Chemical Composition and Nutritional Evaluation of Variously Treated Defatted Rice Polishing for Broiler Feeding

  • The study was conducted to improve the nutritive value of defatted rice polishing (DRP). DRP was treated with various concentrations of HCl, NaOH, $H_2O_2$ and Kemzyme-H $F^{(R)} and the effect on its chemical composition and nutritive value in broiler chicks was observed. The treatments levels of 0.4 N HCl, 0.2 N NaOH and 6% $H_2O_2$ were selected from many concentrations of HCl, NaOH and $H_2O_2$ tried earlier on DRP. The selection was made on the basis of release of nutrients from DRP. The Kemzyme-H $F^{(R)} was used at rate of 0.1% of DRP. The selected concentrations of HCl, NaOH, and $H_2O_2$ were then used for treatment of DRP that was used in biological experiments. Two hundred and forty, day-old Hubbard male broiler chicks (38-40 g) were randomly divided into 48 experimental units with five chicks each. Each chemically treated DRP was incorporated into broiler diets at 10, 20 or 30% levels replacing yellow corn from the control feed and thus sixteen experimental feeds were prepared. These feeds were randomly assigned to 48 experimental units such that there were three replicates of chicks on each diet. The results of the study suggest that DRP can be effectively used in broiler diets at 20% level. The best weight gain and feed conversion ratio were observed with diet containing 20% level of DRP treated with 6% $H_2O_2$. The diets containing 30% levels of treated DRP were uneconomical, as excess use of oil was required to compensate the energy needs of the birds.

매생이와 생강나무잎 에탄올 추출물이 첨가된 통밀빵을 섭취한 랫트의 혈당과 지질수준에 미치는 영향 (Effect of Diet Containing Whole Wheat Bread with Capsosiphon fulvescens and Lindera obtusiloba Ethanol Extracts on Plasma Glucose and Lipid Levels in Rats)

  • 이 연구는 일반식빵, 통밀빵, 그리고 생강나무 잎 추출물, 매생이 추출물, 매생이 냉동 건조물을 첨가한 통밀빵을 제조하고, 4중 동안 랫트에게 공급한 뒤 혈당과 혈중지방질 관련 지표에 미치는 영향을 조사하였다. 시험기간 동안 모든 통밀빵섭취 그룹에서의 체중증가는 일반식빵섭취 그룹과 비교하여 유의적으로 낮게 나타났으며, 음수, 식이 섭취량은 그룹간의 유의적인 차이를 보이지 않았다. 간과 신장의 무게, AST, ALP, ALT는 모든 그룹간의 유의적인 차이가 없는 것으로 보아 일반식빵과 통밀빵 식이제공은 간과 신장의 스트레스 증가와 무관함을 확인할 수 있었다. 공복혈당 수준은 NC 그룹은 1주차에서부터 4주차까지 혈당이 완만하게 상승했던 것에 비해 모든 통밀빵 그룹은 혈당 증가폭이 감소하는 경향을 나타내었다. 4주차에서는 모든 통밀빵 그룹은 NC 그룹과 비교하여 유의적인 차이를 보이며 혈당이 감소하는 것을 확인할 수 있었다. 경구당부하검사(OGTT)시 90분대 혈당치에서 모든 통밀빵 그룹은 NC 그룹($152.13{\pm}19.90mg/dL$)의 혈당치와 비교하여 유의적인 차이를 보이며 감소하는 것을 보여주었으며(p<0.05), AUC에서 WDC 그룹을 제외한 W 그룹, WL 그룹, 그리고 WC 그룹의 area value가 NC 그룹과 유의적인 차이를 보이며 적어졌다. WL 그룹과 WC 그룹의 경우 NC 그룹과 W 그룹과도 유의적인 차이를 보이며 면적이 감소하는 것을 확인하였다. 혈당 곡선아랫면적과 경구당부하 검사 결과를 종합해 볼 때 생강 추출물과 매생이 추출물이 첨가된 식빵은 일반식빵에 비해 내당능에 도움을 줄 수 있을 것으로 판단된다. 당화헤모글로빈의 함량을 측정한 결과 NC 그룹과 비교하여 모든 통밀빵 그룹에서 유의적인 차이를 보이며 함량이 감소하였고, 특히 WL 그룹과 WC 그룹은 W 그룹과도 유의적인 차이를 나타내며 당화 헤모글로빈 수치 관리에 좋은 효과를 나타내었다. 혈청 인슐린 농도를 측정하였을 때 모든 그룹에서 유의적인 차이를 보이지 않았으나 수치적으로 보았을 때 NC 그룹 대비 WL 그룹에서 39.9%, WC 그룹에서 37.2% 인슐린 함량이 증가하는 것을 확인하였다. 혈중 지방 질 지표를 측정한 결과 총 콜레스테롤과 LDL-콜레스테롤 수치에서는 그룹간 유의적인 차이를 나타내지 않았고, 중성지방의 경우 NC 그룹과 비교해 모든 통밀빵 그룹에서 유의적인 차이를 보이며 중성지방의 수치가 감소하였다. 혈중 HDL-콜레스테롤 수치는 WL 그룹이 NC 그룹과 유의적인 차이를 보이며 증가하는 것을 확인할 수 있었다. TBARS assy를 이용한 지방질과 산화 산물인 MDA를 측정한 결과 NC 그룹과 비교하여 모든 통밀빵 그룹에서 유의적인 차이를 나타내며 농도가 낮아졌고, WC 그룹에서 가장 낮은 수치를 나타내었다. 지방질과 관련된 유전인자인 $PPAR-{\alpha}$ mRNA 발현을 측정한 결과 모든 그룹간의 유의적인 차이는 나타나지 않았고, $PPAR-{\gamma}$ mRNA 발현의 경우 WL 그룹, WC 그룹에서 NC 그룹과 유의적인 차이를 나타내며 가장 적은 발현을 나타내었다. 이러한 결과를 미루어 보아 일반식빵에 비해 통밀빵을 섭취한 모든 그룹은 몸무게, 공복혈당, 경구당부 하 검사(OGTT), 당화헤모글로빈(HbA1c), 중성지방(TG), 동맥경화지수(AI) 측정 항목에서 통계적으로 유의적인 차이를 나타내었고 이것으로 통밀은 식빵 섭취시의 혈당 증가 완화 효과를 가지며 혈중 지방질수준 개선과 비만의 예방에 기여할 것으로 판단된다. 또한 생강나무 잎 추출물과 매생이 추출물을 첨가한 통밀빵 그룹은 당화헤모글로빈(HbA1c) 측정 항목에서 통밀빵 그룹과도 유의적인 차이를 나타내며 수치가 감소한 것으로 보아 통밀의 효능에 기능성 소재인 생강나무 잎과 매생이 추출물의 효능이 더 해지면서 상당한 혈당강하효과를 나타낸 것으로 판단된다.

저장조건에 따른 녹차음료의 기능성성분 변화 및 안정성 (Changes in Functional Constituents and Stability of Green Tea Beverage during Different Storing Conditions)

  • 제주산 건조녹차를 $55^{\circ}C$에서 15분간 추출하여 여과한 다음 PET병에 밀봉하여 $4^{\circ}C$ 저온과 실내 및 실외에 각각 보관하면서 보존안정성을 측정하였다 저장기간에 따라 L값은 거의 변화가 없었지만 a값이 감소하고 b값이 증가하면서 전체적인 색깔이 짙어졌으며, 실내에서 저장한 경우가 실외저장이나 저온저장보다 저장기간이 길어질수록 더 짙어졌다. 녹차추출물 중의 주요 카테킨류로는(-)-epigallocatechin, (-)-epigallocatechin gallate, epicatechin, (+)-catechin, epicatechin gallate순이었으며, EGC의 함량비율이 전체의 53.8%이었다. 총 카테킨 함량은 저온저장에서는 큰 차이가 없었지만, 실내 및 실외 저장한 경우에는 $148.87{\mu}g/mL$에서 14주일 후에는 각각 133.98, $141.87{\mu}g/mL$로 10%, 4.7%가 감소하였다. 카페인 함량은 저장조건에 관계없이 전체적으로 증가하였으며, 총 폴리페놀 함량은 저장조건 및 저장기간에 따른 차이가 크지 않았다. 전자공여능은 저장방법과 저장기간에 관계없이 60% 이상의 효과가 있었다. 아질산염 소거능은 pH 1.2에서 90% 이상으로 가장 높았으며, pH가 증가할수록 감소하다가 pH 6.0에서는 아질산염 소거효과가 없어졌다. 3개월 이상 경과하더라도 색깔, 폴리페놀성분, 항산화작용등의 변화가 크지 않았다. $95^{\circ}C$에서 10분간 살균으로 보관기간 중에 미생물의 증식이 인정되지 않아 저장안전성을 유지하는 것으로 판단되었다.

Synthesis and Anticancer Activity of Some Novel 1,3-Diaryl/heteroarylprop-2-en-1-one Derivatives

  • In the present investigation, a series of some novel 1,3-diaryl/heteroarylprop-2-en-1-one derivatives (3a-j) have been synthesized and evaluated for their in vitro cytotoxicity against three cancer cell lines, two hepatocarcinoma cell lines HUH-7, Hep-3b and one leukemia cancer cell line MOLT-4. Based on these results, structure-activity relationship (SAR) was studied on modification of $R^1$ and $R^2$ to identify the compound with maximum potency. Amongst the compounds, 3b and 3d strongly inhibited the growth of Hep-3b and MOLT-4 cells with $IC_{50}$ value of 3.39 and $3.63{\mu}M$ respectively. The results obtained from the inhibitory study had further been supported by the reactive oxygen species (ROS) measurement using flow cytometry in MOLT-4 cells. These observations collectively reveal that compounds comprising 1,3-diarylprop-2-en-1-one framework with pyrazole ring at position-3 and heteroaryl/aryl substituents at position-1 can be used as promising anticancer agents.

An Intercultural Experiment to Build Life Science Innovation in Korea

  • The establishment of Institut Pasteur Korea (IP-K) has been an intercultural experiment, which transplanted a French research organization with many foreign researchers into Korea to grow a new institution as a long-term collaboration. The Mission of the newly founded institute has been to develop more effective ways of generating value with basic life science research in the face of a world-wide Pharma crisis. The challenges have been i.) to invent new technologies and approaches in drug discovery, ii.) to convince the Korean stakeholders of their inherent value, iii.) to induce Pharma industry to adopt the new technologies and iv.) to create a context in the Korean R&D landscape where the new institute could contribute tangible benefits. If Institut Pasteur Korea has succeeded in all counts, then due to a somewhat skewed and unlikely set of cultural complementarities between Korea and France. The abstract and conceptual French approach was matched by Korean pragmatism, linearity and relentless improvement towards the defined development goal. IP-K has become an example for innovation made in Korea, which is now re-imported into Europe. As the project could arguably not have succeeded in either partner county alone, it highlights the benefits of longterm, in depth international collaborations.

Bacillus subtilis Fermentation for Enhancement of Feed Nutritive Value of Soybean Meal

  • In order to increase the nutritional quality of soybean meal (SBM) as an animal feed, Bacillus subtilis TP6, a previously isolated strain from an Indonesian traditional fermented soybean food, Tempeh, was used as a starter organism for solid-state fermentation. In the pre-treated SBM with water content of 60% (v/w), B. subtilis TP6 was grown to a maximum viable cell number of $3.5{\times}10^9CFU/g$. Compared to control, crude protein in Bacillus fermented SBM was increased by 16%, while raffinose, stachyose, and trypsin inhibitors were reduced by 31, 37, and 90%, respectively. The Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that proteins in the fermented SBM were remarkably hydrolyzed into smaller molecular masses, resulting in a decrease in large sized proteins. Our data suggested that B. subtilis fermentation could increase the nutritive value of SBM through reduction of anti-nutritive factors and improvement of protein quality by hydrolysis of soy protein. In addition, B. subtilis TP6 produced a functional ingredient, poly-${\gamma}$-glutamic acid which has various health benefits.

Breeding of four-leaf white clover (Trifolium repens L.) through 60Co gamma-ray irradiation

  • Four-leaf white clover is not found easily in nature due to its low appearance rate (1 in 10,000). Because people believe that it brings good luck and like to either keep it or present it to a loved one, it has commercial and ornamental value. To breed four-leaf clover, we exposed its flowers to ${\gamma}-rays$ at the pollination stage. The $M_1$ seeds produced following doses at 25-100 Gy showed an approximately 74% germination rate, with seedling survival at 46%. In the $M_1$ generation of plants irradiated within that dose range, we found an increased frequency of four leaflets. One of them, Jeju Lucky-1 (JL-1), had a frequency of about 60%. To see whether that mutation was somaclonal or genetic, we observed its $M_2$ generation and found that such a phenotype reappeared. Although our results demonstrated that the irradiation of fully mature flowers led to a higher frequency of 4-leaflets, we could not clearly explain the genetic mechanism involved. We suggest that JL-1 is valuable as a new variety, without further genetic fixation, because white clover can be propagated vegetatively by stolons.

Bacillus ginsengihumi sp. nov., a Novel Species Isolated from Soil of a Ginseng Field in Pocheon Province, South Korea

  • A Gram-positive, aerobic or facultative anaerobic, non motile, endospore-forming bacterial strain, designated Gsoil $114^T$, was isolated from a soil sample of a ginseng field in Pocheon Province (South Korea), and was characterized taxonomically by using a polyphasic approach. It grew well on nutrient agar medium and utilized a limited number of organic substrates as sole carbon sources, including D-xylose and some other carbohydrates, but did not utilize L-amino acids and organic acids. The isolate was positive for oxidase test but negative for catalase, and negative for degradation of macromolecules such as starch, cellulose, xylan, casein, chitin, and DNA. The G+C content of the genomic DNA was 41.8 mol%. The predominant isoprenoid quinone was menaquinone 7 (MK-7). The major fatty acids were $anteiso-C_{15:0}$ (32.1%), $iso-C_{15:0}$ (30.5%), and $anteiso-C_{17:0}$ (30.2%). Comparative 16S rRNA gene sequence analysis showed that strain Gsoil $114^T$ fell within the radiation of the cluster comprising Bacillus species and joined Bacillus shackletonii LMG $18435^T$ with a bootstrap value of 95%. The highest 16S rRNA gene sequence similarities were found with Bacillus shackletonii LMG $18435^T$ (97.6%), Bacillus acidicola DSM $14745^T$ (96.9%), Bacillus sporothermodurans DSM $10599^T$ (96.5%), and Bacillus oleronius DSM $9356^T$ (96.5%). The phylogenetic distance from any other validly described species within the genus Bacillus was less than 96%. DNA-DNA hybridization experiments showed that the DNA-similarities between strain Gsoil $114^T$ and closest phylogenetic neighbors were less than 39%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Gsoil $114^T$ (=KCTC $13944^T$=DSMZ $18134^T$) was classified in the genus Bacillus as the type strain of a novel species, for which the name Bacillus ginsengihumi sp. nov. is proposed.

홀스타인 육성우의 계절별 분뇨 배설량 및 특성에 관한 연구 (Quantity and Characteristics of Manure Produced by Holstein Heifer at Different Seasons)

  • 우리나라에서 사육되고 있는 젖소 육성우의 분뇨배설량을 구명하기 위하여 육성우 6두를 공시하여 축산연구소 낙농우사에서 봄, 여름, 가을, 겨울 등 4계절에 걸쳐 각각 10일간 시험을 수행하였으며, 그 결과는 다음과 같다. 1. 건물섭취량은 평균 6.7kg으로 봄, 여름, 가을 및 겨울이 각각 4.6, 8.3, 7.1, 6.8kg으로 봄이 가장 낮은 섭취량을 보였으며, 음수량은 평균 $19.3{\ell}$로, 여름이 $21.8{\ell}$로 가장 많았고 봄과 겨울이 $18.3{\ell}$로 낮았으나 계절별 유의차는 보이지 않았다. 2. 분뇨배설량을 계절별로 조사한 결과, 봄 13.7kg, 여름 23.8kg, 가을 25.0kg, 겨울 20.2kg으로 봄이 가장 낮았으며(p<0.05), 전체 평균 분뇨배설량은 20.3kg이였고, 체중대비 분뇨의 비율은 5.6%였다. 3. 체중과 분뇨배설량간($R^2=0.7816$), 체중과 사료섭취량간($R^2=0.7296$)의 상관관계가 비교적 높았으나, 음수량과 분뇨배설량, 사료섭취량과 분뇨배설량간의 상관관계는 비교적 낮았다. 4. 분뇨의 수분 및 pH를 분석해 본 결과, 분의 뇨의 평균 수분 함량은 각각 83.5, 94.6%였으며, pH는 각각 7.4, 7.6 수준이었다. 5. 분뇨중 오염물질 농도는 분의 $BOD_5,\;COD_{Mn}$, SS, T-N 및 T-P는 각각 18,048, 50,114, 119,833, 2,519, $427mg/{\ell}$였고, 뇨는 각각 5,434, 6,550, 825, 3,616, $28mg/{\ell}$였다. 6. 분뇨중 비료성분은 분의 N, $P_2O_5,\;K_2O$가 각각 0.25, 0.10, 0.14%였고, 뇨는 각각 0.36, 0.006, 0.31%였다.도는 3m 지점에서 $446.6\;{\mu}g/m^3$ 이었으나 20 m 지점에서는 $156.3\;{\mu}g/m^3$로 34.9% 수준이었고 PM10 34.9%, PM2.5 48.7%, PM1.0 86.8% 수준이었다.

락톤류로부터 Hydrogenophaga pseudoflava와 Ralstonia eutropha H16 두 세균에 의한 공중합 PHA의 합성 및 미세구조적 특성 연구 (Synthesis of Copolymeric PHA by Hydrogenophaga pseudoflava and Ralstonia eutropha H16 from Vari-ous Lactones and Their Microstructural Studies)

  • Two typess of copolyesters, poly(3-hydroxybutyric acid-co-4-hydroxy-butyric acid)[P(3HB-co-4HB] and poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid)[P(3HB-co-3HV)], with various monomer ratios and different degree of microstructural heterogeneity were synthesized from Ralstonia eutropha H16 and Hydrogenophaga pseudoflava by using ${\gamma}$-butyrolactone and ${\gamma}$-valerolactone, respectively. The two bacteria showed a large difference in the utilization of ${\gamma}$-butyrolactone for cell growth and PHA synthesis. H. pseudoflava synthesized P(3HB-co-4HB) copolyesters with a wide range of 4HB content from 13 to 96 mol% depending on culture conditions, whiel R. eutropha H16 was able to synthesize the copolyesters containing less than 20 mol% of 4HB. An increase in the 4HB content in the P(3HB-co-4HB) copolyesters synthesized by H. pseud-oflava induced an lowering of their melting temperatures as well as their enthalpies of fusion. The increase in the 4HB content, however, increased the rate of degradation by an extracellular P(3HB) depolymerase. NMR spectros-copy and differential scanning calorimetry showed that the P(3HB-co-4HB) copolyesters from H. pseudoflava were generally microstructurally heterogeneous. The P(3HB-co-4HB) copolyesters) synthesized by R. eutropha H16 were rather random copolymers showing less microstructural heterogeneity than those synthesized by H. pseudoflava. The NMR D value analysis suggested that the monomer distribution of the P(3HB-co-3HV) copolymers from the two bacteria were relatively random.
