Molecular Prevalence of Cryptosporidium spp. in Breeding Kennel Dogs

  • Cryptosporidium is a common intestinal protozoan that can lead to diarrhea in humans and dogs. The predominant species of infection are C. hominis and C. parvum in humans, and C. canis in dogs. However, C. canis can infect immunocompromised humans. Considering the close contact with humans, dogs have the potential to be reservoirs for human cryptosporidiosis. Breeding kennels are the major supply source of puppies for pet shops. The present study is to determine the molecular prevalence and characteristics of Cryptosporidium spp. found in breeding kennel dogs. A total of 314 fecal samples were collected from young and adult dogs kept in 5 breeding kennels. A polymerase chain reaction targeting the small subunit rRNA gene was employed for the detection of Cryptosporidium spp. To determine the species, the DNA sequences were compared to GenBank data. Overall, 21.0% of the fecal samples were positive for Cryptosporidium spp. infection. Cryptosporidium spp. was detected in all 5 facilities. A sequencing analysis demonstrated that all isolates shared 99-100% similarity with C. canis. The results suggest that Cryptosporidium spp. infection is present at a high-level in breeding kennel dogs. However, because dominant species in this survey was C. canis, the importance of breeding kennel dogs as reservoirs for Cryptosporidium spp. transmission to humans is likely to be low in Japan.

Bacteriological and serological investigation of Brucella canis infection of dogs in Taegu city, Korea (대구지역 개의 Brucella canis 감염에 대한 세균학적 및 혈청학적 조사)

  • A bacteriological and serological investigation was conducted in a commercial breeding kennel in Taegu city in which canine abortion caused by Brucella canis occurred, and the family dogs in this area were also surveyed for Br canis infection during a period from March 1999 to May 2000. Of 195 dogs in the breeding kennel, 50(25.6%) were found to be bacteremic and 82(42.1%) were shown to be positive for canine brucellosis by both bacteriological and serological test. Of 357 family dogs examined, 17(4.8%) had an agglutinin titer of 1 : 160 or higher. Of these 17 dogs, 5(2.8%) were from indoor dogs and 12(6.7%) were from outdoor dogs. Only 2 mongrel dogs(1 female and 1 male) of 17 serologically positive dogs showed an agglutinin titer of 1 : 2560 or higher and Br canis was isolated from their blood.

Biochemical characterization and PFGE pattern of Brucella canis isolated from kennels in Gyoengbuk province (경북지역 애견 번식장에서 분리한 Brucella canis의 생화학적특성 및 PFGE 양상)

  • A biochemical characterization and antimicrobial drugs susceptibility study was conducted in four breeding kennel which was canine abortion caused by Brucella canis in Gyeongbuk province in 2003-2006. Total of 267 dogs domesticated in the four kennel were examination. Among them, 143 (53.6%) dogs were sero-positive and 25 of blood samples were isolated to Brucella canis. At amplification of 35KDa-BCSP gene using PCR, 711 bp DNA fragment was same visible in 25 isolates and B canis RM6/66. Biochemical characterization of B canis isolated was non-hemolytic, no production of $H_2S$, no fermentation of carbohydrates, catalase-positive, oxidase-positive, indol-negative, hydrolyzation of urea, reduction of nitrate and development of thionin dye medium. Using disk-diffusion method, all of 25 strains tested were found to be highly susceptible to tetracycline, aminoglycoside, quinolone, macrolide antibiotics, rifampin and ampicillin in vitro. Using PFGE with restriction enzyme Smi I, 25 isolates tested were typed to 2 pattern, S1 and S2.

Investigation of Brucella canis infection in public animal shelters and kennels in Incheon

  • This survey was performed to investigate the seroprevalence of antibodies to Brucella canis in dogs from public animal shelters and breeding kennels in Incheon. A total of 402 dogs selected randomly were tested serologically by using immunochromatographic antibody test kit. None of 289 dogs in public animal shelters were sero-positive and 10 (8.9%) of 113 dogs in breeding kennels were sero-positive. 5 (4.4%) strains of Brucella canis were isolated from 10 sero-positive dogs' blood. Antimicrobial susceptibility test was carried out by Disk diffusion method. They were susceptible to tetracyclines, quinolones, aminoglycosides and combination amoxicillin with clavulanic acid.

Investigation of Injury Types and Frequency of Bearded Dragons According to Stocking Density (사육밀도에 따른 턱수염 도마뱀(Bearded dragon)의 부상 형태와 빈도 조사)

  • This study was conducted to determine the injury type and frequency of bearded dragons during single and group breeding according to stocking density, for 16 weeks. A total of 14 bearded dragons compromising three groups were selected for this study. Six and four bearded dragons bred in a cage of 45 cm × 45 cm × 45 cm were designed as group A and B, respectively. Group C was made of four partitions (20 cm × 20 cm × 20 cm) of equal size in the kennel, and the dragons, four in number, were reared alone. Injury type criteria were given 1 score each in 6 stages. Most of the injury types in group A were primary tail cutting, which was 1 point, depending on the score, except for 4 scores. The injury type of group A varied, that is 1, 2, 3, and 5 scores, and the pattern lasted for 2 to 9 weeks. After 10 weeks, the dragons showed no response to injury type, except 4 scores at 11 weeks and 2 scores at 14 weeks with a frequency of one, respectively. The representative injury types of the group B were primary tail cutting, which was 1 point, and the scores of the injury type from 2 to 11 weeks were the highest; the frequency of occurrence at 1 and 2 scores was 5 times and 3 times, respectively, and the frequency of occurrence at 3 scores was twice. In addition, 5 scores were found to have the lowest frequency of occurrence. Solitary breeding showed no response to the type of injury for 16 weeks. Consequently, single breeding with an appropriate stocking density did not affect stress, and it could be expected a positive aspect for pet welfare.

Mastitis with Proteus mirabilis in Brittany Spaniel, a case (Brittany Spaniel에서 Proteus mirabilis에 의한 유방염 발생: 증례)

  • The dog breeding kennel bacterial infection are very significant in perinatal mortality. In many case, Staphylococcus aureus, Streptococcus, and Escherichia coli were infected in intra-uterine or by the genital tract to the puppies, and they are cause of septicemic death of the puppies and clinical mastitis of bitch, leading to septicemic death of newborn puppies. Severe mastitis due to bacterial infection was diagnosed in a 2 year-old female Brittany Spaniel which loss 6 puppies with odor small rice sharp white diarrhea of nine puppies. Bright curd milk and intestinal sample were inoculated on MacConkey agar, blood agar and brain heart infusion agar, and incubated at $37^{\circ}C$ for 24-48 hrs. Gram negative colonies isolated from these sample which were characterized as Glu, Ure, $H_2S$, Orn, Cit, and Cl, and were identified by Microscan Walk-Aways Baxter, American Type Culture Collection, USA) as Proteus mirabils. The isolate was more sensitive to ampicillin, gentamicin, cefoxitin, cefuroxime, and cefazidime. In this results, we confirmed that cause bacteria of septicemic death in puppies was P. mirabilis.

Serological and bacteriological study on canine brucellosis in the large kennel farms in Gyeongbuk province (경북지방 개 브루셀라병 다두 발생농장의 혈청학적 및 세균학적 조사)

  • Canine brucellosis is a contagious disease of the reproductive tract that cause mainly abortion and infertility in dog. A serological and bacteriological survey was conducted for breeding kennels which were suffered from frequent outbreak of canine brucellosis in Gyeongbuk province in 2009. Among 138 samples, 45 serum samples were sero-positive. Brucella canis was isolated from 30 blood samples of the seropositive cases, and from 2 samples of 62 sero-negatives. The biochemical properties of 32 isolates were characterized with no production of H2S, no fermentation of carbohydrates, hydrolyzation of urea, and development of thionin dye medium. At amplification of BCSP and 16S-rRNA gene using PCR, 711bp and 905bp DNA fragments were detected in agarose. Three tandem repeat pattern was shown in genotyping by Multi-locus VNTR assay (MLVA).

Etiologic Survey on the Acute Diarrhea of Puppies in Seoul (서울지역에서 문제되고 있는 강아지의 급성설사증에 관한 병인학적 연구)

  • In Seoul area, there are so many kennel clubs, veterinary hospital, pet establishment and breeding confers that various problems have occurred. They are crowed pet houses, poor sanitation, stress to puppies, sudden environmental changes to puppies and unvaccination against parvovirus, canine distemper virus, parainfluenza virus, infectious hepatitis caused by Adenovirus type I and Leptospira. Several studies were made to survey the infectious agents involved in acute diarrhea of poppies in Seoul are, such as history taking, physical examination, complete blood count and serum chemistry, histopathological finding, bacterial isolation and identification, and hemagglutination test in feces and hemagglutination inhibition test in serum against parvovirus, respectively. The results obtained are summarized as followed. 1. The percent of PCV (30.5$\pm$5.6) and concentration of Total protein(5.0$\pm$0.8) resulted from statistical analysis are significantly lower than normal values (p<0.05), respectively. In addition, fibrinogen (505$\pm$326) was significantly higher an normal value (p<0.001), Band neutrophil (22.9$\pm$12.7) showed signifiant difference (P<0.01). decreased monocyte (3.2$\pm$2.1) and eosinophil (0.7$\pm$0.8) values appeared statistically significant (p<0.001), lymphocyte (16.7$\pm$9), as well. 2. The concentrations of calcium .(8.0$\pm$2.8), glucose (40.1$\pm$31.4), and albumin (2.0$\pm$0.39) were lower than normal values (p<0.01, p<0.01, p<0.001), respectively. Also Inorganic phosphate (7.1$\pm$2.4), pH (p<7.9f:0.2), and Blood Urea Nitrogen (40.41=37.1) were significantly higher than normal values (p<0.001, p(0.001, p<0.05), respectively. 3. Simple and mixed infections occupied 18% and 82% in the distribution of causes in puppies with acute diarrhea, respectively. 4. As puppies got older, incidence of acute diarrhea caused by Staphylococcus aureus was decreased to 13% and infection of canine distemper virus was increased to 53%, but E coli and canine parvovirus always showed high frequency of outbreak in the body weight ranged from 35g to 7.8Kg. 5. As showed in table 5, infections of E coli and Canine Parvovirus showed high outbreak regardless of the age which is classified into three stages, 35~50 day, 60 day and 75~day to 1 year, canine distemper virus appeared increased, but in case of Staphylococcus aureus, visa versa. 6. In comparison wi methods for the laboratory diagnosis diagnosis parvovirus, Hemagglutination test showed positive reaction in 25% and mean serum antibody titer measured by Hemagglutination inhibition test showed 2779 (n=20). In addition, positive reaction was 90% (18/20). 7. In histopathological studies, enteritic and pneumonic lesions were indicated in 53.7% and 39.5% of samples, respectively. 8. Etiologic diagnosis based on the history taking, clinical signs and histopathological findings in puppies with acute diarrhea and vomiting indicated that 50% of puppies were infected by canine parvovirus and distemper virus. 9. In the parasitological examination made by simple flotation with saturated zinc sulfate tour parasites found were Isospora canis, Toxocara canis, Ancylostoma spp and Toxocara spp. Isospora canis and Toxocara canis were more frequently found among those parasites of diarrhoeic causes in puppies ranged from 35 days to 1 year. Their infestation rates were 15% and 13% respectively.
