  • Microorganisms causing bovine mastitis, swine diarrhea and swine pneumonia were isolated from farms in Chungbuk and Kyunggi Province and the isolates were identified using microbial identification system(MIS). The most common isolates from bovine mastitis were Staphylococcus sp.,Streptococcus sp., and Corynebacterium sp., those from swine diarrhea were Escherichia coli, Salmonella sp., and Clostridium perfringens, those from swine pneumonia were Bordetella bronchiseptica and Pasteurella multocida. Examination of 16 antibiotics against these pathogens revealed that the incidence of antibiotic-resistant microoganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. These results suggest that the use of antibiotics in a farm should be controlled in order to decrease the number of antibiotic-resistant microorganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. Development of a Rapid PCR Test for Identification of Streptococcus agalactiae in Milk Samples Collected on Filter Paper Disks

  • Streptococcus (Strep.) agalactiae is one of the major pathogens of bovine mastitis and is the main cause of subclinical infection. This study attempted to develop a rapid PCR diagnosis procedure using milk samples collected on filter paper disks. Chromatographic filter paper was employed as the preservation media and kept at room temperature for one to four weeks. The revival rate of Strep. agalactiae kept on dried filter paper disks was affected by the pretreatment preservation time. The revival test suggested that not all the bacteria in artificially contaminated milk samples on the filter paper disks could be recovered. After that, a PCR based on the 16-23S intergenic spacer region of Strep agalactiae was performed. The results distinguished the strep. agalactiae from major pathogens of bovine mastitis at a $2{\times}10^2$ colony forming units (CFU)/ml level, which showed similar sensitivity to the results from liquid milk samples. The results also showed that milk samples collected on filter paper disks could be kept at room temperature for one to four weeks with little negative effect on sensitivity and specificity. The field test showed that the diagnostic sensitivity and specificity was 96.15% and 98.60%, respectively. In conclusion, the protocol will provide a rapid and economic procedure for the detection of bovine mastitis.

  • Because Staphylococcus aureus (S. aureus) has variable number of short sequence repeat region in coagulase gene, it has been used to investigate the relatedness of S. aureus isolates. In this study, we isolated S. aureus strains from 20 dairy farms with bovine mastitis from September 2000 to August 2001. PCR-RFLP analysis of coagulase gene revealed 10 different patterns. Most of the S. aureus isolates showed only one coagulase gene RFLP pattern per farm. However, there were several S. aureus clones spreading between dairy farms. All the farms showed poor management conditions of milking machine and milker, indicating that managements for mastitis control program include use of proper milking matching, premilking sanitation, and segregation in the S. aureus infection herd. Our data suggest that PCR-RFLP analysis of coagulase gene might be applicable for the epidemiological investigations of S. aureus isolated from bovine mastitis cows.

Antibacterial Effect of Caesalpinia sappan Extract against Mastitis Pathogens from Dairy Cows (소목추출물의 젖소 유방염 원인균에 대한 항균효과)

  • To find alternative agents for bovine mastitis, the extracts of 30 selected herbal medicine were screened for their antibacterial activity against mastitis pathogens. The extracts of 9 herbal medicine, Terminaliae chebula, Cinnamonum loureirii, Paeonia japonica, Inula britannica, Caesalpinia sappan(C. sappan), Salix babylonica, Alpinia katsumadai, Polygonum aviculare and Coptidis japonica showed strong antibacterial activity. Among of them, C. sappan extract was found to be the most active against all bacteria in this experiment. Methanol extract of C. sappan and brazilin, the main component of C. sappan, had inhibitory activity against a variety of pathogens, however they had less of an inhibitory effect on gram negative bacteria. Minimal inhibitory concentrations of brazilin against gram positive pathogens was lower than that of methanol extract. These results showed that the extract of C. sappan has a potential as a therapeutic agent for bovine mastitis.

Therapeutic effects of honeybee (Apis Mellifera L.) venom injection on bovine mastitis (젖소 유방염에 대한 국내산 봉독의 치료 효과)

  • The therapeutic effect of honeybee venom collected using be venom collector on bovine mastitis was investigated. Mastitic cows from four farms were selected in the Yang-pyeong areas. Chronic mastitic cows were injected with the various concentrations of honey-bee venom per day. There was a significant difference in the reduction rates of somatic cell counts (SCC) according to treatment concentration and method of bee venom. The milk SCC were significantly decreased in all concentrations of bee venom 3 days after treatment. The reduction rates of SCC in treatment of 3, 6, 12 and 24mg honeybee venom were 20, 43, 63.3 and 65.8% respectively. Honeybee venom treatment consisted of two methods, a syringeful and a Bovivet Spenstift. The treatment with Bovivet Spenstift was more effective in the reduction rates of SCC compared with the syringeful. Thirty two out of 53 quarters were cured by Bovivet Spenstift with 12mg bee venom per day for 14 days. The venom cure rates of bovine mastitis by Escherichia coli, Stapylococcus aureus, Gram positive bacteria and Gram negative bacteria were 33.3, 75, 75 and 43.8% respectively. These results suggested that bee venom treatment (by Bovivet Spenstift with 12mg) might be effective for treatment of bovine mastitis.

Studies of Infection Rate and Causative Agents of Bovine Mastitis in Chungnam Area (충남지역(忠南地域) 유우유방염(乳牛乳房炎)의 감염율(感染率) 및 원인균(原因菌)에 관한 연구(硏究))

  • A total of 820 dairy cattle were examined formastitis by California mastitis test method and 181 dairy cattle were examined bacteriologically according to the method of Connecticut mastitis control program. The results obtained were as followings: 1. Milk samples from 1.231(38.1%) of 3,225 quarters and 568(69.3%) of 820 dairy cattle were positive for mastitis by California mastitis test method. 2. Among 181 cattle examined bacteriologically by the method of Connecticut mastitis control program 159(22.9%) quarters of 102(56.4%) cattle were infected. Clinical mastitis was found in 5 cattle. 3. Important causative organisms were found to de Staphylococcus aureus(44.8%), Streptococcus agalactiae(21.5%), and non-str, agalactiae Streptococcus(26.4%). Corynebacterium ssp., Pseudomonas spp., Coliforms and Yeasts were also occasionally found to be causative organisms. 4. Strains of Staphylococcus aureus isolated from bovine udders were tested for sensitivity to antibiotics. The rates of resistant strain were 13.7%, 11.8%, 8.8%, 5.9%, 100%, 4.9% and 4.9% for penicillin G, streptomycin, terramycin, erythromycin, chloramphenicol, ampicillin and kanamycin, respectively.

Evaluation of Bactericidal Capacity of Silver Ion against Bovine Mastitis Pathogens (소 유방염 원인균에 대한 은이온의 향균력 평가)

  • Bovine mastitis, defined as an inflammation of the mammary gland, is usually associated with bacterial infection. Thus, treatment and control of mastitis relies primarily on antimicrobial therapy. This study investigated the bactericidal actions of silver ion against causing various bovine mastitis pathogens. Minimal inhibitory concentrations (MICs), minimum bactericidal concentrations (MBCs) and bactericidal activity times and concentrations of silver ion against pathogens were determined. The effect of silver ion on bacterial morphology was studied by scanning electron microscopy (SEM). The MICs and MBCs of silver ion for various bacteria strains ranged from 1.9-15.6 ${\mu}g$/ml. SEM images indicated formation of a pit, distortion and disruption of cell walls in silver treated Staphylococcus aureus and Escherichia coli. The results demonstrate that silver ion has a bactericidal capacity against causing various pathogens of bovine mastitis and suggest that silver ions may be exploitable as a therapeutic/preventative tool of bovine mastitis.

Survey of mastitis management and incidence of mastitis in high somatic cell count of bulk milk at dairy farms in the Gyeongnam (경남지역의 체세포수 문제 목장에서의 젖소 유방염 관리실태 및 발생양상 조사)

  • Survey of mastitis management and incidence of mastitis in the Gyeongnam was started in May to September 2009 to solve mastitis problem statistically valid data for use in estimating mastitis management, isolation and antimicrobial drug susceptibility in 30 dairy farms having over 350,000/ml somatic cell count. In investigation on recognition of farmer about bovine mastitis, the ratio of understanding of differences between infectious and environmental origin, understanding of correlation between superbacteria and using indiscriminate, necessity of pathogen identification, and necessity of antimicrobial sensitivity tests were 80.0%, 73.3%, 33.3%, and 53.3%, respectively. In survey of mastitis management type, regular california mastitis test (CMT), conducting CMT test and empirical self-treatment, when detecting suspected cows, were 30.0%, 40.0%, and 46.7%, respectively. Checking and cleaning pulsators biweekly, cleaning vacuum system and replacing liners every 3~6 month, and getting milking system checked by engineers showed 80.0%, 76.7%, and 76.7% in the questionnaires, respectively. In recognition of farmer about milking hygiene for prevention of bovine mastitis, using individual towels, separated milking (milking order of cows), and teat-dipping disinfection after milking exhibited 13.3%, 86.7%, and 93.3%, respectively. In conclusion, through the questionnaires and laboratory test, we suggest that recognition of farmer about management and incidence of mastitis was very low, thus systemic educational program and public relations about mastitis management were need for dairy farmers.
