Efficient In Vitro Labeling Rabbit Bone Marrow-Derived Mesenchymal Stem Cells with SPIO and Differentiating into Neural-Like Cells

  • Mesenchymal stem cells (MSCs) can differentiate into neural cells to treat nervous system diseases. Magnetic resonance is an ideal means for cell tracking through labeling cells with superparamagnetic iron oxide (SPIO). However, no studies have described the neural differentiation ability of SPIO-labeled MSCs, which is the foundation for cell therapy and cell tracking in vivo. Our results showed that bone marrow-derived mesenchymal stem cells (BM-MSCs) labeled in vitro with SPIO can be induced into neural-like cells without affecting the viability and labeling efficiency. The cellular uptake of SPIO was maintained after labeled BM-MSCs differentiated into neural-like cells, which were the basis for transplanted cells that can be dynamically and non-invasively tracked in vivo by MRI. Moreover, the SPIO-labeled induced neural-like cells showed neural cell morphology and expressed related markers such as NSE, MAP-2. Furthermore, whole-cell patch clamp recording demonstrated that these neural-like cells exhibited electrophysiological properties of neurons. More importantly, there was no significant difference in the cellular viability and $[Ca^{2+}]_i$ between the induced labeled and unlabeled neural-like cells. In this study, we show for the first time that SPIO-labeled MSCs retained their differentiation capacity and could differentiate into neural-like cells with high cell viability and a good cellular state in vitro.

Is the panoramic mandibular index useful for bone quality evaluation?

  • Purpose: The aim of this study was to determine whether the panoramic mandibular index (PMI) is useful for assessing bone mineral density. We also analyzed the potential correlations between PMI parameters and patient age. Materials and Methods: Four observers measured the PMI of both sides of the mental foramen using a picture archiving and communication system and images in the Digital Imaging and Communications in Medicine format. They studied 300 panoramic radiographic images of patients belonging to the following age groups: 40-49 years, 50-59 years, 60-69 years, 70-79 years, and 80-89 years. The observers were allowed to zoom in or out and to adjust the contrast of the images. Further, they were instructed to record the reasons for any measurements that could not be made. Then, we conducted a reliability analysis of the measured PMI and assessed the correlations between different patient age groups and the 3 parameters used for determining the PMI from the available data. Results: Among the 600 data items collected, 23 items were considered unmeasurable by at least 1 observer for the following 4 reasons: postoperative state, lesion, unidentified mental foramen, and alveolar bone loss. The intraobserver reproducibility of the measurable data was 0.611-0.752. The mandibular cortical width (MCW) decreased significantly as patient age increased. Conclusion: PMI had limited usability when the margin of the mental foramen was not clear. In contrast, MCW, a parameter used for determining the PMI, had fewer drawbacks than the PMI with respect to bone mineral density measurements and exhibited a significant correlation with patient age.

CLINICAL STUDY ON THE IMMEDIATE IMPLANTATION WITH GTR THERAPY, INCLUDING BONE HEALING OF EXTRACTION SOCKETS (유도조직재생술에 의한 발치창의 골치유 및 즉시 임프란트 매식에 대한 임상적 연구)

  • Early implantation before sufficient ossification has taken place usually results in osseointegartion failure due to reduced bone-fixture interface area. However, various studies have shown successful osseointegration results following immediate implants concurrently with GTR. The clinical trends have been to shorten the patients' edentulous state by immediate implantation, and reduce the alveolar bone resorption. However, it may be difficult to attain the complete soft tissue coverage of the sites, increasing the chance of infection. Furthermore, there may be more studies needed on the clinical behaviors of e-PTFE membranes, various modofications in the membrane materials and bone graft materials. Various animal and clinical studies have been reported on the successful osseointagration following immediate implantation, but the long-term follow-up studies are limited. The present study investigated 16 immediately-implanted implants with GTR therapy with or without calcium carbonate grafting on 11 patients 3 years after installation and 24-30 months after functional loading. Based on the clinical, radiographic and histologic findings, the following results have been attained. 1. Clinically, stability has been shown on all 16 implants throughout the investigated periods. 2. Radiologically, the alveolar bone loss has progressed up to the polished neck portion but not beyond it, suggesting the progressive osseointegration from the GTR therapy. 3. The GTR method used in the present study is easy to use clinically, and may be appied in the regeneration of ossoeous defects around implants and in the immediate implantation. 4. The difficulty in complete tissue coverage may be avoided by delaying the installation for 2 to 3 weeks after the extraction allowing certain degree of soft tissue healing.

  • According to the bone-standard measuring, the length between the elbow and the wrist has the span of 1.25 cheoks whereas it has the span of 1.2 cheoks or 1.2 cheoks today, the breadth of the chest has the span of 9.5 chons whereas it has the span of 8 chons today, and the length from celestial pivot to transverse bone has the span of 6.5 chons whereas it has the span of 5 chons, and the length from free rib region to thigh pivot has the span of 6 chons whereas it has the span of 9 chons today. It is said that all of the acupuncture points are correctly prescribed by the bone-standard measuring irrespective of men and women of all ages, fat and lean, and whether large or small in height. This lies at the root of the selecting acupuncture points. The bone standard in Spiritual Pivot and that in common use at present have a little difference and the bone standard of today is based upon A-B Classic. Spiritual Pivot named as Acupuncture Classic was in good preservation until the mid-eleventh century, but the book lost a lot in contents was the incomplete edition. In the 8th year of the king Cheoljong's reign of the North Song Dynasty in 1093, though the nine-volume Spiritual Pivot drafted from the Goryo Dynasty was published, the book was wanting in consistency. While on the other, I think that A-B Classic which has been in a state of perfection had a great influence on the healers of the day.

Midfacial degloving approach for management of the maxillary fibrous dysplasia: a case report

  • Background: Fibrous dysplasia (FD) is a benign bone lesion characterized by the progressive replacement of normal bone with fibro-osseous connective tissue. The maxilla is the most commonly affected area of facial bone, resulting in facial asymmetry and functional disorders. Surgery is an effective management option and involves removing the diseased bone via an intraoral approach: conservative bone shaving or radical excision and reconstruction. Case presentation: This case report describes a monostotic fibrous dysplasia in which the patient's right midface had a prominent appearance. The asymmetric maxillary area was surgically recontoured via the midfacial degloving approach under general anesthesia. Follow-up photography and radiographic imaging after surgery showed the structures were in a stable state without recurrence of the FD lesion. Furthermore, there were no visible scars or functional disability, and the patient reported no postoperative discomfort. Conclusions: In conclusion, the midfacial degloving approach for treatment of maxillary fibrous dysplasia is a reliable and successful treatment option. Without visible scars and virtually free of postoperative functional disability, this approach offers good exposure of the middle third of the face for treatment of maxillary fibrous dysplasia with excellent cosmetic outcomes.

Health and Safety at Work: Analysis from the Brazilian Documentary Film Flesh and Bone

  • Background: The objective of this article is to make some analysis on the process of work and accidents occurring in slaughterhouses, evidenced in the Brazilian documentary film called Flesh and Bone. As such, it was necessary to discuss an alternative theoretical concept in relation to theories about health and safety at work. This alternative discussion focuses on the concepts of biopower and biopolitics. Methods: The use of audiovisual elements in research is not new, and there is already a branch of studies with methodological and epistemological variations. The Brazilian documentary Flesh and Bone was the basis for the research. The analysis of this documentary will be carried out from two complementary perspectives: "textual analysis" and "discourse analysis." Results: Flesh and Bone presents problems related to health and safety at work in slaughterhouses because of the constant exposure of workers to knives, saws, and other sharp instruments in the workplace. The results show that in favor of higher production levels, increased overseas market sales, and stricter quality controls, some manufacturers resort to various practices that often result in serious injuries, disposal, and health damages to workers. Conclusion: Flesh and Bone, by itself, makes this explicit in the form of denunciation based on the situation of these workers. What it does not make clear is that, in the context of biopolitics, the actions aimed at solving these problems or even reducing the negative impacts for this group of workers, are not efficient enough to change such practices.

Miniscrew insertion sites of infrazygomatic crest and mandibular buccal shelf in different vertical craniofacial patterns: A cone-beam computed tomography study

  • Objective: To identify optimal areas for the insertion of extra-alveolar miniscrews into the infrazygomatic crest (IZC) and mandibular buccal shelf (MBS), using cone beam computed tomography (CBCT) imaging in patients with different craniofacial patterns. Methods: CBCT reconstructions of untreated individuals were used to evaluate the IZC and MBS areas. The participants were divided into three groups, based on the craniofacial pattern, namely, brachyfacial (n = 15; mean age, 23.3 years), mesofacial (n = 15; mean age, 19.24 years), and dolichofacial (n = 15; mean age, 17.79 years). In the IZC, the evaluated areas were at 11, 13, and 15 mm above the buccal cusp tips of the right and left first molars. In the MBS, the evaluated areas were at the projections of the first molars' distal roots and second molars' mesial and distal roots, at a 4- and 8-mm distance from the cementoenamel junction. Intergroup comparisons were performed with analysis of variance and the Tukey test. Results: There was no statistically significant difference in the IZC bone thickness among the groups. For MBS bone availability, some comparisons revealed no difference; meanwhile, other comparisons revealed increased MBS bone thickness in the brachyfacial (first molars distal roots) and dolichofacial (second molars mesial and distal roots) patterns. Conclusions: There was no significant difference in the IZC bone thickness among the groups. The facial skeletal pattern may affect the availability of ideal bone thickness for the insertion of extra-alveolar miniscrews in the MBS region; however, this variability is unlikely to be clinically meaningful.

Limb Salvage with Tumor prosthesis for Osteosarcoma of Distal Tibia and Fibula (경골 및 비골 원위부 골육종의 종양삽입물을 이용한 사지구제술)

  • Recently limb-salvage operation is widely used for the treatment of malignant bone tumor. But distal leg is out of range of limb-salvage operation due to its technical problem. We report satisfactory limb-salvage operation with tumor prosthesis in 3 cases of osteosarcoma of the distal tibia and fibula. Two cases involved in the tibia and 1 case in the fibula. Average age at operation was 23 years. Neoadjuvant and adjuvant chemotherapy were performed in all 3 cases. We used custom made tumor prosthesis which is designed by Seoul National University Orthopaedic Department. Overall Functional Evaluations by Enneking rating system were good in all 3 cases. All cases are CDF(continuosly disease free since the surgical procedure) state at mean follow-up 2 year and 9 months. In conclusion limb-salvage with tumor prosthesis is useful treatment modality for malignant bone tumor of distal tibia and fibula. Good functional results and few complications suggests limb salvage of distal lower leg may be replaceable with B-K amputation.

The Difference of Predictor of Bone Mineral Density in Pre and Postmenopausal Women (폐경에 따른 골밀도 예측인자의 차이)

  • We studied the relationship between prediction parameters and bone mineral density for pre-and-post menopausal women. We measured BMI%Fat by BIA, blood pressure and lipid profiles for 483 adult women who are in NPO state. SBP, TC, TG, LDL have significant statistical value in the postmenopause women group and postmenopause woman. The value of postmenopause women of these parameters are lower than premenopause woman. BMD has the most strongest relationship with LBM. The BMD and LDL level of postmenopause women have statistically negative relationship. The results show that for the premenopause cases, weight, BF, and HDL level were the major factors which affect the BMD. For postmenopause cases, however, weight, age, and LDL level turned out to be the most significant factors.

Analysis of Correlations among Bone Mineral Density, Serum Lipid Levels, and Cognitive Function in the Elderly with Dementia (치매노인에서 골밀도 및 혈중 지질농도와 인지기능과의 상관관계 분석)

  • Purpose : The purpose of this study was to analyze the correlations among bone mineral density(BMD), serum lipid levels, and cognitive function in the elderly with dementia. Methods : We recruited seventy elderly with dementia(men=35, women=35) to participate in the Korean mini mental state examination(K-MMSE). Their T-scores and serum lipid levels were analyzed for correlation analysis. Results : The results of this study showed that there are significant correlations between cognitive function and three factors BMD, low-density lipoprotein cholesterol(LDL-C) level, and total cholesterol(TC) level. The cognitive function scores increased proportionally with BMD but were inversely proportional to LDL-C and TC levels. There were no significant relations among cognitive function, high-density lipoprotein cholesterol(HDL-C) level, and triglyceride(TG) level. Conclusion : These results indicate that there is a direct proportionality between cognitive function and BMD and inverse proportionalities between cognitive function and LDL-C level and between cognitive function and TC level. Therefore, these levels can be indices for preventing and predicting dementia.
