In this paper, we present a novel integrated cell processor to handle individual embryos. Its functions are composed of transporting, isolation, orientation, and immobilization of cells. These functions are essential for biomanipulation of single cells, and have been typically carried out by a proficient operator. The purpose of this study is the automation of these functions for safe and effective cell manipulation using a MEMS based cell processor. This device is realized with a relatively simple design and fabrication process. Experimental results indicate that it can act as an efficient substitute for essential but very tiresome and repetitive manual work while contributing significantly to the improvement of speed and success rate of operation by facilitating cell manipulation. The cell viability test for the device is studied through the distribution of mitochondria in mice embryos and cultivation of cells for 86h.
This study was conducted to determine removal effect on phytoplankton (chlorophyll-${\alpha}$ as whole algae) and cyanobacteria by a fish Pseudorasbora parva and macroinvertebrate Palaemon paucidens in September 2006. Three treatments with 25 (T1), 50 (T2) and 100 (T3) individuals along with control (C1, no input fish), and two treatments with 25 (T4) and 50 (T5) individuals along with control (C1) were made for fish and macroinvertebrate, respectively. The initial concentrations of chlorophyll-${\alpha}$$(Chl_i)$ in each 10L test tank were set up for the levels of $95{\sim}100{\mu}g\;L^{-1}$ and the daily values were monitored in the test tank during 7 days. In the lab tests, P. parva did not show Chl-${alpha}$ removal effect; the removal rate of Chl-${alpha}$ for P. parva was -58% in T1, -56% in T2, and 61% in T3 during the test period. In contrast, P. paucidens. in the treatments of T4 and T5 removed the phytoplankton effectively and the removal effect were appeared to be 33% and 22%, respectively. Also, P. paucidens showed high feeding efficiency in the removal of cyanobacteria. The levels of cyanobacteria were greatly lowed from 6,048 to 927 cells $mL^{-1}$ in T4 and from 6,539 to 1,053 cells $mL^{-1}$ in T5, resulting in 85% and 84% in the removal effect, respectively. Our results for biomanipulation tests suggest that P. paucidens may be used as a potential candidate organism for algae control in spite of the preliminary results by laboratory tests.

Bottom-up approaches to control of Microcystis aeruginosa blooms were comparatively investigated through an in situ mesocosm experiment using aquatic plants (Iris pseudoacorus). In the mesocosm experiments, floating I. pseudoacorus, seemed to be effective in controlling massive M. aeruginosa blooms in an agricultural reservoir, exhibiting a close coupling with temporal variations in Chl-a and DO concentration. Shading by floating I. pseudoacorus resulted in a reduced phytoplankton abundance inhibiting light energy availability. Moreover, I. pseudoacorus may suppress phytoplankton growth through the excretion of chemical substances, likes a allelopathy, that inhibit phytoplankton photosynthetic activity. The $^{15}N$ atom % of I. pseudoacorus showed higher values than POM, suggesting that I. pseudoacorus assimilates DIN predominantly compared to phytoplankton, which was mostly M. aeruginosa. This result strongly suggests that the M. aeruginosa bloom should be regulated by aquatic plants, like I. pseudoacorus, this approach can affect zooplankton composition. This is the first study that has used stable isotope tracers to evaluate the biomanipulation efficiency through floating I. pseudoacorus.
To evaluate the influences of stocking and removal of silver carp on plankton communities and physicochemical parameters, fish biomanipulation was conducted in six enclosures constructed on the coast of a hypertrophic lake during the summer of 1997. Of these, three enclosures were established for the fish-stocking experiment; one fishless enclosure used as a control and two enclosures with high and low densities of fish. The other three enclosures for the fish-removal experiment were prepared in an entirely converse process compared to that for fish stocking. The results of randomized intervention analysis (${\alpha}=0.05$) showed significant changes in several physicochemical parameters and plankton communities after fish stocking and removal. Fish stocking decreased water transparency, DO, pH and chi-a (> $40\;{\mu}m$), while increasing the concentration of nitrogen and chi-a (<$2\;{\mu}m$). However, water transparency did not increase by fish removal. Small plankton communities, such as picocyanobacteria, eukaryotic picoplankton and ciliates (<$2\;{\mu}m$) showed significant reciprocal changes after both fish treatments: increase by the presence of fish and decrease by the absence of fish. No count of large sized phytoplankton and zooplankton showed significant change in the presence of fish, while large zooplankton, Diaphanosoma, Cyclopida and nauplii were significantly increased by fish removal. Although the frequent heavy-winds and endless supply of phosphorus from the lake sediment strongly disturbed the fish behavior and enclosure management, the stocking and removal of silver carp showed an obvious reciprocal influences on plankton communities and its physico-chemical parameters in a shallow-hypertrophic lake.
Most South Korean lakes are middle/small size artificial reservoirs, which are almost agricultural reservoirs (17,956). A total of 67 species (21 families) were recorded and collected from 65 agricultural reservoirs though field samplings and literature surveys. Dominant species was Pseudorasbora parva (relative abundance 24.5%), and Carassius auratur (41 sites) was the highest frequency. Feeding group of fish communities in the reservoirs was as follows: carnivorous (16.2%), omnivorous (79.5%) and herbivorous fish (4.3%). The number of individuals (P=0.024), species number (P=0.047) and carnivores number (P=0.024) were significantly correlated with reservoir ages. Reservoirs were classified into 3 groups according to feeding patterns of carnivore, omnivore and herbivore groups. The omnivores were dominant group in agricultural reservoirs. Detailed studies on fish community will be a base for the understanding of food web structure and biomanipulation in reservoir systems.
Water quality and plankton community dynamics after the introduction of two domestic freshwater shellfishes (Unio douglasiae Griffith & Pidgeon and Cipangoplaudina chinese malleata Reeve), were monitored daily in enclosures, which constructed in the tidal zone of eutrophic agricultural reservoir (Shingu reservoir, Korea) for one week between July 31 and August 6, 2007. This biomanipulation study to improve the water quality of eutrophic lake, comprised eight enclosures (duplicate x four kinds), enclosure had no mussels (Control), stocked only with U. douglasiae at density of 60 individuals (UD), stocked only with C. chinese malleata at density of 60 individuals (CCM), and combined-stocked with 30 individuals of UD and 30 individuals of CCM (MIX), respectively. Our results clearly indicate that UD strongly decreased the concentration of chlorophyll-a and increased the water transparency, whereas CCM and MIX (included CCM) did increase algal density or decrease transparency, due perhaps to the disturbance of C. chinese malleata. Therefore, the field application of shellfish to enhance the water quality of eutrophic agricultural reservoir, should consider the grazing characteristics of biological control agents, especially active moving animals like snail.
To examine the algicidal effect of co-treatment of biological control agent on centric diatom Stephanodiscus hantzschii in the filtered water, an endemic algicidal bacterium (Pseudomonas putida) and a heterotrich ciliate (Stentor roselli) were iso-lated from Pal'tang reservoir, Korea. Bacterial isolate and ciliate removed 98% and 80% of the diatom for 7 days of cultivation. Co-treatments of these two agents perfectly inhibited the diatom growth, compared to the single treatment of each agent. This synergistic interaction of the bacterium and ciliate could provide an effective tool in the biomanipulation to control the diatom bloom in freshwater lakes and streams.

In a natural water body, a useful ecotechnology to reduce standing crops of phytoplankton is to strengthen the top-down force of zooplankton. However, the predation of fish for zooplankton can make the force weak. This study was conducted to find out the effect of fish on the grazing pressure of zooplankton in the mesocosms established in a eutrophic stream (Kyongan Stream) from October to November in 2010. In the corral with fish, chlorophyll a concentration increased, and a small size cladoceran Bosmina longirostris was dominant. In the corral without fish, chlorophyll a concentration decreased along with the domination of a large cladoceran Daphnia galeata and a large copepod Eudiaptomus japonicus. The size-selective predation of fish appeared to miniaturize the zooplankton community, to narrow their food-size spectrum, and to weaken the top-down force.
Stable isotope tracers were first applied to evaluate the Microcystis cell assimilation efficiency of bivalves, since the past identification method has been limited to tracking the changes of each chl-a, clearity, and nutrient. The filter-feeders (Sinanodonta woodiana and Unio douglasiae) were assessed under the condition of cyanobacteria (Microcystis aeruginosa) blooms through an in mesocosm experiment using $^{13}C$ and $^{15}N$ dual isotope tracers. chl-a concentration in the treatment mesocosm was dramatically decreased after the beginning of the second day, ranging from 116 to $66{\mu}g\;L^{-1}$. In addition, the incorporated $^{13}C$ and $^{15}N$ atom % in the S. woodiana bivalve showed higher values than U. douglasiae bivalves. The results demonstrate that U. douglasiae has less capacity to assimilate toxic cyanobacteria derived from diet. Our results therefore also indicate that S. woodiana can eliminate the toxin more rapidly than U. douglasiae, having a larger detoxification capacity.
This study was to evaluate microcystin production by Microcystis aeruginosa in response to three different levels of indirect (0, 10, 50% of fish cultured media filtrate; control, FCMF1 and FCMF2) exposures to omnivorous and planktivorous fish (Carassius gibelio langsdorfi and Hypophthalmichthys molitrix, CCMF and HCMF, repectively). The cell biomass, intracellular microcystin (MC) and extracellular MC were measured everyday. The intracellular MC contents of all treatments were significantly increased than the controls (CCMF1, P=0.015; CCMF2, P<0.001; HCMF1, P<0.001; HCMF2, P<0.001). The intracellular MC contents of M. aeruginosa were significantly higher in CCMF2 than in CCMF1 (P=(0.023), Those of M, aeruginosa in HCMF2 were significantly higher than that in HCMF1 (P<0.001). The extracellular MC contents were not significantly different between control and CCMFs but those of M, aeruginosa in HCMF1 and HCMF2 were significantly higher than that in control (HCMF1, P=0.003; HCMF2, P<0.001). This study strongly supports that induced-defensive MC production (intra and extracellular MC) of potentially toxic cyanobacteria in response to kairomone concentration and this results can consider the biomanipulation of eutrophic waters as well as an information concerning strategies for recovering eutrophic waters.
