Antioxidant Activity of Black Panax ginseng (흑삼의 항산화 활성)

  • This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.

Physicochemical Properties of Soybean Curd Residue Powder by Different Soybean and Drying Methods (콩의 종류와 건조 방법에 따른 비지 분말의 이화학적 특성)

  • This study compared the physicochemical properties of soybean curd residue and black soybean curd residue produced by hot air-drying and freeze-drying. Regardless of drying method, the crude protein, crude ash, crude fiber contents, pH, L, a, b color values and water soluble index were higher in soybean curd residue, whereas total polyphenol contents and antioxidant activity were higher in black soybean curd residue. Significant differences in water absorption index, oil absorption capacity and emulsion activity were observed between soybean curd residue and black soybean curd residue in freeze-drying. On the other hand, the emulsion stability was not significant difference in both hot-air drying and freeze-drying. The crude protein and crude fiber contents of soybean curd residue were not significant difference between hot-air drying and freeze-drying. Freeze-drying resulted in higher crude ash contents, pH, water absorption index, water soluble index, oil absorption capacity, emulsion activity and emulsion stability than hot-air drying. Hot-air drying have caused significantly higher water contents, water activity, total polyphenol contents and antioxidant activity in soybean curd residue than freeze-drying. In conclusion, soybean type and drying methods affect the physicochemical and quality characteristics of soybean curd residue, which could be important factors in the manufacture of processed foods.

Physicochemical Properties of Black Doraji (Platycodon grandiflorum) (흑도라지의 이화학적 품질특성)

  • The physicochemical properties of raw and black doraji were examined with the goal of increasing the utilization of doraji. Raw and Black doraji contained similar amounts of crude ash, crude fat, and crude protein, but raw doraji had higher level of fiber. Arginine was the major free amino acid in both types of doraji. Free sugar and organic acid contents of doraji increased after steaming heat treatment. Potassium was a prominent mineral in both raw and black doraji, constituting 85% of total minerals. Black doraji contained almost twice as much crude saponin as did raw doraji. Black doraji showed lower values of L (lightness), a (redness), and b (yellowness). The hardness and chewiness of raw doraji were higher than those of black doraji. Black dorai showed a higher amount and extraction rate of saponin, even though nutrient value of doraji slightly decreased after steaming heat treatment. Therefore, black doraji can be a valuable ingredient in functional foods.

Antioxidant and antimicrobial activities of black Doraji (Platycodon grandiflorum) (흑도라지의 항산화 및 항균 활성)

  • This study was conducted to investigate the antioxidant and antimicrobial activities of the hot-water and methanol extracts of raw and black Doraji to increase its utilization. In order to prepare the black Doraji, it was steamed for 15 days at a temperature of $60^{\circ}C$ and then it was dried at a temperature of $30^{\circ}C$ for 3 hr. The methanol extract from the black Doraji (BM) contained the highest levels of total polyphenols among the extracts, and the total polyphenol content of the extract from a black Doraji was higher than that of the extract from a raw Doraji. The total flavonoid contents of the hot-water extract from a black Doraji (BW) was the highest (7.94 mgQE/g) among the samples. The DPPH and ABTS radical scavenging activities increased according to the increase in the concentrations of the Doraji extracts. The BM has the highest radical scavenging activity among the extracts. Each extract showed a slight difference in the antibacterial activity according to the tested strains. The Black Doraji showed a higher antimicrobial activity compared to the raw Doraji. The hot-water extracts demonstrated higher activities than the methanol extracts, and the BW revealed the strongest activity. In this study, the black Doraji showed more effect of the antioxidant and anti-microbial activities than the raw Doraji. These results will provide fundamental data for improving the sitological value and the black Doraji can be used as a valuable resource for the development of nutraceutical foods.

  • Heterocyclic aromatic amines (HCAs) are established genotoxic human cancer risks, that display their activity also in a number of animal models and in vitro. They are formed during the trying or broiling of creatinine-containing foods, mainly fish or meat. We established that mixing soy protein with ground meat blocks the formation of HCAs. we also observed that coating the surface of meat with polyphenols green or from black tea gives a dose - related reduction of the formation of HCAs.(omitted)

The Diversity of Reproductive and Foraging Behaviors on Breeding Season of Eurasian Oystercatcher (Haematopus ostralegus) (검은머리물떼새의 번식행동 및 번식기 섭식행동의 다양성)

  • This study was conducted in three sites, Si-Hwa Lake, Dongman and Seoman island and Janguyeop island, from march, 1999 to september, 2002. The behaviors of pre-breeding season, territorial behaviors, reproductive ecology, foraging sites and behaviors, and the competition of reproduction and foods between intraspecific or interspecific of Eurasian Oystercatcher (Haematopus ostralegus) were observed in each studying sites. The breeding of Eurasian Oystercatcher started on the middle of April in Si-Hwa Lake and on the middle of May in Dongman and Seoman island and Janguyeop island. For intension of pair bond on pre-breeding season, Eurasian Oystercatcher foraged with pair and behaved male-female chasing flight behavior. The pair foraged with male and female before copulation. If other pairs and individuals approached in feeding site of pair, this pair attacked them with piping calling and intruder chasing flight. If continuos serial behaviors were not observed, the discrimination of male-female chasing flight and intruder chasing flight was difficult. Territorial behaviors classified four types; butterfly flight, calling behavior, chasing behavior, fight behavior. The important foraging sites in Si-Hwa Lake are the land place in Daeboo island, tidal flat of Bangameori, tidal flat a front of a stationary net for catching fishes and tidal flat a front of a view station for bird watching. Eurasian Oystercatcher foraged at tidal flat on low water of the tide and foraged at feeding sites near island on flood tide in Dongman and Seoman island. Eurasian Oystercater in Janguyeop island usually foraged feeding sites near island, because water level was not different between low water of the tide and flood tide. Eurasian Oystercatcher competed on foods of intraspecific and interspecific. They chased for taking foods by force in feeding sites and drove out intruders in feeding sites. The foods interspecific competition happened with Black-tailed Gull (Larus crassirostris). Eurasian Oystercatcher was robbed of foods and attacked by Black-tailed Gull. The individual of food competition with Black-tailed Gull was low foods intake rate comparison with other feeding sites and this individual flied out other feeding sites.

Antioxidant Effect of Chungkukjang Supplementation against Memory Impairment induced by Scopolamine in Mice (Scopolamine으로 유도된 기억 손상 마우스에서 청국장 식이의 항산화 효과)

  • In this study, the antioxidant effect of Chungkukjang supplementation against memory impairment and oxidative stress in scopolamine (2 mg/kg i.p)-injected mice was investigated. The experimental animals were divided into five groups and fed experimental diets for 12 weeks; normal diet group (C), scopolamine + normal diet group (S), scopolamine + 63.0% soybean Chungkukjang supplementation group (SS), scopolamine + 45.0% Yakkong Chungkukjang supplementation group (SY), and scopolamine + 50.0% black foods such as black rice, black sesame seeds, and sea tangle added Yakkong Chungkukjang group (SYB). For the results of food intake, body weight gain, and brain weights, levels of scopolamine-injected groups were lower than the levels of the control group. The reduced brain weight of the scopolamine-injected group (S) was regulated to control level by supplementation of three types Chungkukjang. In the oxidative stress indicator, nitric oxide and malondialdehyde levels in serum of scopolamine-injected mice were higher than those of other groups. However, supplementation of soybeans, Yakkong and black foods added Yakkong Chungkukjang was proven to regulate them. Antioxidant enzyme activities such as superoxide dismutase (SOD) and glutathione-S-transferase (GST) in serum showed no significant differences among the groups. The reduced levels of vitamin A and vitamin E in serum and brain tissue of scopolamine-injected mice were controlled by supplementation of three types of Chungkukjang. Total antioxidant capacity (TAC) of scopolamine-injected group was lower than those of other groups. However, TAC was significantly elevated by Chunggukjang supplementation. Therefore, antioxidative effects of soybeans, Yakkong, and black foods added Yakkong Chungkukjang supplementations against oxidative stress in scopolamine-injected in mice could expected.

A Study on the Intake Patterns of Health Food of the Elderly Aged over 60 Years in the Chuneheon Area (춘천지역 60세 이상 노년층의 건강식품 섭취실태 조사)

  • This study was carried out to investigate the intake patterns of health food according to the kind of disease of the elderly aged over 60 years living in the Chuncheon area. This study was conducted by using a questionnaire from December 1999 to the March 2000. The data was analyzed using a SPSS program. The results were summarized as follows : The poorer the self- perceived health condition, the lower the level of activity of daily living(ADL) and instrumental activities of daily living(IADL) were the higher. the frequency score of health food intake. The incidence of disease, such as arthritis(44.7%), hypertension(18.8%), digestive diseases(17.1%), diabetes(10.5%) were found to be higher. than other chronic diseases among the elderly. The frequency score of health food intake was not significantly different according to self-perceived health status, disease status, or the level of ADL and IADl.. The must important source of information for health food intake was the recommendation of friends and relatives(68.7%). Twenty eight point two percent of the elderly considered health foods to be effective, however 8.3% of the elderly considered them to be ineffective. The elderly with chronic diseases had taken health floods in the descending order of neuralgia-arthritis, hypertension, digestive disease, diabetes. The elderly with digestive diseases, diabetes, renal disease, neuralgia- arthritis and respiratory disease were inclined to habitually take health floods once or twice per year. Intake of Korean traditional folk food was as high as intake of registered health food and Chinese tonic medicine. The most frequently taken health food was tonic medicine, which is fo11owed by pumpkin, ginseng products, herbal tea formula with black goat, deer antler and Kye-so-ju. Intakes of health food such as tonic medicine, blood of the deer, royal jelly, aloe were found to be higher among the elderly with chronic diseases such as diabetes, hypertension, and digestive disease. However, intakes of ginseng products, deer antler, pumpkin, herbal tea formula with black goat and Kye-so-ju were found to be higher among the healthy elderly. The elderly with neuralgia-arthritis had taken sixteen kinds of health foods. From these results, a wide consumer education program should be developed to convince people of the importance of well-balanced diet and to choose proper health foods according to the elderly's health conditions. Also, comprehensive and scientific research into Korean traditional folk foods are needed for the correct use.

Determination of Tocopherol and Tocotrienol Contents in Rice Cooked with Various Cereals (밥의 종류에 따른 토코페롤 및 토코트리에놀 함량 분석 -연구노트-)

  • The objective of this study was to characterize and determine contents of vitamin E isomers (alpha-, beta-, gamma-, delta-tocopherols and tocotrienols) in different steamed rice dishes, with or without other grains. Five different rice dishes were evaluated for the vitamin E nutritional value as major Korean staple foods. They were plain steamed rice (SR) and steamed rice mixed with barley (SRBa), red bean (SRRB), black bean (SRBB), or multi-grains (SRMG) containing a mixture of black rice, barley, red beans, and black beans. Vitamin E isomers were extracted from five grams of freeze-dried samples with hexane after saponification. An analytical method, using a normal-phase HPLC with a UV detector, was developed and used to determine the amount of each vitamin E component. The results showed that SR contained three vitamin E isomers (alpha-tocopherol, alpha-tocotrienol, and gamma-tocotrienol). Alpha-tocopherol and gamma-tocotrienol were de-tected from all samples while only SRBB contained beta-tocopherol and beta-tocotrienol. SRMG showed the highest (3.9$\mu$g/g dry wt) and SRRB showed the lowest alpha-tocopherol (1.3$\mu$g/g dry wt) contents. SRBB contained about 5 to 16 times more gamma-tocopherol (19.7$\mu$g/g dry wt) than othe.5. These results suggested that adding black bean or multi-grains can dramatically improve the vitamin E nutritional values compared to the plain steamed rice (SR). Information obtained from this study can be directly related to the amount of vitamin E intake and can be used to balance the diet for Koreans.

Antioxidative and Fibrinolytic Activity of Extracts from Soybean and Chungkukjang(Fermented Soybeans) Prepared from a Black Soybean Cultivar (검은콩의 품종에 따른 콩과 청국장 추출물의 항산화능 및 혈전용해능)

  • We sought to develop natural preservatives or functional health foods from black soybeans (Yakkong, Seoritae and Chungkukjang). These materials were extracted with distilled water or 70% (v/v) ethanol, and the extracts sanalyzed for polyphenol content and tested for antioxidative and fibrilytic activities. The polyphenol contents of water extracts from Yakkong and Yakkong Chungkukjang was 316.23 mg/100 g, 896.01 mg/100 g, respectively, whereas those from Seoritae and Seoritae Chungkukjang were 304.28 mg/100 g and 875.23 mg/100 g. The polyphenol contents of water extracts from Chungkukjang were 2.8-fold higher than those from black soybean. The electron-donating abilities (EDAs) and superoxide dismutase (SOD)-like activities of water extracts from black soybean Chungkukjang were higher than those of water extracts from black soybean, but both extracts had fibrinolytic activity, the highest activity was present in water extracts of Yakkong Chungkukjang. The polyphenol contents, and antioxidative and fibrinolytic activities of extracts from black soybean Chungkukjang were higher than those of other black soybean extracts from black soybeans. These results indicate that black soybean Chungkukjang can be used for development of a health food or as a natural antioxidant.
