  • Flavonoids with oxygen atoms are known to have potent biological effect.They have been studied long as major antioxidants which protect cell membranes. Recent medical surveys show that increased intake of selenium decreases the risk of breast, colon, lung and prostrate cancer by preventing free radical generation. The flavonoids, isoflavonoids, and coumarins which form the bulk of these compounds are very polar and have limited use as drugs which have to pass through BBB(Brain Blood Barrier)The non-polar property is increased by exchange oxygen to selenium as a part of heterocyclic compound.   • Citrus unshiu Marc., a family of Rutaceae, has been used as antitussives, antiemetics, expectorants, spasmolytics, stomachics and digestants in oriental medicine. But there is few reports on the pharmacological effects of Citrus unshiu pericarpium. Present experiment was performed to observe the effects of the water extract of Citrus unshiu on isolated auricle and intestine in rabbits and blood pressure in rats and also to study on the active component on the cardiovascular system by means of thin layer chlomatography and spectrophotometer. 1. The water extract showed positive inotropic and positive chronotropic effect in the concentration of $1{\times}10^{-4}g/ml$, $5{\times}10^{-4}g/ml$, and $1{\times}10^{-3}g/ml$. 2. The motility of isolated rabbit intestine was significantly depressed by the water extract. 3. The blood pressure was markedly increased by the water extract. 4. The blood vessel of rabbit ear was constricted by perfusion of the ether extract. 5. The purified alkaloid of Citri pericarpium by ion exchange resin showed the same Rf value with l-synephrine on thin layer chromatography. 6. The purified alkaloid of Citri pericarpium by thin layer chromatography showed the same absorption curve with l-synephrine in the ultraviolet scanning.

Detection of Phagocytosis-Promoting Factor of Culture Supernatant from Feline Peripheral Blood Mononuclear Cells Cultured with Egg White Derivatives (계난백유래물질로 배양한 고양이 말초혈액 단핵구세포 배양상층액중의 탐식촉진인자 검출)

  • The aim of this study is to determine the phagocytosis-promoting factor(s) for feline peripheral blood polymorphonuclear cells (PMN) by culture supernatant from mono-nuclear cells (MNC) treated with egg white derivatives (EWD). The phagocytic activity of PMN was analyzed by a flow cytometry system. The EWD did not show direct effect on the phagocytic response of PMN. The phagocytic activity of PMN was enhanced by culture supernatant from MNC but not PMN treated with EWD. Therefore, it was suggested that the enhanced phagocytic activity of feline PMN could be mediated by humoral factor(s) released from MNC treated with EWD. Thus, the phagocytosis-promoting factor(s) in supernatant fraction from MNC culture treated with EWD were isolated by reverse phase high pressure liquid chromatography. The resulting supernatant fraction on 29.02 minutes of retention time showed high phagocytic activity of PMN. The molecular weight of this supernatant fraction was 16 to 18 kDa when analyzed by capillary electrophoresis. The isoelectric point was pH 5.76 when assessed by ion-exchange chromatography. These results suggest that EWD stimulates feline MNC to elaborate a phagocytosis-promoting factor, 16 to 18 kDa of molecular weight, which could be an important mediator for the enhancement of phagocytic activity of feline peripheral blood phagocytes. Further study will be needed to elucidate this phagocytic factor.

  • A preterm female infant born at 27 weeks of gestation with a birth weight of 990 g developed acute hypotonia, apnea, hypotension and bradycardia mimicking septic shock syndrome at 14h after birth. Laboratory tests indicated a severe hypermagnesemia of 45 mg/dL. The renal function, complete blood count and maternal blood concentrations of magnesium were normal, and the blood cultures were negative. The patient recovered with treatment including exchange transfusion. However, the etiology of the severe hypermagnesemia remains unknown.

  • The OXYREX hollow fiber membrane oxygenator developed by joint work of KIST and Green Cross Medical company has been evaluated by experimental investigation and clinical application, In this oxygenator gas exchanges occur through small pores of 0.1pm size which are distributed on 70% of surface of polypropylene hollow fiber. The Oxyrex membrane oxygenator consists of 36 thousand hollow fibers and it has 3.3m2 of gas exchange surface. The Oxyrex membrane oxygenator has unique blood flow path: blood enters the oxygenator passes between the hollow fibers and exits through outlet ports, that provides low transmembrane pressure drop. In the animal experiment and in vitro investigations of Oxyrex oxygenator, it showed low transmembrane pressure difference, effective heat exchanger performance, stable gas transfer function and less blood trauma. The Oxyrex oxygenator been used from March, 1990, to October, 1990, in 40 patients undergoing open heart operations. In the clinical applications of Oxyrex, adequate oxygenation[PaO2, 283$\pm$70mmHg] and carbon dioxide removal[PaCO2, 27\ulcorner6mmHg]were maintained under the condition of FiO2: below 0.6, Hct; 25%, perfusion flow; 2.4 L/min, gas flow: 2.1 L/min. During maximum 365 minutes of cardiopulmonary bypass[CPB] time period, the Oxyrex oxygenator maintained stable condition of PaO2, PaCO2 respectively and it also kept low plasma hemoglobin level. The complement proteins C3 and CH50 were not significantly changed pre to post CPB. There were no complications related to the oxygenator during and after the CPB.

Albumin Fractions from Different Species Stimulate In Vitro Progesterone Production by Granulosa Cells in Buffalo

  • The ovarian follicular fluid was found to contain steroidogenesis stimulatory protein similar to albumin from human and buffalo. Therefore, the albumins from various species, commercial and purified, were studied for their steroidogenic effect on progesterone secretion by granulosa cells from buffalo ovaries, during culture. A dose of $20{\mu}g$ of bovine serum albumin was optimum to exhibit maximum progesterone secretion on day 6 of culture, in medium ($350{\mu}l$) containing $10^5$ cells. Among commercial albumins, chicken albumin showed highest effect on progesterone secretion, which was followed by albumins from goat, bovine, human, sheep and rat, respectively at day 6 of culture. The albumins were also purified from blood serum of buffalo, goat and rat using salt fractionation, ion-exchange chromatography, gel filtration and SDS-PAGE. The highest stimulatory effect on progesterone secretion was shown by albumin purified from buffalo blood serum and lowest by that from rat blood. Comparatively the buffalo and goat albumins were more biologically active than commercial albumins. The presence of some active molecules conjugated with freshly purified albumins may be responsible for better stimulatory effect.

Effect of Several Adsorbents on The Gastrointestinal Absorption of Paraquat

  • The effect of several adsorbents on paraquat poisohing was investigated (1) by measuring the saturatd amount of the poison adhered on the adsorbents in vitro and (2) by assaying the blood level of paraquat in the rat in in situ intestinal absorption experiments. Activated charcoal powder, natural aluminum silifonate) were used as adorbents. The steady-state blood level of paraquat in its absorption experiment with the cationic exchange resins was markedly lower than those without the resins or with other adsorbents. A good relationship was achieved between the calculated AUC or adsorptioin rate (in situ) and the saturated adsorption amount (in vitro). The rank order of the effect was sodium polystyene sulfonate > calcium polystyene sulfonate>natural aluminum silicate>activated charcoal powder. The effect of sodium polystyrene sulfonate after intestinal washing with physiological saline ws also measured, and a synergistic effect (marked decrease in blood paraquat level0 was found as compared with the intestinal washing alone. The simultaneous use of G.I. weshing and powerful adsorbent was scientifically proven to be most benefical.

Quantitative Speciation of Selenium in Human Blood Serum and Urine with AE- RP- and AF-HPLC-ICP/MS

  • Various separation modes in HPLC, such as anion exchange (AE), reversed-phase (RP), and affinity (AF) chromatography were examined for the separation of selenium species in human blood serum and urine. While RP- and AE-HPLC were mainly used for the separation of small molecular selenium species, double column AF-HPLC achieved the separation of selenoproteins in blood serum efficiently. Further, the effluent of AF-HPLC was enzymatically hydrolyzed and then analyzed with RP HPLC for selenoamino acid study. The versatility of the hybrid technique makes the in-depth study of selenium species possible. For quantification, post column isotope dilution (ID) with $^{78}Se$ spike was performed. ORC ICP/MS (octapole reaction cell inductively coupled plasma/mass spectrometry) was used with 4 mL $min^{-1}$ Hydrogen as reaction gas. In urine sample, inorganic selenium and SeCys were identified. In blood serum, selenoproteins GPx, SelP and SeAlb were detected and quantified. The concentration for GPx, SelP and SeAlb was $22.8{\pm}3.4\;ng\;g^{-1}$, $45.2{\pm}1.7\;ng\;g^{-1}$, and $16.1{\pm}2.2\;ng\;g^{-1}$, respectively when $^{80}Se/^{78}Se$ was used. The sum of these selenoproteins ($84.1{\pm}4.4\;ng\;g^{-1}$) agrees well with the total selenium concentration measured with the ID method of $87.0{\pm}3.0\;ng\;g^{-1}$. Enzymatic hydrolysis of each selenium proteins revealed that SeCys is the major amino acid for all three proteins and SeMet is contained in SeAlb only.

Comparative study of 82Sr separation/purification methods used at Brookhaven National Laboratory and ARRONAX

  • Nuclear imaging is one of the most powerful measures for non-invasive diagnosis of myocardial vascular disease. Radionuclide such as 13N, 15O, 201Tl and 82Rb is used for the measurement of cardiac blood flow. 13N, 15O and 201Tl are produced in cyclotrons while 82Rb is obtained from generator. Rubidium (Rb), an alkali ion, behaves biologically like potassium, and accumulates in myocardial tissue. Rb has rapid blood clearance profile which allows the use of 82Rb with a short physical half-life of 75 s for non-invasive evaluation of regional myocardial perfusion. There are several advantages of 82Rb over other radioisotopes. An ultra-short half-life significantly reduces the exposure of patients to radiation and allows to repeat injections for studying the effects of medical intervention. As a positron emitter, 82Rb allows positron emission tomography (PET) imaging which have shown superior diagnostic performances. 82Rb can be produced from generator by decay of its parent 82Sr. However, the preparation of 82Sr is difficult, because appropriate purity is required to meet the specification of the product. Recently reported procedure from ARRONAX research institute showed that a Chelex-100 resin is sufficient for this purpose and additional column is not necessary. Whereas Brookhaven National Laboratory (BNL) procedure contains three ion exchange resin separation, including Chelex-100 resin. Currently, since 82Sr production site is non-existent in Korea, Korea Atomic Energy Research Institute (KAERI) has plan to produce 82Sr within specifications. We compared 82Sr purification procedures reported from ARRONAX and BNL to investigate the most suitable procedure for our conditions.

Effect of Ionic Polymers on Sodium Intake Reduction (이온성 고분자를 이용한 나트륨 섭취 감소 효과)

  • Sodium chloride is present in our body fluids, and the blood contains approximately 0.9 wt% salt, which plays an important role in maintaining the osmotic pressure. However, the amount of salt intake has consistently increased, and an excessive intake can be the cause of high blood pressure, etc. In this study, it was investigated in vivo and in vitro whether biocompatible ionic polymers with K or Ca ions can be replaced by Na ions through an ion exchange process to be excreted. Among the polymers, Ca-polystyrene sulfonate, K-polystyrene sulfonate, Ca-carrageenan, and Ca-tamarind had an excellent Na exchange ability in the body temperature, simulated gastric fluid and also simulated intestinal fluid. The mechanism of Na removal by absorption and excretion without changing food taste in the mouth through the insolubility properties of these polymers is expected to be a solution for the current problems related with excess sodium intake.
