Lactococcal plasmid pGKV21의 SSB-coated 229-nt ssi signal 상에서 E. coli RNA polymerase에 의한 시발체 RNA 합성 (Primer RNA Synthesis by E. coli RNA Polymerase on the SSB-coated 229-nt ssi Signal of Lactococcal Plasmid pGKV21)

  • 플라스미드 pGKV21에는 229-nt single-strand DNA initiation (ssi) signal이 존재한다. Asymmetric PCR 기법으로 합성된 229-nt ssDNA 단편을 이용하여 실제로 RNA polymerase에 의한 priming ability와 protein interaction을 확인하였다. in vitro primer RNA 합성 실험 결과, 229-nt ssDNA 단편은 filamentous M13 phage의 주형 DNA에서와 비슷한 효율로 시발체 RNA를 합성하였으며, 이 반응은 strand-specific하게 이루어졌다. DNase I footprinting과 gel retardation 실험 결과, RNA polymerase와 SSB 단백질은 229-nt ssDNA 단편에 stable interaction을 하며, 시발체 RNA를 합성하였다. 또한, in vivo 조건 하에서 RNA polymerase의 저해제인 rifampicin을 처리하여 세포내에 ssDNA 중간체가 집적되는 정도를 비교하여 본 결과, 플라스미드 pGKV21은 rifampicin-sensitive RNA polymerase가 상보가닥 합성에 관여 함을 보여 주었다.

SOCS1 counteracts ROS-mediated survival signals and promotes apoptosis by modulating cell cycle to increase radiosensitivity of colorectal cancer cells

  • As negative regulators of cytokine signaling pathways, suppressors of cytokine signaling (SOCS) proteins have been reported to possess both pro-tumor and anti-tumor functions. Our recent studies have demonstrated suppressive effects of SOCS1 on epithelial to mesenchymal signaling in colorectal cancer cells in response to fractionated ionizing radiation or oxidative stress. The objective of the present study was to determine the radiosensitizing action of SOCS1 as an anti-tumor mechanism in colorectal cancer cell model. In HCT116 cells exposed to ionizing radiation, SOCS1 over-expression shifted cell cycle arrest from G2/M to G1 and promoted radiation-induced apoptosis in a p53-dependent manner with down-regulation of cyclin B and up-regulation of p21. On the other hand, SOCS1 knock-down resulted in a reduced apoptosis with a decrease in G1 arrest. The regulatory action of SOCS1 on the radiation response was mediated by inhibition of radiation-induced Jak3/STAT3 and Erk activities, thereby blocking G1 to S transition. Radiation-induced early ROS signal was responsible for the activation of Jak3/Erk/STAT3 that led to cell survival response. Our data collectively indicate that SOCS1 can promote radiosensitivity of colorectal cancer cells by counteracting ROS-mediated survival signal, thereby blocking cell cycle progression from G1 to S. The resulting increase in G1 arrest with p53 activation then contributes to the promotion of apoptotic response upon radiation. Thus, induction of SOCS1 expression may increase therapeutic efficacy of radiation in tumors with low SOCS1 levels.

  • Autosomal dominant polycystic kidney disease (ADPKD) is characterized by formation of multiple fluid-filled cysts that expand over time and destroy renal architecture. The proteins encoded by the $PKD1$ and $PKD2$ genes, mutations in which account for nearly all cases of ADPKD, may help guard against cystogenesis. Previously developed mouse models of $PKD1$ and $PKD2$ demonstrated an embryonic lethal phenotype and massive cyst formation in the kidney, indicating that $PKD1$ and $PKD2$ probably play important roles during normal renal tubular development. However, their precise role in development and the cellular mechanisms of cyst formation induced by $PKD1$ and $PKD2$ mutations are not fully understood. To address this question, we presently created $Pkd2$ knockout and $PKD2$ transgenic mouse embryo fibroblasts. We used a mouse oligonucleotide microarray to identify messenger RNAs whose expression was altered by the overexpression of the $PKD2$ or knockout of the $Pkd2$. The majority of identified mutations was involved in critical biological processes, such as metabolism, transcription, cell adhesion, cell cycle, and signal transduction. Herein, we confirmed differential expressions of several genes including aquaporin-1, according to different $PKD2$ expression levels in ADPKD mouse models, through microarray analysis. These data may be helpful in $PKD2$-related mechanisms of ADPKD pathogenesis.

Chlorosis of Ogura-CMS Brassica rapa is due to down-regulation of genes for chloroplast proteins

  • Cytoplasmic male sterility (CMS) is a maternally inherited trait leading to loss of the ability to produce fertile pollen and is extensively used in hybrid crop breeding. Ogura-CMS was originally generated by insertion of orf138 upstream of atp8 in the radish mitochondrial genome and transferred to Brassica crops for hybrid breeding. Gene expression changes by dysfunctional mitochondria in Ogura-CMS result in pollen developmental defects, but little is known about gene expression patterns in vegetative tissue. To examine the interaction between nuclear and organellar regulation of gene expression, microarray and subsequent gene expression experiments were conducted with leaves of $F_1$ hybrid Chinese cabbage derived from self-incompatible (SI) or Ogura-CMS parents (Brassica rapa ssp. pekinensis). Out of 24,000 genes deposited on a KBGP24K microarray, 66 genes were up-regulated and 26 genes were down-regulated by over 2.5 fold in the CMS leaves. Up-regulated genes included stress-response genes and mitochondrial protein genes, while genes for ascorbic acid biosynthesis and thylakoid proteins were down-regulated. Most of the major component genes for light reactions of photosynthesis were highly expressed in leaves of both SI and CMS plants, but most of the corresponding proteins were found to be greatly reduced in leaves of CMS plants, indicating posttranscriptional regulation. Reduction in thylakoid proteins and chlorophylls led to reduction in photosynthetic efficiency and chlorosis of Ogura-CMS at low temperatures. This research provides a foundation for studying chloroplast function regulated by mitochondrial signal and for using organelle genome introgression in molecular breeding.

Implementation of the Wearable Sensor Glove Using EDA Sensor and Conducting Fabric

  • The wearable sensor glove was developed using EDA sensors and conducting fabric. EDA(Electro-dermal Activity) signal is an electric response of human skin. There are SIL(Skin Impedance Level) and SIR(Skin Impedance Response) in EDA. SIL consists mostly of a DC component while SIR consists of an AC component. The relationship between drowsiness and the EDA signal is utilized. EDA sensors were made using a conducting fabric instead of AgCl electrodes, for a more suitable, more wearable device. The EDA signal acquisition module was made by connecting the EDA sensor gloves through conductive fabric lines. Also, the EDA signal acquisition module can be connected to a PC that shows the results of the EDA signal processing analysis and gives proper feedback to the user. This system can be used in various applications to detect drowsiness and prevent accidents from drowsiness for automobile drivers.

  • EEG signal is analyzed by two methods, analysis by visual inspection of EEG recording sheets and analysis by quantative method. Generally visual inspection method is used in the clinical field. But this method has its limitation because EEG signal is random signal. Therefore it is necessary to analyze EEG signals quantatively to obtain more precise and objective information of neural and brain. In this paper, power spectrum of EEG signal was estimated by AR(AutoRegressive) model in the frequency domain. This process is useful as a preprocessing stage for tomographic brain mapping (TBM) at each frequency, band. As a method for estimating power spectral density of EEG signals, periodogram method, autocorrelation method. covariance method, modified covariance method, and Burg method are tested in this paper.

Treatment of Bone Repair by Inductively Magnetic Fields

  • An inductively coupled magnetical signal (pulse wave, 0.7 to 60Hz, eighteen volts peak to Peak) that was applied non-invasively on the skin surface overlying the approximate site(measure position). In the group with unipolar pulse signal currents produced smaller than in the group with bipolar pulse signal. The signal was transmitted to the active coil, including a time-varying magnetic field: this in turn induced a the-varying electrical field in the field in the bone. It is very important to determine system parameters due to treatment time(healing) and the simplicity. This paper investigation was designed to compare the relative effects of pulsed unipolar currents with the effects of an identical pulsed bipolar currents. Since Inductive coupling is non-invasive and involves portable equipment, it is easy to apply and requires precise localization, it has distinct advantages and field characteristics along the bone for each different signal.

  • This paper proposes a method called linear prediction (a high performant technique in digital speech processing) for analyzing digital ECG signals. There are several significant properties indicating that ECG signals have an important feature in the residual error signal obtained after processing by Durbin's linear prediction algorithm. The ECG signal classification puts an emphasis on the residual error signal. For each ECG's QRS complex. the feature for recognition is obtained from a nonlinear transformation which transforms every residual error signal to set of three states pulse-cord train relative to the original ECG signal. The pulse-cord train has the advantage of easy implementation in digital hardware circuits to achive automated ECG diagnosis. The algorithm performs very well feature extraction in arrythmia detection. Using this method, our studies indicate that the PVC (premature ventricular contration) detection has a at least 90 percent sensityvity for arrythmia data.

A Study on a Healthcare System Using Smart Clothes

  • Being able to monitor the heart will allow the diagnosis of heart diseases for patients during daily activities, and the detection of burden on the heart during strenuous exercise. Furthermore, with the help of U-health technology, immediate medical action can be taken, in the case of abnormal symptoms of the heart in daily life. Therefore, it appears to be necessary to develop the corresponding technology to monitor the condition of the heart daily. In this study, a novel wearable smart system was proposed, to monitor the activity of the heart in daily life, and to further evaluate the rhythm of arrhythmia. The wearable system includes three modified bipolar conductive fiber electrodes in the chest part, which can resolve the reduction problem of the magnitude of the signal, by magnifying the signal and removing the noise, to obtain high affinity and validity for medical-type usage (<0.903%). The biological signal acquisition and data lines, and the signal processing engine and communication consist of a conductive ink, and the pic18 and ANT protocol nRF24AP2, respectively. The proposed algorithm was able to detect a strong ECG, signal and r-point passing over the noise. The confidence intervals were 96 %, which could satisfy the requirement to detect arrhythmia under the unconstrained conditions.

위전도신호의 측정 및 분석시스템 개발 (Development of Detection and Analysis System for Electrogastrographic Signal)

  • 위전도는 피부전극을 이용하여 위에서 발생하는 전기신호를 측정하여 위운동상태를 측정하는 비관혈적인 검사방법이다. 본 연구에서는 임상적으로 유용한 위전도신호 측정 및 분석시스템을 개발하였으며, 개발시스템은 크게 하드웨어(생체증폭기, 필터)와 소프트웨어(유저인터페이스, 분석알고리듬, 환자데이터베이스)로 구성된다. 개발시스템의 생체신호증폭기는 신호분석에 용이하도록 3채널로 구성되어 있으며, 신호의 저장 및 분석은 PC에서 수행하도록 구성되어 있다. 위수축 정보의 획득을 위해서 분석신호는 16Hz로 샘플링하였으며, 위전도신호분석은 임상이용에 유용한 여러 가지 분석 파라메터를 추출할 수 있도록 하였다. 개발된 시스템은 동물실험을 통하여 성능을 평가하였으며, 현재 환자를 대상으로 임상시험이 진행중이다.
