Laboratory maintenance of field-collected Lymnaea viridis for use as an intermediate host for Fasciola hepatica (야외에서 채집한 간질의 중간숙주인 애기물달팽이의 실험실 사육)

  • 전남대학교 수의과대학에서는 조류 blue-green algae를 먹이로 하여 간질의 중간숙주인 애기물달팽이를 실험실에서 유지시켜 왔으나 조류의 배양에 어려움이 많아서 보다 간편하면서 달팽이의 번식에도 좋은 방법을 고안하고자 노력하였다. $28.5{\times}17{\times}18cm$ 크기의 유리수조에 모래를 1.5cm 높이로 깔고 10개의 꼬막껍질을 그 위에 올려놓은 다음 4리터의 증류수를 부어넣었다. 야외에서 채집한 달팽이의 난괴를 부화시켜 얻은 0.63cm 크기의 어린 달팽이 45마리를 세 그룹으로 나누어 세 개의 수조에서 $20^{\circ}C$의 온도를 유지하면서 사육하였고 먹이로 상추잎을 넣어주었다. 매주 수조바닥의 물 $2{\ell}$를 흡인하여 갈아주면서 상추잎을 바꿔넣었고, 계속해서 공기를 통과시켜 산소를 공급하였다. 이와 동일한 방법으로 실험실에서 조류로 사육하던 17대의 달팽이를 사육하여 그 성적을 비교하였던 바 야외에서 채집한 달팽이의 성장률이 실험실에 적응된 것들에 비하여 약간 떨어졌으나 생존율이나 산란율에서는 큰 차이가 인정되지 않았다. 그래서 1년이 넘도록 이 방법으로 달팽이를 사육하면서 간질의 피낭유충을 생산하고 있다. 꼬막껍질은 서서히 분해하면서 달팽이의 각을 형성하는 물질을 제공하여 생존율을 높게 한 것으로 생각된다. 그리고 수조를 햇볕이 드는 창문 가까이에 놓아두었기 때문에 조류가 자생하여 적은 양이나마 달팽이의 먹이가 되었을 것으로 보인다.

Strain Identification and Comparative Analysis of Toxigenic Cyanobacteria Determined by PCR

  • Microcystis aeruginosa is common form of cyanobacteria (blue-green algae) capable of producing toxic heptapeptide (microcystin) that cause illness or death. The comparison of molecular genetic method with the morphological characteristics of cyanobacteria was conducted. We have designed PCR primers (JJM98F, JJM1141R) for cyanobacterial 16S rRNA and phycocyanin intergenic spacer (PC-IGS) gene domain. To confirm the production of microcystins, PCR primers for the N-methyltransferase (NMT) domain of microcystin synthetase gene mcyA were designed using 21 cyanobacteria strains Most of isolated strains from the Nakdong River was classified as Microcystis aeruginosa and the similarities were $99\%$ with M. aeruginosa AF 139292. $38.1\%$ of isolated strains contained microcystin synthesis gene. NMT (N-methyltransferase) were not detected in isolated strain in several strains, which means non-toxic. However, the NMTs of the strains were detected during the cultivation.

  • 국내 이상기후로 인해 여름철 하천과 호소에서 빈번하게 발생되는 조류의 과대성장이 매년 문제가 되고 있다. 하천 조류는 일차생산자로서 매우 중요한 역할을 하지만, 하천조류 중 유해남조류가 생장하면서 발생하는 악취 유발 물질과 독성물질의 배출로 문제를 야기하고 있다. 국내에서는 조류경보제와 수질예보제를 시행하여 국민의 안전을 도모하기 위해 최선을 다하고 있으며, 발령 기준은 유해남조류세포수에 따라 발령이 되기 때문에 유해남조류 측정은 매우 중요하다. 현재 조류의 분석방법은 현장에서 조류샘플을 채취하고 실험실에서 현미경을 통해 조류샘플을 검경하고 녹조류, 남조류, 규조류의 세포수 또는 우점종을 산정한다. 조류검경은 개인의 역량에 따라 오차가 생길수 있고 시간이 많이 소요된다. 최근 많은 연구자들이 이런 문제를 해결하기 위해 인공위성, 광학영상, 초분광영상 등을 통해 녹조류와 남조류 대체 인자인 Chlorophyll, Phycocyanin을 통해 조류농도를 추정하고 있으나, 조류세포수 분석에 대한 연구는 미비한 실정이다. 본 연구에서는 매년 조류 발생으로 문제가 되는 물금취수장 인근 하천에서 발생한 조류를 채취하고 조류검경을 통해 얻은 남조류세포수와, LISST-HAB를 통해 얻은 Phycocyanin농도, 초분광영상을 활용한 조류스펙트럼 데이터를 통해 남조류세포수 추정하고 남조류세포수와 비교분석을 진행하였다. 본 연구를 통해 조류측정 원격탐사 연구의 기초자료로 제공하고자 한다.

  • This study was made to investigate the effect of pesticides on microflora to nitrogen metabolism, nitrification and nitrogen fixing activity in the submerged soil. The results are summarized as follows; Pesticides treatment leaded to the inhibition of $NH_4{^-}$-oxdizers, $NO_3{^-}$-reducer, and denitrifying bacteria population. $NO_2{^-}$-oxdizers were inhibited by cabamate compounds, carbofuran and MIPC. Simetryne seemed to stimulate the denitrifying bacteria at 60 days after incubation. Generally, formation of $NO_2{^-}$ and $NO_3{^-}$ tended to decrease by pesticides application. Pesticides application stimulated Azotobacter and Clostridia populations, while simetryne inhibited Athiorhodaceae and Thiorhodaceae. However acephate seemed to be stimulatory to blue-geen algae. $C_2H_2$-reducing activity by acephate was clearly appeared. The change of $C_2H_2$-reducing activity did not seems to be affected by pesticides application.

The Dynamic of Phytoplankton Community in Unmun Dam (운문댐의 식물플랑크톤 군집 동태)

  • The seasonal changes in phytoplankton species composition, standing crops, dominant species, species diversity and physico-chemical characteristics in the Unmun dam were studied, from August, 2009 to April, 2010. The phytoplanktons of a total of 121 taxa were identified, the composition of phytoplankton community was characterized by a green algae and diatoms, and the quantity composition of standing crops was dominated by diatoms and dinophyceae. The diatoms Aulacoseria granulata and blue-green alga Anabaena planktonica in Summer, Peridinium voltzii in Autumn and Asterionella formosa in Winter to Spring were, especially, prominent. The seasonal changes of the biomass varied from 578 cells $mL^{-1}$ to 12,938 cells $mL^{-1}$, and the maximum algal density was observed in April, Asterionells formosa contributed to 88% of the total cell numbers. The species diversity and richness were highest during autumn, and dominance index was highest in the spring season.

Domestic Test Species for Aquatic Toxicity Assessment in Korea (수생태계 독성평가에 적용 가능한 국내 시험종 선정)

  • The use of aquatic species in ecotoxicity research is well established in developed countries. However, there are limitations of using the species that are not native to Korea, and the toxicity data produced by domestic test species are significantly needed to reflect the domestic situation. The purpose of this study was to investigate the domestic species that can be applicable for the aquatic toxicity assessment in Korea. Aquatic toxicity data were collected in the framework of the project 'Development of integrated methodology for evaluation of water environment' to obtain a range of test species used for aquatic toxicity assessment internationally. The test species collected were evaluated in terms of domestic distribution based on the reliable references and the advices of experts. We figured out the 71 test species native to Korea. They included 7 fish, 26 invertebrates (2 annelids, 2 bryozoa, 13 crustaceans, 3 insects, 4 mollusc, 1 platyhelminth, and 1 protozoan), 26 plants (9 diatoms, 14 green algae, 3 macrophytes), and 12 others (2 amphibians, 3 bacteria, 6 blue-green algae, and 1 fungus). The result of this study should be a very useful information for ecotoxicity assessment in aquatic ecosystem, especially in choosing the test species applicable for the ecotoxicity in Korea hereafter.

The Characteristics of Spatio-Temporal Distribution on Phytoplankton in the Nakdong River Estuary, during 2013-2015 (낙동강 하구역에서 2013-2015년 식물플랑크톤의 시·공간분포 특성)

  • To understand the characteristics of the spatio-temporal distribution of phytoplankton after barrage construction in the Nakdong River Estuary, this study investigated relevant environmental parameters and phytoplankton status based on bi-monthly samples collected from the Nakdong River Estuary itself from February 2013 to December 2015. Environmental parameters did not differ significantly across these years but did vary between zones and seasons. The results suggested that the upper zone was dominated by fresh-water diatoms, green algae, and blue-green algae, whereas the lower zone was mostly dominated by dinoflagellates. The presence of Stephanodiscus spp., Asterionellopsis formosa, and Microcystis spp. in the upper zone was related to the inflow of freshwater discharge by artificial control of dyke gates. The dominant phytoplankton species in this zone were dependent on temperature, wind speed, DIP, and DIN, while those in the lower zone were mostly dependent on nutrients and wind speed. In addition, at the lower zone, there were negative correlations between Prorocentrum donghaiense, DIN, and wind speed, with its abundance being higher during the summer than other seasons. Analysis of temporal variations did not indicate any significant differences in the upper zone but did reveal variations among seasons at the lower zone. Except in 2014, the lower zone could be divided into periods dominated by diatoms (October-April) and dinoflagellates (June-August). These results suggest that the characteristics of the phytoplankton community were influenced by changes in the inflow of freshwater species and nutrients given the difference in the range affected by freshwater discharge.

Benthic Marine Algal Flora and Community Structure of Yongho-dong Area in Pusan, Korea (부산 용호동 일대의 저서 해조상 및 군집구조)

  • Benthic marine algal flora and community structure of Yongho-dong area in Pusan, Korea were investigated seasonally in intertidal and subtidal zones by a quadrat method along 5 transect lines from July 1996 to April 1997. In this area, a total of 99 species including 3 blue-green, 13 green, 26 brown and 57 red algae was found. Among them, 33 taxa, together with dominant Ulva pertusa and Corallina pilulifera, occurred throughout the year. Enteromorpha spp. and Ulva spp. vertically distributed in upper and middle zone, while Simphyocladia latiuscula, Chondria crassicaulis, Corallina pilulifera and Sargassum sup. in lower zones. Biomass per unit area exhibited a wide range of variation, from 1,241 g/$m^2$(spring) to 1,648 g/$m^2$(summer) in fresh weight. Maximum and minimum species diversity were recorded in spring and autumn, respectively. Results of the UPGAM cluster analysis suggest that the examined 5 sites can be divided into two groups, sites exposed to open sea (transects 1, 2, 3) and comparatively sheltered sites (transects 4, 5) faced with the Pusan Harbor. The former group is represented by large brown algae, Sargassum and Hizikia, while Ulva pertusa and Lomentaria species are mainly found in the latter group. A calcareous alga, Corallina pilulifera occurred dominantly in both groups throughout the year. In conclusion, number of species in this area was remarkably reduced as compared to the previous data. This result may suggest probably more change of algal vegetation in future, considering the physical and chemical pollutions loaded in the coastal marine environment of this area.

The Analysis of Phytoplankton Community Structure in the Middle-Lower Part of the Nakdong River (낙동강 중·하류의 식물플랑크톤 군집구조 분석)

  • A seasonal variation of phytoplankton community in the middle-lower part of the Nakdong River was studied in four sampling stations at monthly intervals from January to December 2012. We identified 40 genera 72 species of phytoplankton. Among these, diatoms were 36 species (50.0%), green algae 20 species (27.8%), blue-green algae 9 species (12.5%) and others 7 species (9.7%), respectively. The phytoplankton standing crops were recorded a maximum of 29,640 cells/mL at the Mulgeum (St. 4) in June and a minimum of 236 cells/mL at the Goryung (St. 1) in October. Also, Standing crops were increased with proceeding from middle part to lower part. Ecological important species were Stephanodiscus hantzschii, Cyclotella meneghiniana, Synedra acus, Aulacoseira granulata, Pediastrum sp. and Microcystis aeruginosa. Seasonal succession of phytoplankton represented that Stephanodiscus hantzschii was dominant species in winter, Cyclotella meneghiniana, Synedra acus was dominant species in spring, Microcystis aeruginosa, Cyclotella meneghiniana, Fragilaria crotonensis, Synedra acus, Aulacoseria granulata was dominant species in summer, and Aulacoseria granulata, Cyclotella meneghiniana, Fragilaria crotonensis was dominant species in autumn. In the community analysis, diversity index and dominant index were higher May~July and December~February, respectively. Also, diversity index and dominant index were decreased and increased with proceeding from middle part to lower part.

Characteristics of Cyanobacterial Occurrence and Concentration Distribution of Cyanotoxins in Hoeya Reservoir (회야호의 남조류 발생 특성과 남조류 독소의 농도분포특성)

  • Algae blooms have soared recently in the lakes across the nation due to eutrophication. Blue-green algae cause unpleasant scene, produce taste and odor problem, and hinder processes in drinking water treatment. Algae toxicity monitoring has been strengthened, because the damages of wild lives and livestocks by algal toxins have been reported. Investigation on the characteristics of cyanobacterial occurrence and concentration distribution of Cyanotoxins in Hoeya reservoir have been conducted. Physical and chemical influences of water environment on cyanobacterial occurrences have also been studied. Movements of four species of Microcystin and five species of Anatoxin-a among Cyanotoxins were observed by LC-MS/MS analysis. Microcystis spp. among the cyanobacteria have mainly dominated in the Hoeya reservoir during the investigating period. The density of cyanobacteria were positively correlated with temperature and pH of water. Highest concentrations of Microcystin-LR and Microcystin-RR were $0.424{\mu}g/L$ and $0.117{\mu}g/L$ at the sampling points. Total concentration of Cyanotoxins in water coming into the water treatment plant was $0.182{\mu}g/L$, and they were not detected in treated water.
