홍삼 및 생약재의 가공방법에 따른 항산화활성 특성 및 향미 효과 비교 (Comparison of Antioxidant Activity and Flavor Effect According to Processing Method of Red ginseng and Herbal Medicine)

  • 본 연구에서는 인삼 및 생약류의 전통적 건조 가공방법 대비 증숙 가열건조 과정을 거쳐 홍삼화로 기능성 성분 함량 및 활성 증대로 홍삼 및 생약류에 다량 함유된 polyphenols 및 flavonoids 등 기능성 물질이 항산화활성 특성에 미치는 영향 및 향미 효과를 비교하였다. 실험 결과에 의하면, 사전 열처리 과정을 추가하는 것에 의해 DPPH 라디컬 소거능, polyphenols 및 flavonoids 등 항산화성이 현저하게 증가되었다. 또한 수삼을 증숙 가열건조 홍삼 제조과정 중에 amino-carbonyl 갈색화 반응이 촉진되어 갈색도가 증가되고 구수한 향미를 갖게 되었다. 그러나 홍삼에 함유된 주효 약효성분인 ginsenosides의 함량은 쓰고 텁텁한 맛의 주된 요인이 되었다. 또한 홍삼의 로스팅 처리 후 분말 제조로 쓰고 텁텁한 맛이 상당히 개선되었으나 한편 한방에서 십전대보탕 및 사물탕 등 자양강장 처방에 주요 약재로 사용되는 당귀, 천궁, 작약은 생약 특이의 향미가 매우 강하고 쓰고 텁텁한 맛이 매우 강하여 즉석 추출용 재료로 사용하기 위해서는 제조과정에서 증숙가열 제조방법이 필요한 것으로 검토되었다.

Monosodium Glutamate의 맛표현 용어와 기본맛 성분과의 상호작용에 관한 연구 (Studies on the Taste Describing Terms of Monosodium Glutamate and the Interactions Between MSG and Other Basic Taste Substances)

  • MSG의 맛 표현용어를 설문조사를 통하여 수집분석하고 관능검사를 통한 MSG의 감응역가를 다른 기본 맛의 역가와 비교하였으며, MSG가 다른 기본 맛 성분에 첨가되었을때 일어나는 맛의 강도변화를 관능적 방법으로 측정하였다. 96명의 설문 응답자로부터 수집분석한 자료에 의하면 MSG 자체의 맛은 느끼한 맛(58%)과 역겨운 맛(24%)으로 주로 표현되었으나 음식에 첨가됨으로써 기대 되는 맛은 감칠 맛(79%)으로 주로 표현(表現)되었다. 관능검사자들이 MSG 수용액을 직접 맛보고 표현한 용어로는 최소 감각농도 부근의 낮은 농도에서는 느끼한 맛, 쓴맛, 단맛 등이 감지되었으나 최소 감미농도 이상에서는 느끼한 맛과 짠맛이 주로 표시되었다. MSG 수용액의 최소 감각농도는 0.002mole 수준이었으며 최소 감미농도는 0.006mole 수준이었다. MSG와 기본 맛 성분(成分)과의 상호작용을 보면 MSG가 식염용액에 첨가되면 짠맛의 상승작용을 나타내었다. 그러나 MSG가 설탕용액에 첨가되면 첨가된 MSG 농도가 낮은 때(0.01M)에는 다소 단맛의 상승효과를 나타내었으나 MSG 농도가 높아지면 단맛의 억제효과를 나타내었다. MSG를 구연산용액에 첨가했을 때는 전반적으로 신맛의 억제효과가 나타났다.

랫드를 이용한 여주 추출물의 안전성 평가 (Safety Evaluation of Ethanol Extract from Unripe Fruit of Bitter Melon (Momordica Charantia L.) in Sprague-Dawley Rats)

  • 본 연구에서는 항당뇨에 유효성이 있는 천연물질인 여주 추출물을 이용하여 여주에 포함된 독성물질인 vicine의 분석과 경구투여에 따른 독성을 조사하기 위하여 단회 경구투여 독성시험과 13주 반복 경구투여 독성시험을 진행하였다. 본 시료는 여주 미숙과 열매추출물로 vicine의 분석 결과 존재하지 않거나 극미량일 것으로 판단된다. 단회 경구투여 독성 시험과 시험물질에 의한 이상증상과 사망동물은 발생하지 않아 여주 추출물의 ALD는 암수 모두 5,000 mg/kg/d 이상으로 판단된다. 13주 반복 경구투여 독성시험의 투여용량을 결정하기 위하여 0, 1,250, 2,500 및 5,000 mg/kg/d의 투여용량으로 2주 반복투여 용량결정시험을 실시한 결과, 모든 시험군에서 시험물질에 의한 이상증상 및 독성 변화가 관찰되지 않아 동일한 용량으로 13주 반복 경구투여 독성시험을 진행하였다. 실험기간 동안 사망률, 일반증상, 체중 변화, 사료섭취량, 안검사, 요검사, 혈액학적 검사, 혈액응고시간 검사, 혈액생화학적 검사, 부검소견, 장기중량 및 조직병리학적 소견을 관찰한 결과, 시험물질에 의한 전신적인 독성학적 변화는 관찰되지 않았다. 따라서 여주 추출물의 NOAEL은 5,000 mg/kg/d로 판단되었고, 표적장기는 관찰되지 않았다. 본 연구의 결과로 볼 때 여주 추출물은 투여 가능한 최대 용량에서도 독성이 나타나지 않는 안전한 천연물로 확인하였고, 본 시험 결과를 바탕으로 ADI는 3,000 mg/man으로 판단된다. 따라서 기능성 식품으로서의 개발 가능성을 확인하였다.

  • Impulse of ki refers to palpitation around the navel, it is mostly caused by impairment of yang due to erroneous sweating, deficiency of the spleen yang. Behcet's syndrome caused by prolonged damp-heat and pathogenic germs. Its symptoms are characterized by conjunctival congestion, blue canthus and ulceration of the oral cavity, restlessness all the time, etc. It is advisable to follow the therapeutic principles of clearing away heat and drying damp, detoxicating and sterilizing. Lily disease is one of the emotional disease. It caused by yin deficiency of the heart and lung, found in mental depression or convalescence after a critical disease. Its symptoms are characterized by restlessness, reticence, insomnia, failure to walk, poor appetite, a subjective hot or cold, bitter taste and dark urine. It is advisable to follow the therapeutic principle of nourishing yin to clear away heat. Cholera refers to the disease marked by sudden severe vomiting and diarrhea, and colic of the heart and the abdomen. It is mostly caused by deficiency of the ki in the middle-energizer, emotional stress, etc. It is characterized by such symptoms as sudden severe vomiting and diarrhea, restlessness, etc. It is advisable to follow the therapeutic principles of warming yang and removing toxic substances, and strengthening the spleen and stomach. Heat invasion of blood chamber due to affection of the exterior pathogens, pathogenic heat invading the blood in deficiency and fighting with the blood. Its symptoms are marked by fever, feeling of the fullness, etc. There are deficiency of blood, hot blood and stasis of blood. The different therapeutic treatment should be taken according to different types thereof.

Studies on Anti-cancerous and Anti-malarial Substances from Simaroubaceae Plants

  • Cancer is a general term subjected to a series of malignant tumor diseases which may affect many different parts of the human body. These cancer diseases are characterized by a rapid and uncontrolled formation of abnormal cells in the body. Cancer chemotherapeutic agents can often provide the prolongation of life and occasionally cures. To date many kinds of compounds have been obtained from plants kingdom as anti-neoplastic and anti-cancerous agents. However, there is no special type of compounds for cancer therapy. In our laboratory, anti-tumor and cytotoxic screenings on higher plants collected in Japan, China, Korea, Southeast Asia and South America have been done by using Sarcoma 180 ascites in mice, P388 lymphocytic leukemia in mice, Chinese hamster lung V-79 cells, P388 cells and nasopharynx carcinoma (KB) cells. The family, Simaroubaceae consists of about 20 genera and 120 species, mainly shrubs and trees, distributed in tropical and subtropical country. Simaroubaceae is classified as RUTALES, together with Rutaceae, Burseraceae, Meliaceae, Malpighiaceae and Polygalaceae. The members differ from the Rutaceae in not containing oil glands. Bitter principles are a characteristic of the family, Simaroubaceae. The genera include Quassia (Simarouba) (40 spp.), Picrasma (Aeschrion) (6 spp.), Brucea (10 spp.), Soulamea (10 spp.), Ailanthus (10 spp.) and Perriera (1 spp.) etc.. Surinam quassia derived from Quassia amara growing in Guianas, north Brazil and Venezuela is used in traditional medicines for stomachic, anti-amoebic, anti-malarial and anti-anaemic properties. Also, various parts of a number of plants of the family Simaroubaceae have been used in traditional medicine for the treatment of a variety oi diseases including cancer, amoebic, dysentery and malaria. Then, the research has established that it is the quassinoid content of these plants that is responsible for above activities. In this meeting, I will present on anti-tumor and anti-malarial activities and their active principles of Simaroubaceae plants, Eurycoma longifolia, Ailanthus vilmoriniana, Simaba cedron and Brucea mullis which have been studied in our laboratory.

맥주 공장 부산물 효모의 최적 자가 소화 조건 결정 (Optimization for Autolysis of Brewers Yeast Slurry)

  • 맥주 공장 부산물 효모의 용도를 다양화하기 위하여 효모 슬러리를 자가 소화시키고 상등액을 농축하여 조미료로 이용할 수 있는 효모추출물을 제조하였다. 맥주 효모의 세포벽에 부착되어 있는 호프 쓴맛 성분을 제거하기 위하여 효모 슬러리에 20%(w/v) NaOH 용액을 가하여 pH 9.8로 맞추고, 약 5분간 방치한 후 즉시 원심분리하면서 물로 세척하였다. 세척 후 효모 쓴맛 정도는 24.1 bitterness unit(BU)로서 threshold value 25.0 BU 미만이었다. 세척한 효모를 자가 소화시킨 후 수율[(수득한 효모추출물의 고형분 무게)/(투입 효모슬러리의 고형분 무게)${\times}100$]을 조사한 결과, 효모슬러리 농도를 $9{\sim}10%(w/w)$로 하고, 온도 $53^{\circ}C$, pH 6.8에서 20시간 이상 자가 소화시키면 34%(w/w)이상, 최대 38%(w/w)까지의 수율로 효모 추출물을 얻을 수 있었다.

PC12 cell에서 tert-butyl hydroperoxide로 유도된 산화적 손상에 대한 치자의 신경보호효과 (Neuroprotective Effect of Gardeniae Fructus against Oxidative Damage Induced by tert-Butyl Hydroperoxide in PC12 Cells)

  • Objective : Gardeniae Fructus (GF) is the ripe fruit of Gardenia jasminoides Ellisa with a bitter taste and cold properties. Ingredient compounds including geniposide are known to have anti-inflammatory, antioxidant, and neuroprotective effects. The purpose of this study was to investigate the neuroprotective effect of GF on tBHP-induced PC12 cells. Methods : Cell viability was measured by the MTT assay, and apoptosis was determined by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The expression level of each protein was monitored by Western blot analysis, and reactive oxygen species (ROS) were analyzed using DCFH-DA. Results : In PC12 cells, tBHP induced cell death through apoptosis with caspase activation and PARP inactivation. Cells treated with tBHP showed an increase in intracellular ROS and depletion of GSH. Pretreatment with GF prevented tBHP-induced apoptosis, reduced ROS, and increased GSH. GF also maintained increased Nrf2 expression in the presence of tBHP. Phosphorylation of JNK and p38 MAPK was increased by tBHP, whereas phosphorylation of ERK was decreased. GF restored changes in ERK and p38 phosphorylation, but not JNK phosphorylation. Conclusion : These results indicate that GF has neuroprotective effects through anti-apoptotic and antioxidant effects mediated by regulation of Nrf2 expression and phosphorylation of ERK and p38. It also demonstrates the potential use of GF as a source of antioxidant and neuroprotective substances.

염전의 함수로 제조한 천일식제조소금의 물리화학적 특성 (The Physical and Chemical Properties of Salt Manufactured by New Process with Brine Produced in Korean Salt-farms)

  • 천일염은 자연 환경에서 제조되기 때문에 불용분 사분과 같은 이물이 함유될 가능성이 있다. 본 연구는 여과와 증발을 통하여 소금의 불순물 등 이물질을 효율적으로 제거하면서도 미네랄이 풍부한 소금을 생산하기 위해 수행되었다. 함수를 여과한 후 $90^{\circ}C$에서 회전식진공농축기를 이용하여 소금결정을 제조한 후에 탈수하였다. 이렇게 제조된 소금을 천일식제조소금이라 명명하였다. 소금의 수율은 물의 증발량에 의해 결정되는데, 증발량이 40%, 50%, 60%일 때 각각 40S, 50S, 60S로 표기하였다. 천일식제조소금의 수율은 40S, 50S, 60S가 각각 7.22%, 10.79%, 15.06%로 일반 천일염의 제조 수율인 4~8%보다 유사하거나 높았다. 천일식제조소금의 염도는 40S, 50S, 60S가 각각 90.38%, 91.16%, 68.16%를 나타냈다. 관능검사 결과 전체기호도가 가장 높은 소금은 40S이며, 쓴맛이 강함 소금은 60S를 나타냈다. Na, Mg, K, Ca의 미네랄 함량은 40S와 50S가 Mg 함량이 낮다는 것을 제외하면 천일염과 유사하였다. 불용분 함량은 천일염이 0.023%로 천일식제조소금보다 높고, 사분분석 함량은 천일염과 40S, 50S는 유의차가 없었다. 용해속도(g of salt/100 mL $H_2O$/sec)는 40S, 50S, 60S가 각각 0.69, 0.70, 0.69로 천일염의 용해속도(0.47)보다 높았고, 상대습도 70%, 저장온도 $20^{\circ}C$에서의 수분 재흡수 속도는 천일식제조소금 40S, 50S가 정제염보다는 빠르지만 천일염보다는 약 3~5배 낮은 결과를 보여주었다. 본 연구에서 개발한 천일식제조소금은 미네랄, 불순물 함량, 용해속도 및 수분 재흡수 등의 물리화학적 특성이 천일염에 비해 우수한 것으로 판단된다.

성동구내 어린이집 원아 부모의 소아 한방치료에 대한 인식 및 이용에 관한 조사 연구 (A Survey on Parent's Recognition and Utilization Patterns of Oriental Medical Care of Preschool Students in Seong-Dong district)

  • Objectives: The purpose of this study is to examine the parental recognition and utilizing patterns of oriental medical care in children who live in Seong-Dong district. Methods 802 day-care center children's parents participated in the survey on recognition and utilizing patterns of oriental medical care, and 702 of the surveys were analyzed. Results: 1. 38.33% has received previous oriental medical treatments and most were received at the children's age under 12-24 months. Majority of the treatments took place in the clinical settings. By recommendation, some also received their treatment at the medical centers. One of the reasons why they receive oriental medical treatments was that they prefer to balance their treatment with the orental medical treatments along with the western's. 2. 53.85% of those who received oriental medical treatments were satisfied with their treatments. The effective treatment results were the major reason for the satisfaction. 3. 49.56% of those who had oriental medical treatments chose specific oriental medical facilities for the children's ailment. Most specific oriental medical facilities were local oriental medical clinics. Reputation fame were the main reason in choosing the clinics. 4. In 2009, 55.26% had received care in the specific oriental medical clinics for the diseases, such as respiratory symptoms, systemic symptoms, gastrointestinal symptoms, skin problems, neurological and psychiatric disease for infants, and respiratory symptoms, gastrointestinal symptoms, systemic symptoms, skin problems, neurological and psychiatric symptoms, urinary symptoms and musculoskeletal symptoms for toddlers. 5. The average duration of herbal treatment were 7-14 days, preferably 4-6 days, and preferred number of medical visits were once a week on Saturdays and preferred day for night cares were on Friday nights. Average treatment fee per treatment was less than 10,000 won which coincided with the preferred treatment fee per visit. 6. The very first medical institutions chosen by parents once their children are ill are the following; Western medical institutions, western medical clinics, pharmacies, oriental medical clinics, health centers, and lastly, oriental medical institutions. The major concerns in receiving oriental medical cares include pesticides in herbs and hazardous substances in heavy metals. Preferred form of oriental medicine was decoction, the negative feedback of oriental medicine was mostly due to the bitter taste of the oriental herbal decoctions. Preferred frequency of intake was twice a day with less than 10cc~20cc per single intake. Conclusions: Taken all together, we conclude that the growing recognition of Oriental Pediatric medicine is needed among parents.
