Effects of Vitamin A on Carcass Composition Concerning Younger Steer Fattening of Wagyu Cattle

  • Regarding the fattening of younger cattle that the Ministry of Agriculture Forestry and Fisheries (MAFF) recommends in Japan, this study looked at vitamin A control in feed and blood and its effect on performance of production and carcass composition of Wagyu steers. Five sets from 10 Wagyu artificial identical twins were divided to into 2 groups, a restricted group (Test) and a supplied group (Control). The body weight at the finishing time of the Test was significantly lower than that of the Control (p<0.05). The daily gain from 13 to 21 months old, as the animals in the Test were fed the concentrated feed without Vitamin A, was significantly different (p<0.05) between the Test and the Control. The total daily gains for the Test and the Control for the fattening period were 0.82 kg/day and 0.93 kg/day, respectively, which showed a significant difference (p<0.01). Regarding the rib thickness, the Test was thinner than the Control. The Beef Marbling Scores of the Test and Control were 3.60 and 2.80, respectively. The muscle weight of the Test was significantly smaller than that of the Control (p<0.01). However, regarding the ratio to the carcass, the Test was significantly higher than the Control (p<0.05). For the fat weight, the Test was smaller by about 15 kg than the Control (p<0.01). Furthermore, for the fat ratio to the carcass, the Test was significantly lower than the Control (p<0.05). The bone weights of the two groups were approximately the same. Regarding the ratio of bone to carcass, the Test was higher than the Control (p<0.01). For the younger fattening method, the low level vitamin A in the serum had the effect that the muscle ratio to the carcass weight was greater and the fat was less, but the carcass and muscle weight were less.

선발육종기술을 이용한 북방전복의 성장 (Growth of Pacific abalone, Haliotis discus hannai, using selection breeding techniques)

  • 속성장을 위한 선발 1세대 전복 (Selected abalone : SA)의 성장률을 조사하기 위하여 동일 사육 환경 조건에서 대조구 전복 (Control abalone : CA) 과 비교실험을 하였다. 그 결과 각 성장형질 (각장, 각폭 및 중량) 은 SA구가 CA구보다 유의하게 빠른 것으로 나타났다. 45일째 및 90일째의 평균 각장에 있어 SA구가 CA구 보다 각각 31.7% 및 17.8% 빠르게 나타났으며, 중량에 있어서도 각각 43.7% 및 25.6% 빠른 것으로 나타났다. 더욱이 90일째의 각장과 중량의 상대성장 비교에 있어서도 SA구가 CA구 보다 유의적으로 높은 상관관계를 나타내었다. 이러한 결과는 선발육종기술을 이용하여 양식 전복의 성장률을 증대할 수 있는 가능성을 시사하고 있다.

Generation of Newly Discovered Resistance Gene mcr-1 Knockout in Escherichia coli Using the CRISPR/Cas9 System

  • The mcr-1 gene is a new "superbug" gene discoverd in China in 2016 that makes bacteria highly resistant to the last-resort class of antibiotics. The mcr-1 gene raised serious concern about its possible global dissemination and spread. Here, we report a potential anti-resistant strategy using the CRISPR/Cas9-mediated approach that can efficiently induce mcr-1 gene knockout in Escherichia coli. Our findings suggested that using the CRISPR/Cas9 system to knock out the resistance gene mcr-1 might be a potential anti-resistant strategy. Bovine myeloid antimicrobial peptide-27 could help deliver plasmid pCas::mcr targeting specific DNA sequences of the mcr-1 gene into microbial populations.

Development of the Insect Smart Farm System for Controlling the Environment of Protaetia brevitarsis seulensis

  • 이 연구는 식품원료로 등록되고 건강기능성 효능이 널리 알려지면서 생산과 수요가 점차 증가하고 있는 흰점박이꽃무지 유충의 사육환경 제어를 위한 식용곤충 스마트팜 공조시스템을 설계하여 제안하고자 수행하였다. 제안된 곤충 스마트팜 공조 시스템은 사육실과 공조실을 구분하여 사육에 최적화된 환경을 공조실에서 만들어 사육실로 보급하는 시스템으로 기존 곤충 사육농가에서 냉난방기, 가습기 등을 통한 환경 제어를 할 때 사육실 내 부분별로 온도 및 습도 등이 매우 상이하여 식용곤충을 판매할 때 균일한 크기 및 무게의 흰점박이꽃무지 유충을 생산하기 어렵다는 문제점의 해결책으로 제시될 수 있다. 곤충 스마트팜 공조 시스템을 사용함으로써 기존 곤충농가의 환경제어에 비해 온도의 차를 6℃, 습도의 차를 24.7%를 감소할 수 있으며 사육실 내 부분별로 온·습도가 다른 점을 개선하여 흰점박이꽃무지 유충의 사육 최적 환경을 계절에 상관없이 상시 제공함으로써 연중 생산을 통한 생산량 증대와 곤충 사육농가의 소득증대를 도모할 수 있다. 또한 제안된 곤충스마트팜 공조시스템은 설정된 최적 온도, 습도 및 CO2를 효율적으로 제어함으로써 갈색거저리 등 기타 식용곤충의 사육 및 버섯류의 생육에 필요한 환경제어 시스템으로도 활용할 수 있을 것이다.

Selection of 5-Methyltryptophan and S-(2-Aminoethyl)-L-Cysteine Resistant Microspore-Derived Rice Cell Lines Irradiated with Gamma Rays

  • Microspore-derived cell lines resistant to 5-methyltryptophan (5MT, a tryptophan analog) or S-(2-aminoethyl)-L-cysteine (AEC, a Iysine analog) were selected in rice by in vitro mutagenesis. For selection of 5MT or AEC resistant cell lines, suspension-cultured cells were irradiated with gamma rays. Thirteen 5MT resistant cell lines were selected and they were able to grow stably at 2 times higher 5MT concentration. A feedback insensitive form of anthranilate synthesis, the pathway specific control enzyme for tryptophan synthesis, was detected from the 5MT resistant lines. Contents of the free amino acids in five resistant lines (MR12-1 to MR12-5) showed a 7.4 to 46.6 times greater level than that in the control culture. Tryptophan, phenylalanine, and tyrosine levels in the shikimate pathway were 28.1 and 22.5 times higher in MR12-3 and MR12 4, respectively, than that measured in the control cells. Four AEC resistant cell lines were isolated from cultures grown on medium containing 1 mM AEC, They were able to grow stably with 2 mM AEC, while sensitive calli were inhibited by 0.5 mM AEC. Aspartate kinase activities of the resistant lines were insensitive to the natural inhibitor, Iysine, and accumulated 2.2 to 12.9-fold higher levels of free Iysine than that of the control cells. Especially, the levels of aspartate, asparagine, and methionine in the aspartate pathway showed higher accumulation in the AEC resistant lines than that in the control cells.

백로와 왜가리 집단번식이 소나무군집에 미치는 영향 (Effects of Group Breedling of Herons of Pine Community)

  • Effects of group breeding of herons on pine community were studied at Pomaeri, in Yangyang, Kangwon Province, Korea. This site has been protected as a Natural Monument (No. 229) since 1970. Herons have used this habitat as a breeding site from] anuary to October every year. In 1995, more than 500 herons were observed in this habitat. Many big pine trees are dying or already dead due to group inhabitation of herons, and no pine saplings were found at forest floor in this habitat. Nutrient contents of soil in this habitat were much higher than those in control plot. This must be due to the addition of feces from herons and of thin twigs and other organic materials from the canopy and bird nests. Species composition of herb layer in this habitat was quite different from that in control plot. Breeding site was dominated by Humulus japonicus. Persiearia perJohata, Persicaria thunbergii. and Commelina communis. which are indicator species of soil eutrophication.

Relationship between porcine miR-20a and its putative target low-density lipoprotein receptor based on dual luciferase reporter gene assays

  • Objective: Mutations in low-density lipoprotein receptor (LDLR), which encodes a critical protein for cholesterol homeostasis and lipid metabolism in mammals, are involved in cardiometabolic diseases, such as familial hypercholesterolemia in pigs. Whereas microRNAs (miRNAs) can control LDLR regulation, their involvement in circulating cholesterol and lipid levels with respect to cardiometabolic diseases in pigs is unclear. We aimed to identify and analyze LDLR as a potential target gene of SSC-miR-20a. Methods: Bioinformatic analysis predicted that porcine LDLR is a target of SSC-miR-20a. Wild-type and mutant LDLR 3'-untranslated region (UTR) fragments were generated by polymerase chain reaction (PCR) and cloned into the pGL3-Control vector to construct pGL3 Control LDLR wild-3'-UTR and pGL3 Control LDLR mutant-3'-UTR recombinant plasmids, respectively. An miR-20a expression plasmid was constructed by inserting the porcine premiR-20a-coding sequence between the HindIII and BamHI sites in pMR-mCherry, and constructs were confirmed by sequencing. HEK293T cells were co-transfected with the miR-20a expression or pMR-mCherry control plasmids and constructs harboring the corresponding 3'-UTR, and relative luciferase activity was determined. The relative expression levels of miR-20a and LDLR mRNA and their correlation in terms of expression levels in porcine liver tissue were analyzed using reverse-transcription quantitative PCR. Results: Gel electrophoresis and sequencing showed that target gene fragments were successfully cloned, and the three recombinant vectors were successfully constructed. Compared to pMR-mCherry, the miR-20a expression vector significantly inhibited wild-type LDLR3'-UTR-driven (p<0.01), but not mutant LDLR-3'-UTR-driven (p>0.05), luciferase reporter activity. Further, miR-20a and LDLR were expressed at relatively high levels in porcine liver tissues. Pearson correlation analysis revealed that porcine liver miR-20a and LDLR levels were significantly negatively correlated (r = -0.656, p<0.05). Conclusion: LDLR is a potential target of miR-20a, which might directly bind the LDLR 3'-UTR to post-transcriptionally inhibit expression. These results have implications in understanding the pathogenesis and progression of porcine cardiovascular diseases.

Rice Insects : The Role of Host Plant Resistance in Integrated Management Systems

  • Insects are among the most important abiotic and biotic constraints to rice production. National rice research programs are in various stages in the development and implementation of integrated pest management (IPM) stratagies for rice insect control. Among the various control tactics, insect resistant cultivars are sought as the major tactic in rice IPM. Through the activities of interdisciplinary teams of scientists significant progress has been made in the development and release of insect resistant cultivars to farmers. Because of its compatibility with other control tactics insect resistance has proven to fit well into the IPM approach to rice insect control agents and minimize the need for insecticide applications. The development of biotypes which overcome the resistance in rice plants has been a significant constraint in the breeding of rice for resistance to insects. Most notable examples in Asia are the green leafhopper, Nephotettix virescens, brown planthopper, Nilaparvata lygens and the Asian rice gall midge, Orseolia oryzae. The current breeding stratege is to develop rice cultivars with durable resistance on which virulent biotypes cannot adapt. In spite of the significant progress made in the breeding of insect resistant cultivars there are still numerous important rice insect species for which host plant resistance as a control tactic has not been fully utilized. Advances in biotechnology provide promise of solving some of the problems that have limited the use of host plant resistance as a major tactic in the integrated management of rice insect pests.

Advances towards Controlling Meiotic Recombination for Plant Breeding

  • Meiotic homologous recombination generates new combinations of preexisting genetic variation and is a crucial process in plant breeding. Within the last decade, our understanding of plant meiotic recombination and genome diversity has advanced considerably. Innovation in DNA sequencing technology has led to the exploration of high-resolution genetic and epigenetic information in plant genomes, which has helped to accelerate plant breeding practices via high-throughput genotyping, and linkage and association mapping. In addition, great advances toward understanding the genetic and epigenetic control mechanisms of meiotic recombination have enabled the expansion of breeding programs and the unlocking of genetic diversity that can be used for crop improvement. This review highlights the recent literature on plant meiotic recombination and discusses the translation of this knowledge to the manipulation of meiotic recombination frequency and location with regards to crop plant breeding.
