Effect of Different Forage Source and Nutrient Levels in Diet on in vitro Goat Rumen Fermentation and Methane Production (조사료의 종류와 사료 내 영양소 수준이 흑염소의 반추위 in vitro 발효성상과 메탄생성에 미치는 영향)

  • The present study investigated the effect of forage sources and their inclusion levels in diet on the rumen fermentation of Korea traditional goat. Timothy and alfalfa were used as forage sources. Forages were mixed with concentrate diet in different ratios. The ratios of forage to concentrate diets were varied to 1:9, 5:5 and 9:1. The rumen fluid of goat was gathered from slaughter house. Dry matter digestibility was decreased and methane production was increased as forage levels in diet was increased. When forage sources, timothy and alfalfa, were compared, groups with timothy showed greater methane production than the groups of alfalfa. Molar ratio of produced acetate and valerate were increased when forage level in diet was increased. In the case of propionate and butyrate, they were decreased as elevated forage levels in diet. The result of this study provided a basic information for rumen fermentation of Korean traditional goat and these information could be applied in the development of nutritional and feeding strategy.

  • A field trial on selection for increased live weight in Black Bengal (BB) goat was conducted over two generations. Parents of both sexes were selected (mass selection) based on mature body weight criteria set against age. A parallel randomly mated line was maintained to compare the response (R) of selection at birth. 3, 6, 9 and 12 months of age in both generations. Lactation yield (LY), average per day yield (APDY) and lactation length (LL) were evaluated in the selected line in comparison to randombreds as a result of correlated response. Live weights were significantly improved at onward bir:th in first generation and only at birth in second generation. Improvements (%) in live weight at 3, 6. 9 and 12 months in first generation were 17.6, 18.4, 16.6, and 12.0 at birth in second generation. Significant correlated R were found in LY and APDY. Results suggest that there may be a positive relationship between live weight and lactational traits in BB goats of Bangladesh. It was also concluded that such a field scheme can be effectively used for genetic improvement of goats in subsistence farming, at least for short term gain.

Laparoscopic Transabdominal Transfer of Blastocysts in Korean Black Goats

  • As a part of the effort to improve post-transfer survival rate of embryos in Korean black goats, a technique for laparoscopic uterine transfer of blastocysts was carried out. A total of 26 transferrable embryos (morula to expanded blastocysts) were transferred to 13 recipient goats via transabdominal laparoscopic method. In consequence of our hormone protocol, 65% of the recipients (13/20) were found to have synchronized estrus. After confirmation of corpus luteum in each recipient goat, a Babcock laparoscopic forceps was inserted into the lower abdominal cavity to hold a uterine horn and fasten it near the peritoneum without causing injury. Then 7.5cm long 16G IV catheter was inserted directly into the uterine lumen through the abdominal wall. After removal of the stylet of the IV catheter, the embryo transfer tube (identical in size to the stylet and loaded with blastocysts) was inserted into the uterine lumen through the catheter to unload the embryos. Of the 13 estrus synchronized recipients, 9 were transferred blastocysts and 4 were transferred molurae (2 embryos in each recipient) in uterine ipsilateral to the ovary with corpus luteum. Four of the 9 recipients which blastocysts were transferred using this method has been confirmed pregnant (44.4% pregnancy rate).

Effects of Different Levels of Concentrate in the Diet on Physicochemical Traits of Korean Native Black Goat Meats

  • The effects of feeding of diets based on roughage supplemented with concentrate on the carcass and meat quality attributes of Korean black goats were investigated. The 40 male goats at 5 mon age were divided into four treatment groups; T1 (1.5%), T2 (2.0%), T3 (2.5%) and T4 (ad libitum) feeding of concentrate with ad libitum rice straw. Forty bucks at the end of the feeding were fasted for 12 h before slaughter and were then slaughtered. For carcass measurement, cold buck carcasses were examined after 24 h of chilling at $5^{\circ}C$. The carcasses were chilled in refrigerator at $2{\pm}2^{\circ}C$ for 24 h, then the longissimus dorsi muscle was removed from the carcasses. The samples were separately vacuum packaged, frozen and stored at $-20^{\circ}C$ for up to 1 mon, prior to physicochemical evaluations. Carcass yields were greatly affected by rate of concentrate feeding. T4 showed the heaviest live and carcass weights and the highest dressing and fat percentages (p<0.05). The moisture contents were lower in T4 compared with other treatments, while the crude protein and fat contents were higher (p<0.05). mono-unsaturated fatty acid in T4 was significantly higher than others, which showed the highest percentages of oleic acid (C18:1). For the sensory test, T4 was tenderer and produced better flavor scores than others. Increasing the level of concentrate in the diet resulted in an improvement in growth performance, and carcass and physicochemical characteristics of goat meats.

Amperometric Biosensor for Hydrogen Peroxide Determination Based on Black Goat Liver-Tissue and Ferrocene Mediation (흑염소 간-조직과 Ferrocene 매개체를 사용한 과산화수소정량 전류법 바이오센서)

  • The response characteristics of the bioelectrode developed by the co-immobilization of black goat liver tissue and ferrocene in a carbon paste matrix for the amperometric determination of hydrogen peroxide were evaluated. In the range of electrode potential examined ($-0.3{\sim}+0.0\;V$ vs. Ag/AgCl), the response time was relatively short (t95%=12 s) and it responded in the wide range of pH. The detection limit was 2.25${\times)10^{-6}M$ and a relative standard deviation of the measurements which were repeated 15 times using 1.0${\times}10^{-2 }$M hydrogen peroxide was 1.87%. The bioelectrode sensitivity decreased to 50% of the original value in 19 days of continuous use.

A morphological study of vomeronasal organ of Korean black goat (Capra aegagrus hircus) (한국흑염소 보습코기관의 형태학적 관찰)

  • The vomeronasal organ (VNO) plays an important role in reproduction and social activities in ruminants including goats. A morphological study on the structure of VNO and its epithelial cells was carried out in Korean black goats. Grossly, the VNO of Korean goats opens into mouth through incisive ducts. Microscopically, the epithelium of VNO consisted of medial sensory epithelium and lateral non-sensory epithelium. Several blood vessels and nerve bundles were observed in the lamina propria encased by vomeronasal cartilage. Immunohistochemical staining showed that protein gene product (PGP) 9.5 was immunostained in the receptor cells of the sensory epithelium and in some cells of the non-sensory epithelium. Galectin-3 was mainly observed in the supporting cells of sensory and non-sensory epithelium. Lectins including wheat germ agglutinin, Ulex europaeus agglutinin, Bandeiraea simplicifolia lectin Isolectin B4, Dolichos biflorus agglutinin and soybean agglutinin used in this study were bound in VNO sensory, non-sensory epithelia as well as in the lamina propria with varying intensity. Collectively, this is a first descriptive morphological study of VNO of Korean black goat with special reference to lectin histochemistry.

Effect of Feeding Systems on Feed Intake, Eating Behavior, Growth, Reproductive Performance and Parasitic Infestation of Black Bengal Goat

  • The experiment was conducted to find out the effect of 4 feeding systems on growth, feed intake, eating behavior, lactation performance, gestation period, post-partum heat period, conception rate and parasitic infestation of Black Bengal goat. Twenty four does of approximately 1 year of age were randomly selected for 4 treatment (feeding systems) groups having 6 replications in each. Treatments were stall feeding ($T_1$), tethering ($T_2$), restricted grazing ($T_3$) and grazing ($T_4$). $T_1$ group was housed continuously and adequate amounts of natural grass were supplied for ad libitum feeding. $T_2$ group was tethered for grazing natural grass from 8 a.m. to 4 p.m. being moved at one hour intervals. Goats of $T_3$ group were allowed grazing from 8 a.m. to 1 p.m. $T_4$ group was grazed from 8 a. m. to 4 p.m. Concentrate supplement was given at the rate of 150 g per day per goat for all of the treatment groups. Duration of experiment was 219 days. Daily live weight gain was significantly (p<0.05) higher in case of stall fed goats than that of others. DM intake also significantly (p<0.05) differed among the treatment groups and was 3.40 3.95, 3.76 and 4.05 per cent of their live weight for stall feeding, tethering, restricted grazing and grazing groups, respectively. Rate of rumination was significantly (p<0.05) higher in case of tethering group of goats than that of others. Birth weight of kids, milk yield, lactation period and post-partum heat period were significantly higher in case of stall fed goats than others. Tethering group showed significantly (p<0.05) higher litter size than others. Infestation rate of Fasciola was significantly (p<0.05) higher in the grazing group. In conclusion, it may be stated that among these feeding groups overall performance of stall fed goats was more satisfactory, and that the tethering group showed better performance than the others.

Effects of Feeding Herb Resources Powder on Meat Quality and Sensory Properties in Korean Native Black Goat (한방제재 급여가 재래 흑염소육의 육질 및 관능적 특성에 미치는 영향)

  • This study was carried out to investigate the effect of different herb resource powders on meat quality and sensory properties in Korean native black goat for 500 d. The experimental treatment was arranged with 24 heads (3 treatment/8 heads) by control group, T1 and T2 group respectively (Control, not added herb powders; T1, 1% added herb powders; T2, 2% added herb powders). Total weight gain for the Korean native black goat was the highest in T2 group feeding for 500 days, and daily gain tended to be similar to the total weight gain. The total feed intake were the highest as 519.20 kg in T2 group, although feed conversion showed 18.35 in the T2 group, which means it had the best feed efficiency compared to the other treatment groups. The carcass rate was higher in the T1 group (51.10%) than in the other groups (p<0.05). The cooking loss and drip loss of Korean native black goat was the highest as 34.72% and 3.83% in the control group (p<0.05). However, total cholesterol amounts in the treatment group were not significantly different from, although tended to be higher than, the control group (p>0.05). Also, the overall sensory evaluation of the treatment group revealed low scores, meaning more meat flavor than those of the control in tenderness, flavor, texture, and black goat off-flavor and overall evaluation (p<0.05). Total synthesis evaluation was higher for the treatment group (3.71, 3.90 point) than that of the control group (4.82 point) (p<0.05). The MUFA/SFA ratio of the treatment group was not significantly different from, although tended to be higher than, the control group

Comparative Quality Traits, Flavor Compounds, and Metabolite Profile of Korean Native Black Goat Meat

  • Although goat meat has several health benefits than other red meats but comsumers reluctant it due to its unpleasant flavor. This study aimed to investigate the odorant of goat meat as well as compare the quality traits of meat regarding sex status. The loin meats [non-castrated male (NCM), castrated male (CM), and female (FM)] were collected and stored at 4℃ in a laboratory refrigerator and analyzed on the 1st, 5th, and 8th consecutive days. The moisture content was the lowest and the protein content was highest in FM (p<0.05). Fat and ash content in NCM and FM were similar while lowest in CM. The CIE L* was significantly higher in NCM, but there were no significant differences of CIE a* and CIE b* within groups at initial day. The color intensity increased on the 5th storage day and decreased again after the 8th storage day, except in NCM. NCM displayed the highest thiobarbituric acid reactive substances value (p<0.05), whereas CM displayed a higher pH value than other groups throughout the storage period. Indole levels were the highest in NCM (0.031 mg/kg); however, skatole levels were not significant differences across all treatments (p>0.05). No significant difference was observed in the fatty acid composition between NCM and CM (p>0.05), whereas a significant difference was observed in FM (p<0.05). Most of the water-soluble metabolites showed significant differences between sexes. Overall, sex status effects on quality properties of meat and castration can improve the overall sensory acceptance by reducing goaty flavor of Korean native black goat meat.

Application of artificial insemination and pregnancy diagnosis kit for Korea native black goats (한국 재래흑염소의 계통별 인공수정과 임신진단 키트의 활용)

  • This study investigated the application of artificial insemination and pregnancy diagnosis kit for Korean native black goats. Semen was collected by electrical ejaculation, followed by semen analysis and artificial insemination in three goat strains (Dangjin, Jangsu and Tongyoung). Pregnancy was confirmed using a cow pregnancy test kit (IDEXX Rapid Visual Pregnancy Test kit) and ultrasound diagnosis. Analysis revealed that semen collected from male Korean native black goats by electrical ejaculation was about 1~1.5 ml in volume, $18{\sim}25{\times}10^8/ml$ concentration, and having > 97% motility. Furthermore, confirmation of pregnancy by pregnancy test kit and ultrasound diagnosis after artificial insemination were similar. In addition, the efficiency of pregnancy was 20~40% for all three strains: Tongyoung was the highest with 44%, followed by Dangjin (%), and Jangsu (20%). This study determines the artificial fertilization efficiency and the feasibility of using a cow pregnancy test kit for early pregnancy diagnosis in Korean native black goats. Although further research is required for validation, the results of the current study contribute to the breeding and improvement of Korean native black goat in research institutions as well as in general farms.
