백서 뇌 피질 Homogenate 내 ATPase 활성도에 미치는 수종 최면제 및 안정제의 영향 (Effects of Various Hypnotic and Tranquilizer on the Homogenate ATPase Activity of the Rat Brain Cortex)

  • The activity of Mg and Na-K activated ATPase of homogenate from rat brain cortex was measured in vitro under the variety of conditions. The effects of various hypnotic and tranquilizer such as phenobarbital, amobarbital, diazepam, promazine and chlorpromazine on the activities of both ATPase was investigated and the results was summarized as follows. 1. Na-K ATPase was slightly inhibited by phenobarbital and amobarbital while Mg ATPase was moderately activated by these drugs. 2. Both Mg and Na-K ATPase activities were markedly inhibited by diazepam. 3. Promazine and chlorpromazine markedly inhibited both Mg and Na-K ATPase activities. These findings indicate that remarkable correlation between hypnotic or tranquilizing potency and ATPase inhibition could be observed.

활성산소에 의해 유도된 흰쥐 뇌조직의 지질산화에 대한 식물체 추출물의 효과 (Effect of Plants Extracts on Lipid Peroxidation of Rat Brain Tissue Induced by Reactive Oxygen Species)

  • Sprague-Dawley 계열의 흰쥐를 perfusion시킨 후 분리한 뇌조직을 이용하여 뇌의 산화적 손상을 억제할 수 있는 식물체 및 항산화제의 활성을 조사하였다. 뇌의 산화적 손상 유도를 위해서는 활성산소원으로 $2.5{\mu}M$ ferrous sulfate와 7.5mM hydrogen peroxide를 뇌조직 homogenate에 첨가하였으며 손상의 정도는 malondialdehyde와 4-hydroxyalkenal 총 함량으로 표현되는 지질 과산화도(LPO)를 이용하였다. $37^{\circ}C$에서 10분간의 처리 시 LPO는 4.1에서 6.9nmol/mg protein 증가하였다. 한편, 식물체들의 뇌 손상 억제 효과를 분석하기 위해, 68종의 식용식물체로부터 수용성 추출물을 조제한 후 이를 활성산소 처리 전에 뇌 조직 homogenate에 첨가함으로써 지질산화 억제 정도를 비교하였다. 그 결과 엽채류에서는 아욱, 열무잎, 비름, 무순, 셀러리 등이, 근채류에서는 조림감자, 생강, 고구마 등이 80% 이상의 LPO억제 효과를 나타냈다. 과채류와 과일에서는 풋고추, 꽈리고추, 쥬니키, 가지, 아보카도, 오렌지, 바나나 등이 60% 이상의 효과를 보였으며, 버섯류에서는 팽이버섯, 양송이버섯, 표고버섯, 새송이버섯, 느타리버섯 순으로 효과가 높게 나타났다. 한편, 32종의 항산화제 중에서는 (+)catechin(91%), (-)epigallocatechin gallate(85%), (-)epicatechin gallate(83%), kaempferol(83%), chlorogenic acid(78%), melatonin(75%), ${\alpha}-tocopherol(69%)$ 등이 산화적 뇌 손상 방어에 우수한 것으로 나타났다.

Protein Phosphatase 2A의 활성화에 미치는 Lipid Bilayer Membrane의 저해 효과 (Inhibitory Effect of Lipid Bilayer Membrane on Protein Phosphatase 2A)

  • protein phosphatase 2A는 bovine brain homogenate의 세포질 fraction에서 얻어졌다. 기질로서 인산화된 histione H1을 이용하여 측정한 phosphatase 의 활성은 dipalmitoyIphophatidylcholine(DPPC) 혹은 phosphatidylserine/DPPC의 혼합물로 구성된 liposome의 존재하에서 저해되었다. Protein phosphatase 2A의 lipid membrane에의 결합은 다중층 지질막의 혼합물 계에서 liposome 의 양이 증가함에 따라서 상등액 중의 phosphatase의 활성이 감소하는 것으로 확인할 수 있었다. 또한 [$^{125}I$]protein phosphatase 2A가 liposome과 동시에 용출되는 것으로도 확인되었다. 그러나 liposome에 대한 protein phosphatase의 친화력은 높지 않았다. 한편, okadaic acid와 liposome은 협동으로 phosphatase의 활성을 감소시켰다. 이것은 okadaic acid가 lipid membrane이나 membrane에 결함한 phosphatase에는 결합하지 않는다는 것을 의미한다. 그러므로 lipid membrane에 의한 protein phosphatase 2A의 활성 저해 효과는 phosphatase 2A와 lipid membrane과의 결합에 의한 것이라고 설명될 수있다.

Production and Characterization of Monoclonal Antibodies to Bovine Brain Succinic Semialdehyde Reductase

  • Monoclonal antibodies against bovine brain succinic semialdehyde reductase were produced and characterized. A total of nine monoclonal antibodies recognizing different epitopes of the enzyme were obtained, of which two inhibited the enzyme activity and three stained cytosol of rat spinal cord neurons as observed by indirect immunofluorescence microscopy. When unfractionated total proteins of bovine brain homogenate were seperated by gel electrophoresis and immunoblotted, the antibodies specifically recognized a single protein band of 34 kDa, which comigrates with purified bovine succinic semialdehyde reducatase Using the antisuccinic semialdehyde reductase antibodies as probes, we investigated the cross-reactivites of brain succinic semialdehyde reductases from some mammalian and an avian species. The immunoreactive bands on Western blots appeared to be the same in molecular mass-34 kDa-in all animal species tested, including humans. The result indicated that brain succinic semialdehyde reductase is distinct from other aldehyde reductases and that mammalian brains contain only one succinic semialdehyde reductase. Moreover, the enzymes among the species are imunologically very similar, although some properties of the enzymes reported previously were different from one another.

Heat Processing of Edible Plants Grown in Korea Has Differential Effects on Their Antioxidant Capacity in Bovine Brain Homogenate

  • Oxidant radicals are implicated as a causal factor in the pathogenesis of neurobiological disorders and neuro-degenerative diseases. The objective of this study was to investigate the antioxidant activity of edible plants against oxidative stress in bovine brain tissue. Fifty five kinds of edible plants grown in Korea were dried either by freeze-drying or hot-air drying (7$0^{\circ}C$), and evaluated for their antioxidant activity by measuring TBARS (thiobarbituric acid-reactive substances) in brain homogenates subjected to Fe$^{+2}$_mediated lipid peroxidation with or without the addition of botanical extracts. Heat-drying decreased the antioxidant activity of most plant extracts by 10~81%, compared with freeze-drying. However, Aruncus americanus, Ligularia stenocephala, Artemisia princceps var. orientalis, Petasites japonicus and Aster scaber showed very strong antioxidant activities regardless of processing, with or without heat treatment. The $IC_{50}$/ values of the methanol extracts from these edible plants were in the range of 0.093~0.379 mg/$m\ell$, which was lower than that of ascorbic acid (0.79 mg/$m\ell$). Thermal processing of some edible plants enhanced their antioxidant activity.

Regulation of Genetic Aggression by Central Dopamine System - Plurality of Dopamine Receptor -

  • Two types of aggressive behavior were produced by selective breeding in ICR mimce. NC900 line mice exhibited high level of species-typical, isolation-induced aggression, conversely, NC100 line mice exhibited little aggression. The present study tested the hypothesis that these differences involved brain monoamine systems. Comparisons of microdissected samples from various brain regions showed that NC100 line mice had significantly lower concentrations of dopamine. DOPAC and HVA in the nucleus accumbens (NAB) and caudate nucleus (NCU) than NC900 line. Homogenate binding studies demonstrated that NC100 mice had significantly increased density of $D_1$ dopamine receptor, but not $D_2$ dopamine receptor in the caudate nucleus. These results support the hypothesis that central dopamine pathways play an important role in modulating the genetically selected differences in aggressive behavior, and of which intensity differs from TEX>$D_1$\;and\;$D_2$ dopamine receptors.

야생초 추출물에 의한 간장내 활성산소 생성과 항산화 효소계 조절에 관한 연구 (Modulation of Hepatic Lipid Peroxidation and Antioxidant Defenses by Wild Plants Extracts)

  • This study was performed to elucidate the possible antioxidative effects of several wild plant extracts. Wild plants were extracted with methanol or water using general method. In first experiments, antioxidative effects were measured by lipid peroxidation using rat brain homogenate. Coptis japonica extract showed the highest antioxidative activity among the 15 wild plant extracts. In second experiments, rats were fed on the semipurified diets with or without Coptis japonica extracts at the level of 0.5% for 4 weeks. MDA production of liver homogenate were significantly lower in the rats fed Coptis japonica extracts (P<0.05). Cytosolic catalase. GPX, and SOD activities were not changed, whereas the activities of GST and glutathione level were significantly higher in rats fed Coptis japonica extracts (P<0.05). These results suggest that Coptis japonica extract has an antioxidative effect through increasing GST activity and glutathione level and decreasing MDA production.

자기면역성 뇌척수염 조직에서 CPP32의 면역조직화학적 관찰 (Immunohistochemical study of CPP32 (Caspase-3) in the spinal cords of rats with experimental autoimmune encephalomyelitis)

  • The aim of this study was to evaluate the involvement of CPP32 (caspase-3), one of the death-related enzymes, in the course of experimental autoimmune encephalomyelitis (EAE). EAE was induced in Lewis rats immunized with an emulsion of rat spinal cord homogenate with complete Freunds adjuvant supplemented with Mycobacterium tuberculosis (H37Ra, 5mg/ml). The expression of CPP32 in the spinal cords of rats with EAE was studied. In normal rat spinal cords, CPP32 is constitutively, but weakly, expressed in neurons and some neuroglial cells. In the EAE spinal cords, many inflammatory cells were positive for CPP 32, and the majority of CPP32(+) cells were identified as ED1(+) macrophages. During this stage of EAE, the number of CPP32(+) cells in brain cells, including neurons and astrocytes, increased, and these cells also had increased CPP32 immunoreactivity. CPP32 immunor eactivity was not always matched with apoptosis of inflammatory cells in EAE lesions. We speculate that CPP32, which is constitutlvely expressed in brain cells, increases in response to neuroimmunological stimulation in both brain neuronal cells and inflammatory cells. The functional role of CPP32 in neuroimmunological disorders is discussed.

수종 식물추출물의 항히스타민작용 (Antihistaminic Action of the Several Medicinal Plant Extracts)

  • The antihistaminic action of eighteen herbal medicines was investigated by the radioligand binding and functional assays. The hexane fractions of Trichosanthis radix, Mori cortex radicis and Evodiae fructus dosedependently inhibited [$^3$H] mepyramine binding to H$_1$, receptor in guinea-pig brain homogenates and histamine-induced contraction of isolated guinea-pig ileum. Antihistaminic action of the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus was more potent than their antimuscarinic action evaluated from the inhibition of [$^3$H]QNB binding and carbachol response. The ethyl acetate and chloroform fractions from Scutellariae radix also inhibited histamine-induced contraction, but antihistaminic potencies of these fractions were almost identical with their antimuscarinic potencies. The hexane fractions of Mori cortex radicis and Evodiae fructus inhibited selectively the increase of histamine-induced cutaneous vascular Permeability in the rat dorsal skins. However, the ethyl acetate fraction from Scutellariae radix inhibited eqipotently the effects of histamine and serotonin on the vascular permeability. These results demonstrate that the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus have the selective histamine H$_1$receptor blocking activity.
