The study was carried out to evaluate the effect of exogenous bovine somatotropin on water metabolism in relation to mammary function in early lactation of crossbred Holstein cattle. Ten, 87.5% crossbred Holstein cattle were divided into two groups of 5 animals each. At day 60 of lactation, the control group was given placebo while animals in the experimental group were given recombinant bovine somatotropin (rbST) by subcutaneous injection with 500 mg of rbST (14-days prolonged-release rbST). In rbSTtreated animals, milk yield increased 19.8%, which coincided with a significant increase in water intake (p<0.01), while DM daily intake was not different when compared to the control animals. Water turnover rate as absolute values significantly increased (p<0.05), while the biological half-life of water did not change in rbST-treated animals. Total body water (TBW) and total body water space (TOH) as absolute values significantly increased (p<0.01) in rbST-treated animals, while it was decreased in the control animals. Absolute values of empty body water (EBW) markedly increased (p<0.05), which was associated with an increase in the extracellular fluid (ECF) volume. Absolute values of plasma volume and blood volume were also significantly increased (p<0.05) in rbST-treated animals. The increase in mammary blood flow in rbST-treated animals was proportionally higher than an increase in milk production. The plasma IGF-1 concentration was significantly increased (p<0.01) in rbST-treated animals when compared with those of control animals during the treatment period. Milk fat concentration increased during rbST treatment, while the concentrations of both protein and lactose in milk were not affected. The present results indicate that rbST exerts its effect on an increase in both TBW and EBW. An increased ECF compartment in rbST-treated animals might partly result from the decrease in fat mass during early lactation. The action of rbST on mammary blood flow might not be mediated solely by the action of IGF-1 for increase in blood flow to mammary gland. An elevation of body fluid during rbST treatment in early lactation may be partly a result of an increase in mammary blood flow in distribution of milk precursors to the gland.
뇌 영상분석 알고리즘 적용에 따른 뇌혈류의 변화를 관찰하기 위해 정상 성인 13명(평균연령 39세)을 대상으로 뇌혈류 단 광자 단층촬영(Single Photon Emission Computed Tomography, SPECT)을 시행하였다. 획득된 영상을 여과후 역투영법(Filtered Back Projection, FBP)과 반복적 방법(Ordered Subset Expectation Maximization, OSEM)으로 영상을 재구성하여 통계학적 파라미터 뇌지도법으로 비교하여 방사성 의약품의 뇌 분포양상을 확인하였으며, 혈류의 변화는 크러스트(Cluster)로 표현시켰다. 이에 대한 결과로는 여과후 역투영법이 반복적 방법보다 섭취가 증가된 부위는 우측 전두엽, 대뇌회전하부, 외핵, 좌측대뇌 변엽과 대상이랑이며, 섭취가 감소된 부위는 좌측전두엽, 중간전두이랑, 하측전두이랑, 중심전이랑, 하측전두이랑, 중심전이랑이었다. 이는 영상재구성시 적용되는 알고리즘에 따라 뇌 혈류분포가 다르게 나타난 것을 크러스트(Cluster)로 표현시켰고, 명확한 시각적 표시가 가능하도록 뇌 확룔 지도로 보여주는데 의의가 있다.

For each subject, CFD simulations were carried out assuming two different non-Newtonian rheology models Carreau and Ballyk models) and rescaled Newtonian viscosities based on characteristic shear rates to account for the shear-thinning property of blood. The sensitivity of WSS and oscillatory shear index (OSI) were contextualized with respect to the reproducibility of the reconstructed geometry and to assumptions regarding the inlet boundary conditions. We conclude that the assumption of Newtonian fluid is reasonable for studies aimed at quantifying the distribution of WSS-based extrema in an image-based CFD model of carotid bifurcation.

These paper was attempted to investigate the mechanism of increased blood level of sulfisomidine by cimetidine pretreatment pharmacokinetically. Especially, effect of cimetidine pretreatment on both renal clearance and biliary clearance of sulfisomidine was studied in rabbits. The results are as follows. The blood level of sulfisomidine administered intravenously in dose of 25mg/kg was elevated significantly by cimetidine pretreatment. Relative bioavailability and biological half-life were increased significantly by cimetidine pretreatment. Overall elimination rate constant ($betha$) and distribution rate constant ($K_{13}$) of sulfisomidine were decreased significantly by cimetidine pretreatment. The renal and biliary clearance of sulfisomidine were decreased significantly compared with those of control rabbits by cimetidine pretreatment. The results may be also related to the inhibition of sulfisomidine metabolism enzyme activity or reduction of blood flow in the liver.
건강한 정상인의 복부대동맥과 장골동맥 분기관의 다이콤 의료 영상으로부터 구성된 혈관 모델을 대상으로 전산혈류해석을 수행하였다. 본 연구에서는 CT로 촬영한 혈관영상으로부터 단면영상을 읽고 각 단면의 경계 좌표를 매끄럽게 추출하는 프로그램을 개발하여 적응하였다. 해석용 3차원 메쉬 생성과 혈류해석은 상용프로그램을 사용하였다. 해석을 통해 얻어진 압력, 속도 및 유량 분포는 정상인의 생리학적 범위 내에 있었다. 초음파로 측정된 장골동맥 중심부의 심장 수축기 속도는 해석에 비해 20% 정도 작은 값을 보였으며 한 주기 동안 속도 변화의 경향은 유사하였다. 본 연구의 접근방법은 환자 개인의 실제 혈관 형상에 기반한 혈류해석으로서 좁아진 동맥 또는 시술 될 동맥에서의 압력, 속도값, 속도분포에 대한 정보를 제공한다. 이 결과는 의사로 하여금 환자의 혈류역학적 변수의 변화를 파악하게 하여 환자의 상태를 더 잘 이해하고 수술의 필요성을 판단하도록 도움을 줄 수 있으며 나아가 혈관 수술의 기초자료로 사용될 수 있을 것이다. 이 연구는 향후 동맥경화와 밀접히 연관된 혈류역학적 변수의 분포를 혈관 내에 보여줌으로서 동맥경화를 조기 진단하는 프로그램으로 발전할 것으로 기대된다.
The model of an isolated organ system has been constructed to simulated the behavior of drug in the circulatory system of an acting organ or site. The model is developed on the following assumptions : The drug in the microcirculatory system cannot permeate the capilary walls. The capilary bed is modeled as a simple ideal plug flow system with and without radial concentration gradient. The mathematical model is developed from basic considerations of drug distribution with hemodynamical and pharmacokinetical meanings. It is considered that a nonmetabolic drug substance is injected into the arterial inflow site of an isolated organ at a constant rate. The concentration of the drug in the outflow site is mathematically expressed as a function of time.
목적: 요소분석법. 독립성분분석법 등이 PET을 이용하여 심근혈류를 비침습적으로 측정하기 위하여 사용되어 왔다. 이론적으로 뛰어나고 새로운 방법인 앙상블 독려성분분석법을 이용하여 $H_2^{15}O$ 동적 심근 PET데이터의 정량분석방법을 개발하였다. 이 연구에서 사용한 앙상블 독려성분분석법을 이용하여 환자의 혈류를 정량화 하였다. 대상 및 방법: 관동맥질환이 의심되어 관류 SPECT를 시행한 환자 20명을 대상으로 $H_2^{15}O$ 동적 심근 PET을 시행한 후 앙상블 독립성분분석법을 이용하여 심근 독립성분영상을 추출하였으며, 좌심실영역과 심근영역에 대한 영상대조도를 조사하였다. 앙상블 학습은 독립성분과 가중치 행렬에 대한 확률분포를 가정하고 베이지안 이론에 의해서 혼합자료에 대한 확률분포를 추정한다. 이렇게 추정한 혼합자료의 확률분포와 실제 분포간의 차이인 Kullback-Leibler 발산치가 최소가 되도록 독립성분과 가중치 행렬을 순차적으로 변화시켜가며 최종 해를 찾는 방식이다. 이 연구에서 사후확률분포는 동적 핵의학 영상에 적합한 비음성제약조건과 함께 수정된 가우시안 분포를 이용하여 최적화 하였다. 혈류량은 심첨부, 중벽 네 부분, 하벽 네 부분의 9개 영역으로 나누어 측정하였으며, 측정결과에 대해 관류 SPECT 소견과 관동맥조영술의 소견과 비교하였다. 결과: 전체 20명의 휴식기 및 부하기 영상에서 5명을 제외한 15명의 데이터에 대해 심근혈류를 측정할 수 있었다. $H_2^{15}O$ 동적 심근 PET에서 앙상블 독립성분분석법을 이용하여 정량화한 휴식기 혈류량은 $1.2{\pm}0.40$ ml/min/g, 부하기 혈류량은 $1.85{\pm}1.12$ml/min/g이었다. 같은 영역에 대해 두 번 측정했을 때 측정된 심근혈류값의 상관계수는 0.99로 재현성이 높았다. 분리된 독립성분영상에서 영상대조도는 좌심실에 대한 심근영역의 비는 평균 1:2.7이었다. 관동맥 조영술을 시행한 9명에서 협착이 없는 분절과 협착이 있는 분절의 혈류예비능에 유의한 차이가 있었다(P<0.01). 또한, 관동맥조영술에서 협착이 확인된 66분절의 심근관류 SPECT 소견에서 가역적 혈류감소를 보인 분절의 혈류예비능이 더 많이 감소되는 경향을 보였으나 통계적 유의성을 보이지는 않았다. 결론: 앙상블 학습을 이용한 독립성분분석방법을 이용하여 심근혈류가 측정이 되었다. 앙상블 독립성분분석법을 이용한 $H_2^{15}O$ 동적 심근 PET 분석방법이 관상동맥 질환의 분석 및 동적 핵의학 영상 데이터의 연구에 도움이 될 것으로 기대된다.

Taurine, a ${\beta}-amino$ acid, plays an important role as a neuromodulator and is necessary for the normal development of the brain. Since de novo synthesis of taurine in the brain is minimal and in vivo studies suggest that taurine dose not cross the blood-brain barrier, we examined whether the choroid plexus, the blood-cerebrospinal fluid (CSF) barrier, plays a role in taurine transport in the central nervous system. The uptake of $[^3H]-taurine$ into ATP depleted choroid plexus from rabbit was substantially greater in the presence of an inwardly directed $Na^+$ gradient taurine accumulation was negligible. A transient in side-negative potential gradient enhanced the $Na^+-driven$ uptake of taurine into the tissue slices, suggesting that the transport process is electrogenic, $Na^+-driven$ taurine uptake was saturable with an estimated $V_{max}$ of $111\;{\pm}\;20.2\;nmole/g/15\;min$ and a $K_M\;of\;99.8{\pm}29.9\;{\mu}M$. The estimated coupling ratio of $Na^+$ and taurine was $1.80\;{\pm}\;0.122.$$Na^+-dependent$ taurine uptake was significantly inhibited by ${\beta}-amino$ acids, but not by ${\alpha}-amino$ acids, indicating that the transporter is selective for ${\beta}-amino$ acids. Since it is known that the physiological concentration of taurine in the CSF is lower than that in the plasma, the active transport system we characterized may face the brush border (i.e., CSF facing) side of the choroid plexus and actively transport taurine out of the CSF. Therefore, we examined in vivo elimination of taurine from the CSF in the rat to determine whether elimination kinetics of taurine from the CSF is consistent with the in vitro study. Using a stereotaxic device, cannulaes were placed into the lateral ventricle and the cisterna magna of the rat. Radio-labelled taurine and inulin (a marker of CSF flow) were injected into the lateral ventricle, and the concentrations of the labelled compounds in the CSF were monitored for upto 3 hrs in the cisterna magna. The apparent clearance of taurine from CSF was greater than the estimated CSF flow (p<0.005) indicating that there is a clearance process in addition to the CSF flow. Taurine distribution into the choroid plexus was at least 10 fold higher than that found in other brain areas (e. g., cerebellum, olfactory bulb and cortex). When unlabelled taurine was co-administered with radio-labelled taurine, the apparent clearance of taurine was reduced (p<0.0l), suggesting a saturable disposition of taurine from CSF. Distribution of taurine into the choroid plexus, cerebellum, olfactory bulb and cortex was similarly diminished, indicating that the saturable uptake of taurine into these tissues is responsible for the non-linear disposition. A pharmacokinetic model involving first order elimination and saturable distribution described these data adequately. The Michaelis-Menten rate constant estimated from in vivo elimination study is similar to that obtained in the in vitro uptake experiment. Collectively, our results demonstrate that taurine is transported in the choroid plexus via a $Na^+-dependent,saturable$ and apparently ${\beta}-amino$ acid selective mechanism. This process may be functionally relevant to taurine homeostasis in the brain.

The oxygen distribution at steady state is analyzed mathematically in a hexagonal cylinder. The domain is penetrated by parallel cylindrical capillaries of different oxygen squirt. Asymptotic solution is used to determine the effect of axial diffusion. Oxygen concentration profiles are displayed at some positions of capillary-beds.
