ELECTRON MICROSCOPIC STUDY OF RESIN-DENTIN HYBRID ZONE PRODUCED BY THE MOISTENING OF ACID CONDITIONED DENTIN SURFACE (산 표면처리 후 상아질 표면의 습윤이 하이브리드층 형성에 관한 전자현미경적 연구)

  • The effect of moistening and air-drying of acid-conditioned dentin before priming on the formation of resin-dentin hybrid zone was investigated, Freshly extracted human molars were used and divided at random into 5 groups, Groups 1 - 3 consisted of specimens conditioned with 10 % phosphoric acid for 20 seconds; Group 1 served as a control in which the conditioned dentin was simply blot-dried with a damp facial tissue; Group 2 was air dried for 30 seconds ; Group 3 was air dried for 30 seconds and immediately remoistened for 10 seconds with air-water syringe. and then the specimen was blot-dried with a damp facial tissue. Groups 4-5 were not acid conditioned ; In group 4, the smear layer on the dentin was blot dried before primer placement; Group 5 was air dried only for 30 seconds, The acetone-based primer and bonding agent of All Bond 2 (Bisco. Inc., USA) and composite resin (Z-100, 3M Dental products, USA) were applied for acid conditioned dentin and non-conditioned dentin. The morphologic ultrastructure of resin-dentin hybrid zone was examined by the use of SEM and TEM. and the existence of inorganic material and analysis of Ca/P weight-percent ratio in the resin-dentin hybrid zone were revealed by the EDAX, The results were as follows : 1. In the moistened specimens from acid-conditioned groups, the resin penetrated about 3-$4{\mu}m$ into dentin and the denatured collagen smear layer was not present at the surface. The resin tag was formed to a thickeness of 3-$4{\mu}m$ at the upper part of dentinal tubule and compactively connected to each other by means of many lateral branching. 2. In the air-dried specimens from acid-conditioned groups, the resin penetrated about 2.0-$2.5\;{\mu}m$ into dentin and an upper thin black layer to a thickness of 30-35nm was identified between adhesive resin and demineralized collagen layer. The resin tag to have a diameter of $2.5{\mu}m$ was formed at the upper part of dentinal tubule. However the funnel shape of the tag was not notable compared to the moistened specimens. 3. In the remoistened specimens from acid conditioned groups, the resin penetrated about 2.0-$2.5{\mu}m$ into dentin and an upper black layer was not present. The resin tag at the upper part of dentinal tubule was formed less than $2{\mu}m$ and was weakly connected to each other by means of few lateral branching. 4. In the non-conditioned groups, the smear layer was formed to a thickness of $0.5{\mu}m$ at dentin surface. However, the resin-dentin hybrid zone was not identified by TEM. The evidence of resin penetration into intertubular and intratubular dentin did not show. 5. All the acid-conditioned groups showed that the detected calcium and phosphorus weight percent ratios at the $2{\mu}m$ upper portion from the resin-dentin interface into the resin were much higher than that at the $2{\mu}m$ lower portion from the resin-dentin interface to dentin. Study on the Characteristics of DNA Comet Assay for Irradiated Vegetables and Grains (방사선조사된 채소류 및 곡물류의 DNA Comet Assay 특성 연구)

  • The possibility of DNA comet assay as a rapid method for screening the irradiated vegetables and grains was evaluated. Vegetables (spring onion, garlic, and tomato) irradiated at $0{\sim}3$ kGy and grains (rice flour and black soybean) irradiated at $0{\sim}9$ kGy were used as samples. Optimum DNA comet assay conditions, such as elution, sedimentation of suspension, and lysis time of cell, were established. The optimum conditions for vegetables were 10 min for the elution time, 0 min for the sedimentation time, and 5 min for the lysis time. The optimum conditions for grains were 15 min for the elution time, 60 min for the sedimentation time, and 30 min for the lysis time. For the food application of DNA comet assay, it was possible to screen various food samples irradiated at the following doses: spring onion at 2 kGy, garlic at 3 kGy, tomato at 1 kGy, rice flour at 9 kGy, and black soybean at 3 kGy. Each sample showed different forms and sizes in DNA comet. Therefore, further studies on various methods using comet shape, concentration, or area in DNA comet assay are necessary.

The Optomotor Response of Killifish and Yellowtail (송사리와 방어의 시각운동반응)

  • The authors carried out an experiment to find the optomotor response of killifish, Orizias latipes(TEMMINCK et SCHLEGEL) and Yellowtail, Seriola quinqueradiata (TEMMINCK et SCHLEGEL) according to the colors of the netting pattern on the visual screen and the revolving velocities of the visual screen. The experimental water tank was made of 0.5 cm thick transparent acryl in the cylindrical shape (100R$\times$42H cm). The water level in the tank was maintained 30cm high from the bottom. The colors of the netting pattern (mesh size: 19.1cm, width of netting twine: 1.5cm, hanging ratio: 84%) on the three visual screens were black, red and green respectively. The revolving velocities of the visual screen were controlled by pulley, bevel gear and variable speed motor in three steps; slow (15.0cm/sec), middle (37.4cm/sec) and high (62.9cm/sec). The fish was put into the water tank before each experiment and released in it for 30 minutes in order to acclimatize itself to the tank. The visual screen was revolved for 4 minutes per each experiment, at first the fish was released for 1 minute, and then the behavior of the fish was observed for 3 minutes. In the course of clockwise and counter clockwise experiments, 10 minutes-pause was given for the rest. The behavior of the fish was observed by video system, and rounding number and swimming speed of the fish were analysed. The results obtained are as follows: (1) Optomotor response rate of Killifish and yellowtail were 95% and 94% respectively. (2) Response of the fish according to the colors of the netting pattern on the three visual screens was best in black, and second in red and third in green. (3) Response of the fish according to the revolving velocities of the visual screen was best in high speed, and second in middle speed and third in slow speed.

  • The digestive tract of the black sea bream, Acanthopagrus schlegeli is composed of esophagus, stomach, intestine, anus and four or five pyloric caeca. Pyloric caecum is a blind sac in shape and originated from pyloric portion of the stomach. Relative length of But (RLG), that is length of digestive tract to standard length, is 1.04 (n=10). Histological layer of the digestive tract is composed of serous membrane, muscular layer, undeveloped submucosal layer and mucosal layer. The mucosal folds of the esophagus are regular branched form, Esophageal muscularis mucosae is well-developed. Mucosal epithelial layer is composed of cuboidal or columnar epithelium and mucous secretory cell. Microvilli are absent in the free surface of mucosal epithelium. The mucosal folds of the stomach are regular unbranched form. The stomach has a well-developed muscular layer and muscularis mucosae. Microvilli are present in the free surface of mucosal surface epithelium. The fundic portion of the stomach have a well-developed gastric gland and more numerous secretory granules than the other parts. The mucosal folds of the pyloric caeca and the intestine are irregular branched form, Intestine is divided into the anterior, mid and posterior intestines with length of mucosal folds and histological features, Posterior intestine has a more developed striated border and goblet cells than the other parts. Mid intestine has a more abundant absorptive cells than the other parts in the intestine and pyloric caeca.

Sclerotinia Rot on Water Cress (Oenanthe javanica) Caused by Sclerotinia sclerotiorum (Sclerotinia sclerotiorum에 의한 미나리 균핵병)

  • A sclerotinia rot of Water cress (Oenanthe javanica) occurred in the commerical farmers field at Garye-myon, Uiryeong-gun, Gyeongnam Province, Korea, 2002. The typical symptoms appeared on leaves and stems. At first, the infected leaves or stems turned dark green later become watery soft rotted; white fluffy mycelia grew from the lesion, later formed black sclerotia. Sclerotia on the infected plants and PDA medium were globose to cylindrical or irregular in shape and 1.0~10.7 ${\times}$1.0~7.6 mm in size. Cup-shaped aphothecia with numerous asci were formed from sclerotinia and the size were 0.4~1.6 cm in diameter. Asci with 8 spores were cylindrical and 74~236 ${\times}$ 4.2~24.8 m in size. Ascospores of one cell were hyaline, ellipsoid to ovoid in shape, and 8.3~12.4 ${\times}$ 3.6~7.2 m in size. The optimum temperature for mycelial growth was $25^{\circ}C$ , and sclerotinia forma-tion was between 15~$20^{\circ}C$. On the basis of mycological characteristics and pathogenecity test to host plants, the fungus was identified as Sclerotinia sclerotiorum. This is the first report of Sclerotinia sclerotiorum caused sclerotinia rot on Oenanthe javanica caused by in Korea.

A basic study on the mixing bar interaction efficiency in shield TBM chamber (Shield TBM 챔버 내 mixing bar 교반 효율에 대한 기본연구)

  • This study is the basic study for improving the range of influence and potency of mixing bars in the chamber of Shield TBM. Currently, there are many studies on disk cutters, cutter bits and segments in the study of the domestic Shield TBM. However, studies that mix soil and rocks that come from the membrane during the Shield TBM excavation and scatter them with screw conveyors are not as good as those abroad. In this study, the existing Shield TBM Chamber was manufactured as a miniature and the experiment. Inside the chamber, different sizes (4 mm, 6 mm, 8 mm, 10 mm) and colors (black, white, red, and blue) were used to form layers. This experiment was carried out by different shapes and sizes of RPM and mixing bars. In addition, the difference between a miniature model and a reclining one was checked to determine the effect of the direction of gravity on the mixing efficiency. This was done in the same way for all other conditions other than differences in the direction of gravity. Through this experiment, we identified the orientation of the chamber model, the size and shape of the mixing bar inside, and the mixing effect and torque depending on RPM. A comparative review of the mixing effect and torque confirmed that the shape and size of the mixing bar affect the mixing of samples, and that the direction of gravity affects torque.

TYME: Interactive Typography for a poetic expression in Multimedia Environment (TYME: 멀티미디어 환경에서 시적 표현을 위한 인터랙티브 타이포그래피)

  • TYME is an interactive typography program based on the ephemeral nature of time. It is a text based poetic tool, built with Processing. This project is presented as a performance that utilizes a computer with a display screen, a projector, and speakers. As the user types, white characters appear, flow on the score on the screen and trigger jazz sounds, then characters disappear into black space in several seconds on the screen like smoke. Typography from this invented instrument is evocative and wistful, and allows the user to associate with ephemeral time. While typing the characters as though playing an instrument at intervals of time, the user can freeze the motion and print out or save otherwise fleeting moment with a button. I intend to contain both characteristics: the amorphous shape of smoke and the elusive attribute of smoke for the expression of intangible and ephemeral time. Every alphabetic shape is derived from the video dips that I shot of smoke. The resulting alphabetic images are then programmed using the Processing scripting language and which can then be typed on the screen with a keyboard. TYME could be a model as a project that reflects the unfixed quality of digital typography, and as a design approach for interactive expressive typography by scripting code. This project also represents the characteristics of typographic play, which can be realized in an computational environment like this model.

Characteristics and breeding of a new variety 'Sootari' in Pleurotus ostreatus (느타리버섯 신품종 '수타리'의 육성 및 특성)

  • 'Sootari', a new variety of oyster mushroom, was bred by mating with monokaryons isolated from 'Suhan' and 'Gonji-7ho'. The optimum temperature for the mycelial growth was $20{\sim}25^{\circ}C$ on PDA medium and those for the primordia formation and growth of fruiting body of 'Sootari' were $16{\sim}18^{\circ}C$ on sawdust substrate. In bottle cultivation, mycelial growth required about 25 days. In addition, primordia formation and growth of fruiting body required 4 days each. Regarding characteristics of the fruiting body, the shape and color of pileus were round type and black, respectively, and stipe color and shape were white and short and thin, respectively. The yield of fruiting bodies was $131.3{\pm}26.0g$ per 1,100 mL bottle, which was 2% higher than that of Gonji-7ho.

  • Black and White aerial photogrphs are much useful to obtain the information on the channel-bed fluctuation in the following aspects. 1. In the decision of river width, the linear regression formula between the value of aerial photograph interpretation and that of field surveying is Y=1.0+0.94X(the decision coefficient is $r^2=0.98$). Therefore, aerial photographs are proved effective for the measurement of river width. 2. Aerial photograph interpretation makes it possible to classify the plane channel and the deposits in river, and suggests the situation of the plane distribution of deposits, the size of channel and the course of channel formation. 3. The periodical channel situation can be figured out through the interpretation of aerial photographs pictured in different times. Also, the comparing and analyzing each interpretated information can be able to guess the course of the variation of channel influencing powerfully channel - bed fluctuation. 4. The microtopographic map of river can be made through the decision of river with, the interpretation of the plane shape of channel - bed and the analysis of variation of channel. On the basis of this map, the plane analysis of deposit is possible.

The Actual Wearing Conditions and Preferred Design of Tailored Jackets for Obese Women in Their 20s and 30s (20~30대 비만여성의 테일러드 재킷 착용실태 및 선호디자인)

  • This study provides basic data for the development of jacket patterns which reflect designs corresponding to the body shape features and preferences of obese women in their 20s and 30s by understanding problems they may encounter in wearing a ready-made jacket by researching the reality of ready-made tailored jacket wearing and their design preferences. A survey was conducted by sampling 82 obese women (over $25kg/m^2$ BMI) between the ages of 20 to 39. The details of the survey consists of general information, interest rate and importance rate on clothes, reality of wearing and purchasing jackets, measurement fits per each part, and preferred designs. SPSS 14.0 was used to analyze the data. The results showed that 76.8% of obese women wore their jackets less than 5 times a month. Those in the 20s wore the jackets more frequently than those in the 30s. The reasons for not wearing jackets frequently included uncomfortableness in activities and unavailability of the correct size. A total of 55% of the total preferred the fitting to be relatively smaller than the body size or tightly fitting. Although in the past, there was a tendency to wear a loose fit to cover the body shape, currently even obese body shapes avoided the loose fit as the tight fit is the trend. In terms of the satisfaction rate for the measurements per each part of jackets, there was a low satisfaction rate for most of the items. For the circumference items such as waist length and chest size, people responded that they were too small and for the length of items such as the length of jacket and sleeve length, people responded that they were too long. This was because the readymade clothes brands increase the length of items and circumference items in uniform in the same intervals when producing large sizes. Both those in the 20s and the 30s preferred a tight fit. In terms of preferred fabric, a wool/spandex mix was most popular and then cotton/spandex. This shows that they prefer those fabrics which allow an excellent activity while maintaining the exterior well. In addition, they preferred black color with no patterns because they wanted their body size to look reduced and did not want to receive any attention.
