Toxicity of Chloramine-T on the Flounder, Paralichthys olivaceus, and Bactertcidal Activity Against Fish Pathogenic Bacteria and Blue Green Algae, Tetraselmis suecica (Chloramine-T의 넙치에 대한 독성과 어병세균 및 Blue Green Algae에 대한 살균 효과)

  • CT(Sodium N-chloro-para toluenesulfonamide, Chloramine-T) known to be a strong oxidative agent was investigated to use as a candidate of disinfectant in the marine aquaculture industry by the analysis of the bactericidal activity against different aquatic microorganisms. One hundred percent mortality appeared at and above 16 ppm CT in sea water in flounder, the predominant species in the marine aquaculture of Korea, when exposed for 48hr. However, bactericidal activity was appeared to be very effective, and all different species of the fish pathogenic bacteria exposed to CT of less than 2 ppm in sea water were dead within 15 min. It allowed us to confirm that CT could be a very effective disinfectant to protect the spread of fish pathogenic bacteria derived from diseased fish or sea water in marine aquaculture. High concentration(10 ppm) with longer exposing time(24 hrs) was required for the algicidal activity of CT, at which concentration might induce the acute toxicity against fish, however, restricted the expanded use of CT for the elimination of zooplanktons at phytoplanktons in marine farms. Moreover the bactericidal activity of CT inhibited almost completely in the present of more than 10 ppm of organic materials pointed out that quality of the culturing sea water should be considered carefully for the application CT to the aquatic farms as a therapeutic agent.

Growth Characteristics of Blue-green Algae (Anabaena spiroides) Causing Tastes and Odors in the North-Han River, Korea (북한강 수계에서 이취미를 유발하는 남조류(Anabaena spiroides)의 증식 특성)

  • Blue-green algae blooms occurred during early winter in the North-Han River, Korea. Among blue-green algae, Anabaena spiroides were observed for approximately 33 consecutive days, between 28 November and 30 December, 2011. A. spiroides emerged from Lake Uiam to Lake Paldang, depending on the flow of the river has spread downstream. Changes of physical water environment like rising water temperature and increasing hydraulic retention time influenced the A. spiroides bloom. The A. spiroides bloom showed a very rapid increase in cell density, and a slow decrease: the cell density increased to a maximum of $11,325cells\;mL^{-1}$ in Lake Paldang (st. 5), and was completely disappeared after the water temperature dropped below $4^{\circ}C$. A decrease in water temperature was the most influential factor among all environmental parameters, for the reduction of A. spiroides cell density. The A. spiroides bloom was accompanied with the occurrence of very high concentrations of the odor metabolite geosmin. Geosmin reached the peak value of $1,640ng\;L^{-1}$ in Lake Paldang (st. 4). The geosmin concentration was very strongly correlated with cell numbers of A. spiroides.

Determination of Optimum Coagulant Dosage for Effective Water Treatement of Chyinyang Lake - The Effect of Coagulant Dosing on Removal of Algae- (진양호소수의 효과적인 정수처리를 위한 최적응집제 주입량 결정 -조류제거를 위한 응집제 주입효과-)

  • This study was performed to determine the optimum coagulant dosing for effective treatment of raw water in Chinyang lake. Removal rates of algae and characteristics of the water according to coagulants dosage were investigated by treatment with Microcystis aeruginosa, which is a kind of blue-green algae, to the raw water below 5NTU. The coagulants dosage for maximum removal rate of algae were 30 mg/$\ell$ of Alum, 30 mg/$\ell$ of PAC and 10 mg/$\ell$ of PACS, respectively. The removal rate of algae in 30 mg/$\ell$ of PAC was highest as 85% compared with the other treatments. At the point of maximum removal rate of algae, the removal rates of turbidity were 34%, 66% and 22% in Alum, PAC and PACS, respectively. Residual Al was decreased depend upon decreasing turtidity in water by treatment of Alum or PAC, but decreased depend upon increasing turbidity in water by treatment of PACS. The removal rate of ${Mn}_{2+}$ in water was high in the order of Alum, PAC and PACS treatment. And ${Fe}_{2+}$ in water was not changed by treatemnt of these coagulants. Particle numbers distributions according to the particle size of suspended solids that were not precipitated at 8 min. of settling time after treatment of coagulants dosage for the maximum removal rate of algae were investigated. Most of the particle sizes were below 30 $\mu$m and particle numbers distributions below 10 $\mu$m were 64%, 56% and 66% by treatment of Alum, PAC and PACS, respectively. Zeta potential was in the range of -6.1~-9.7 mV at optimum coagulants dosage for algae removal.

Electroencephalography (EEG) based Toxicity Test of Algae Organic Matter on Zebrafish (조류기인 유기물질의 제브라피쉬에 대한 뇌파측정기반 독성평가)

  • Harmful algae blooms have become a serious environmental problem in major river basins in Korea. They are known to produce various algal organic matters (AOMs) including intracellular organic matters (IOMs) and extracellular organic matters (EOMs). Generally AOMs cannot be easily removed by coagulation/flocculation process in conventional drinking water plants. AOMs produced by blue-green algae also include various toxins such as Microcystins, Anatoxin-a, and Saxitoxin known to have harmful effects on living organisms in aquatic environment. In this study, toxic effects of EOMs produced by three different algae species (Microcystis sp., Anabaena sp., and Oscillatoria sp.) on zebrafish were investigated using electroencephalography (EEG) recording method, a technology for recording brain activity. Electroencephalographic changes in zebrafish revealed that a low EOM had a negative effect on zebrafish compared to both Anabaena sp. and Oscillatoria sp. at 30 ppm EOM exposures. This result might be due to Microcystins present in EOMs produced by Microcystis sp. As a result of power spectrum density anallysis, exposure to EOMs produced by Microcystis sp. caused a state of vigilance in zebrafish. This EEG based toxicity test can be used to examine effects of harmful materials at low levels on living organisms in an aquatic system.

  • 자연계에 있어서 질소는 대기중의 분자상질소를 비롯하여 초산, 암모니아와 같은 무기태질소, 단백질, 핵산 등의 유기태질소 등 다양한 형태로 존재하며 생물권내에서 흡수, 고정, 대사, 분해되는 등 다양한 순환을 거듭하고 있다. 대기중의 분자상질소는 Rhizobium, Azotobacter, Klebsielle, Clostridium, Blue-green algae 및 광합성세균 등에 의해 고장되어 암모니아의 형태로 환원된다. 한편 대부분의 식물들은 초산이나 암모니아 형태의 질소를 흡수 동화하여 핵산, 단백질을 만들고 이들 구성물은 사후 암모니아로 재분해 된다. 또한 동식물의 유체내지는 배설물들도 각기 분해되어 암모니아의 형태로 변화되는데 이와같은 일련의 질소순환(nitrogen cycle)은 초화세균, 탈질세균 내지는 질소고정균등 대부분의 미생물에 의해 크게 지배를 받고 있다.

Water-Blooms (Green-Tide) Dynamics of Algae Alert System and Rainfall-Hydrological Effects in Daecheong Reservoir, Korea (대청호 조류경보제의 녹조현상 동태와 강우-수문학적 영향)

  • Daecheong Reservoir has suffered eutrophication and water-blooms by blue-green algae from initial impoundment, and algae alert system (AAS) was introduced in 1997. The purpose of this study was to investigate the effect of rainfall and hydrological factors in increase or decrease variability of green-tide and prolonged AAS, studied and analyzed the current situation of AAS has been operating for 19 years (1997~2015) in Daecheong Reservoir. The total issued number of AAS was 46 times, the most frequent period in August and September were 22 times (752 days) and 16 times (431 days), respectively, it accounted for 82.6%. Many number and frequency during this period were significantly associated with rainfall, various discharge and water level. Rainfall and hydrological events are associated with the rainy season of monsoon-Changma and the typhoon, it was concentrated in June~September, total rainfall in this period accounted for 69.9% of the annual rainfall. An increase in inflows was dependent on the intensity, frequency and the amount of rainfall. Accounted for 68.4% of the total annual inflow, it was a time when the most rapidly changing hydrological variability in the reservoir. The total outflow was closely related to rainfall, and compared the distinctive characteristics of hydropower generation and watergate-spillway discharge. In addition, the upreservoir zone of Daecheong Reservoir could be vulnerable to green-tide by regulating discharge of the upstream dam. The issue of AAS was strongly related to the with and without of watergate-spillway discharge. The watergate-spillway discharge had a total of 25 times, it was maximum 17 days from July to September in the year. And the opening times and each duration of the watergate were 1~4 times and the range of 3~37 days, respectively. When the watergate opened, the issue of AAS was maintained to 13 years and the movement of water bodies and green-tide was great about five times than that of non-open, had a profound effect on prolonged AAS within reservoir. In Daecheong Reservoir, Chusori (CHU) area of the So-ok Stream was still showing serious symptoms green-tide levels in the summer, but Janggye (JAN) waters of the main reservoir was pointed out that more important. AAS will be operated by an absolutely consider the rainfall and hydrological effects around the watergate-spillway discharge. The measures of green-tide will be included in the limnological studies more suited to the characteristics of the watershed and reservoir of the our country. Finally, from now on, we will prepare the systematic management and guidelines for vulnerable zone water-blooms that are the source within the reservoir before the monsoon rather than waiting for the arrival of green-tide on the operating stations of AAS.

Red and Blue Photons Can Enhance the Production of Astaxanthin from Haematococcus pluviatis

  • The unicellular green alga, Haematococcus pluvialis, accumulates the highest level of astaxanthin among knownastaxanthi.n-producing organisms. Light is the most important factor to induce astaxanthin by H. pluvialis. BIue andred LEDs, whose ${\lambda}_{max}$'s are 470 and 665 nm, respectively, were used for internally illuminated light sources.Fluorescent lamps were also used for both internal and external illumination sources. The astaxanthin levels in thesevarious lighting systems were analyzed and compared each other. The cultures under internally illuminated LEDsaccumulaled 20% more astaxanthin than those under fluorescent lamp. Furthermore, LEDs generated much lessheat than the fluorescent lamps, which gives one more reason for the LEDs being a suitable internal Light source forastaxanthin induction. The results reported here would lead novel designs of photobioreactors with improvementsof illumination methods for high level of astaxanthm production. The maximum astaxanthin concentrations as wellas the astaxanthin yield per supplied photon were increased by at least 20% when blue or red LEDs were supplied.

The Community of Cryophilic and Mountain Periphyton at High Altitude Streams in the Han-river System (한강수계 상위고도 하천의 저온성 및 산악성 부착조류군집)

  • The community of periphyton was studied at high altitude streams of the Han-river System, including Buk-stream and Hangae-stream from October to November 2010. The study had special emphasis on the distribution of the cryophilic and mountain periphyton. In this research, 117 taxa of periphyton appeared and 80 taxa of diatom were included within these periphytons. Saproxenous taxa, such as Achnanthes convergens and Hannaea arcus var. subarcus were appeared in most sites, 7 sites were dominant by filamentous taxa in green and blue green algae. The DAIpo value was from 48.8 to 92.5 for the water system of this research. Most sites showed a good water quality, receiving a B class or above except the Woljeong-stream and the Daeki-stream. Cryophilic and mountain periphyton appeared 9 taxa which composed by diatoms Cymbella affinis, Cymbella tumida, Diatoma mesodon, Diatoma vulgare, Eunotia pectinalis, Hannaea arcus, Hannaea arcus var. subarcus and Synedra inaequalis and green algae Ulothrix zonata. Among these taxa, Hannaea arcus var. subarcus, found in 29 out of 33 sites, had the highest frequency of apperance. The relative frequency of Hannaea arcus var. subarcus was positively correlated with altitude($r^2$=0.539) and negatively correlated with water temperature($r^2$=0.338). As a result, Hannaea arcus var. subarcus showed the characteristics of cryophilic and mountain periphyton in the research site.

Prediction of cyanobacteria harmful algal blooms in reservoir using machine learning and deep learning (머신러닝과 딥러닝을 이용한 저수지 유해 남조류 발생 예측)

  • In relation to the algae bloom, four types of blue-green algae that emit toxic substances are designated and managed as harmful Cyanobacteria, and prediction information using a physical model is being also published. However, as algae are living organisms, it is difficult to predict according to physical dynamics, and not easy to consider the effects of numerous factors such as weather, hydraulic, hydrology, and water quality. Therefore, a lot of researches on algal bloom prediction using machine learning have been recently conducted. In this study, the characteristic importance of water quality factors affecting the occurrence of Cyanobacteria harmful algal blooms (CyanoHABs) were analyzed using the random forest (RF) model for Bohyeonsan Dam and Yeongcheon Dam located in Yeongcheon-si, Gyeongsangbuk-do and also predicted the occurrence of harmful blue-green algae using the machine learning and deep learning models and evaluated their accuracy. The water temperature and total nitrogen (T-N) were found to be high in common, and the occurrence prediction of CyanoHABs using artificial neural network (ANN) also predicted the actual values closely, confirming that it can be used for the reservoirs that require the prediction of harmful cyanobacteria for algal management in the future.

The Water Quality and the Phytoplankton Communities in the Dong River of Kangwon Province, Korea (강원도 동강의 수질과 식물플랑크톤군집)

  • The present study is to examine the water quality and to analyze the plank tonic and benthic phytoplankton communities at 6 stations in the Dong River and its tributary streams of Kangwon Province from November, 2001 to March, 2002. During the studies, water temperature ranged from 3.5 to 12.8℃; pH, 6.5-7.9; DO, 9.29-11.36 mg·l^(-1); BOD_5, 0.20-2.38 mg·l^(-1); TN, 1.2842-3.1871 mg·l^(-1); TP, 0.0052-0.0576 mg·l^(-1); and SS, 0.85-9.62 mg·l^(-1). The standard of water quality according to the Korean Environmental Preservation Law showed the first class except St. 6 through the survey. Six taxa of plank tonic phytoplankton identified were poor flora in November, 2001. The representative species frequently observed were the diatoms Achnanthes lanceolata, Achnanthes minutissima, Cymbella minuta, Cymbella parva, Diatoma vulgare, Fragilaria construens, Gomphonema pervulum, Navicula cryptocephala, Nitzschia palea., Scenedesmus sp. of green algae and Stephanodiscus hantzschii. Monthly dominant species of phytoplankton were Achnanthes lanceolata and A. minutissima in both February and March, 2002, but the blue-green algae, Oscillatoria sp. and the diatom, Stephan discus hantzschii were predominant at some stations in March. Phytoplankton standing crops ranged from 9.84 ${\times}$ 10$^3$ to 3.56 ${\times}$ 10$^4$ cell·l$^{-1}$ in November and 1.68-2.99 ${\times}$ 10$^5$ cell·l$^{-1}$ in February, while it changed 4.52-8.01 ${\times}$ 10$^5$ cell·l$^{-1}$ at St. 1, 2, 3 and 1.03-1.71 ${\times}$ 10$^6$ cell·l$^{-1}$ at St. 4, 5, 6 in March. Benthic phytoplankton communities was composed of 38 taxa in November, 31 taxa in February and 23 taxa in March. It showed a contrary tendency to planktonic phytoplankton diversity. Benthic diatoms which were more than 25% of the total populations were Cymbella turgida, Diatoma vulgare, Cocconeis placentula, Navicula cryptocephala var. intermedia in November; Achnanthe lanceolata, Cocconeis placentula, Achnanthes minutissima in February; and Achnanthes lanceolata and Cyclotella meneghiniana in March.
