  • Eight natural or semistynthesized monoterpenes were examined for their effects in rat brain monoamine oxidase(MAO) using benzylamine as substrate. Thujone and 3-carene were found to have the inhibition effects on rat brain MAQ activity, 38% and 95% inhibition at 103M respectively. The kinetic study on 3-carene, the most potent inhibitive type. But (+) pulegon and (-) isopulegon was found to activate MAO slightly.

  • The effects of monosodium glutamate (MSG) on the activities if tyrosine hydroxylase (TH), dopamine ${\beta}$-hydroxylase (DBH), tryptophan hydroxylase (TPH) and monoamine oxidase (MAO) in various regions (cerebral cortex, striatum, midbrain, pons and medulla of nat brain have been determined. It was observed that up to 1mM MSGhad no significant effects on the activities of brain tyrosine hydroxylase, dopamine ${\beta}$-hydroxylase, tryptophan hydroxylase and monoamine oxidase in all regions of rat brain. These results indicated that MSG itself exerted no direct effect on the important enzymes synthsizing and metabolizing the monoaminergic neuronal system.

  • This study was undertaken in order to evaluate effects of methanol extracts of Stachys sieboldii MIQ and its related enzyme activities in brain tissues of rats. Sprague-Dawley(SD) male rats were fed within a control group, which is a basic diet group. The experimental diet group was given 100 and 200 mg/kg to supervise 100 and 200 mg/kg body weight per day for 20 days. Lipid peroxide levels and acetylcholine esterase activity in brain tissues were slightly decreased at a dose dependent manner, in vitro. Lipid peroxide levels were also decreased at a dose dependent manner; methanolic extracts of Stachys sieboldii MIQ demonstrated significant inhibitory effects, in vivo. Monoamine oxidase and xanthine oxidase activities were significantly inhibited in the brain tissues of experimental group compared to control group and the ratio of type conversion of xanthine oxidase were decreased.

  • The inhibitory effects of MeOH extracts of 100 medicinal herbs on monoamine oxidase (MAO) activity were investigated. MAO was purified from mouse brain and its activity was determined by fluorospectrophotomer using kynuramine as a substrate. Nine kinds of MeOH extracts of herbs including Artemisia iwayomogi showed a mild inhibitory effect with ${100}-{200}\;{\mu}/ml$ in their $IC_{50}$ values on MAO activity. Seventeen MeOH extracts including Juglans mandshurica exhibited a weak inhibition of MAO activity with ${200}-{300}\;{\mu}/ml$ in their $IC_{50}$ values.

  • The age-related changes of monoamine contents and monoamine oxidase (MAO) activities in the whole brain and blood pressures were measured and compared every week during the ages of 5 weeks to 12 weeks in two types of animals; (1) spontaneously hypertensive rats (SHR) and (2) normotensive Sprague-Dawley rats (NR). Blood pressures in SHR began to rise at the ages of 7 or 8 weeks and were significantly higher thereafter than in NR. Abnormally low MAO activities in the brain of SHR were detected from the ages of 8 weeks. Comparisons were also made between brain monoamine-norepinephrine, dopamine and 5-hydroxytrytamine contents of SHR and those of NR at the same ages throughout the test period. In SHR, none of those monoamine values was consistently higher or lower than in NR during the test period. Protein contents of the brain between SHR and NR were not significantly different at any ages. These observations suggest that the abnormally low MAO activities in SHR brain may be one of the underlying neurological factors for the susceptibilities to hypertension and the deficit of MAO activities may be due to the changes in properties rather than in amount of this enzyme.

  • The effects of evodiamine on monoamine oxidase (MAO) activity were investigated. MAO was purified from mouse brain and the $K_m\;and\;V_{max}$ values of MAO were $78.5{\pm}5.28{\mu}M$ and $0.68{\pm}0.07$ nmol/min/mg protein, respectively (n=4). Evodiamine at $30-120{\mu}M$ showed an inhibitory effect on MAO activity using a substrate kynuramine with an $IC_{50}$ value of $104.2{\mu}M$ (n=4). Evodiamine also exhibited a non-competitive inhibition on MAO. The $K_i$ value for evodiamine was $72.5{\pm}10.8{\mu}M$ (n=4). These results suggest that evodiamine partially contributes to the regulation of monoamine content.

  • The effects of MeOH extracts from 28 herbal medicines on monoamine oxidase (MAO) activity were investigated. MAO was purified from mouse brain and its activity was determined by fluoro-photometry using kynuramine as a substrate. Three MeOH extracts, Coptis japonica, Cinnamomum cassia and Poncirus trifoliate from the herbal medicines showed a strong inhibitory effect with less than $100\;{\mu}g/ml$ in their inhibitory amounts of $50\%$ ($IC_{50}$ values) on MAO activity. Four MeOH extracts including Evodia officinalis exhibited a mild inhibition of MAO activity with $100-200\;{\mu}g/ml$ in their $IC_{50}$ values.

  • The effects of coptisine on monoamine oxidase (EC 1.4.3.4; MAO) activity in mouse brain were investigated. Coptisine showed an inhibitory effect on MAO activity with a concentration-dependent manner. Coptisine exhibited 51.0% inhibition of MAO activity at $9\;{\mu}M$. The $IC_{50}$ value of coptisine was $8.7\;{\mu}M$. Coptisine inhibited MAO activity competitively with kynuramine as a substrate. The $K_i$ value of coptisine was $4.1\;{\mu}M$. These results indicate that coptisine functions to regulate the catecholamine content at biologically active sites.

  • We examined the inhibitory activities against monoamine oxidase (MAO) of Prunella vulgaris in vitro and in vivo methods. Methanolic extract of P. vulgaris showed significantly Inhibitorγ activities on MAO-A and MAO-B that were prepared from rat brain and liver in vitro. The inhibitory activities were measured by serotonin and benzylamine as substrates, respectively. MAO-A and MAO-B activities were potently inhibited by ethylacetate extracts of P. vulgaris in vitro tests. It was observed that those activities in vivo tests have different tendency each other. MAO-A activity was increased by the oral administration of methanolic extract of P. vulgaris while MAO-B activity was decreased. Consequently, we suggest that P. vulgaris may have the effects on the inhibitory activities against MAO both in vitro and in vivo.

  • Objectives: This study was undertaken to investigate the action mechanism of Holotrichia (HT) at parameter related to dementia. Methods: HT was tested for the effects on acetylchonine esterase and monoamine oxidase activities, lipid peroxidation, antioxidation in brain of lead acetate-treated rats. Lead acetate were treated firstly into samples for 10 days, and then lead acetate and HT were set with them for 20 days. Results: The level of lipid peroxide, acetylcholine esterase and monoamine oxidase activities, enzyme activities and ratio of type conversion of xanthine oxidase increased in lead acetate-treated rats were decreased as highly as normal group by HT. Superoxide dismutase, catalase and glutathione peroxidase activities, the level of acetylcholine decreased in lead acetate-treated rats were increased as lowly as normal group by HT. Conclusions : These results suggest that HT might have an effect on treatment of dementia according to decreasing the activities of acetylcholine esterase, monoamine oxidase and level of lipid peroxide in brain.
