  • Bone maintains its homeostasis through balance between bone resorbing osteoclasts and bone forming osteoblasts. Thus, unusual balance between osteoclasts and osteoblasts leads to pathological bone diseases, such as osteoporosis, rheumatoid arthritis, autoimmune arthritis, periodontitis. Schisandra chinensis well known traditional herbal has been used for treatment of diseases in China, Korea, Japan, andothers. Recently, research studies have demonstrated that the lignans found in Schisandra chinensis stimulate osteoblasts and suggest that it may be helpful against osteoporosis. However, the inhibitory effect of water extract of Schisandra chinensis on osteoclast differentiation remains largely unknown. In this study, Water extract of Schisandra chinensis markedly suppressed RANKL-induced osteoclast differentiation in cultures of BMMs without cytotoxicity. The mRNA expression of c-Fos, NFATc1, and TRAP induced by RANKL was inhibited by water extract of Schisandra chinensis. It also suppressed c-Fos and NFATc1 protein expression. Taken together, these results suggest that water extract of Schisandra chinensis has the potential to serve as a treatment of bone disease such as osteoporosis.

Proliferative and Differentiative Effects of Trachelogenin Isolated from Germinated Safflower (Carthamus tinctorius) Seeds on Calvarial Bone Cells

  • Germination is well-known to enhance the digestibility, functionality, and palatability of plant seeds. To examine the functionality of germinated-safflower seed (GSS), proliferative and differentiative effects of GSS extract on the mouse calvarial bone cells were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolinbromide (MTT) assay and alkaline phosphatase activity, respectively. Water extract of GSS increased dose-dependently proliferative and differentiative effects on calvarial bone cell, and its effects were stronger than those of ungerminated-safflower seeds (UGSS) extract. One major component was isolated from GSS extract by a series of purification procedure of solvent fractionation, Diaion HP-20, and Sephadex LH-20 column chromatographies. Its chemical structure was identified as trachelogenin (TC) by nuclear magnetic resonance (NMR) and mass spectrometry (MS) spectral analysis. Trachelogenin showed significant proliferative (125.7%) and differentiative (132.1%) effects on calvarial bone cells at $10^{-8}M$, and its effects were significantly higher than those of $17{\beta}-estradiol\;(E_2)$. TC was found to be a major active compound responsible for high proliferative and differentative effects of the water extract of GSS. Therefore, these results suggest that TC in GSS may be useful as potential therapeutic agent for the prevention and treatment of bone loss.

Effects of gangliosides from deer bone extract on the gene expressions of matrix metalloproteinases and collagen type II in interleukin-1β-induced osteoarthritic chondrocytes

  • BACKGROUND/OBJECTIVES: We investigated the anti-osteoarthritic effects of deer bone extract on the gene expressions of matrix metalloproteinases (MMPs) and collagen type II (COL2) in interleukin-$1{\beta}$-induced osteoarthritis (OA) chondrocytes. MATERIALS/METHODS: Primary rabbit chondrocytes were treated as follows: CON (PBS treatment), NC (IL-$1{\beta}$ treatment), PC (IL-$1{\beta}+100{\mu}g/mL$ glucosamine sulphate/chondroitin sulphate mixture), and DB (IL-$1{\beta}+100{\mu}g/mL$ deer bone extract). RESULTS: The results of the cell viability assay indicated that deer bone extract at doses ranging from 100 to $500{\mu}g/mL$ inhibits cell death in chondrocytes induced by IL-$1{\beta}$. Deer bone extract was able to significantly recover the mRNA expression of COL2 that was down-regulated by IL-$1{\beta}$ (NC: 0.79 vs. DB: 0.87, P < 0.05) and significantly decrease the mRNA expression of MMP-3 (NC: 2.24 vs. DB: 1.75) and -13 (NC: 1.28 vs. DB: 0.89) in OA chondrocytes (P < 0.05).CONCLUSIONS: We concluded that deer bone extract induces accumulation of COL2 through the down-regulation of MMPs in IL-$1{\beta}$-induced OA chondrocytes. Our results suggest that deer bone extract, which contains various components related to OA, including chondroitin sulphate, may possess anti-osteoarthritic properties and be of value in inhibiting the pathogenesis of OA.

  • The aim of this study was to find out the effectiveness on germinated seed of Rhynchosia Volubilis for Female Bone Morphogenesis. For this purpose, We compared two methods. water extract and alcohol extract using germinated Rhynchosia Volubilis(GRV) according to germinating days were conducted to measure the polyphosphate contents and investigate HOS-TE85 propagation rate. Both water and alcohol extract two methods were not toxicant. And if not excessively treated, alcohol extract rate were more about $5{\sim}15$ times than water extract rate. So usually water extract were better than alcohol extract. but in case of osteoporosis, alcohol extract were effective.

상기생이 ethanol을 장기 투여한 흰쥐의 골 대사에 미치는 영향 (Effects of Taxilli Ramulus Extract on Bone Metabolism of Ethanol Treated Rats)

  • Objectives : To investigate the effect of Taxilli Ramulus (TR) extract on bone metabolism of ethanol-treated animal model. Methods : The changes of serum calcium, calcitonin, estrogen level, a1ka1ine phosphatase activity, osteocalcin, parathyroid hormone content and urine calcium level were observed with ethanol treatment for 60 days. The results were compared with an ethanol- TR extract double treatment group. Results : We observed increment of serum osteocalcin, parathyroid hormone content, alkaline phosphatase activity and urine calcium level by chronic ethanol feed and they were recovered to near normal level with Taxilli Ramulus extract treatment. Weight gain, serum calcium level, calcitonin and estrogen content were remarkably reduced with ethanol treatment and their levels were normalized by Taxilli Ramulus extract. Conclusions : These results showed that Taxilli Ramulus extract have the ability to recover to normal in the body an abnormal calcium metabolism process due to external factors. These results suggested that Taxilli Ramulus extract have preventive effects on calcium concentration loss and osteoporosis.

천마가 RANKL에 의해 유도된 파골세포의 분화에 미치는 효과 (Effects of Gastrodia elata Blune Water Extract on RANKL-induced Osteoclast Differentiation)

  • Impairment of balance between bone-resorbing osteoclasts and bone-forming osteoblasts result in bone disease. Especially, increased osteoclast formation and activity are responsible for bone diseases such as osteoporosis, rheumatoid arthritis, periodontal disease. Natural metabolites of plants have recently received much attention as an alternative tools for the development of novel therapeutic strategy. The aim of this study was to search the natural products to inhibit osteoclast differentiation and was to evaluate of its mechanism. Water extract of Gastrodia elata Blune significantly inhibited receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation in bone marrow macrophages (BMMs) in a dose dependent manner. However, water extract of Gastrodia elata Blune did not affect cytotoxicity when compared with control. The mRNA expression of c-Fos, NFATc1, and TRAP induced by RANKL was inhibited by water extract of Gastrodia elata Blune treatment. Also, water extract of Gastrodia elata Blune inhibited the protein expression of c-Fos and NFATc1 expression in BMMs treated with RANKL. Water extract of Gastrodia elata Blune suppressed the phosphorylation of p38 induced by RANKL. In general, RANKL considerably inhibited the expression level of Id2 and MafB known as negative regulators of osteoclastogenesis, but RANKL did not inhibit Id2 and MafB expression in BMMs when it was co-treated with Gastrodia elata Blune. Taken together, these results suggest that Gastrodia elata Blune may be a useful drug in the treatment of bone-related disease.

흑염소 육골액의 적정 추출시간 및 성분 분석에 관한 연구 (A Study on the Extraction Time and Component Analysis of Goat Meat with Bone Extract)

  • An attempt was made in this study to investigate the optimum extracting time from meat with bone of goat and the nutritional component of its extract. for the trials, the mixtures of meat with bone and water were adjusted to the ratios of five to four by weight and extracted for 6, 9 and 12 hours at 120$\^{C}$ under autoclave. Judging from the content of mineral and amino acid, nonenzymatic browning and yield, the optimum extracting time was 9 hours. The major components of mineral were composed of 47.7mg% potassium, 12.7mg% calcium, 150.0mg% sodium, 105.3mg% phosphorus and 0.5mg% iron, and of amino acids composed of 1,308.0mg% glutamic acid, 1,464.2mg% glycine, 750.2mg% alanine and 828.lmg% proline in extract. The yield of extract was 32.1 percentage by dry basis.

Effect of Ethanol Extract of Safflower Seed on Bone Loss in Ovariectomized Rat

  • We investigated whether ethanol extracts of the safflower seeds containing phenolic compounds were responsible for the bone-protecting effects. Crude ethanol extract (CEE) of the safflower seeds was fed for 4 weeks at the level of 1% in diet to female Sprague-Dawley rats that had been subjected to bilateral ovariectomy (OVX). The CEE effects (OVX+CEE) were evaluated by comparing results obtained from OVX, Sham, and OVX injected with $17{\beta}$-estradiol ($OVX+E_2$) groups. OVX resulted in a dramatic reduction in the trabecular bone mass of the proximal tibia (approximately 40% of the Sham group) and an increase in fat deposition in bone marrow. In $OVX+E_2$ group, the bone loss was completely prevented as well as marrow adiposity. In OVX+CEE group, approximately 80% of the bone mass was maintained compared with Sham group and fat deposition in the bone marrow was prevented. Meanwhile, the partially purified ethanol extract containing the phenolic compounds stimulated proliferation of the ROS 17/2.8 osteoblast-like cells in a dose-dependent manner, as potently as positive controls of $E_2$ and genistein. The present data demonstrate that the ethanol extracts of safflower seeds reduced bone loss caused by estrogen deficiency. The bone-protecting effect of safflower seeds seems to be mediated, at least partly, by the stimulating effect of the phenolic compounds on the growth of osteoblasts.

한약이 난소제거 흰쥐와 SAM P6 생쥐의 골소주에 미치는 영향 (Effects of Herbal Medicine on Tibia Bone Changes in Ovariectomized Rats and SAM P6 Mice)

  • The effects of herbal medicine on travecular bone area were studied using ovariectomized rat as an animal model of Type I osteoporosis and SAM P6 as that of Type II. We counted red blood cells(RBC), hemoglobin(Hb), and hematocrit(Hct) using Couter`sR method. Each traditional boiling water extract of Achyrathis Radix, Psoraleae Fructus, Rehmanniae Radix Preparat, and Cornii Fructus and a systemic water extract of Astragali Ractiex was given 5g/kg/day, p.o., for 30 days in a group of 4-5 ovariectormized rats. One ml of blood was taken by tail vein at day 0, 7, 14, 21, and 30 days after administration of the extract. The traditional hot water extract of Cervi parvumn Corni (Cervi) was given the same dose as described above for 14 days in a group of 10 SAM P6 mice and systemic water extract of Astragali Radix was administered as the same dose as above for 30 days in 10 SAM P6 mice. Travecular bone area was measured 5 mcm decalcified and stained thin bone slice by image analysis using a digitalizer. In Type I, ovariectomized rats, administration of Astragali Radix, Rhemanniae Radix Preparat, and Corni Fructus decreased in RBC, Hb and Hct. In Type II, administration of Cervi increased in RBC and Hct and that of Astragali Radix was also elevated RBC. In Type I, any administration of herbal medicine used in this study did not elevate travecular bone area significantly except Corni Fructus showed a trend of increase in travecular bone area. However, Type II, Cervi and Astragali Radix increased in both mean and total travecular bone area. Thus, there are significant difference in response of herbal medicine in different types of osteoporosis.

녹용추출물이 성장기 흰쥐의 혈중 IGF-I 농도, 골격성장 및 비장세포 증식능에 미치는 영향 (Effects of Deer Antler Extract on Serum IGF-I, Bone Growth and Splenocyte Proliferation in Growing Rats)

  • Although it has traditionally known that deer antler and medicinal herbs extract contain some functional components for health promotion, the nutritional significance remains to be elucidated. This study examined the efficacy of deer antler extract (DA) , medicinal herbs extract (MH) and their mixture (DAMH) on serum IGF-I, bone growth with growing rats in vivo and splenocyte proliferation with spleen cells in vitro. Three week-old young female rats (Sprague-Dawley) were divided into 4 groups and then fed basal diet (AIN-93G) or experimental diets containing DA, MH, DAMH, respectively, for 7 weeks. We collected blood, liver, kidney, spleen, femur and tibia from rats. There was no significant difference in weight gain, but food intake increased in DA- and MH-fed groups. There were no signs of liver and kidney damage in the DA, MH and DAMH-fed groups compared to basal diet group. In femur and tibia, wet weights: breaking forces and bone minerals (Ca, Mg and Zn) were significantly higher in the DA-fed group than in the other groups. Serum alkaline phosphatase (ALP) , bone-specific alkaline phosphatase (BALP) activities were significantly lower in the DA, MH, DAMH-fed groups than in basal diet group. Also, serum insulin-like growth factor-I (IGF-I) concentrations were significantly increased in DA-fed group compared to the other groups. Therefore DA was shown to have an activity of bone growth promotion by increasing the IGF-I, a major bone growth factor. The deer antler extract showed an enhanced immune action on the primary cultured-cells from spleen of rats, representing that splenocytes were proliferated by lipopolysaccharide (LPS), but not by concanavalin A (Con A). These results indicate that deer antler extract has beneficial effects on bone growth via IGF-I and on splenocyte activation.
