Effects of Bacillus polyfermenticus SCD Administration on Fecal Microflora and Putrefactive Metabolites in Healthy Adults

  • Probiotics have been suggested to improve gastrointestinal health in humans. To investigate the effects of Bacillus polyfermenticus SCD administration on fecal microflora and putrefactive metabolites in humans, Bacillus polyfermenticus SCD (4.00${\times}$10$\sub$5/ CFU/mg) was administrated to ten healthy subjects (5 men and 5 women, average age 24 years) three times a day for 2 weeks. Fecal samples were collected before (1st and 2nd weeks, control), during (3rd and 4th weeks), and 2 weeks after the administration. The fo11owing microbial groups were evaluated in the feces: aerobic and anaerobic bacteria, Bacillus polyfermenticus SCD, Lactobacillus, Bifidobacterium, total lactic acid bacteria, Salmonella, Clostridium, Clostridium perfringens, Eubacterium, Staphylococcus, Coliform bacteria, Pseudomunas, and Yeast. Fecal concentrations of total aerobic bacteria (p<0.05, p<0.01, 3rd and 4th weeks), total lactic acid bacteria (p<0.01, 3rd, 4th and 5th weeks), and Bifidobacteria (p<0.05, 4th and 5th weeks) were significantly increased in all subjects, compared to the control, from the 3rd week after the administration of the products. Clostridium (p<0.01, 4th week), Clostridium perfringens (p<0.05, p<0.01, 3rd and 4th weeks), and coliform (p<0.01,5th week) were significantly reduced from the 3rd week of administration. No significant changes in the fecal concentrations of Pseudomonas, Lactobacillus, Eubacterium, Staphylococcus, yeast, and total anaerobes were observed. Six weeks after the administration, the concentration of all rnicroorganlsrns returned to the basal level. Bacillus polyfermenticus SCD was significantly maintained from the 3rd week to 6th week of the study. Despite the absence of a statistical significance, the putrefactive metabolites (ammonia, indole, skatole, and $\rho$-cresol) and the pH value tended to be lower during and after the test periods than the base line. These results show that this probiotic preparation is able to colonize the intestine, and suggest that it may be useful as a beneficial probiotic in humans.

Characterization and Evaluation of Melanocortin 4 Receptor (MC4R) Gene Effect on Pork Quality Traits in Pigs (돼지 Melanocortin 4 Receptor (MC4R) 유전자의 육질연관성 분석)

  • This study aimed to investigate the single nucleotide polymorphisms (SNPs) of the porcine MC4R gene and validate the effect of the MC4R genotype for marker assisted selection (MAS). Six amplicons were produced to analyze the entire base sequences of the porcine MC4R gene and six SNPs were detected (c.-780C>G, c.-135C>T, c.175C>T-Leu59Leu, c.707A>G-Arg236His, c.892A>G-Asp298Asn, and c.*430A>T). Linkage disequilibrium (LD) of the six SNPs was analyzed by performing haploid analysis. There was a perfect linkage disequilibrium in c.-780C>G, c.-135C>T, c.175C>T-Leu59Leu, c.707A>G-Arg236His, and c.*430A>T. Only the c.892A>G (Asp298Asn) SNP showed a very low LD with an $r^2$ value of 0.028 and the D' value of 0.348. As a result, the two SNPs-c.707A>G (Arg236His) and c.892A>G (Asp298Asn)-were selected to extract the genotype frequencies from the 5 pig breeds by using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) genotype analysis method. The SNP frequency of c.707A>G (Arg236His) indicated the presence of the A (His) allele only in Yorkshire, while the G allele was fixed in the KNP, Landrace, Berkshire, and Duroc. Association analysis was carried out in 484 pigs with the c.707A>G (Arg236His) SNP and the meat quality traits of four different pig cross populations: a significant association was noted in crude fat, sirloin moisture, meat color, and the degree of red and yellow coloration. The frequency of the c.892A>G(Asp298Asn) SNP genotype varied among the breeds; while Duroc showed the highest frequency of the A (Asn) allele, KNP showed the highest frequency of the G (Asp) allele. Association analysis was carried out in 1126 pigs with the c.892A>G (Asp298Asn) SNP and the meat quality traits of four pig populations: a highly significant linkage was noted in the back-fat thickness (P<0.002). It was found that the back-fat thickness was higher in individuals with the AA genotype than in those with the AG or GG genotype. Thus, in this study, we verified that the c.892A>G (Asp298Asn) SNP in the pig MC4R gene has a sufficient effect as a gene marker for MAS in Korean pork industry.

Effects of Carcass Processing Method and Curing Condition on Quality Characteristics of Ground Chicken Breasts (분쇄 계육 가슴살의 도체가공방법과 염지조건이 품질특성에 미치는 영향)

  • This study was carried out to investigate the effect of hot boning and curing condition on the quality characteristics of ground chicken breast. Treatments were cured by four conditions follows; control (general curing method), T1 (after hot-boning and then immediately cured), T2 (after hot-boning and immediately cured, then frozen), and T3 (after hot-boning, immediately frozen, refrigerated and then cured). The pH of chicken breast in the state of pre-rigor was 6.22. The pH of cold storage or freezing chicken breast meat respectively were 5.70 or 5.61. The pH of T1 and T2 treatments were significantly higher than those of control and T3 treatment (p<0.05). After stored for 1 wk, the pH value of T1 treatment had a higher value than those of other treatments. T1 treatment had the highest water holding capacity and the lowest cooking loss among all treatments, regardless of the cooking methods. The reduction in diameter for T1 and T2 treatments was lower than those of control and T3 treatment (p<0.05). T1 treatment had the lowest fat loss and moisture loss among all treatments, and the emulsifying capacity of T1 treatment was the highest. The protein solubility of T1 treatment was significantly lower than that of T3 treatment.

Characterization of Nutritional Value for Twenty-one Pork Muscles

  • A study was conducted to evaluate nutritional value for twenty-one pork muscles. Ten market-weight crossbred pigs (five gilts and five barrows) were used for evaluating proximate chemical composition, cholesterol, total iron, calorie and fatty acid contents. As preliminary analysis revealed no noticeable sex effect, pooled data from both sexes were used for the final analysis. M. rectus femoris had the highest moisture content, while m. latissimus dorsi was lowest in moisture content (p<0.05). Protein content was highest for m. longissimus dorsi and lowest for m. supraspinatus (p<0.05). The tensor fasciae and latissimus dorsi muscles contained the highest intramuscular fat (p<0.05), while rectus femoris, adductor and vastus lateralis were lowest in intramuscular fat content. When simple correlations between chemical values were computed for the pooled dataset from all muscles, intramuscular fat had significant (p<0.05) negative linear relationships with moisture (r = -0.85) and protein (r = -0.51) contents. Calorie levels were not significantly affected by fat content, while rectus femoris and latissimus dorsi muscles showed lowest and highest calorie contents, respectively (p<0.05). Polyunsaturated fatty acid content was highest (p<0.05) for both m. adductor and m. rectus femoris, while it was lowest for m. longissimus dorsi. Collectively, the current study identified a large amount of variation in nutritional characteristics between pork muscles, and the data can be used for the development of muscle-specific strategies to improve eating quality of meats and meat products.

Biogas Production from Vietnamese Animal Manure, Plant Residues and Organic Waste: Influence of Biomass Composition on Methane Yield

  • Anaerobic digestion is an efficient and renewable energy technology that can produce biogas from a variety of biomasses such as animal manure, food waste and plant residues. In developing countries this technology is widely used for the production of biogas using local biomasses, but there is little information about the value of these biomasses for energy production. This study was therefore carried out with the objective of estimating the biogas production potential of typical Vietnamese biomasses such as animal manure, slaughterhouse waste and plant residues, and developing a model that relates methane ($CH_4$) production to the chemical characteristics of the biomass. The biochemical methane potential (BMP) and biomass characteristics were measured. Results showed that piglet manure produced the highest $CH_4$ yield of 443 normal litter (NL) $CH_4kg^{-1}$ volatile solids (VS) compared to 222 from cows, 177 from sows, 172 from rabbits, 169 from goats and 153 from buffaloes. Methane production from duckweed (Spirodela polyrrhiza) was higher than from lawn grass and water spinach at 340, 220, and 110.6 NL $CH_4kg^{-1}$ VS, respectively. The BMP experiment also demonstrated that the $CH_4$ production was inhibited with chicken manure, slaughterhouse waste, cassava residue and shoe-making waste. Statistical analysis showed that lipid and lignin are the most significant predictors of BMP. The model was developed from knowledge that the BMP was related to biomass content of lipid, lignin and protein from manure and plant residues as a percentage of VS with coefficient of determination (R-square) at 0.95.This model was applied to calculate the $CH_4$ yield for a household with 17 fattening pigs in the highlands and lowlands of northern Vietnam.

A Study on the Technology Evaluation of Development of Separation and Purification Techniques of Flavonoid in Citrus (감귤 플라보노이드 성분의 추출 및 정제방법 개발의 기술가치평가)

  • This study was carried out to evaluate the economic value of development of separation and purification techniques of flavonoid in citrus using IRR, NPV. The separation and purification techniques of flavonoid of citrus was developed by national institute of horticultural and herbal science in Korea rural development administration. This technology could be used to improve efficiency and practicality of cultivation of citrus. The research results show that the technological value of separation and purification techniques of flavonoid of citrus was evaluated 3,155(scenario 1)~6,518(scenario 2) million won. IRR was 28.3%~61.2%, which was greater than discount rate(11.9%). NPV is 5,541~18,773 million won. Therefore, the economic validity of development of separation and purification techniques of flavonoid of citrus is identified by the results of technology evaluation.

Study of the Technology Evaluation of Development of Environment-friendly Controlling Method of Mushroom (길항 미생물 HC5를 이용한 버섯의 친환경방제 기술개발의 가치평가)

  • This study evaluated the economic value of the development of an environment-friendly controlling method of mushroom using IRR and NPV. The environment-friendly controlling method of mushrooms was developed by the national institute of horticultural and herbal science in Korea rural development administration. This technology can be used to improve the efficiency and practicality of cultivation of mushroom. The research results show that the technological value of the development of an environment-friendly controlling method of mushroom was evaluated at 135.4 billion won (average), 182.6 billion won (maximum), and 88.1 billion won (minimum) based on the discount rate of 7.3%. The IRR was 1,958.5% - 4,054.4%, which was greater than the discount rate (6%~10%). The NPV was evaluated at 134.8 billion won (average), 218.1 billion won (maximum) and 87.5 billion won (minimum) based on the discount rate of 7.3%. Therefore, the economic validity of the development of an environment-friendly controlling method of mushrooms was identified by the results of a technology evaluation.

  • This study was researched to purify and characterize variety bioactive material acethylmannan from Aloe vera. Purified acethylmannan was mannose (67%), acetyl group (23%) and the rest glucose, galactose that consisting of long chain polydispered ${\beta}-1,4$ linked mannan polymers. The sugar and acetyl group in molecular were linked molar ration one third. $IC_{50}$ value (i.e that concentration which exhibits 50% more enzyme inhibition than control) on angiotensin converting enzyme were 0.58 mM. This compound were found to be a competitive inhibition of Angiotensin Converting Enzyme with apparent Ki values of 0.068 mM.

Patterns of Offensive Odor Compounds According to Blocks in Shiwha Industrial Complex (시화산업단지의 블록 별 악취유발물질 특성)

  • This research was conducted on characteristic of offensive odors in Shihwa industrial complex. Result of blocks distribution of TVOC indicates that mechanic block, site D, was the highest concentration (74 ppb). Chemistry block, site A, was the second highest concentration (50 ppb). Also, mixed blocks, metal blocks and park etc. were measured almost similar concentration about 30 ppb, but mixed block, site F, was the place where concentrations were the smallest. Average of TVOC was shown about 35 ppb concentration. Aldehydes including acetaldehyde, butyraldehyde and hydrogen sulfide concentrations were prevalent among offensive odors in Shihwa industrial complex. Comparing the offensive odor intensity mostly about acetaldehyde, butyraldehyde and hydrogen sulfide which contain high offensive odor intensity showed results that sites A, B (chemistry block) and site D, I (mechanic block) site H (metal block) have showed the intensity over 1. In the case of acetaldehyde, relatively the high odor intensities over '2' were able to obtain in many cases. The correlation coefficient (r) for hydrogen sulfide was 0.91, so that high positive correlation exists between offensive odor intensity and the hydrogen sulfide element. Butyraldehyde also showed high positive correlation coefficient, as 0.82. Correlation coefficient of acetaldehyde that had the highest value as offensive odor substance was 0.62, had somewhat correlation with offensive odor intensity.

Study on the Optimum Range of Weight-Age Data for Estimation of Growth Curve Parameters of Hanwoo (한우의 체중 성장곡선 모수 추정을 위한 체중 측정 자료의 최적 범위에 관한 연구)

  • Mature weight (A) and rate of maturing (k) estimated by nonlinear regression were studied to determine the optimum age range over which the estimate of growth curve parameters can be estimated. The weight-age data from 1,133 Hanwoo bulls at Hanwoo Improvement Center of N.A.C.F. were used to fit the growth curve using Gompertz model. All available weight data from birth to the specific age of months were used for the estimation of parameters: the six specific ages used were 12, 14, 16, 18, 20 22 and 24 months of age. The mean estimates of mature weight (A) were 966.5, 1,255.9, 1,126.2, 916.5, 842.2, 780.9 and 767.0kg for ages 12 through 24 months, respectively. The mean estimates of mature weight (A) to 22 and 24 months of age were not different from each other. However, they were different from the estimates based on the data to other ages. Mean estimates of rate of maturing (k) were 3.362, 3.595, 3.536, 3.421, 3.403, 3.409 and 3.411 for ages 12 through 24 months, respectively. The mean estimates of maturing rate (k) for ages 18 through 24 months of age were not significantly different from each other. However, they were different from the estimates based on the data to other ages. Correlations among estimates of A at various ages showed the highest value of 0.93 between 22 and 24 months. Correlations among estimates of k at various ages were highest ranging from 0.91 to 0.99 among 18 to 24 months. The correlations between A and k were positive and tended to decrease with the increase of the age from 0.84 for the age of 12 months to 0.10 for the age of 24 months. Thus, the estimates of growth curve parameters, A and k, suitable for genetic studies can be derived from accumulated Hanwoo bulls after 22 months of age.
