Development of Adsorbent for Heavy Metals by Activation of the Bark (활성화 수피를 이용한 중금속 흡착제 개발)

  • The objective of this research was to develop the adsorbent far heavy metals by activating the bark sample. Barks from pine tree with diameters of $2{\sim}4\;mm$ were activated in the muffle furnace under a high relative humidity condition at temperatures of $600{\sim}900^{\circ}C$. The removal efficiency of the activated bark (ACTBARK) for Cu and Cd was temperature dependent showing the order of $900^{\circ}C$ > $800^{\circ}C$ > $700^{\circ}C$ > $600^{\circ}C$. The critical temperature was considered to be $900^{\circ}C$ to become an efficient adsorbent for Cu and Cd. The bark samples activated at temperatures lower than $700^{\circ}C$ showed a less removal efficiency than the crude bark. The ACTBARK activated at $900^{\circ}C$ removed more Cu and Cd from solution than the commercial activated carbon and charcoal. The ACTBARK (activated at $900^{\circ}C$) adsorbed all of the Cu and Cd in solution with concentrations less than 150 mg/L. The selectivity of the ACTBARK was in the order of Cu > Zn > Ni > Pb > Fe > Cd > Mn.

  • The purification methods of bacterial protease have been published by many workers, especially by the using of ion exchange resins. But in the practical application to obtain a comparatively purified enzyme, the known methods do not give always a satisfiable results. Here we developed an industrially applicable method for purification of bacterial protease with the using of tannin. By the adaptation of the optimal conditions of this method on the purification, a 150000 unit/g. (Fuld Gross unit) of protease sample could obtained.

  • With an attempt to obtain a glucose isomerizing enzyme producing microorganism, one hundred and thirty-three strains of microorganism were isolated from soil samples. After screening, a strain K-17 which belonging to actinomyces family, was finally selected. Using this strain of K-17, sugars produced from glucose by the reaction of sugar isomerizing enzyme were tested with paper chromatography. Only a kind of resulting sugar, fructose, was detected from enzyme reaction sample and other sugars were never detected. By these results, the enzyme produced by strain K-17 is classified as a glucose isomerase.

Physiological Characteristics of Biosurfactant-Producting Bacillus subtilis TBM 3101 (Biosurfactant를 생산하는 Bacillus subtilis TBM 3101의 생리학적 특성)

  • A biosurfactant-producing strain, Bacillus subtilis TBM 3101 was isolated from the soil sample at Tae-Baek Mountain through an antifungal test and emulsification assessment. The strain was assessed, regarding to the microbial growth, by physical and chemical test, surface tension, emulsification activity and stability. The surface tension of the isolate sharply decreased to the minimum 29mN/m at 48 h growth. Of note, its emulsification was stabilized to the highest degree when tributyrin was utilized as a substrate, indicating that in comparison to a variety of synthetic surfactants, the biosurfactant produced by the isolate was significantly similar to synthetic surfactant, tween 20. In addition, the biosurfactant showed high emulsification activity when soybean oil, crude oil and tetradecane were used as a substrate. Thus, these studies could contribute to the detection and development of biosurfactant beneficial to the environment and humans.

First Report of Tobacco mild green mosaic virus Infecting Pepper in Korea

  • A rod-shaped virus was isolated from pepper showing mild mosic during the winter growing seasons of 2001 and 2002 in Korea. Based on its biological reactions, serological relationships, reverse transcription-poly-merase chain reaction (RT-PCR) using specific primers, and nucleotide sequence analysis of coat protein (CP) gene, the isolated virus was identified as Tobacco mild green mosaic virus (TMGMV) and designated as Korean pepper isolate (TMGMV-KP). Crude sap from infected tissue was mechanically transmitted to various indicator plants, which produced characteristic symptoms of tobamovirus infection. However, no symptom was observed in Gomphorena globosa. In RT-PCR assays with specific primers toy respective detection of TMGMV, Tobacco mosaic virus (TMV), Pepper mild mottle virue (PMMoV), and Tomato mosaic virus (ToMV), a single strong band of about 500 bp in length was produced from the sample used only with TMGMV primers. The amplified DNA was cloned and the nucleotide sequence was determined. Sequence comparisons with the CP gene of other tobamoviruses indicated that TMGMV-KP shared 99.3% identity with TMGMV Japanese isolate and only 59.1, 58.6, and 58.1% identity with TMV, PMMoV and ToMV, respectively. This is the first report of TMGMV in Korea.

A Novel Recombined Potato virus Y Isolate in China

  • This study reports the findings of a distinct Potato virus Y (PVY) isolate found in Northeast China. One hundred and ten samples (leaves and tubers) were collected from potato plants showing mosaic symptoms around the city of Harbin in Heilongjiang province of China. The collected tubers were planted and let to grow in a greenhouse. New potato plants generated from these tubers showed similar symptoms, except for one plant. Subsequent serological analyses revealed PVY as the causing agent of the disease. A novel PVY isolate (referred to as HLJ-C-44 in this study) was isolated from this sample showing unique mild mosaic and crisped leaf margin symptoms. The complete genome of this isolate was analyzed and determined. The results showed that HLJ-C-44 is a typical PVY isolate. Phylogenetic analysis indicated that this isolate belongs to the N-Wi strain group of PVY recombinants ($PVY^{N-Wi}$) and also shared the highest overall sequence identity (nucleotide and amino acid) with other members of this strain group. However, recombination analysis of isolate HLJ-C-44 revealed a recombination pattern that differed from that of other $PVY^{N-Wi}$ isolates. Moreover, biological assays in four different potato cultivars and in Nicotiana tabacum also revealed a different phenotypic response than that of a typical $PVY^{N-Wi}$ isolate. This data, combined, suggest that HLJ-C-44 is a novel PVY recombinant with distinct biological properties.

Osteoblast and Bacterial Culture from Cryopreserved Skull Flap after Craniectomy : Laboratory Study

  • Objective : Cranioplasty using a cryopreserved skull flap is a wide spread practice. The most well-known complications of cranioplasty are postoperative surgical infections and bone flap resorption. In order to find biological evidence of cryopreserved cranioplasty, we investigated microorganism contamination of cryopreserved skulls and cultured osteoblasts from cryopreserved skulls. Methods : Cryopreserved skull flaps of expired patients stored in a bone bank were used. Cryopreserved skulls were packaged in a plastic bag and wrapped with cotton cloth twice. After being crushed by a hammer, cancellous bone between the inner and outer table was obtained. The cancellous bone chips were thawed in a water bath of $30^{\circ}C$ rapidly. After this, osteoblast culture and general microorganism culture were executed. Osteoblast cultures were done for 3 weeks. Microorganism cultures were done for 72 hours. Results : A total of 47 cryopreserved skull flaps obtained from craniectomy was enrolled. Of the sample, 11 people were women, and the average age of patients was 55.8 years. Twenty four people had traumatic brain injuries, and 23 people had vascular diseases. Among the patients with traumatic brain injuries, two had fracture compound comminuted depressed. The duration of cryopreservation was, on average, 83.2 months (9 to 161 months). No cultured osteoblast was observed. No microorganisms were cultured. Conclusion : In this study, neither microorganisms nor osteoblasts were cultured. The biological validity of cryopreserved skulls cranioplasty was considered low. However, the usage of cryopreserved skulls for cranioplasty is worthy of further investigation in the aspect of cost-effectiveness and risk-benefit of post-cranioplasty infection.

Complete genome sequencing of Pseudomonas fluorescens NBC275, a biocontrol agent against fungal pathogens of plants and insects (식물 및 곤충의 곰팡이 병원균에 항균력을 가진 Pseudomonas fluorescens NBC275 균주의 유전체 염기서열)

  • Pseudomonas fluorescens NBC275 (Pf275) isolated from soil sample collected at riverside of Nakdonggang showed antagonistic activity against fungal pathogens of plants and insects. Here we present complete genome sequence of Pf275. The genome comprises of 6,610,362 bp with GC content of 60.9%, which includes 5,869 predicted protein-coding genes, 16 rRNAs, and 65 tRNAs. Genome analysis revealed gene clusters encoding antimicrobial secondary metabolites such as pyoverdine, 2, 4-diacetylphloroglucinol, and phenazine, which are known to play essential roles in biocontrol of diseases.

Factors Associated with Smoking Frequency and Suicidal Attempt in Korean Adolescent Smokers (흡연 청소년의 흡연 빈도와 자살 시도에 영향을 미치는 요인)

  • Objectives : Among adolescents, the proportion of daily smoking tends to increase with age, and does not return to the previous level. Therefore, we examined the factors associated with smoking frequency in a nationwide sample of Korean adolescents to help prevent the progression of smoking. Methods : Among 127,804 adolescents who participated in the 2016 and 2017 Korean Youth Risk Behavior Web-Based Survey, data from 7,781 adolescents who replied that they currently smoked were analyzed. According to the frequency of cigarette smoking, participants were classified into daily and non-daily smokers. Participants' socio-demographic characteristics, smoking behavior, and psychological problems including perceived stress, depressive mood, and suicidality were assessed. Results : There were 3,627(47.4%) daily smokers and 4,154(52.6%) non-daily smokers. Low parental educational level, and early smoking experience were positively associated with daily smoking. High academic achievement, and high socio-economic status were negatively associated with daily smoking. Daily smoking was positively associated with lower happiness, lower sleep satisfaction, and severe stress during the past 12 months. Factors affecting suicide attempts of smokers were sex, residential type, socioeconomic status, and early smoking experience. Conclusion : The results indicated that socially disadvantaged families, and smoking experience with young age influenced daily smoking among Korean adolescents. Daily smoking was associated with psychological problems including lower happiness, lower sleep satisfaction, and severe stress. Early smoking experience influenced suicidal attempts among Korean adolescent smokers. Early intervention for smoking cessation in this high-risk group may contribute to prevention of these psychiatric complications of smoking.

Correlation between Knowledge, Attitude, and Compliance of Preventive Behaviors regarding Middle East Respiratory Syndrome among Nursing Students (간호대학생의 중동호흡기증후군에 대한 지식, 태도 및 예방행위 실천 간의 관계)

  • Purpose: This study aimed to investigate the relationships between the level of knowledge, attitude and compliance of preventive behaviors of the Middle East respiratory syndrome (MERS) among nursing students. Methods: The study sample consisted of 219 nursing students. Through the use of a structured questionnaire, data collection was conducted from June 1st to June 30th 2015. The descriptive statistics, independent t-tests and Pearson's correlation coefficient were employed to analyze the data. Results: The score of the MERS-related knowledge was 9.15 out of 13. The participants had a positive attitude toward the MERS. The level of MERS-related knowledge was high in the case of temporary school closure or break in clinical practice because of the clinical training in hospitals that MERS occurred (t= 2.42, p= .016). The compliance level of the preventive behavior for MERS was high in female students (t= -2.11, p= .036), in the case of temporary school closure or break in clinical practice due to the clinical training in hospitals that MERS occurred (t = 3.29, p= .001), and in students that had MERS-related education for prevention (t = 2.80, p= .006). The MERS-related knowledge was positively correlated with the level of compliance for preventive behavior (r= .18, p= .009). Conclusion: To prevent MERS infection in nursing students, the level of knowledge on MERS should be enhanced so that they can practice preventive behaviors against it. Additionally, the MERS infection control education should include etiology and treatment products based on the MERS response guideline issued by the Centers for Korea Disease Control and Prevention.
