Improvement of Virus Safety of an Antihemophilc Factor IX by Virus Filtration Process

  • Viral safety is an important prerequisite for clinical preparations of plasma-derived pharmaceuticals. One potential way to increase the safety of therapeutic biological products is the use of a virus-retentive filter. In order to increase the viral safety of human antihemophilic factor IX, particularly in regard to non-enveloped viruses, a virus removal process using a polyvinylidene fluoride membrane filter (Viresolve NFP) has been optimized. The most critical factor affecting the filtration efficiency was operating pH and the optimum pH was 6 or 7. Flow rate increased with increasing operating pressure and temperature. Recovery yield in the optimized production-scale process was 96%. No substantial changes were observed in the physical and biochemical characteristics of the filtered factor IX in comparison with those before filtration. A 47-mm disk membrane filter was used to simulate the process performance of the production-scale cartridges and to test if it could remove several experimental model viruses for human pathogenic viruses, including human hepatitis A virus (HAV), porcine parvovirus (PPV), murine encephalomyocarditis virus (EMCV), human immunodeficiency virus type 1 (HIV), bovine viral diarrhea virus (BVDV), and bovine herpes virus (BHV). Non-enveloped viruses (HAV, PPV, and EMCV) as well as enveloped viruses (HIV, BVDV, and BHV) were completely removed during filtration. The log reduction factors achieved were $\geq$6.12 for HAV, $\geq$4.28 for PPV, $\geq$5.33 for EMCV, $\geq$5.51 for HIV, $\geq$5.17 for BVDV, and $\geq$5.75 for BHV. These results indicate that the virus filtration process successfully improved the viral safety of factor IX.

한우 송아지에서 급성 BVDV-1b 발생보고 (Acute BVDV-1b Outbreak in Korean Indigenous Calves)

  • 2011년 경북 영주의 여러 농가에서 이병율과 치사율이 높은 소 바이러스성 설사 바이러스(BVDV)가 발생하였다. 유전자 분석 결과 두 개의 유전자형 BVDV-1b (n = 21)와 BVDV-2a (n = 7)이 확인되었다. 검사결과, 이 지역의 농가에서는 BVDV-1b 유전자형이 가장 많이 검출되었고, 또한 몇 농가에서 BVDV-2a 유전자형도 검출되었다. 이들 농가에서 발생한 BVDV-1b 감염은 BVDV-2 감염과 유사한 중증의 급성 임상증상을 보여 주었다. 이 결과는 한우 송아지에서 급성이면서 치명적인 BVDV-1b 발생을 보고한다.

Identification of Korean native cattle persistently infected with BVDV using Ear-notch method

  • Bovine viral diarrhea Virus (BVDV) infections cause respiratory, gastrointestinal, and reproductive problems, such as infertility, abortion, stillbirth, and sickly offspring. Many countries have reduced the economic damage through the application of different control programmes, and some have successfully eradicated BVD. Detection and elimination of cattle persistently infected (PI) with BVDV is important for BVD eradication because PI cattle are a main source of BVD transmission. In this study, the prevalence of Korean native cattle persistently infected (PI) with BVDV was investigated and determined in 49 farms with 3,050 cattle. The all samples were collected by ear notch sampling. Korean native cattle with initial positives on antigen-ELISA (Ag-ELISA) were sampled again after 3~4 weeks and cattle with second positives in both Ag-ELISA and immunohistochemistry (IHC) were identified as PI cattle. Among the 49 farms, 14 (28.6%) farms had at least more than one PI cow and 21 (0.69%) of 3,050 cattle were determined as PI cattle. As a result of this work, it is suggested that national BVD eradication program is required to reduce economic losses by BVDV infection in Korean cattle industries.

광주지역 한우 분변 내 설사병 병원체 조사 (Prevalence of enteropathogens in the feces from diarrheic Korean native cattle in Gwangju area, Korea)

  • Calf diarrhea is a common disease in young claves and is still a major cause of productivity and economic loss in livestock farms. Fecal samples from Korean native cattle (n=100) with diarrhea from 64 farms in Gwangju area, Korea from september 2017 to December 2018 were examined for shedding of important protozoan parasitic, viral and bacterial pathogens using culture, rapid test kit and PCR methods. Of 57 (89.1%) of the 64 Korean native cattle farms examined had samples infected with at least one of the investigated pathogens. Among 100 fecal samples, 88 samples were positive for at least one the twelve pathogens and 51 samples were simultaneously positive for two or more pathogens by culture and PCR assay. Bovine group A rotavirus (BRV) was the most common pathogen, found in 43/100 (43.0%) samples on 32/64 (50.0%) farms. Subsequently, kobuvirus (30.0%), pathogenic E. coli (29.0%), bovine parvovirus (17.0%), Giardia spp. (13.0%), Eimeria spp. (10.0%), Clostridium perfringens type A (8.0%), bovine torovirus (8.0%), bovine viral diarrhea virus (6.0%), bovine coronavirus (5.0%), bovine norovirus (2.0%) and Cryptosporidium spp. (2.0%) were detected. Nebovirus, kırklareli virus, bovine adenovirus, Salmonella spp. and intestinal parasites were not detected. Of the 72 calves sampled in this age group, 64 (88.9%) samples were positive for at least one enteropathogen. BRV was identified in 34/72 (47.2%) samples from 27/48 (56.3%) farms. Subsequently, pathogenic E. coli (30.6%), kobuvirus (29.2%), BPaV (22.2%), Giardia spp. (15.3%), Eimeria spp. (9.7%), BVDV (6.9%), Cl. perfringens type A (6.9%), BCoV (4.6%) and Cryptosporidium spp. (2.8%) were detected in fecal samples. A total of ninety-six strains of E. coli were isolated from one hundred fecal samples collected from Korean native cattle with diarrhea. The presence of stx1, stx2, eaeA, LT, STa, STb, ehxA, saa, F4, F5(K99), F6, F17, F18 and F41 genes in the isolates was investigated by PCR. Out of ninety-six E. coli isolates screened for specific genes, 30 strains E. coli were identified to harbor shiga toxin-producing E. coli (STEC) 7 (7.3%), enterohemorrhagic E. coli (EHEC) 8 (8.3%), enteropathogenic E. coli (EPEC) 6 (6.3%), enterotoxigenic E. coli (ETEC) 2 (2.1%) and STEC/ETEC hybrid 7 (7.3%). This study provides epidemiological estimates of the prevalence of Korean native cattle's enteropathogens in Gwangju area, Korea, which would be used for cattle farmers and veterinarians to select appropriate therapeutic method.

재래산양의 설사병 병원체 감염률 조사 (Status of diarrhea pathogens from Korean indigenous goat feces)

  • The purpose of this study was to survey on infection status of pathogens of diarrhea from Korean indigenous goat. A total of 800 fecal samples was collected from 50 farms from January to November 2016 and was tested by automatic biochemical machine and polymerase chain reaction (PCR). The overall infection rates of parasitic, bacterial and viral pathogens was 13.0%, 23.0%, 11.3% and the rates of coccidia, Escherichia coli (E. coli), bovine viral diarrhea virus (BVDV), rotavirus and coronavirus were 13.0%, 23.0%, 5.3%, 8.8% and 2.6%, respectively. In the rates of mixed detection, single was 29.8%, double 5.1%, triple 2.8%, quadruple 1.1% in each sample, respectively.

Dry-Heat Treatment Process for Enhancing Viral Safety of an Antihemophilic Factor VIII Concentrate Prepared from Human Plasma

  • Viral safety is a prerequisite for manufacturing clinical antihemophilic factor VIII concentrates from human plasma. With particular regard to the hepatitis A virus (HAV), a terminal dry-heat treatment ($100^{\circ}C$ for 30 min) process, following lyophilization, was developed to improve the virus safety of a solvent/detergent-treated antihemophilic factor VIII concentrate. The loss of factor VIII activity during dry-heat treatment was of about 5%. No substantial changes were observed in the physical and biochemical characteristics of the dry-heat-treated factor VIII compared with those of the factor VIII before dry-heat treatment. The dry-heat-treated factor VIII was stable for up to 24 months at $4^{\circ}C$. The dry-heat treatment after lyophilization was an effective process for inactivating viruses. The HAV, murine encephalomyocarditis virus (EMCV), and human immunodeficiency virus (HIV) were completely inactivated to below detectable levels within 10 min of the dry-heat treatment. Bovine herpes virus (BHV) and bovine viral diarrhea virus (BVDV) were potentially sensitive to the treatment. However porcine parvovirus (PPV) was slightly resistant to the treatment. The log reduction factors achieved during lyophilization and dry-heat treatment were ${\geq}5.55$ for HAV, ${\geq}5.87$ for EMCV, ${\geq}5.15$ for HIV, 6.13 for BHV, 4.46 for BVDV, and 1.90 for PPV. These results indicate that dry-heat treatment improves the virus safety of factor VIII concentrates, without destroying the activity. Moreover, the treatment represents an effective measure for the inactivation of non-lipid-enveloped viruses, in particular HAV, which is resistant to solvent/detergent treatment.

경남 남부지역 젖소 사육 농가의 소바이러스성설사병(BVD) 감염실태 조사 (Prevalence of bovine viral diarrhea virus from dairy cattle farms in Gyeongnam southern area, Korea)

  • Bovine viral diarrhea virus (BVDV) is one of the most important disease viruses in cattle that can cause severe economical losses due to decreased fertility, abortion, diarrhea, and respiratory symptoms. Therefore, this study was aimed to investigate prevalence of BVDV infection (Transiently infection, Persistently infection) in dairy cattle in Gyeongnam southern area, Korea and use this data as the basis for establishing an eradication program and policy. A total of 44 bulk-tank milk samples (farms) collected in milk collecting center were tested for BVDV antibody using an ELISA. As the result, out of a total of 44 bulk-tank milk samples (farms), 38 (86.4%) samples were BVDV antibody positive. Blood samples (17 farms, n=543) were collected from BVDV antibody positive farms in bulk-tank milk, tested for BVDV antigen with ELISA and PCR. BVDV infected farms were 47% (8/17) and BVDV infected head were 2.2% (12/543). Persistently infected cattle (PI) were detected at 6 (35.3%) farms out of 17 farms and a total of 6 (1.1%) out of 543 head of cattle were identified as PI. The seropositive of BVDV antibody at farms and head were 100% (17/17) and 49.45% (91/184), respectively. The seroprevalence of BVDV antibody in PI infected farms (67.35%) much higher than that of BVDV antibody in transiently infected cattle (TI) infected farms (45%) and uninfected farms (34.48%). For eradication of BVDV infection in cattle populations, First of all, we should remove PI and need vaccination.

The prevalence of causative agents of calf diarrhea in Korean native calves

  • Infectious calf diarrhea is one of the most significant diseases of neonatal calves. This study is conducted to identify the prevalence of pathogens in calf diarrhea for 2 years. A total of 544 feces samples from Korean native beef calves were obtained to investigate selected seven pathogens causing calf diarrhea: bovine rotavirus, bovine coronavirus, Cryptosporidium parvum, bovine viral diarrhea virus, Eimeria species, Escherichia coli K99, and Salmonella species. The presence of diarrhea, the number and species of detected pathogens, and the calves' ages were analyzed using various statistical methods depending on the case. Of the 544 calves, 340 calves (62.5%) had normal feces and 204 calves (37.5%) had diarrhea. The presence of pathogens was significantly associated with diarrhea (p < 0.01) and fecal scores and the number of detected pathogens showed a significant linear trend (p < 0.001). Of the 7 target pathogens, 6 were detected in samples, but only C. parvum (p = 0.001) and bovine rotavirus (p < 0.001) were found at significantly higher rates in diarrheic calves than in non-diarrheic calves. Only Eimeria spp. showed a significant linear trend between the detection rate of the pathogen and the age groups (p < 0.05).

광주(光州)·전남지역내(全南地域內) 소의 바이러스성 질병(疾病)에 관한 혈청학적(血淸學的) 연구(硏究) (Seroepidemiological studies on virus-borne diseases of cattle in Kwangju and Chonnam area)

  • Sera from 304 Holsteins or Korean native cattle were collected from slaughterhouse in Kwangju area to study the infection of major virus-borne diseases. Serum antibody titers against infectious bovine rhinotracheitis virus(IBRV), bovine viral diarrhea virus(BVDV), parainfluenza type-3 virus(PI-3V), bovine ephemeral fever virus(BEFV), bovine Ibaraki virus(BIV), bovine Akabane virus(BAKV), bovine rotavirus(BRV), bovine coronavirus(BCV) were measured by serum neutralization tests. Results which obtained were as follows. Sera from 280 cattle(92.1%) contained antibodies against BRV which rate was the highest among the 8 viruses, and serum antibodies against BCV, BVDV, BIV, BAKV, BEFV, IBRV and PI-3V were detected from 266(87.5%), 149(49%), 108(35.5%), 94(30.9%), 80(26.3%), 32(10.5%) and 24(7.9%) cattle, respectively. Prevalence of seropositives to BVDV, BIV, BAKV, BEFV were higher among Holsteins than among the Korean native cattle(P<0.05). Prevalence of antibody titers against BVDV, BIV and BEFV in Korean native cattle were higher among females than males, while more males contained antibodies to BAKV, IBRV and PI-3V than females in their blood(P<0.05). Seropositives to BVDV, BIV, BAKV, BEFV and IBRV in Holsteins were higher among females than males(P<0.05). In Korean native cattle, serum antibody titers against IBRV and PI-3V ranged from 1:2~1:32 and 1:2~1:64, respectively, while serum antibody titers against the rest 6 viruses ranged from $1:2{\sim}1:{\geq}256$. In Holsteins, serum antibody titers against IBRV and PI-3V ranged from 1:2~1:64 and 1:2~1:32, respectively, while serum antibody titers against the rest 6 viruses ranged from $1:2{\sim}1:{\geq}256$.

원화추출물(PB-81)의 소 로타바이러스 설사병에 대한 항바이러스 및 치료효과 (Antiviral and Therapeutic Effects of Extracts (PB-81) of Daphne Genkwa (Siebold & Zucc.) on Bovine Rotavirus)

  • 원화(Daphne genkwa (Siebold & Zucc))의 50% 에탄올 추출물인 PB-81이 소 로타바이러스 설사병 환축에서 설사병의 치료효과와 바이러스 증식 억제 효과가 나타나는가를 확인하였다. PB-81에 의해 상피세포주인 A549 세포와 혈액세포주인 NK92 세포에서 각각 IFN-β와 IFN-γ가 유도되는 것을 확인하였다. PB-81의 바이러스의 증식억제 효과를 확인하기 위해 PB-81을 MBDK 세포에 바이러스의 감염 전, 동시, 감염 후에 투여하는 세 가지 경우에서 바이러스 억제효과를 확인한 결과 PB-81을 투여한 모든 경우에서 바이러스가 억제되었으며, 바이러스 감염 전에서 투여하는 경우에서 가장 좋은 바이러스 억제효과가 나타났다. 마우스에서의 독성검사에서는 투여최대용량인 20 mg/mL에서도 독성에 따른 부작용이 나타나지 않았다. 소 로타바이러스 설사병 환축 16두와 대조군 4두를 대상으로 한 치료효과 검증에는 20 mg/5 mL 용량의 PB-81을 투여한 결과, PB-81을 투여한 모든 소 로타바이러스 설사병 환축이 완치되었으며 PB-81의 투여 후에 완치까지 소요된 기간은 PB-81투여군이 평균 2.25일이었고, PB-81을 투여하지 않은 대조군이 6.5일로 PB-81의 투여에 의한 소 로타바이러스 설사병의 치료효과가 나타나는 것을 확인하였으며 PB-81을 투여한 모든 환축에서 부작용이 검출되지 않았다.
