamycin, spectinomycin and hygromycin at different concentrations (0, 25, 50, 100, 200, and $400mg{\cdot}L^{-1}$) were examined. Hygromycin was favorable for selecting the transformants in the genetic transformation of Phalaenopsis as PLBs of four cultivars were all dead at even $25mg{\cdot}L^{-1}$ hygromycin. Responses of PLBs of P. 'Maki Watanabe' and P. 'Brother Lawrence' to DL-phosphinothricin (PPT) were determined at different concentrations (0, 0.1. 0.25, 0.5, 1.0, 2.0, 2.5, and $5.0mg{\cdot}L^{-1}$) and $0.5mg{\cdot}L^{-1}$ PPT was thought to be suitable for selecting the transformants of Phalaenopsis. The optimum conditions for Agrobacterium cocultivation with Phalaenopsis PLBs were examined using a two-step cocultivation method in Dtps. 'City Girl' and A. tumefaciens LBA4404. In the first infection period in a 1 : 10 suspension of Agrobacterium to a VW medium, 1 hr infection showed the highest PLB survival ratio. And then, PLBs were cocultivated with a bacterial strain and a 3-day cocultivation period was better for Phalaenopsis PLBs than a prolonged period. Agrobacterium tumefaciens strains LBA4404 (pTOK233) and EHA105 (pGA643) were used to compare their efficiency on the genetic transformation of Phalaenopsis PLBs. The PLBs infected with EHA105 survived more than those infected with LBA4404 after two days in a dark condition and two weeks in light condition on a selective medium. About 1,000 PLBs for each of P. 'Maki Watanabe' and P. 'Brother Lawrence', and each bacterial strain of AGL1 (pCAMBIA3301) and LBA4404 (pTOK233) were used for the regeneration of transgenic plants. The bacterial strain AGL1 had a higher genetic transformation efficiency than LBA4404, with no significant difference between cultivars. In this study, 11 hygromycin-resistant plantlets and 32 PPT-resistant plantlets were produced, but these putative transgenic plantlets need further examinations.
This experiment was performed to investigate the flowering habit of sesame (Sesamum indicum L.). Sesame varieties tested could be classified into 8 different plant types by their morphological traits such as capsule shape, capsule setting habit and branching types among sesame gene pool of Crop Experiment Station, ORD. The first flower was appeared at the lowest node on main stem. Flowers were appeared progressively toward the tip of the main stem and also toward the tips of branches. The interval of flowering for a node was about one day, but 3 to 8 days for the flowers on the tips. Side flowers started at 4 to 5 nodes lower than those of center flower at the same day. Flowers were beared 2 by 1 node on the middle part of flower setting node (7-9) in mono capsule setting habit in spite of its normal is 1 by 1 node on the other nodes. Flowers were beared opposite direction on each node of stem and flowering toward the tip of main stem composed of cross shape between nodes and spiral, reverse of clockwise direction. We called this habit as cross spiral flowering order and cross spiral phyllotaxis. The first flower on branches was appeared when center flower on the 5th node of main stem began to flower. The branches produced at higher nodes on main stem showed larger flowering periods and more number of flowers than that at lower parts. BTB (Branch, Tricapsule, Bicarpels, 4 Loculi) type showed three capsule setting habits and same flowering period both on main stem and branches while BTQ (Branch, Tricapsule, Quadricarpels, 8 Loculi) type showed three capsule setting habit on main stem and mono-capsule setting habit on branches. In BTQ type, the period of flowering was much shorter on branches than on main stem. Branching type was considered more promising than non branching type for the breeding of early maturing high yielding variety because branching type has the advantage of bearing a lot of flowers in comparatively short flowering period.

This study estimated the effect of sowing and harvesting dates on dry matter (DM) yield and feed value of forage oats at Sancheong, Korea. The forage oats (Darkhorse vs. Highspeed) were used in this experiment. The experimental main plots consisted of the different sowing and harvesting dates at 2 seasons as follows: spring oats of sowing (February 25, March 3 and March 13) and harvesting (May 27, June 6 and June 16); and fall oats of sowing (August 15, August 25 and September 4) and harvesting (October 15, October 25 and November 4). On spring oats, Highspeed sown on March 3 and then harvesting on June 6 had the highest (p<0.05) plant length and DM yield. Crude protein content decreased (p<0.05) in seed of the delayed-harvesting Highspeed. On fall oats, plant length and DM yield with the delayed-harvesting date increased (p<0.05), while crude protein content of the delayed-harvesting Highspeed decreased (p<0.05). This study concluded that the spring oat sown in early March and then harvesting in early June was recommended to increase dry matter and feed value although the fall oat sown in end August and then harvesting in early November was recommended for fall period.

The specific antibody response of olive flounder, Paralichthys olivaceus to different water temperature were tested by enzyme-linked immunosorbent assay (ELISA). In the rearing temperature of $15^{\circ}C$, first anti-bovine serum albumin (BSA) antibody titer was appeared after 14 days of immunization, whereas 24~48 days post-immunization (PI) resulted maximum antibody titer in all 5 experimental fish with optical density (OD) values 1.94~3.04. At the end of the experiment (84 days), 0.03~1.28 OD values were observed. In the rearing temperature of $12{\sim}13^{\circ}C$, first antibody titer was found 28 days PI in 2 out of 5 fish. Three fish shown high OD titer (1.88~2.68) between 56 and 70 days and OD values of 0.49 to 2.35 were observed at 84 days. However, the anti-BSA antibodies of two fish showed less than 0.8 OD values until 84 days. In the rearing temperature of $10^{\circ}C$, specific antibody appeared at 56 days, maximum antibody titer was observed at 70 days in 2 out of 5 fish (OD values: 1.37~1.53) and 1.00 to 1.11 OD values were observed at 84 days. Rest 3 fish showed OD values of 0.12 to 0.68 much below to that of other 2 fish, throughout the experimental period. In conclusion, specific antibody response of olive flounder at high temperature was much faster, higher and longer than that at lower temperature.
This study was carried out to clarify the effect of dietary calcium, casein and suet on intestinal absorption of cadmium in mice. It was performed for 30 days, from April 11, to May 10, 1988. Ninety mice were devided into 4 experimental groups and control group with 6 mice each dietary group, and measured survival ratio, body weight and weight ratio of organ to body. The contents of cadmium in liver, kidney, spleen, muscle and skin with hair, and feces were analyzed by atomic absorption spectrophotometer after sacrifice by anesthesia. The ratio of survival after 30 day breeding was $100\%$ in control group, but $66.7\%$ in group IV(basal diet + Cd + Ca) with $100{\mu}g$ one dose and with 50ppm cadmium containing fluid free-intake, and group V (basal diet + Cd + suet) with free-intake of 50ppm containing cadmium fluid. The ratio of body weight gains in cases of single dose $100{\mu}g$ of cadmium was highest as $42.3\%$ in group IV and lowest as $26.0\%$ in group V, in cases of free-intake of 50ppm containing cadmium fluid was highest as $24.0\%$ in group IV and lowest as $11.6\%$ in group II (basal diet + Cd). The body weight, in the case of single dose of $100{\mu}g$ of cadmium showed no increase untill 5th day from acute poisoning. While in case of free-intake of 50ppm containing cadmium, it showed very slight increase through all breeding period. The weight ratio of organ to body were lowest in liver of group II in both occasions. The most of cadmium administered was excreated in feces within 2 days after single dose $100{\mu}g$ cadmium. The contents of cadmium in all tissues were significantly high in each group comparing to control group as liver, kidney, spleen and muscle of group II which showed the highest level in both occasions of $100{\mu}g$ one dose and free-intake of 50ppm containing cadmium fluid, and higher in group II of the occasion with $100{\mu}g$ single dose, and in group V of the occasion of 50ppm containing cadmium fluid free-inatke for skin with hair. And the content of cadmium in tissues is generally higher in the occasion of 50ppm containing cadmium free-intake than $100{\mu}g$ single dose. From the above results I conclude that the addition of casein, calcium and suet in cadmium containing diet is effective to inhibtion of intestinal absorption of cadmium by particularly, calcium.
Pear cultivar 'Supergold' (Pyrus pyrifolia var. culta Nakai) was originated from the cross between 'Chuwhangbae' and 'Manpungbae' with the aims of improving the fruit quality of 'Chuwhangbae' cultivar at Pear Research Station of National Institute of Horticultural & Herbal Science, Rural Development Administration in 1994. 'Supergold' was preliminarily selected in 2002 and named in 2008. The tree shows a vigorous growth habit and semi-spread characters like as 'Manpungbae'. Furthermore, it has sufficient flowers and carries abundant pollen grains, so it can also be used as a pollinator. 'Supergold' is highly resistant to black leaf spot (Alternaria kikuchiana) in the field condition. The optimum harvest time is around Sep. 11th, which is ahead of 'Whangkeumbae' about 5 days in the harvest period. The fruit shape is oblate and fruit skin color is greenish-white at harvesting time. The average weight of fruit is 570 g, and the soluble solids content is $13.6\;^{\circ}Brix$. The flesh is very soft and juicy, and renders good eating quality. Shelf life is about 6 months under the cold storage condition. To determine the self-incompatibility (SI) genotype of 'Supergold' pear cultivar, it was crossed with other cultivars of which SI genotypes have already known. The result of cross-pollinations of 'Supergold' with other cultivars showed relatively high rates of fruit set from 64.5% to 91.0%, except for the cross with pollens of 'Nijisseiki' that represented only 28.8% of fruiting rate. Although sometimes the stigma of 'Supergold' crossed with 'Hayatama', 'Chojuro', and 'Nijisseiki' showed malformed pollen tube tips, 'Supergold' is generally supposed to have cross-compatibility with all other pollen donor cultivars. It is considered that the S-allele of 'Supergold' is $S_3S_4$, which is based on the result of PCR-RFLP.
