이식 및 수확시기에 따른 쪽의 생엽수량 및 색소함량의 변화 (Changes of Fresh Leaf Yield and Colorant Level with Different Transplanting and Harvest Time in Persicaria tinctoria H. Gross)

  • 쪽 두 품종, 나주재래종과 나람블루를 사용하여 높은 생엽수량 및 색소함량을 얻기 위한 적정한 이식시기와 수확시기를 알아보고자 수행한 재배시험 결과, 대체적으로 두 품종 모두 비슷한 경향을 나타냈다. 이식시기가 늦을수록 생엽수량은 감소하는 경향을 보였으며, 이식시기 시험구간 다소 차이가 있었지만 니람이나 색소함량은 유의한 차이를 보이지 않았다. 수확시기가 늦어질수록 초장, 분지수, 생엽수량은 증가하는 경향을 보였지만, 니람함량은 8월 20일까지 증가하다가 이후 정체하거나 감소하는 경향을 보였다. 수확시기에 따른 인디고 함량 변화는 8월 5일에 가장 높은 함량을 보였고 이후 감소하는 경향을 보였다. 따라서 쪽 재배에서 높은 생엽수량과 색소함량을 얻기 위해서는 가능한 이르게 이식하는 것이 좋으며 8월 초순경에 수확하는 것이 유리한 것으로 판단되었다.

씀바귀 추출물이 인체유방암세포의 활성 산소 및 Bcl-2 Family에 미치는 영향 (Effects of Ixeris dentata Extract on Radical Oxygen Species and Bcl-2 Family in Human Breast Cancer Cells)

  • 본 연구는 인체유방암세포인 MDA-MB-231세포를 이용하여 IDE를 처리하였을 때 세포사멸에 미치는 영향을 세포화학적 방법으로 확인하였다. IDE를 각각 0, 20, 30 및 $40{\mu}g/mL$ 첨가하여 24시간 배양한 후, 세포증식 억제, 막투과성, 세포내 ROS 분석 및 세포사멸 단계의 특성을 FACS 분석하고, RT-PCR에 의한 사멸관련 유전자 중 Bax/Bcl-2 ratio 분석을 통하여 IDE의 항암작용의 조절작용을 밝히고자 하였다. MTT의 세포 증식 억제는 첨가된 IDE의 농도에 따라 유의적으로 감소하였다(p<0.05). 이와 동시에, trypan blue에 대한 염색성과 DCF-DA 형광 분석의 결과는 각각, 막투과성과 세포내 ROS 농도가 모두 농도 의존적으로 증가됨을 보였다(p<0.05). 이와 같은 IDE 농도에 따른 세포화학적 변화 중에서 세포의 초기사멸에서 후기사멸 과정으로 급격한 사멸단계의 변화가 특히, IDE 농도 30과 $40{\mu}g/mL$에서 일어났다. RT-PCR 분석에 의한 Bax/Bcl-2 ratio도 IDE 농도 30과 $40{\mu}g/mL$에서 급격히 증가하였다(p<0.05). 이와 같은 세포화학적 결과와 RT-PCR 결과를 종합해 볼 때, IDE의 유방암세포(MDA-MB-231)에 대한 세포사멸작용은 막 투과성의 증가와 ROS 증가를 통하여 세포에 점진적인 손상을 일으키며, 이는 세포의 생화학적 변화도 초래하여 세포 증식 억제를 감소시켜, 결국 세포내의 사멸관련 유전자 지표인 Bax/Bcl-2 ratio를 크게 변화시키는 일련의 세포사멸을 점진적으로 유도시켜 항유방암의 효과의 가능성을 제시하였다.

Dye removal from water using emulsion liquid membrane: Effect of alkane solvents on efficiency

  • Effect of different alkane based solvents on the stability of emulsion liquid membrane was investigated using normal alkanes (n-hexane, n-heptane, n-octane and n-decane) under various operating parameters of surfactant concentration, emulsification time, internal phase concentration, volume ratio of internal phase to organic phase, volume ratio of emulsion phase to external phase and stirring speed. Results of stability revealed that emulsion liquid membrane containing n-octane as solvent and span-80 (5 % (w/w)) as emulsifying agent presented the highest amount of emulsion stability (the lowest breakage) compared with other solvents; however, operating parameters (surfactant concentration (5% (w/w)), emulsification time (6 min), internal phase concentration (0.05 M), volume ratio of internal phase to organic phase (1/1), volume ratio of emulsion phase to external phase (1/5) and stirring speed (300 rpm)) were also influential on improving the stability (about 0.2% breakage) and on achieving the most stable emulsion. The membrane with the highest stability was employed to extract acridine orange with various concentrations (10, 20 and 40 ppm) from water. The emulsion liquid membrane prepared with n-octane as the best solvent almost removed 99.5% of acridine orange from water. Also, the prepared liquid membrane eliminated completely (100%) other cationic dyes (methylene blue, methyl violet and crystal violet) from water demonstrating the efficacy of prepared emulsion liquid membrane in treatment of dye polluted waters.

Efficient Recovery of Lignocellulolytic Enzymes of Spent Mushroom Compost from Oyster Mushrooms, Pleurotus spp., and Potential Use in Dye Decolorization

  • This study was conducted in order to perform efficient extraction of lignocellulolytic enzymes amylase (EC 3.2.1.1), cellulase (EC 3.2.1.4), laccase (EC 1.10.3.2), and xylanase (EC 3.2.1.8) from spent mushroom compost (SMC) of Pleurotus ostreatus, P. eryngii, and P. cornucopiae. Optimal enzyme recovery was achieved when SMCs were extracted with 50 mM sodium citrate (pH 4.5) buffer at $4^{\circ}C$ for 2 hr. Amylase, cellulase, and xylanase activities showed high values in extracts from P. ostreatus SMC, with 2.97 U/g, 1.67 U/g, and 91.56 U/g, respectively, whereas laccase activity and filter paper degradation ability were highest in extracts from P. eryngii SMC, with values of 9.01 U/g and 0.21 U/g, respectively. Enzymatic activities varied according to the SMCs released from different mushroom farms. The synthetic dyes remazol brilliant blue R and Congo red were decolorized completely by the SMC extract of P. eryngii within 120 min, and the decolorization ability of the extract was comparable to that of 0.3 U of commercial laccase. In addition, laccase activity of the SMC extract from P. eryngii was compared to that of commercial enzymes or its industrial application in decolorization.

Evaluation of black glutinous rice (Oryza sativa L) extract as a novel nuclear stain for human sperm head assessment by microscopic examination

  • Objective: To compare black rice (Oryza sativa L) extract with three different staining methods for human sperm head assessment. Methods: Semen samples were collected from 34 volunteers. Four smears of each ejaculate were prepared for staining using the rapid Papanicolaou (PAP) stain, SpermBlue, DipQuick, and black rice extract. The percentage of defective sperm heads (mean${\pm}$standard deviation) was compared. Results: Black glutinous rice extract, a natural dye, was used instead of hematoxylin to stain the nuclei of the sperm heads. The percentage of defective sperm heads showed a significant difference between black rice extract and DipQuick (p= 0.000). In contrast, black rice extract and rapid PAP showed no statistically significant difference (p= 0.974). A strong correlation (r = 0.761) was found between the findings obtained using rapid PAP and black rice extract. In contrast, a weak correlation (r = 0.248) was obtained between DipQuick and black rice extract for the percentage of defective sperm heads. Conclusion: The results showed good agreement and a strong correlation between the rapid PAP and black rice extract stains. The advantages of black rice extract as a novel substitute for hematoxylin for nuclear staining include ease of preparation, local availability, and favorable nuclear staining properties. Further studies could also focus on comparing staining techniques in clinical samples.

  • The purpose of the study was to investigate weaving, traditional dyeing, fiber material, pattern how carpet developing according to Turkey area for using the data that can rear to the export strategic industry for carpet production and development that is correct in culture. The result are followed: 1) Life of the age was expressed in color and pattern using according to geographical environment, climate, lifestyle, religion etc. 2) In the case of weaving, there is Kilim, Soumak that appears only weft on the surface as plane weaving without knot and carpet of knotted pile weaving and knot of carpet is duplex knot difference with carpet of the other country. 3) In the case of textile material, there is use most wool fiber that can get easily from breeded sheep by nomads. 4) In the case of dyeing, did the local traditional color to use dye extracting in dyeing material that can get easily in the area. Red that can extract in madder that can get easily in which area of Turkey, dark navy blue of indigo dye that indigo plant fermentation and cream beige that is wool's natural color were exposed representative traditional color of Turkey carpet. 5) Pattern was advanced uniquely as culture of the area; weaving person's sensitivity and desire are reflected through centuries. Amulet, riches & honors, fecundity and happiness appeared most pattern in any area.

Decolorization of Dyehouse Effluent and Biodegradation of Congo Red by Bacillus thuringiensis RUN1

  • A dye-decolorizing bacterium was isolated from a soil sample and identified as Bacillus thuringiensis using 16S rRNA sequencing. The bacterium was able to decolorize three different textile dyes, namely, Reactive blue 13, Reactive red 58, and Reactive yellow 42, and a real dyehouse effluent up to 80-95% within 6 h. Some non-textile industrially important dyes were also decolorized to different extents. Fourier transform infrared spectroscopy and gas chromatography-mass spectrometer analysis of the ethyl acetate extract of Congo red dye and its metabolites showed that the bacterium could degrade it by the asymmetric cleavage of the azo bonds to yield sodium (4-amino-3-diazenylnaphthalene-1-sulfonate) and phenylbenzene. Sodium (4-amino-3-diazenylnaphthalene-1-sulfonate) was further oxidized by the ortho-cleavage pathway to yield 2-(1-amino-2-diazenyl-2-formylvinyl) benzoic acid. There was induction of the activities of laccase and azoreductase during the decolorization of Congo red, which suggests their probable role in the biodegradation. B. thuringiensis was found to be versatile and could be used for industrial effluent biodegradation.

아로니아 추출물에 의한 직물의 천연염색과 염액의 Spectrum 분석 (Analysis of naturally Dyed Textile Fabrics by using Aronia Extract)

  • This study examined the dyeing characteristics and spectrum of cotton and silk by using dyeing solution extracted from aronia. The value $L^{\ast}$ which indicates the brightness of cotton regardless of mordant increases with increasing pH from pH 3.5 to pH 10.5. The color difference value(${\Delta}E$) of the cotton was generally high in pH 3.5 regardless of mordant existence. The silk showed the highest ${\Delta}E$ value in pH 3.5 dye solution as cotton did. On the other hand, the silk with mordant dyed showed the highest ${\Delta}E$ value in pH 7. Silk fabrics dyed with Aronia solution turns out red in pH 3.5, blue in pH 7 and yellow in pH 10.5. This is because of the amino acid, one of the silk ingredients, combines with a part of anthocyanin to show blue. As the result of the spectrum measurement, the maximum absorption wavelength of Aronia solution was increased in the order of pH 10.5, pH 3.5 and pH 7, regardless of extraction temperature and mordant. The measurement results of color fastness to washing and color fastness to light are generally low. Therefore, there is a need for further study to improve color fatness in the future.

A Portulaca oleracea L. extract promotes insulin secretion via a K+ATP channel dependent pathway in INS-1 pancreatic β-cells

  • BACKGROUND/OBJECTIVE: This study was designed to investigate how a Portulaca oleracea L. extract (POE) stimulates insulin secretion in INS-1 pancreatic ${\beta}-cells$. MATERIALS/METHOD: INS-1 pancreatic ${\beta}-cells$ were incubated in the presence of various glucose concentrations: 1.1 or 5.6, 16.7 mM glucose. The cells were treated with insulin secretagogues or insulin secretion inhibitor for insulin secretion assay using an insulin ELISA kit. In order to quantify intracellular influx of $Ca^{2+}$ caused by POE treatment, the effect of POE on intracellular $Ca^{2+}$ in INS-1 pancreatic ${\beta}-cells$ was examined using Fluo-2 AM dye. RESULTS: POE at 10 to $200{\mu}g/mL$ significantly increased insulin secretion dose-dependently as compared to the control. Experiments at three glucose concentrations (1.1, 5.6, and 16.7 mM) confirmed that POE significantly stimulated insulin secretion on its own as well as in a glucose-dependent manner. POE also exerted synergistic effects on insulin secretion with secretagogues, such as L-alanine, 3-isobutyl-1-methylxanthine, and especially tolbutamide, and at a depolarizing concentration of KCl. The insulin secretion caused by POE was significantly attenuated by treatment with diazoxide, an opener of the $K{^+}_{ATP}$ channel (blocking insulin secretion) and by verapamil (a $Ca^{2+}$ channel blocker). The insulinotropic effect of POE was not observed under $Ca^{2+}$-free conditions in INS-1 pancreatic ${\beta}-cells$. When the cells were preincubated with a $Ca^{2+}$ fluorescent dye, Fluo-2 (acetoxymethyl ester), the cells treated with POE showed changes in fluorescence in red, green, and blue tones, indicating a significant increase in intracellular $Ca^{2+}$, which closely correlated with increases in the levels of insulin secretion. CONCLUSIONS: These findings indicate that POE stimulates insulin secretion via a $K{^+}_{ATP}$ channel-dependent pathway in INS-1 pancreatic ${\beta}-cells$.

Myristicae Semen Extract Protects Excitotoxicity in Cultured Neuronal Cells

  • Myristica fragrans seed from Myristica fragrans Houtt (Myristicaceae) has various pharmacological activities peripherally and centrally. The present study aims to investigate the effect of the methanol extract of Myristica fragrans seed (MF) on kainic acid (KA)-induced neurotoxicity in primary cultured rat cerebellar granule neuron. MF, over a concentration range of 0.05 to $5\;{\mu}g/ml$ inhibited KA $(500\;{\mu}M)-induced$ neuronal cell death, which was measured by trypan blue exclusion test and 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. MF $(0.5\;{mu}g/ml)$ inhibited glutamate release into medium induced by KA $(500\;{\mu}M)$, which was measured by HPLC. Pretreatment of MF $(0.5\;{mu}g/ml)$ inhibited KA $(500\;{\mu}M)-induced$ elevation of cytosolic calcium concentration $([Ca^{2+}]_c)$, which was measured by a fluorescent dye, Fura 2-AM, and generation of reactive oxygen species (ROS). These results suggest that MF prevents KA-induced neuronal cell damage in vitro.
