소 유방염, 돼지설사, 돼지 폐렴의 원인균 분리 동정과 항생제 내성균 분포 (Prevalence of Antibiotic-Resistant Strains among Bacteria Isolated from Bovine Mastitis, Swine Diarrhea, and Swine Pneumonia.)

  • Microorganisms causing bovine mastitis, swine diarrhea and swine pneumonia were isolated from farms in Chungbuk and Kyunggi Province and the isolates were identified using microbial identification system(MIS). The most common isolates from bovine mastitis were Staphylococcus sp.,Streptococcus sp., and Corynebacterium sp., those from swine diarrhea were Escherichia coli, Salmonella sp., and Clostridium perfringens, those from swine pneumonia were Bordetella bronchiseptica and Pasteurella multocida. Examination of 16 antibiotics against these pathogens revealed that the incidence of antibiotic-resistant microoganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. These results suggest that the use of antibiotics in a farm should be controlled in order to decrease the number of antibiotic-resistant microorganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. 한우 송아지의 소바이러스성 설사바이러스 지속감염률 조사 (Prevalence for persistent infection with bovine viral diarrhea virus in Korean native calves)

  • Bovine viral diarrhea (BVD) is very important disease in cattle industry with a worldwide distribution. Detection and elimination of persistently infected calves with bovine viral diarrhea virus (BVDV) was valuable strategy for BVD eradication because those calves were main source for transmission. We surveyed persistent infection with BVDV by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry (IHC) using whole blood and skin. Five hundred thirty nine Korean native calves were tested. Four calves (0.7%) were positive for BVDV antigen for both tests. Those calves remained positive for follow-up by RT-PCR and IHC. Therefore they were identified as persistently infected with BVDV. We confirmed that immunohistochemistry using skin biopsy samples was very useful tool to detect persistently infected calves with BVDV. As far as we know, this would be first report on persistent infection with BVDV in Korea.

  • 25일령의 한우송아지가 10일 간의 호흡기질환과 출혈성 설사의 병역을 나타내다가 폐사하였다. 이 송아지는 다른 송아지들과 비교해보았을 때 극도로 위축되어 있었다. 꼬리와 회음부의 분변 흔적은 만성설사가 지속되었음을 말해주었다. 부검시 다양한 기관(조직)에서 반상출혈이 관찰되었다. 역전사 중합효소연쇄반응에 의하여 소 바이러스성 설사병 바이러스가 진단되었다. 이 증례는 계통발생분석에서 BVDV-2a 그룹에 속하는데, 이것은 고병원성인 미국 균주890 (U18059)과 유사하다. 본 증례는 BVDV-2가 한국에서 유행하고 있다는 증거를 제공한다. 이를 통해 우리는 BVDV-2의 발생을 재확인하였다.

  • Between February 2020 and September 2021, 378 calves with diarrhea were investigated across 96 cattle breeding farms in Korea, using a rapid diagnostic kit. The study examined the infection rates of major pathogens causing diarrhea in calves, which were categorized by season, age, birth month, and region. Additionally, logistic regression analysis was conducted to investigate the factors affecting the infection rate. The study found that the five representative pathogens causing calf diarrhea exhibited differences in infection rates based on season, region, age, and birth month. Bovine rotavirus, bovine coronavirus, Cryptosporidium, and Giardia commonly exhibited varying risks of infection based on season and age. Furthermore, in addition to these risk factors, bovine rotavirus and Cryptosporidium were found to impact the infection risk of each pathogen by region, while Giardia was found to be affected by birth month.

세포배양 유래 생물의약품 제조공정에서 Reovirus, Bovine Viral Diarrhea Virus, Bovine Parainfluenza Virus 동시 검출을 위한 Multiplex Reverse Transcription-PCR (Multiplex Reverse Transcription-PCR for Simultaneous Detection of Reovirus, Bovine Viral Diarrhea Virus, and Bovine Parainfluenza Virus during the Manufacture of Cell Culture-derived Biopharmaceuticals)

  • 동물세포배양 유래 생물의약품 생산 공정에서 다양한 외래성 바이러스가 오염된 사례가 있기 때문에 바이러스 안전성 보증을 위한 바이러스 검출시험이 필수적이다. Reovirus (Reo), bovine viral diarrhea virus (BVDV), bovine parainfluenza virus (BPIV)는 동물 세포주와 동물 세포 배양 공정에 오염되는 대표적인 RNA 바이러스이다. 세포배양 유래 생물의약품의 안전성을 확보하기 위해, 세포주, 원료물질, 제조공정, 완제품에서 Reo, BVDV, BPIV를 동시에 검출할 수 있는 Multiplex Reverse Transcription (RT)-PCR 시험법을 확립하였다. Reo, BVDV, BPIV에 특이적인 primer를 선별하였으며, multiplex RT-PCR 시험법을 최적화하였다. Reo, BVDV, BPIV를 동시에 검출할 수 있는 multiplex RT-PCR 시험법의 민감도는 각각 $7.76{\times}10^2\;TCID_{50}/ml$, $7.44{\times}10^1\;TCID_{50}/ml$, $6.75{\times}10^1\;TCID_{50}/ml$이었다. 확립된 multiplex RT-PCR을 생물의약품 제조공정 검증에 적용할 수 있는지 확인하기 위하여 인위적으로 각 바이러스를 오염시킨 CHO 세포에서 검출 시험을 실시한 결과 각 바이러스를 감염시킨 CHO 세포와 세포배양 상청액에서 각 바이러스를 검출할 수 있었다. 위와 같은 결과에서 확립된 multiplex RT-PCR시험법은 세포주, 원료물질, 제조공정, 완제품에서 Reo, BVDV, BPIV를 동시에 검출할 수 있는 특이성과 민감성이 우수한 시험법임을 확인하였다.

Delayed Clinical Signs Associated with Cerebellar Lesion in Holstein Calf Infected with Bovine Viral Diarrhea Virus

  • A 5-month-old Holstein downer calf was presented, and bovine viral diarrhea virus was detected in the feces and the cerebrospinal fluid. Combined treatment of recombinant bovine somatotropin (rBST) administration and standing rehabilitation had been performed for two months, and the calf could maintain the standing position for a while. However, the symptoms were not improved, and the calf died due to ruminal tympany and aspiration pneumonia. Megaesophagus, abomasal ulcer, and a normal-sized but softened cerebellum were observed in clinical necropsy. Improvement of symptoms was confirmed by rBST, but was not reached the complete recovery of gait and standing position.

Alteration of the gut microbiota in post-weaned calves following recovery from bovine coronavirus-mediated diarrhea

  • Bovine coronavirus (BCoV) is associated with severe diarrhea, dehydration, and depression, which result in significant economic damages in the dairy and beef cattle industries worldwide. However, differences in the gut microbiota structure and their correlations with differing physiological parameters between BCoV-infected calves with diarrhea and recovered calves are not well understood. In this study, fecal specimens were collected from 10 post-weaned calves, before and after 2 months of fluid therapy, and the samples were used for microbiota analysis. Following recovery, the alpha-diversity profiles (observed operational taxonomic units [OTUs], and Chao1, Shannon, and Simpson indices) changed significantly when compared with those of calves with diarrhea. Beta-diversity analysis exhibited significant differences in gut microbiota compositions between calves with diarrhea and those in the recovered state. The abundances of eight phyla and thirteen genera in feces changed markedly after restoration of BCoV diarrhea. In addition, our correlation study clearly revealed that increased abundances of the genera Caproiciproducens, Pseudoflavonifractor, and Oscillibacter negatively correlated with serum glucose, and phosphorus levels, but positively correlated with serum chloride in calves with diarrhea, whereas increased abundances of the genera Peptostreptococcaceae;Clostridium (Clostridium cluster XI), Intestinibacter, Cellulosilyticum, Ruminococcus, Romboutsia, Paeniclostridium, Clostridiaceae;Clostridium and Turicibacter in recovered calves showed the opposite pattern. These results suggest that structural changes of the gut microbiota after recovery from BCoV infection correlate with changes in physiological parameters. In conclusion, our data provide evidence of gut microbiota-composition changes and their correlations with the physical profiles of post-weaned calves, before and after fluid therapy for BCoV-related diarrhea.

An Immunohistochemical Study of Viral Antigen in Aborted Fetuses Naturally Infected by Bovine Viral Diarrhea Virus

  • The tissue distribution and cellular localization of viral antigen in the brain of aborted fetus with bovine viral diarrhea virus(BVDV) infection was studied; BVDV antigens was detected in spleen, kidney, lung, eyelid as well as brain. In the brain, the virus was recognized in neurons and non-neuronal cells in the cerebellum and cerebrum. Many cells in the superficial layer and occasional Purkinje cells had BVDV antigens. As well, BVDV was also found in the perivascular cells, vascular endothelial cells and smooth muscle cells in the vessels and neuroglial cells in the white matter. This finding suggests that BVD virus favors infect progenitor cells in the brain, notably in the superficial layer of cerebellum, and damage normal development of cerebellum, which leads to cerebellar hypoplasia.

Klebsiella pneumoniae infection secondary to bovine viral diarrhea in two prematurely born calves

  • This paper describes the development of neurological signs of two prematurely born calves four days after birth. The pathological examination results indicated fibrinopurulent polyserositis, including meningoencephalitis with suppurative bronchopneumonia. Bovine viral diarrhea virus subtype 2a was detected in most of the internal organs, and the bacterial colonies cultured from the samples were identified as Klebsiella (K.) pneumoniae. Molecular analysis via multilocus sequence typing identified a different K. pneumoniae isolate in each calf-type 14 in calf A and type 65 in calf B. This is the first report identifying K. pneumoniae sequence types 14 and 65 in cattle.
