The Anesthetic Effects of the Epidural Administration of Tiletamin-Zolazepam and Lidocaine in Rats (흰쥐에서 Tiletamine-Zolazepam 및 Lidocaine의 경막외투여시의 마취 효과)

  • This study was performed to evaluate the anesthetic effects of the epidural administration of tiletamin-zolazepam and lidocaine to rats. Blood pressure, heart rate, respiratoty rate and blood chemistry were examined according to the time lapse, after the administration of tiletamine-zolazepam, lidocaine or saline. The results obtained were as follows. 1, Tiletamine-zolazepam group revealed fast anesthesia onset time (p<0.01) and also revealed prolonged ambulation time compared with lidocaine group (p<0.01). 2. In the effects of blood pressure, tiletamine-zolazepam group revealed significantly higher value than lidocaine group or saline group, and revealed the highest value at 20 minutes after administration. According to the time lapse, blood pressure of tiletamine-zolazepam group was recovered and showed similar value with lidocaine group and control group at 90 minutes after administration. 3. In the effects of heart rate, tiletamine-zolazepam group revealed significantly lower value than lidocaine group or saline group and revealed the lowest value at 30 minutes after administration, and recovered similar value with pre-administration at 90 minutes after administration. 4. In the effects of respiratory rate, lidocaine group revealed significantly lower value at 30 minutes administration compared with 0 and 60 minutes after administration (p<0.01). Tiletamine-zolazepam group also revealed significantly lower value at 30 minutes compared with 0 and 60 minutes after administration (p<0.01). The changes at 60 minutes after administration, lidocaine group revealed lower value than saline or tiletamine-zolazepam group, and tiletamine-zolazepam group revealed similar value with 0 minutes. 5. In the effects of tidal volume, lidocaine group revealed significantly lower value than saline group (p<0.001) and tiletamine-zolazepam group also revealed lower value than saline group, at 30 minutes after administration. The values at 60 minutes after administration, revealed similar results with that of 30 minutes after administration. 6. In the blood chemistry, the values of alanine transminase (ALT), aspartate transminase(AST) and creatinine did not reveal significant results at 60 minutes after administration. The values of ALT at 60 minutes slightly decreased compared with pre-administration, and revealed normal level.

  • This experiment was carried out to provide the data of blood chemistry in use of application to clinical diagnosis in racehorses. Blood was collected from 30 racehorses(♀ 17, ♂ 13). Calcium, inorganic phosphorus, blood glucose, blood urea nitrogen, cholesterol, bilirubin, fibrinogen and ketone body in serum were analyzed as follows. 1. The values of calcium and inorganic phosphorus were maintained a certain level without much variation regardless of sex and species, Ca/P ratio was in the range of 3.13~3.35. 2. The value of blood glucose was 60 mg/100ml, Age and sex of racehorses make no difference in the value. 3. The value of blood urea nitrogen and total cholesterol showed a tendency to increase according to the age. Total cholesterol value was somewhat higher in male than in female. 4. The value of total bilirubin showed a tendency to increase according to the age. There was no difference between sexes in the value. 5. The value of fibrinogen showed no apparent difference with age and sex. 6. Ketone body was negative in all of the cases. 7. There was no variation in the species of the racehorses.

  • During the period from june 1994 to february 1995 influence of free stall with concrete floor on profile of blood chemistry and clinico-morphopathogenesis of foot disease on 266 cows were studied. The results obtained as follow. 1. No differences between value of blood chemistry in normal and lame cows with foot disease were observed. 2. 20.7% of the cows were clinically lame. 3. Prevalence of clinical digital disorders were investigated: hyperplasia interdigitalis(45.8%), pododermatitis circumscripta(22.4%), dermatitis interdigitalis(9.4%), erosio ungulae(5.9%), phlegmona interdigitalis(3.5%), pododermatitis septica traumatica(3.5%), dermatitis digitalis(2.4%), white line disease(2.4%), pododermatitis aseptica diffusa(2.4%), dermatitis verrucosa(1.2%), fissura ungulae(1.2%) 4. Most claw lesions were located on lateral hindclaws and interdigital space of hindclaws.

  • The effects of lidocaine on the parameters of haematology and blood chemistry were investigated in the carp(Cyprinus carpio). At low concentrations(100 and 200 ppm) of lidocaine, no significant changes in the parameters of haematology and blood chemistry were observed during recovery period from the treatment of lidocaine. However, at high concentrations(300 and 400 ppm) of lidocaine, red blood cell(RBC) and hematocrit(Ht) value, plasma glucose concentration, and the activity of lactate dehydogenase(LDH) were markedly increased compared with the controls. These increases were maintained for up to 60 min. Based on these results, it is suggested that lidocaine is generally not toxic to fish but has shown the physiological stress responses at high concentrations.

  • Excessive drinking causes 'alcohol hangover' within 8-16 hours. The cause of 'hangover' has not been elucidated exactly until now, but it is reported that it is caused by the creation of blood ethanol and acetaldehyde as ethanol metabolites. In this study vinegar extract of wood (VE) or OC-1, to which the powder extract of green tea leaves extract is added, was administered to the rats 30 minutes before the oral administration of ethanol (3 g/kg) and the blood ethanol and acetaldehyde concentration was measured in order to evaluate the efficacy of the beverage material for detoxification. As a result, the blood ethanol concentration in the group of the VE-1(vinegar crude extract) and VE-2 (double diluted solution) is statistically lower (P,0.05) than the exclusive alcohol administered control group. The blood acetaldehyde concentration of all groups of VE and OC-2, which is the double dilution of OC-1, is statistically low after 7 hours following ethanol administration. Especially, the AUC value of OC-2 group is statistically low compared to the control group. Accordingly, it indicates the conclusion that VE and OC-1, reducing the blood ethanol and acetaldehyde concentration which are two leading factors of 'hangover' after drinking, and worthwhile to be developed as beverage materials to eliminate 'hangover'.

Quantitative Determination of Lipid Hydroperoxide in Human Blood Serum by Ferrothiocyanate Method (Ferrothiocyanate법에 의한 혈청 Lipid Hydroperoxide정량)

  • In order to develope the method of quantitative determination of lipid hydroperoxide in human blood serum, we tried the ferrothiocyanate method to total lipids extracted by Bligh-Dyer method and obtained the results as follows. 1. The maximum absorbance showed at the concentration of Mohr's solution, 0.127M at pH 1.70 and ammonium thiocyanate solution, 3.95M in the ferrothiocyanate method. 2. When hydrogen peroxide, cumene hydroperoxide, and oxidized linoleic acid were added to serum, and extracted them by Bligh-Dyer method to examine the extraction efficiency, we confirmed that cumene hydroperoxide and oxidized linoleic acid were extracted in $CHCI_3$ phase, and hydrogen peroxide in $MeOH-H_2O$ phase, respectively. 3. The concentration of lipid hydroperoxide of total lipids extracted from normal adult serum was $2.0{\times}10^{-5}M$, and increased proportionally the concentration of lipid hydroperoxide by increasing the amount of serum. 4. When we compared the total lipids extracted by Bligh-Dyer method and total lipids extracted after lipoprotein is precipitated by Yagi method in human blood serum, the concentration of lipid hydroperoxide was showed nearly the same value. From our results, we concluded that the concentration of lipid hydroperoxide in human blood serum could be determined quantitatively by ferrothiocyanate method.

  • For examination of antibiotic therapeutic efficacy in canine brucellosis, this examination was carried out two female bitches infected with Brucella canis in Gyeongbuk province, and used combicillin, baytril and doxycycline in susceptible antibiotics at B canis. During 18 month after the termination of antibiotic therapy, blood sample of the two bitches were examined for B canis antibody and antigen. The antibody of one bitch was disappeared at 5 month after antibiotic therapy and the other was continued at 18 month, but two bitches were not detected antigen by blood culture and PCR. Examination of blood chemical value (AST, ALT, urea, creatinine) of two bitches was increased in AST value during antibiotic therapy.

  • Hydrogen cyanide (HCN) is an extremely toxic gas frequently produced during the incineration of plastics, such as acrylonitrile-butadiene-styrene (ABS). A victim of a fire who has inhaled smoke could have cyanide in the blood. Therefore, cyanide could be a good marker for a post-mortem examination of a fire as well as carboxyhemoglobine (COHb) test of blood samples. For a particular fire case, a burned body with a suicide note was found inside a burned vehicle. Even though the COHb value is conclusive evidence, measuring the COHb for denatured blood might be difficult due to severe thermal denaturation or the formation of methemoglobin (MetHb). To overcome this difficulty, cyanide could be used as an indicator when investigating the death of a fire victim. In this study, gas chromatography/mass spectrometry (GC/MS) was adopted to measure the levels of cyanide in the blood through derivatization with pentafluorobenzyl bromide (PFBBr) under cation surfactant by scan and SIM mode. The concentration of cyanide in the blood of heart blood and brain of the victim was found to be 0.36 µg/mL and 1.20 µg/mL respectively, which was higher than the average value (0.041 µg/mL) found in the blood of 14 people who smoked.

Effects of the Semyung-Gangi-tang on the Blood Lipid in Experimentally Induced Hyperlipidemic Rabbits (세명항지탕이 고지혈증 토끼의 혈중 지질변화에 미치는 영향)

  • Etiology and pathogenesis of hyperlipidemia are considered as 'difficiency in origin' of liver, spleen and kidney combined with 'excess in superficiality' of phlegm stagnancy and blood stasis. The prescription of Semyung-Gangji-tang, designed for the treatment of hyperlipidemia and atherosclerosis, consists of herbs effective at treating blood stasis and phlegm stagnancy. It invigorates the liver, kidney and spleen. Its medical nature is not too cold or too hot and it increases human vital energy. The purpose of this study is to examine the currative effects of Semyung-Gangji-tang on hyperlipidemia in rabbits. Twenty-four male NZW rabbits, around 2kg of B.W., were divided into 3 groups. Group I was served as the normal group. Group II was served as the hyperlipidemic control group fed with 1 % cholesterol diet for 8weeks and with normal diet for the next 4weeks. Group III was served as the treatment group, treated the same as the control group and medicated with Semyung-Gangji-tang for the last 4weeks. Blood samples were collected from each of the animals at a, 3, 6, 8, 10, 12 weeks and used for the blood chemical analysis. Animals were sacrificed at 8 and 12weeks and chemical analyses were performed on the collected liver samples. The serum total cholesterol value of treatment group ($938.60{\pm}95.07{\;}mg/dl$) at l2weeks was significantly decreased (P<0.00l) compared with the value of control group ($1375{\pm}37.82{\;}mg/dl$) at 12weeks. The serum LDL-cholesterol value of the treatment group ($838.00{\pm}89.52 mg/dl$) at l2weeks was significantly decreased(p<0.00l) compared with the value of the control group ($1249.60{\pm}37.63{\;}mg/dl$) at l2weeks. These results indicate that Semyung-Gangji-tang has the effects of lowering serum lipid in experimentally induced hyperlipidemic rabbits.
