Immunohistochemical study on the gastro-entero-pancreatic(GEP) endocrine cells of the blue fox, Alopex lagopus (북극여우의 위장췌 내분비세포에 관한 면역조직화학적 연구)

  • The regional distribution and the relative frequencies of endocrine cells were studied in nine portions of the blue fox GI tract, and the distribution pattern and cell types of the pancreatic endocrine cells were also studied in the pancreas by immunohistochemical method. Six kinds of immunoreactive cells were identified in the GI tract, and four kinds of immunoreactive cells were also identified in the pancreas. Although numerous 5-HT- and somatostatin-immunoreactive cells were seen throughtout the GI tract, somatostatin-immunoreactive cells were a few in the intestine. Very numerous Gas/CCK-immunoreactive cells were restricted generally in the pyloric region and duodenum. Numerous glucagon-immunoreactive cells were found in the stomach except the pyloric region, and generally a few in the intestine. Moderate number of BPP-immunoreactive cells were found in the stomach except the pyloric region, and a few in the large intestine. Numerous porcine CG-immunoreactive cells were restricted to the cardiac and fundic region. In the pancreas, four types of pancreatic endocrine cells-somatostatin-, glucagon-, BPP- and insuline-immunoreactive-were identified in the pancreatic islet and exocrine portion. These results suggest that the regional distribution, the relative frequencies and cell types of the GEP endocrine cells in the GI tract and pancreas varies considerably among the species.

A Study of Ku on the costume of the variant races dynasty in China-based on Khitai, Mongols and Manchus costume- (중국 민왕조 복식에서 에 관한 연구)

  • The purpose of this study is to investigate the Ku which was worn by nomadic tribes which was Khitai, Mongols and Manchus. The results of this study can be summarized as follows. 1. The fur clothes were very important clothes that nomadic tribes put on in order to protect them against the cold. The Ku was an outfit for protection against the cold. It was made of ani-mal fur and its style was tight sleeve and long dress. 2. The Ku was worn in all social classes including emperor, nobles and the masses in ancient China and Asia dynasty. After the nomadic tribes estabilished dynasty in China the style of Ku was changed. All kinds of animal fur was used to make the Ku but the volume of fur, kinds and colors were classified severely by their social position. In Khitai dynasty the Ku was worn in same style. But the emperor wore the Ku of silver marten. The noble class wore the Ku of purple, black and blue marten and silver squirrel skin. The masses wore the Ku of the other colors marten, sheep, squirrel and dessert fox skin. In Mongol dynasty, the emperor wore the Ku of purple marten, silver squirrel, silver fox and black fox skin. The nobles wore the Ku of the other colors marten and squirrel. The masses wore the Ku of degraded animal fur. Before the Manchus estabilished the varient races dynasty in China, the rich man preferred the Ku of marten, squirrel, fox, goat furs. But the poorman preferred the Ku of cow, horses, pig, sheep, cat, dog, snake and deer skin. After the manchus conquered the China, the manners of dress were changed. Generally the fur did not appear in right side of garment, but high quality or noble fur appeared outside. Sometimes the inside or outside of Ku was made of silk fabrics and sleeves, neck-band was made of the high quality fur.

Ethanol Extract of Schisandra chinensis (Turcz.) Baill. Reduces AICAR-induced Muscle Atrophy in C2C12 Myotubes (마우스 C2C12 근관세포에서 AICAR로 유도된 근위축에 미치는 오미자 추출물의 영향)

  • Muscle atrophy, known as a sarcopenia, is defined as a loss of muscle mass resulting from a reduction in the muscle fiber area or density due to a decrease in muscle protein synthesis and an increase in protein breakdown. Schisandrae fructus (SF) extract of the fruits of Schisandra chinensis (Turcz) Baillon has been used as a tonic in traditional medicine for thousands of years. Although a great deal of work has been carried out on the therapeutic potential of SF, its pharmacological mechanisms of action in muscle diseases actions remain unclear. In the present study, we investigated the inhibitory effects of SF ethanol extracts on the production of muscle atrophy factors in C2C12 myotubes stimulated with 5-aminoimidazole-4-carboxamide-ribonucleotide (AICAR), an AMP-activated kinase (AMPK) activator, and sought to determine the underlying mechanisms of action. AICAR upregulated atrophy-related ubiquitin ligase muscle RING finger-1 (MuRF-1) and stimulated the levels of the forkhead box O3a (FoxO3a) transcription factor in the C2C12 myotubes. SF supplementation effectively and concentration- dependently counteracted AICAR-induced muscle cell atrophy and reversed the increased expression of MuRF-1 and FoxO3a. Our study demonstrates that SF can reverse the muscle cell atrophy caused by AICAR through regulation of the AMPK and FoxO3a signaling pathways, followed by inhibition of MuRF-1.

High Glucose Induces Apoptosis through Caspase-3 Dependent Pathway in Human Retinal Endothelial Cell Line (인간망막 내피세포주에서 고농도 포도당이 caspase-3 경로를 통해 세포자연사 유도)

  • Diabetic Retinopathy (DR) is a leading cause of blindness among adults in the western countries. Hyperglycemia is a condition, that induces apoptotic cell death in a variety of cell types in diabetes, but the mechanism remains unclear. The aim of the study is to understand the effects of high Glucose on Human Retinal Endothelial Cells. Retinal endothelial cells were cultured in Iscove's Modified Dulbecco's Medium (IMDM) containing 5, 25 and 50 mM Glucose, incubated for 24, 36 and 48 hours in humidified 5 % CO$_2$ incubator at 37$^{\circ}C$. Human Retinal Endothelial Cell Line (HREC) were characterized for morphology with different treatment by phase contrast microscopic analysis. Number of dead and viable cells was counted by trypan blue exclusion and supported by MTT assay. The intracellular Hydrogen peroxide (H$_2$O$_2$), a Reactive Oxygen Species (ROS) generation in high glucose conditions was assessed by FOX II assay and apoptosis by caspase-3 assay. The high glucose treated cells undergoing DNA fragmentation was witnessed by Agarose gel electrophoresis. We found that the cells incubated with 25 and 50 mM glucose containing medium for 48 hours altered the morphology of the cell, induced apoptosis and DNA fragmentation. The dead cell number were high in 25 and 50 mM when compared to the cells incubated with 5 mM glucose for 24, 36, and 48 hours. Also, the H$_2$O$_2$ levels and the activity of caspase-3 were increased in high glucose treated cells. Conclusions/interpretation: Our results demonstrated that elevated glucose induces apoptosis in cultured HREC. The hyperglycemia-induced increase in apoptosis may be dependent on caspase activation. The association between ROS generation and caspase-3 activation on high glucose treated cells is yet to be investigated.
