  • Bacteria and fungi are fundamental biotic components of natural biogeochemical cycles for metals and metalloids, and play important roles in dissolution, precipitation, oxidation and reduction processes. Some processes catalyzed by microorganisms also have important applications in environmental biotechnology in the areas of ore leaching and bioremediation.

수용액 중 영가 철(Zero-Valent Iron)의 특성에 따른 Cr(VI)의 환원 효율 비교 (Reduction Efficiency of Cr(VI) in Aqueous Solution by Different Sources of Zero-Valent Irons)

  • 실험에 사용된 영가 철의 평균 입도는 $85.55{\sim}196.46{\mu}m$의 범위를 나타내었고 비표면적은 $0.055{\sim}0.091m^2/g$으로 나타났다. 영가 철 중 J(93%) 및 PU(88%)를 제외한 PA, F, S, J1는 철을 98% 이상 함유하는 것으로 평가되었다. 영가 철에 의한 Cr(VI)의 환원은 영가 철의 종류에 따라 달랐으며, J 및 PU의 경우 3시간 이내에 98% 이상의 Cr(VI)을 제거하였다. 그러나 PA, F, S, J1의 경우 반응 48시간 동안 각각 74, 65, 29, 11%의 Cr(VI) 만을 제거하였으며 이때 환원 효율은 J > PU > PA > F > S > J1 순으로 나타났다. 영가철에 의한Cr(VI)의 환원 과정에서 용액의 pH는 반응초기 3시간 동안 급격히 증가하였으며 이후 안정화되는 경향을 나타내었다. 영가 철의 종류에 따른 pH의 변화는 PU > J > PA > F > S > J1 순으로 나타났으며 Cr(VI)의 환원효율이 큰 영가 철에서 pH의 상승 폭도 크게 나타나는 것으로 평가되었다. 영가 철의 종류에 따른 산화환원전위 Eh의 변화는 pH와는 반대의 경향을 나타내었으며 Cr(VI)의 환원율이 클수록 Eh의 감소율은 증가하였다. 유기화합물의 경우 반응효율이 영가 철의 물리적 특성에 의해 주로 지배되는 것으로 알려져 있으나 Cr(VI)의 경우 화학적 특성이 반응효율을 지배하는 것으로 판단되었다. 본 실험에 사용된 6종류의 상용 영가 철 중에는 J와 PU가 Cr(VI)의 환원에 가장 효과적인 것으로 조사되었다.

  • 이 논문에서는 식품 또는 식품원료 중의 aflatoxin에 관한 오염현황, 최신 분석법 그리고 저감화 시킬 수 있는 방안에 대해 조명하였다. Aflatoxin은 여러 곰팡이가 생성하는 2차 대사산물로서 곰팡이 독소 중 가장 독성이 강한 것으로 알려져 있다. 발암성을 포함한 독성 관계로 aflatoxin은 인간의 건강 측면 뿐 아니라 식품을 포함한 산업전반에 경제적으로 큰 영향을 끼치고 있다. 우리나라와 같은 식량 수입국에서는 식품의 안전성 확보를 위해 노출량 조사, 위해평가 및 관리가 체계적으로 이루어져야 하며 아울러 새로운 aflatoxin 분석 기술의 개발과 독성 저감화 기술의 개발이 병행되어야 한다고 판단된다.

Marine Ecosystem Response to Nutrient Input Reduction in Jinhae Bay, South Korea

  • We study on the dynamic interaction with a simulated physical-biological coupled model response to nutrient reduction scenario in Jinhae Bay. According to the low relative errors, high regression coefficients of COD and DIN, and realistic distribution in comparison to the observation, our coupled model could be applicable for assessing the marine ecosystem response to nutrient input reduction in Jinhae Bay. Due to the new construction and expansion of sewage treatment plant from our government, we reduce 50% nutrient inputs near Masan Bay and sewage treatment plant. COD achieves Level II in Korea standard of the water quality from the middle of the Masan Bay to all around Jinhae Bay except the inner Masan Bay remaining at Level III. When our experiment reduces 50% nutrient inputs near Masan Bay and Dukdong sewage treatment plant simultaneously, COD decreases to about 0.1-1.2 mg/L $(128^{\circ}30'{\sim}128^{\circ}40'\;E,\;35^{\circ}05'{\sim}35^{\circ}11'\;N)$. The COD from the middle of the Masan Bay to Jinhae Bay achieves Level II.

The Effects of Light Intensity, Inoculum Size, and Cell Immobilisation on the Treatment of Sago Effluent with Rhodopseudomonas palustris Strain B1

  • A study was carried out to determine a suitable light intensity and inoculum size for the growth of Rhodopseudomonas palustris strain B1. The pollution reduction of sago effluent using free and immobilised R. palustris cells was also evaluated. The growth rate in glutamatemalate medium was highest at 4 klux compared to 2.5 and 3 klux. The optimal inoculum size was 10% (v/v). Both the COD and BOD of the sago effluent were reduced by 67% after three days of treatment. The difference in biomass production or BOD and COD removal with higher inoculum sizes of 15 and 20% was minimal. This could be attributed to limited nutrient availability in the substrate. The use of immobilised cells of R. palustris reduced the pollution load 10% less compared to pollution reduction by free cells. Hence, there was no significant difference in using free or immobilised cells for the treatment of sago effluent.

자연발효 과정에서 인디고에 환원력을 지닌 미생물 커뮤니티 분석과 농화배양 (Analysis and Enrichment of Microbial Community Showing Reducing Ability toward indigo in the Natural Fermentation of Indigo-Plant)

  • Indigo is utilized in various industries including textile dyeing, cosmetics, printing and medicinal products and its reduced form, leuco-indigo, is mainly used in these process. Chemical reducing agent (sodium dithionite, sodium sulfide, etc.) is preferred to use for the formation of leucoindigo in industry. In traditional indigo fermentation process, microorganisms can participate in the reduction of indigo and thus it has been known to reduce environmental pollution and noxious byproducts. However, in fermentation method using microorganisms it is difficult to standardize large scale production process due to low yield and reproducibility. In this study, we attempted to develop the indigo reduction process using microbial flora which was isolated from naturally fermented indigo vat or deduced by metagenomic approach. From the results of library analyses of PCR-amplified 16S rRNA genes from the traditional indigo fermentation vat sample (metagenome), it was confirmed that Alkalibacteriums (71%) was distinctly dominant in population. Some strains were identified after confirming that they become pure culture in nutrient media modified slightly. Four strains were separated in this process and each strain showed obvious reducing ability toward indigo in dyeing test. It is expected that the analyzed results will provide important data for standardizing the natural fermentation of indigo and investigating the mechanism of indigo reduction.

  • Geosynthetic Reinforcements - membrane drawn type, warp/knitted type, junction bonded type and composite type geogrids, strip type reinforcement - were used to compare the long-term perfor-mance by total factor of safety with reduction factors during service periods. To evaluate the reduction factors, wide-width tensile property, installation damage, creep deformation, chemical and biological degradation tests were performed. Long-term design strengths of geosynthetic reinforcements were calculated by using GRI standard Test Method GG4.

손 소독제 및 소독방법에 따른 외과적 손 소독 효과 (Comparison of Surgical Hand Scrub Formulations and Scrub Methods for Antimicrobial Efficacy)

  • Purpose: The purpose of this study was to compare three surgical scrub methods-4% chlorhexidine gluconate(CHG) with brush, 4% CHG without brush, and waterless scrub using 1% CHG and 61% ethanol combination(alcohol-based agent)-for antimicrobial efficacy. Method: "Glove-juice technique" was used to evaluate microbial hand counts before surgical scrubs, 1 min and 3 hr after surgical scrubs. Result: Waterless scrub using CHG and ethanol combination resulted in a 4.8-log reduction at 1 min and 4.0-log reduction at 3 hr. CHG without brush resulted in a 4.7-log reduction at 1 min and 3.3-log reduction at 3 hr. The traditional scrub using CHG with brush resulted in a 3.6-log reduction at 1 min and 0.8-log reduction at 3 hr. The waterless scrub and CHG without brush showed significantly higher log reduction(p<.05) than CHG with brush at 1 min and 3 hr after surgical scrub. Waterless scrub showed greater log reductions than CHG without brush, but there was no statistical difference between the two groups. Conclusion: Waterless scrub using alcohol-based agent showed more persistent and immediate antimicrobial efficacy than either CHG with brush or CHG without brush.

Self-Sufficient Catalytic System of Human Cytochrome P450 4A11 and NADPH-P450 Reductase

  • The human cytochrome P450 4A11 is the major monooxygenase to oxidize the fatty acids and arachidonic acid. The production of 20-hydroxyeicosatetraenoic acid by P450 4A11 has been implicated in the regulation of vascular tone and blood pressure. Oxidation reaction by P450 4A11 requires its reduction partners, NADPH-P450 reductase (NPR). We report the functional expression in Escherichia coli of bicistronic constructs consisting of P450 4A11 encoded by the first cistron and the electron donor protein, NPR by the second. Typical P450 expression levels of wild type and several N-terminal modified mutants was observed in culture media and prepared membrane fractions. The expression of functional NPR in the constructed P450 4A11: NPR bicistronic system was clearly verified by reduction of nitroblue tetrazolium. Membrane preparation containing P450 4A11 and NPR efficiently oxidized lauric acid mainly to $\omega$-hydroxylauric acid. Bicistronic coexpression of P450 4A11 and NPR in E. coli cells can be extended toward identification of novel drug metabolites or therapeutic agents involved in P450 4A11 dependent signal pathways.

Fluorescent siderophore 생산균주, TS3-7에 의한 풋마름병 발병 억제 (Suppression of Bacterial Wilt with Fuorescent Pseudomonads, TS3-7 strain)

  • Among the root colonizing and plant growth promoting bacteria isolated from the bacterial wilt suppressive soil, five strains were detected to produce siderophores by CAS agar assay. The most effective isolate, TS3-7 strain induced significant suppression of bacterial wilt disease in tomato and pepper plants. Seed treatment followed by soil drench application with this strain resulted in over 80% reduction of bacterial wilt disease compared with the control. Significant disease suppression by TS3-7 strain was related to the production of siderophore. Besides iron competition, induction of resistance of the host plant with siderophore was suggested to be another mode of action that suppress bacterial wilt, based on the lack of direct antibiosis against pathogen in vitro. According to Bergey's Manual of Systemic Bacteriology and 16S rDNA sequence data, TS3-7 stain was identified as Pseudomonas sp. TS3-7.
