Xanthomonas campestris pv. campestris (Xcc) is a plant pathogen of Brassica crops that causes black rot disease throughout the world. At present, 11 physiological races of Xcc (races 1-11) have been reported. The conventional method of using differential cultivars for Xcc race detection is not accurate and it is laborious and time-consuming. Therefore, the development of specific molecular markers has been used as a substitute tool because it offers an accurate and reliable result, particularly a quick diagnosis of Xcc races. Previously, our laboratory has successfully developed race-specific molecular markers for Xcc races 1-6. In this study, specific molecular markers to identify Xcc race 7 have been developed. In the course of study, whole genome sequences of several Xcc races, X. campestris pv. incanae, X. campestris pv. raphani, and X. campestris pv. vesicatoria were aligned to identify variable regions like sequence-characterized amplified regions and insertions and deletions specific to race 7. Primer pairs were designed targeting these regions and validated against 22 samples. The polymerase chain reaction analysis revealed that three primer pairs specifically amplified the DNA fragment corresponding to race 7. The obtained finding clearly demonstrates the efficiency of the newly developed markers in accurately detecting Xcc race 7 among the other races. These results indicated that the newly developed marker can successfully and rapidly detect Xcc race 7 from other races. This study represents the first report on the successful development of specific molecular markers for Xcc race 7.
In the survey from 1992 to 2000, twenty-eight parasitic diseases were observed in major apple producing areas in Korea. The predominant apple diseases were white rot(Botryosphaeria dothidea), Marssonina blotch(Marssonina mali), Valsa canker(Valsa ceratosperma), Alternaria leaf spot(Alternaria mali), and bitter rot(Collectotrichum gloeosporioides and C. acutatum). Apple scab that reappeared in 1990 after disappearance for 15 years was disappeared again since 1997. A viroid disease(caused by apple scar skin viroid) was newly found in this survey. The five diseases, fire blight(Erwinia amylovora), black rot(Botryosphaeria obtusa), scab(Cladosporium carpophilum), Monochaetia twig blight(Monochaetia sp.), and brown leaf spot(Hendersonia mali), which had once described in 1928 but no further reports on their occurrence, were not found in this survey. However, blossom blight(Monilinia mali), brown rot(Monilinia fructigena), and pink rot(Trichothecium roseum), which did not occur on apple after mid 1970s, were found in this survey.

Black rot caused by Xanthomonas campestris pv. campestris (Xcc) is one of the most serious diseases of crucifers world-wide. To establish the efficient screening method for resistant cabbage to Xcc, different inoculation methods, inoculation positions, growth stages of seedlings, and incubation temperatures after inoculation were investigated with the seven cabbage cultivars showing different resistance degrees to the pathogen. Clipping with mouse-tooth forceps was better inoculation method than piercing with 18 pins or cutting with scissors to distinguish the level of resistance and susceptibility. In inoculation using mouth-tooth forceps, clipping the edges of the leaves near veins is more effective than injuring the veins of the leaves directly. In addition, the inoculated plants kept at $22^{\circ}C$ showed more clear resistant and susceptible responses than those kept at 26 or $30^{\circ}C$. On the basis of the results, we suggest that an efficient screening method for resistance of cabbage cultivars to black rot is to clip the edges of the leaves near veins of the four-week-old seedlings with mouth-tooth forceps dipped in a suspension of Xcc at a concentration of $7{\times}10^7$ cfu/ml and incubate the inoculated plants in a growth room at $22^{\circ}C$ with 12-hr light a day.

A new bacterial disease of broccoli (Brassica oleracea var. italica) was observed on field-grown plants in Pyungchang during 2003 and 2004. Seedling infections first appeared as a blackening along the margins of the cotyledon. Cotyledon shriveled and dropped off. Infected seedlings were stunted and yellowed and eventually died. The disease was easily recognized by the presence of yellow, V-shaped, or U-shaped areas extending inward from margin of the leaf. As the disease progressed, the yellow lesions turned brown and the tissues died. Isolations made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Two bacterial strains were purified and used for further tests. Pathogenicity of strains was confirmed on 3-week-old crucifer (cabbage, Chinese cabbage, kale, radish and broccoli) plants cut by scissors with bacterial suspensions containing $10^8 cfu/ml$ of phosphate buffered saline. The Biolog and fatty acid analyses and 16S rDNA sequencing of two strains (SL4797 and SL4800) from broccoli black rot showed that they could be identified as X. campestris pv. campestris because of their high similarity to the tester strain (X. campestris pv. campestris NCPPB528) with a match probability of 100%. This is the first report of black rot of broccoli in Korea.
Rhizopus soft rot caused by Rhizopus stolonifer on June, 2008 occurred on Rubus crataegifolius Bunge in the box at Jinju City Wholesale Market of Agricultural Products at Gyeongnam province in Korea. The infected fruits were rapidly water-soaked, softened, black and eventually rotted. The symptoms occurred after infecting wounds at harvest time. The colony were white to light brown, formed much sporangiospores. The optimum temperature of the fungus on potato dextrose agar was $30^{\circ}C$. Sporangiophores were $12{\sim}25{\mu}m$ in width. Sporangia were globose or hemispheric, white at first and gradually to black, and $82{\sim}195{\mu}m$ in size. Columella were hemispheric, and $70{\sim}92{\mu}m$ in size. Sporangiospores were irregular round or oval, brownishblack and $9{\sim}21{\times}7{\sim}8{\mu}m$ in size. On the basis of symptom, mycological characteristics and pathogenicity of the fungus, the causal fungus was identified as Rhizopus stolonifer. This is the first report of Rhizopus soft rot by R. stolonifer on R. crataegifolius in Korea.

Sporangiophores were $12{\sim}26\;{\mu}m$ in width. Sporangia were globose or hemispheric, white at first and changed gradually to black, and $83{\sim}195\;{\mu}m$ in size. Columella were hemispheric, and $72{\sim}125\;{\mu}m$ in size. Sporangiospores were irregular round or oval, brownish-black and $9{\sim}20{\times}7{\sim}8\;{\mu}m$ in size. On the basis of symptom, mycological characteristics and pathogenicity to banana, the causal fungus was identified as Rhizopus stolonifer. This is the first report of Rhizopus soft rot by R. stolonifer on banana in Korea.
A Rhizopus soft rot caused by Rhizopus stolonifer occurred on Momordica charantia at Daesan-myon, Chang-won city, Gyeongnam province from 2003 to 2005. The disease usually started from wound on fruit with water-soaking lesions. The lesion rapidly expanded and softened. The fungal mycelia grew vigorously and sporangia, sporangiophores, and stolons were formed on surface of the fruit. Spoyangiophores were $15\~30{\mu}m$ in width. Sporangia were globose or hemispheric and $90\~180{\mu}m$ in size. The color of sporangia was white and cottony at first and turned to brownish black with many spores at maturity. Columella were hemispheric and $80\~150{\mu}m$ in size. Sporangiospores were irregular round or oval, brownish-black streaked and $7\~18{\times}6\~12{\mu}m$ in size. The optimum temperature for mycelial growth of the fungus on PDA was $25^{\circ}C$. On the basis of mycological characteristics and pathogenicity to host plants, the causal fungus was identified as Rhizopus stolonifer (Ehrenberg ex. Fr,) Lind. This is the first report of Rhizopus soft rot on M. charantia caused by R. stolonifer in Korea.
Resistance to various diseases of Korean native varieties were investigated in field. The results obtained are summarized as follows: 1) The varieties Ubangtchio and Osib-eubthio were moderately resistant to black shank, but Suantchio, Usultchio and Gwangtchio were slightly resistant. Dixie Bright 101, $H_2$ and Bright Yellow 4 were more resistant to black shank than any of the Korean native varieties. 2) Mokgitchio, Ubangtchio and Osib-eubtchio were moderately resistant to black root rot, but Useultchio Suantchio and Hoetchio were susceptible than Others. 3) Muktchio, Hoetchio and Ubangtchio were slightly resistant to wild fire. 4) Osib-ebthio was highly resistant to brown spot, but Ubangtchio, Hyangtchio and Mokgitchio were moderately resistant and Useultchio was susceptible to brown spot, but it was resistant than Bright ellow 4 or Bernhart 1000-1. 5) Hyangtchio was slightly resistant to CMV-Y. According to these results, except Virus disease, Ubangtchio was resistant to all of the above mentioned diseases. Osib-eubtchio, Hoetchio, Hyangtchio and Suantchio showed proferable disease resistant and these might be well utilized as breeding materials.
Occurrence of soft rots was observed on wasabi (Wasabia japonica Matsum) grown in Chuncheon and Pyengchang Kangwon province, Korea. The symptoms appeared on the wasabi root, which became mushy and black. This eventually resulted in wilting and death of the aboveground parts of the wasabi. The causal organism was isolated from the infected lesions and was identified as Erwinia carotovora subsp. carotovora based on the morphological, physiological and biochemical characteristics, and on the results of the Biolog program (Biolog Inc., U. S. A.). E. carotovora subsp. carotovora is the first described bacterium which causes bacterial soft rot on wasabi in Korea.
Pathogenic fungal species associated with apple anthracnose were studied with diseased fruits collected over two major apple growing areas from 1986 to 1988. Colletotrichum acutatum Simmonds, c. gloeosporioides(Penz.) Sacc. and Glomerella cingulata(Stonem.) Spauld. & Schrenk were identified based on their mycological characteristics. C. acutatum could be easily differentiated from C. gloeosporioides by producing their fusiform conidia and developing apricot∼salmon colony colors, non-forming an ascigerous stages. Growth rate of C. acutatum were slow in plate culture. The three species of anthracnose fungi tested caused apple rot. Symptoms produced by C. gloeosporioides were larger than those of the others. C. gloeosporioides and G. cingulata produced rot lesions on apples that appeared became brown and later black, but C. acutatum produced rot lesions appeared yellowish brown and later reddish brown. G. cingulata has the major species responsible for apple anthracnose, epidemics in Korea.
