Environmental Studies of the Lower Part of the Han River V. Blooming Characteristics of Phytoplankton Communities (한강하류의 환경학적 연구 V.식물플랑크톤 군집 대발생의 특징)

  • Ecological characteristics of phytoplankton communities were investigated biweekly at 6 stations from Feb. 2001 to Feb. 2002 at the lower part of the Han River. During the study, a total of 267 taxa which were composed of 114 green algae, 104 diatoms, 22 blue-green algae, 13 chrysophytes and 2 dinoflagellates were observed. The standing crops of phytoplankton communities were ranged from 1.30 ${\times}10^6$ cells ${\cdot}l^{-1}$ to 37.47 ${\times}10^6$ cells ${\cdot}l^{-1}$. The dominant species were 11 taxa including Pediastrum duplex, Scenedesmus quadricauda in green algae, Osillatoria limosa, Microcystis aeruginosa, Merisomorpedia sp., Phormidium sp. in blue-green algae, and Asterionella gracillima, Aulacoseira granulata, Fragilaria crotonensis, Stephanodiscus hantzschii f. tenuis, Synedra acus in diatoms. Asterionella gracillima and Stephanodiscus hantzschii f. tenuis were abundant at every station from winter to spring. Stephanodiscus hantzschii f. tenuis occupied the highest dominant ratio and standing crop. From summer to autumn, dominant species were Aulacoseira granulata at all stations and blue-green algae such as Osillatoria limosa, Microcystis aeruginosa, Merisomorpedia sp. at some stations. These 4 species were mostly present at the lower stations 4, 5, 6 and very few at the upper stations 1, 2, 3.

Origins and Paleoceanographic Significance of Layered Diatom Ooze from Bransfield Strait in the Northern Antarctic Peninsula around 2.5 kyrs BP

  • We used diatom and porewater data of two piston cores from the central subbasin and one from the western subbasin in the Bransfield Strait in the northern Antarctic Peninsula to elucidate the depositional mechanism of the layered diatom ooze. The layered diatom ooze is characterized by an abundance of organic carbon, biogenic silica, sulfde sulfur, and lower porewater sulfate concentration. This lack of pore-water sulfate concentration in the diatom ooze interval may reflect development of reducing micro-environment in which bacterially mediated sulfate reduction occurred. The negative relationship between the total organic carbon and sulfate contents, however, indicates that sulfate reduction was partly taking place but does not control organic carbon preservation in this unit. Rather, well-preserved Chaetoceros resting spores in the layered diatom ooze indicate a rapid sedimentation of the diatom as a result of repetitive iceedge blooms on the Bransfield shelf during the cold period (around 2500 yrs BP) when the permanent seaice existed on the shelf, During this period, it is expected that the downslope-flowing cold and dense water was also formed on the Bransfield shelf as a result of sea ice formation, playing an important role for the formation of layered diatom ooze in the Bransfield subbasins.

A Study on Phosphorus Removal Process Using Steel Industry By-Products(Slag) at Dynamic condition (동적(動的) 상태(狀態)에서 산업(産業) 폐기물(廢棄物)을 이용(利用)한 인(燐) 제거(除去)에 관한 연구(硏究))

  • Excessive phosphorus (P as orthophosphate) is one of the major pollutants in natural water that are responsible for algal blooms and eutrophication. P removal by slag is an attractive solution if the P sorption capacity of the slag is significant. To design an efficient land treatment facility, basic information on the behaviour of P in the media-water environment is required. In this study, detailed column experiments were conducted to study the P transport under dynamic condition, and mathematical models were developed to describe this process. The column experiments conducted with dust and cake waste products (slag) from BHP steel industry in Australia as adsorbing media indicated that they had higher sorption capacity of P than that of a sandy loam soil from North Sydney, Australia. P transport in the dust and cake columns exhibited characteristics S-shaped or curvilinear breakthrough curves. The simulated results from a dynamic physical non-equilibrium sorption model (DPNSM) and Freundlich isotherm constants satisfactorily matched the corresponding experimental breakthrough data. The mobility of P is restricted proportionally to the adsorbent's sorption capacity.

Morphological Characteristics of Peridinium bipes f. occcultatum (Dinophyceae) Isolated from Three Geographically Segregated Aquatic Systems of Korea

  • To resolve some of the most pressing uncertainties of Peridinium identity, morphological characteristics of Korean Peridinium were examined using light and scanning electron microscopy. The Peridinium samples were collected from three different regions of Juam, Sang-sa and Togyo Reservoirs, when seasonal blooms occurred. Formula of the epithecal plate was recorded with 7 precingular, 3 intercalary, and 4 apical plates (4', 3a, 7"). An apical pore, 3 ${\sim}$ 5 ${\mu}m$ in size, was apparently present. The cingulum was easily observed under light microscope, and was considerably offset by about 15 ${\mu}m$ (2 ${\sim}$ 3 times per cingulum width). The sulcus was straight longitudinally and widened apparently towards the antapex. None of spine was found on the surface of the thecal plates on scanning electron micrographs. The average body length was 50.4 ${\mu}m$ with a range of 29 ${\sim}$ 63 ${\mu}m$. The geometric dimension, as designated to the body length:width ratio, was found from calculation to be 1.12 with a range of 1.00 ${\sim}$ 1.35, therefore, the cell was shown slightly elongated. Based on their morphology, the causative organisms of red tides in three different Korean waters were identified as P. bipes f. occultatum, which was reported for the first time in Korea.

The Rapid Differentiation of Toxic Alexandrium and Pseudo-nitzschia Species Using Fluorescent Lectin Probes

  • Since toxic Alexandrium catenella and non-toxic A. fraterculus are morphologically similar, they are difficult to discriminate under the light microscope. However, a novel technology, such as fluorescein isothiocyanate (FITC)-conjugated lectin probes enables easy and rapid differentiation. Toxic A. catenella bound seven different lectins, whereas the non-toxic A. fratercuzus did not bind Arachis hypogaea (PNA) lectin. In addition, Pseudo-nitrschia species in this study were also difficult to identify to species level with light microscope techniques, but it was possible to classify them using fluorescent lectins. Pseudo-nitzschia multistriata, P. subfraudulenta and P. pungens bound Canavalia ensiformis (ConA), whereas P. subpaclfica did not, and P. pungens also bound Ricinus communis (RCA). These results imply that lectin could be used as a critical tool in the differentiation of P. multistriata, P. subfraudulenta and P. pungens. However, P. subpacifica was not differentiated by the lectins tested. Therefore, it isconcluded that lectin probes are useful for discriminating toxic A. catenella from non-toxic A. fraterculus, and for the identification of some Pseudo-nitzschia species. In addition, this method has a great potential to speed and detection between non-toxic and toxic harmful algal blooms (HABs) in Korean biotoxin monitoring systems.

Clay-based Management for Removal of Harmful Red Tides in Korea: A Multi-perspective Approach

  • Periodically, harmful algal blooms (HABs) have occurred, with impacts on various areas including public health, tourism, and aquatic ecosystems, especially aquacultured and caged fisheries. To prevent or manage invasions of HABs into fish farms on an emergency basis, many methods have been proposed. Frequently over the past 30 years in coastal countries, treatments of clay and clay mixed with polyaluminum chloride (PAC) and chitosan have been tested for HAB-removal effectiveness in both the laboratory and the field. In Korea, yellow loess clay (hwangto) has been dispersed using electrolytic clay dispensers, both to decrease the amount of yellow loess clay's usage in containers and enhance HAB-removal efficiency. However, this emergency method has limitations, among which is the requirement for more effective controlling agents for field applications. Thus, in this paper, we review technologies for clay-based red tides prevention and control and their limitations, and, further, introduce next-generation algicidal technologies for the emergency protection of fish farms.

Selection of Chemicals for the Dissolved Phosphorus Control by Variations of Alkalinity and Hardness (용존인 제어를 위한 알칼리도와 경도 조절제의 선택)

  • This study was done to find chemicals adequate to control alkalinity and hardness in order to reduce dissolved phosphorus in water bodies like rivers and lakes. Five chemicals were selected for the study: calcite, lime, dolomite, magnesite, and gypsum. Data were obtained from the calculations with MINTEQ model as a function of dosage variations of each chemical. Findings are as follows: Three out of the five chemicals are found to be effective in reducing the dissolved phosphorus, i.e., calcite, lime, and dolomite. Calcite and dolomite are able to lower the phosphorus concentration up to one thousandth fold whereas lime does one hundred thousandths fold. In viewpoint of pH variation, both calcite and dolomite seem to be safe since the pH does not increase over 8.3 even in case of overdose. In the same circumstance, with lime the pH increases beyond 9 which is considered to be the highest pH level for the protection of water ecosystem. Nevertheless it is recommendable to use lime in case where there are some difficulties in water quality control due to algae blooms.

Operation and Diagnosis of DAF Water Treatment Plant at Highly Turbid Raw Water (고탁도시 DAF 정수장의 운영 및 진단)

  • DAF process has been designed considering raw water quality characteristics in Korea. Although direct filtration is usually operated, DAF is operated when the freshwater blooms occut or raw water turbidity become high. Pre-sedimentation iS prepared in case when raw water turbidity is very high by rainstorms. A main feature of this plant is that the operation mode can be changed (controlled) based on the characteristics of raw water to optimize the effluent quality and the operation costs. Treatment capacity (surface loading rate) and efficiency of DAF was found to be better than conventional sedimentation process. Moreover, low-density particles (algae and alum flocs) are easily separated while it is difficult to remove in sedimentation. One of the main concerns in adoption of DAF (Dissolved Air-Flotation) process is a high raw water turbidity problem. That is, DAF is not adequate for raw water, which is more turbid than 100NTU. In order to avoid this problem, pre-sedimentation basins are prepared in DAF plant to decrease the turbidity of DAF influent. For simulation of the actual operation, bench and full-scale tests were performed for highly turbid water conditions. Consequently, DAF process coupled with sedimentation is suggested that pre-sedimentation with optimum coagulation prior to DAF would be appropriate.

Seasonal Timing and Distribution of Charybdis japonica (Decapoda: Portunidae) Larvae off Yeonpyeong-do in the Yellow Sea, Korea (연평해역 민꽃게(Charybdis japonica) 유생의 출현 시기와 분포)

  • The distribution and occurrence of Charybdis japonica larvae were investigated off Yeonpyong-do, Korea, in the Yellow Sea. C. japonica larvae were collected monthly at 15 stations from early June to late October in 2006 and 2007. At each station, a Bongo net with 303 and $505{\mu}m$ mesh was deployed once with a double oblique tow. No larvae were caught in June, in both years. Zoea I was predominant in late July in 2006 and early August in 2007, whereas Zoea I accounted for 84% of all larvae collected and no larval stages later than Zoea III were sampled. Megalopa were the most abundant larval stage at all stations in late August in both years. The timing of larval hatching of C. japonica may be related to that of phytoplankton blooms in the study area. The finding that Zoea I and Megalopa were predominant in the study are may indicate that C. charybdis larvae are carried by advection.

Rapid detection and Quantification of Fish Killing Dinoflagellate Cochlodinium polykrikoides (Dinophyceae) in Environmental Samples Using Real-time PCR

  • The mixotrophic dinoflagellate Cochlodinium polykrikoides was reported to be linked to major fish kills in Korea and Japan since the 1990s. Rapid and sensitive detection of microalgae has been problematic because morphological identification of dinoflagellates requires light microscopic and scanning electron microscopic observations that are time consuming and laborious compared to real-time PCR. To address this issue, a real-time PCR probe targeting the ITS2 rRNA gene was used for rapid detection and quantification of C. polykrikoides. PCR inhibitors in water column samples were removed by dilution of template DNA for elimination of false-negative reactions. A strong association between cell quantification using real-time PCR and microscopic counts suggests that the real-time PCR assay is an alternative method for cell estimation of C. polykrikoides in environment samples.
