Changes in Quality by Drying Methods, Different Storage Conditions and Package Mediums for Established GAP Guide Book in Scutellaria baicalensis Georgi (황금 GAP 지침 설정을 위한 건조방법, 저장조건 및 포장재료에 따른 품질변화)

  • Background: The purpose of improving the quality of oriental medicinal herbs is to contribute to the improvement of the income of farm. The present study investigated the effect of reasonable drying methods, different storage conditions and packageing materials on quality and stability of Scutellaria baicalensis Georgi and for developing GAP (good agricultural practice) guide book. Methods and Results: Three methods of drying Scutellaria baicalensis roots produced over two years were used to estimate loss rate owing to drying, storage, and packaging. The methods of drying were categorized into natural drying (36 - 60 h by sunshine), drying with heat dryer (2 - 10 h), or drying with gas bulk dryer (2 - 10 h). After cleaning, and initial drying for a few days under controlled temperature conditions, the second phase of drying was carried out at 35, 45 and $55^{\circ}C$. Changes in hunter color values and quality under the two studied storage treatments (at $20^{\circ}C$ indoors and at $4^{\circ}C$ in a refigerator) were evaluated. Storage period for 60, 120, and 180 days in three packaging materials, PE (polyethylene package), PP (polypropylene gunnysack), and WP (watertight packing paper) were studied. Conclusions: Initial cleaning, reasonable gas drying and hot air drying, and drying in an oven at 35 and $45^{\circ}C$ after cutting the roots resulted in the lowest loss rates in S. Baicalensis root. Hunter color valuse indicated that stroage of dry roots at room temperature was better than PP packaging, and that cold stroage was better than PE packaging for long-term stability.

  • The organic materials included in excreta of livestock are important resources for organic manure and for improving soil quality, although there is still far from effective using. One reason for this is still unclearly standard of quality for evaluation of manure made from excreta of livestock. Therefore, the objective of this study is to develop rapid and accurate analytical method for analyzing organic compositions of manure made from excreta of livestock, and to establish quality evaluation method based on the compositions predicted by near infrared reflectance spectroscopy (NIRS). Sixteen samples of manure, each eight samples prepared from two treatments, were used in this study. The manure samples were prepared by mixing 560 kg feces of swine,60 kg sawdust with moisture content was adjusted to be 65%. The mixture was then keep under two kinds of shelter, black and clear sheets, as a treatment on the effect of sunlight. Samples were taken in every week (form week-0 to 7) during the process of manure making. Samples were analyzed to determine neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) by detergent methods, and organic cell wall (OCW) and fibrous content of low digestibility in OCW (Ob) by enzymatic methods. Biological oxygen demand (BOD) was analyzed by coulometric respirometer method. These compositions were carbohydrateds and lignin that were hardly digested. Spectra of samples were scanned by NIR instrument model 6500 (Pacific Scientific) and read over the range of wavelength between 400 and 2500nm. Calibration equations were developed using eight manure samples collected from black sheet shelter, while prediction was conducted to the other eight samples from clear sheet shelter. Accuracy of NTRS prediction was evaluated by correlation coefficients (r), standard error of prediction (SEP) and ration of standard deviation of reference data in prediction sample set to SEP (RPD). The r, SEP and RPD value of forage were 0.99, 0.69 and 7.6 for ADL, 0.96, 1.03 and 4.1 for NDF, 0.98, 0.60 and 4.9 for ADF, 0.92, 1.24 and 2.6 for Ob, and 0.91, 1.02 and 7.3 for BOD, respectively. The results indicated that NIRS could be used to measure the organic composition of forage used in manure samples.

The Evolution of Makeup Methods of Korean Women in Response to Changing Standards of Beauty in the Early 20th Century (20세기 초 미의식의 변화에 따른 국내여성들의 화장법)

  • Although the human body is a biological subject with definite and distinctive physical features, its actualization and perception differs among societies. The aesthetics of the human body are based on diverse cultural perceptions that must be considered prior to design development. This study establishes the foundations of newly adopted concepts of beauty that are presumed to have been established in the first half of the twentieth century that continue to affect our mindset even now. The research includes human figures in the articles of women's magazines and cosmetic advertisements in the early $20^{th}$ century. The results are as follows: First, the change of perception in the human body: Instead of being a subject of preservation, the body has become a subject of sculpture with emphasis on health in the 1920's and on beauty in the 1930's. The recognition of the importance of the body has created intensive attention on physical training and an increased sense of hygiene. The body exposed to the public perceives itself through the eyes of others that alter one's own perception of oneself as well as become a target of evaluation. There is an additional emphasis on the exotic eroticism of a passive subordinate. Western culture became the standard for modernization along with the dissociation of traditional standards and values. Through the effect of education and western thinking, the awareness of women's rights and self-appreciation was developed. Second, ideal beauty can be summarized as follows: Unprocessed natural beauty was extolled as ideal in the 1920's, but the 1930's, it highlighted big eyes and an aquiline nose that are the characteristics of western women. Taking care of one's appearance was recognized as an important value for every social class. Cosmetics and skin care treatments promised soft and white skin. In contrast to western cosmetics, dark and shiny hair was highly favored. Exercising and traveling, differing seasonal and regional skin treatments were also widely accepted. In its initial stages, the research had originally assumed that the beginning of the twentieth century would be a time in which traditional concepts of beauty and new, westernized aesthetics coexisted. However, as the research progressed, it was clear that the idea of beauty had already adopted occidental ideals by that time. Thus, it seems necessary to continue the study on the shifting paradigms of beauty that must have occurred in the nineteenth and late twentieth century.

Phylogenetic Relationship of Ligularia Species Based on RAPD and ITS Sequences Analyses (RAPD 및 ITS 염기서열 분석을 이용한 곰취 속(Ligularia) 식물의 유연관계 분석)

  • The genetic relationships in 5 species of $Ligularia$ were investigated using RAPD (Randomly Amplified Polymorphic DNA) and ITS (Internal Transcribed Spacer) sequences analyses. In RAPD analysis, sixty three of 196 arbitrary primers showed polymorphism. The amplified fragments ranged from 0.2 to 1.6 kb in size. The dendrogram was constructed by the UPGMA clustering algorithm based on genetic similarity of RAPD markers. A total of 16 accessions were classified into 5 major groups corresponding each species at the similarity coefficient value of 0.77. In the ITS sequence analysis, the size of ITS 1 was varied from 248 to 256 bp, while ITS 2 was varied from 220 to 222 bp. The 5.8S coding region was 164 bp in lengths. Forty nine sites (10.2%) of the 478 nucleotides were variable, and the G+C content of ITS region ranged from 49.4 to 53.5%. In the ITS tree, five species of $Ligularia$ were monophyletic, and $L.$ $taquetii$ was the first branching within the clade. $Ligularia$ $intermedia$ formed a clade with $L.$ $fischeri$ var. $spiciformis$ (BS=79), and $L.$ $stenocephala$ and $L.$ $fischeri$ were also claded. Two data sets were congruent, except of the position of $L.$ $fischeri$ var. $spiciformis$.

Analysis of Growth Response and Gene Expression by Waterlogging Stress on B73 Maize (침수 처리에 따른 B73 옥수수의 생육 반응 및 유전자 발현 분석)

  • Maize is thought to be an alternative crop to rice in paddy fields for efficient field management and maintenance of rice production at appropriate levels in Korea. Thus efforts to breed waterlogging-tolerant maize cultivars have been ongoing. However, molecular studies related to waterlogging tolerance are limited for developing molecular markers to select waterlogging tolerant maize varieties. In this study, we examined molecular biological changes of B73 in the V3 stage after immersion treatment for 7 days. Overall growth of maize was lower in treated samples compared to non-immersed control samples. The length of leaf and root decreased by 21.3% and 50.6%, respectively and the weight of root reduced by 21.6%. Soil plant analysis development (SPAD) value and chlorophyll content of leaf also decreased by 55.7% and 35.3%, respectively. Reactive oxygen species (ROS) content of root increased by 46.5% at 2 hours in immersion treatment. In addition, immersed roots were 2.5-fold thickened with additional aerenchyma formation in the cortex. In order to identify the causes of these changes, we performed a microarray and found increased expression of genes, such as WIP1, PMIP2, EXPA1, TPS1, and MAS1, in immersed samples. These differentially expressed genes and expression of previously reported genes, including ALDH2, Wusl1032, UP-1, UP-2, and CAT2 were further confirmed with qRT-PCR. Here, we report 7 differentially expressed genes after immersing treatment, which may be utilized as useful resources for breeding waterlogging- tolerant maize.

Control of Bulking and Foaming Caused by Microthrix parvicella (Microthrix parvicella에 의한 슬러지 벌킹과 거품문제 해결)

  • This study was undertaken to control sludge bulking and foaming problems at the biological nutrient removal processes. A sewage treatment plant(STP) had a severe sludge settling problem usually over 90% in 30 min settled sludge volume and 300 mL/g in SVI and also thick and heavy brown foam in the secondary clarifiers. Identification study shows that causative filament was Microthrix parvicella which has not been previously reported in Korea. According to the inspection of processes and other related parameters, excessive growth of this particular filament was associated with high SRT caused by lack of proper capacity in sludge treatment line. After providing an extension of the sludge treatment capability, the plant can decrease SRT by wasting more sludge from the processes and also decreasing SS concentration in recycled stream. Subsequently we were able to control growth of M. parvicella. The SVI value was reduced to half of those observed during the severe bulking, but also sludge blanket and its concentration in the clarifier became compact and dense. However, decrease in population of M. parvicella caused increase of total phosphorus concentration in the effluent.

Characterization of Surface Deterioration for Stone Property around the Hyeonleung (Royal Tomb of Joseon Dynasty) in Guri, Korea (구리 현릉 능상석물의 표면 손상특성 평가)

  • The materials of sculptured stone property around the Hyeonleung Royal Tomb in Guri consist mainly of high weathered of granitic rocks with magnetite-series. Deterioration characteristics occurred highly with microorganism, soil inflow and black contaminants at the burial mound zone. As a result of deterioration evaluation, stone surface around the burial mound zone show serious comprehensive damage of soil inflow (50.5%) and lichen coverage (47.6%) which are the major damaging factors, and there are about 8.6% of biological growth noticeably in the north side. Surface contaminants and the discoloration had the complex causes. Gypsum occurred between joints of stones and the major contaminant type, darkening which was analyzed organic bodies caused discoloration. From result of ultrasonic velocity measurements, there are mean value of 2,195 m/s with highly weathered (HW) grade. Most of the stone properties showed 4 to 5 weathered grade. Therefore, it turned out that sculpture stone properties require conservation treatments. To remove soil accelerating damage factors and lichen occupying high percents, the cleaning process is necessary and consolidation, rejoining and filling are needed as well. Also, consideration on removing conservation materials containing Ca and e fflorescence is required and retreatments need to be considered.

The Study of Driving Fatigue using HRV Analysis (HRV 분석을 이용한 운전피로도에 관한 연구)

  • The job of long distance driving is likely to be fatiguing and requires long period alertness and attention, which make considerable demands of the driver. Driving fatigue contributes to driver related with accidents and fatalities. In this study, we investigated the relationship between the number of hours of driving and driving fatigue using heart rate variability(HRV) signal. With a more traditional measure of overall variability (standard deviation, mean, spectral values of heart rate). Nonlinear characteristics of HRV signal were analyzed using Approximate Entropy (ApEn) and Poincare plot. Five subjects drive the four passenger vehicle twice. All experiment number was 40. The test route was about 300Km continuous long highway circuit and driving time was about 3 hours. During the driving, measures of electrocardiogram(ECG) were performed at intervals of 30min. HRV signal, derived from the ECG, was analyzed using time, frequency domain parameters and nonlinear characteristic. The significance of differences on the response to driving fatigue was determined by Student's t-test. Differences were considered significant when a p value < 0.05 was observed. In the results, mean heart rate(HRmean) decreased consistently with driving time, standard deviation of RR intervals(SDRR), standard deviation of the successive difference of the RR intervals(SDSD) increased until 90min. Hereafter, they were almost unchanging until the end of the test. Normalized low frequency component $(LF_{norm})$, ratio of low to high frequency component (LF/HF) increased. We used the Approximate Entropy(ApEn), Poincare plot method to describe the nonlinear characteristics of HRV signal. Nonlinear characteristics of HRV signals decreased with driving time. Statistical significant is appeared after 60 min in all parameters.

A Study on a Multiresolution Filtering Algorithm based on a Physical Model of SPECT Lesion Detectability (SPECT 이상조직 검출능 모델에 근거한 다해상도 필터링 기법 연구)

  • Amultiresolution filtering algorithm based on the physical SPECT lesion detachability provides and optimal solution for SPECT reconstruction problem. Related to the previous study, we estimated the SPECT lesion detection capability by m minimum detectable lesion sizes (MDLSs), and generated m reconstruction filters which are designed to maximize the smoothing effect at a fixed MDLS-dependent resolution level $\frac{MDLS}{4\sqrt{2In2}}$. The proposed multiresolution filtering algorithm used a coarse-to-fine approach for the m-level resolution filter images obtained from these m filters for a given projection image. First, the local homogeneity is determined for every pixel of the filter images, by comparing the local variance value computed in a window centered at the pixel and the mode determined from the distribution of the local variances. Based on the local homogeneity, the pixels declared as homogeneous are chosen from the filter image of the lowest resolution, and for the other pixels the same process is repeated for the higher resolution filter images. For the non-homogeneous pixels after this pixels after this repetition process ends, the pixel values of the highest resolution filter image are substituted. From the results of the simulated experiments, the proposed multiresolution filtering algorithm showed a strong smoothing effect in the homogeneous regions and a significant resolution improvement near the edge regions of the projection images, and so produced good adaptability effects in the reconstructed images.

Giant Colonial Morphology of 10 Clusters of Rapidly Growing Mycobacteria Including Mycobacterium fortuitum-chelonei Complex on 7H10-Crocin Agar Medium (Mycobacterium fortuitum-chelonei complex 및 신속발육 Mycobacteria 10개 균집합체의 거대집락형태)

  • We developed a giant colony test system with rapidly growing mycobacteria by stab-culture with a loopful inoculum of cells into Middlebrook 7H10 agar medium containing soluble extracts of the fruits of Gardenia jasminoides(7H10-crocin agar medium) and assessed the significance of the giant colony test with 28 strains of 10 clusters of rapidly growing mycobacteria classified by the simple biological 5-test characters. Of the 10 clusters of mycobacteria tested, some of strains which belonged to cluster No. 1a, 5a and 11a did grow as gravis types, whereas most of other clusters gave mitis or intermedius types in their colonial sizes at 12 days culture. By this test, pathogenic strains of M. fortuitum-chelonei complex which belonged to cluster No. 5a, b, 7a and 8a, b could be divided into gravis, intermedius and mitis colony types and the gravis ones were characterized by bluish-white "mushroom-shaped" colonies with central complexes in the texture, whereas the intermedius gave grayish-white "flower-shaped" colonies with radiated folds, but without any central complexes. The mitis colonies were characterized by grayish-white smooth or smooth mucoid colonies and were common among the clusters in their shapes. The colony of M. chelonei was bluish-white mitis type and was characterized by its hilly rhizoid colony. The gravis colony of cluster No. 1a identified as M. phlei was characterized by yellow "round straw- mat-shaped" or "chrysanthemum-shaped" colony with whole complexes in the texture, and the gravis colonies of the cluster No. 11a gave grayish-white "flower-shaped" colonies with central stamens, radiating trough and fine cup-shaped strands in the texture. The four colony types of pathogenic species of M. fortuitum-chelonei complex on 7H10-crocin agar medium were distinctive from those of other clusters of rapidly growing mycobacteria and these results indicated that the giant colony test, in conjunction with the simple 5-test characters, would be of value in the differentiation of M. fortuitum complex from other clusters of rapidly growing mycobacteria.
