Characteristics of Sophorolipid as an Antimicrobial Agent

  • Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm, respectively. Also, 100 ppm of sophorolipid inhibited $50\%$ of cell growth of plant pathogenic fungus, Botrytis cineria. However, sophorolipid showed no effect on Escherichia coli, indicating that its selective antimicrobial activity depended on the cell wall structure. Treatment of B. subtilis with sophorolipid increased leakage of intracellular enzyme, malate dehydrogenase, indicating a possible interaction of sophorolipid with a cellular membrane. Comparing lactone-type and acid-type sophorolipids, the former showed a higher antimicrobial activity. Supplementing other surfactants showed no significant effects on the antimicrobial activity. Animal study showed that 5 g of sophorolipid per kg body weight by oral administration caused no toxicity, and sophorolipid induced no irritation on the skin. These results show potential use of sophorolipid as an active ingredient in healthcare products.

  • Hop cones (Humuli Strobili) are the female inflorescences of hop plants (Humulus lupulus L.) belonging to the family Cannabaceae. They have been used as herbal remedies to treat mood and sleep disturbances, and mainly to add as a bittering ingredient in brewing process. Considerable interests on pharmacological and biological activities of hop cones have been focused on their major constituents, namely, phloroglucinols (humulone, lupulone), terpenes (myrcene, humulene), and prenylated flavonoids (xanthohumol, isoxanthohumol, 6-prenylnaringenin, and 8-prenylnaringenin). The present review describes and discusses biological activity profiles of these major compounds in the hop cones.

  • Aloe vera L.(Aloe barbadensis Miller) has been used for many centuries for various medical, cosmetic and neutraceutical purposes. The gel of A. vera was reported to exert numerous biological activities including wound healing, immunomodulatory, anti-cancer, anti-inflammatory, anti-bacterial, anti-viral, anti-diabetic, and anti-psoriasis activities. Acemannan, found predominantly in the inner leaf gel of A. vera, has been identified as the main active ingredient exerting diverse biological activities. In this review, we summarized the recent findings on the immunomodulatory activities of acemannan. Studies that used purified acemannan demonstrate that acemannan exerts immune-stimulating, anti-cancer, anti-viral, and hematopoiesis-increasing activities. In addition, it is being clear that most of these activities of acemannan are mediated primarily by activating professional antigen presenting cells such as macrophages and dendritic cells.

Cytotoxicity and Antioxidant Activities of the Yeonkyo-san and Its ingredients (연교산과 구성약재에 대한 항암 및 항산화효과)

  • Objectives : The antioxidant activities of Yeonkyo-san and its ingredient were investigated for industrial application. Methods : We experimented radical scavenging effect, superoxide dismutase and cytotoxicity effect of various cancer cell. Results : The result were obtained as follows : Primary testing of cytotoxicity of Yeonkyo-san and ingredient was done for G361. B16F10, MDA and A549 cell lines. Yeonkyo-san and ingredient water extract, ethanol extract showed cytotoxicity of over 90% respectively against cell line. The electron donating ability(EDA) of water extract from the Yeonkyo-san was higher than 60%, ethanol extract from the Yeonkyo-san was over 80% at 1000 ppm. The electron donating ability(EDA) of water extract and ethanol extract from the Yeonkyo-san ingredients were increased as well. SOD-like activity was high as 75% in 700 ppm, it was increased. All the other samples showed less than 40% SOD-like activity. Conclusions : The results indicated that water extract and ethanol extract of Yeonkyo-san and ingredient can be used as a natural ingredient with biological functions in cosmetic and food composition.

New Anti-aging & Moisturizer Ingredients of Exopolysaccharides by Grifola frondosa

  • In this study, in an attempt to search for functional cosmetic ingredients from higher fungal, we have produced exopolysaccharides (GF-l, approximately carbohydrate 75%, protein 25%) and polysaccharide (GF-2) of mycelium extract, by submerged culture of Grifolafrondosa. For applications in anti-aging cosmetic field, we investigated the diverse biological activities. Antioxidant activity and inhibition of Matrixmetalloproteinases (MMPs) were investigated enzymatic assays by measuring the superoxide scavenging activity using xanthine-xanthine oxidase system and the proteolytic activity of MMPs using EnzChek Collagenase/Gelatinase kits, respectively. GF-l polysacchairde showed inhibition of superoxide radical by 90% at a concentration of 0.2% (w/v) and inhibition of collagenase by 45% at 0.2% (w/v). GF-2 polysaccharide of mycelium extract also exhibited good antioxidant activity. However, MMPs inhibition activity was relatively lower level compared to GF-l polysaccharides. The treatment of human dermal fibroblast (HDF) with GF-l and GF-2 polysaccharides increased the proliferation of fibroblast by approximately 23-25% at a concentration of 0.5% (w/v), also showed collagen synthesis increase in HDF by about 50% at 0.5% (w/v) compared to that of untreated control. We also report the moisturizing effects of polysaccharides in cosmetic products (O/W emulation) and its own ingredient, in vitro and in vivo. The GF-1 polysaccharide showed higher moisturizing ability than sodium hyaluronate, which is the most commonly used moisturizers ingredient. These results suggest the GF-l polysaccharide, protein-bound polysaccharide, may be used as an ingredient for new moisturizing and anti-aging cosmeceuticals.

Comparative Study of Protocorm-like body and Multiple Shoots from Dendrobium Candidum on Biological Activities (철피석곡의 원괴체상구체 및 다신초 추출물의 생리활성에 관한 비교 연구)

  • Dendrobium candidum is one of the well-known orchid on traditional and rare medicinal herb in China. We artificially cultured protocorm-like body and multiple shoots using the bioreactor culture system from Dendrobium candidum and experimented an efficacy as a cosmetic ingredient for skin care. Several studies previously have reported anti-tumor, wound healing and immunological function of Dendrobium candidum but other efficacies were not well known. In the present study, we investigated the cosmetic efficacy to know applicable value of protocorm-like body and multiple shoots cultured from Dendrobium candidum as a cosmetic ingredient. The biological activities of extracts from protocorm-like body, multiple shoots, aerial part and underground part of Dendrobium candidum were investigated. Results were found that extracts of protocorm-like body are superior to other extracts (underground part, aerial part and multiple shoots extracts) on anti-oxidant effect. Also, protocorm-like body extract contained the phenolic and flavonoid compounds more than aerial part, underground part and multiple shoots extracts. In addition, we investigated skin whitening effect related to whitening of skin. In tyrosinase activity and melanin synthesis assay, multiple shoots extract is superior to other extracts (aerial part, underground part and protocorm-like body), on inhibitory effect of tyrosinase activity and melanin synthesis respectively. These results indicate that the protocorm-like body and multiple shoots extracts cultured from Dendrobium candidum presents a new possibility of being applicable to skin improvement as a cosmetic ingredient.

Protection of Infection and Eradication Activity of Culture Product by Pediococcus pentosaceus CBT SL4 Showing Antimicrobial Activity against Helicobacter pylori (Helicobacter pylori에 대한 항균활성을 나타내는 Pediococcus pentosaceus CBT SL4 배양물의 감염방어 및 제균활성)

  • New food ingredient was developed to eradicate and protect against re-infection of Helicobacter pylori in fermentation broth of lactic acid bacteria (LAB) showing antimicrobial activity against pathogenic microorganisms such as H. pylori and Listeria monocytogenes. LAB strain CBT SL4 was identified as Pediococcus pentosaceus by 16S rDNA sequencing and its culture broth showed antimicrobial activity of 800 AU/mL against H. pylori in optimized fermentation process. Using thin layer concentration system and spray-typed fluid bed drier system, concentrated powder product showing activity of 12,800 AU/g was harvested. Product showed eradication and protection activities against H. pylori infection on feeding test (50 AU/day) using Mongolian gerbil infection model. After 4 weeks therapy of 8,000 AU/day, ${\Delta}13CO_2$ level (DOB30) decreased about 40% in urea breath test on patient with H. pylori infection. Result show concentrated culture product of P. pentosaceus CBT SL4 has eradicating and protecting activities against H. pylori infection and can be used as food-active ingredient for prevention of gastric and duodenum ulcer caused by H. pylori.

Oxidants and Antioxidants Associated with Commercial Pickle Products and Ingredients (상업적인 pickle product와 ingredient의 oxidant와 antioxidant로서의 역할)

  • Investigations of the effects of pickle product ingredients on lipoxygenase (LOX) and methemoglobin (MHG, a nonenzymatic oxidant) catalyzing oxidation of linolenic acid were conducted. In addition, activities of LOX, peroxidase (POD) and catalase (CAT) in dry spices used in pickle products were determined. Some commercial pickle brines were observed to inhibit oxidation of linolenic acid by LOX and MHG. The ingredients in pickle products, such as dill oil emulsion, onion concentrate, oil cassia, polysorbate 80 and turmeric acid, reduced LOX and MHG catalyzed oxidation. Lipoxygenase activity was present in garlic, mustard seed and red pepper. Only in mustard seed, peroxidase activity was observed. Catalase activity was observed in garlic, black pepper, allspice and red pepper.

  • Since chemical structures of ginsenoside as active ingredient of Panax ginseng are known, accumulating evidence have shown that ginsenoside is one of bio-active ligands through the diverse physiological and pharmacological evaluations. Chemical structures of ginsenoside could be divided into three parts depending on diol or triol ginsenoside: Steroid- or cholesterol-like backbone structure, carbohydrate portions, which are attached at the carbon-3, -6 or -20, and aliphatic side chain coupled to the backbone structure at the carbon-20. Ginsenosides also exist as stereoisomer at the carbon-20. Bioactive ligands usually exhibit the their structure-function relationships. In ginsenosides, there is little known about the relationship of chemical structure and biological activity. Recent reports have shown that ginsenoside $Rg_3$, one of active ginsenosides, exhibits its differential physiological or pharmacological actions depending on its chemical structure. This review will show how ginsenoside $Rg_3$, as a model compound, is functionally coupled to voltage-gated ion channel or ligand-gated ion channel regulations in related with its chemical structure.

Whitening Effect and Skin Regeneration Effect of Red Sea Cucumber Extract (홍해삼 추출물의 멜라닌 형성 억제를 통한 미백효과 및 피부 재생효과에 관한 연구)

  • Recently, several researchers have been developing cosmetics from natural ingredients for skin whitening and anti-aging products. The red sea cucumber (RSC), Apostichopus japonicas, is a species of sea cucumber in the family stichopodiae, which is widely distributed in China, Japan, and Korea. To use Red Sea Cucumber as a cosmetic ingredient, its inhibitory effects on melanogenesis and the anti-aging effects of RSC extracts were investigated. First, a tyrosinase activity assay was performed, which showed that RSC inhibited tyrosinase activity at a concentration of $200{\mu}g/ml$. An MTT assay was carried out to evaluate cell toxicity, and the results showed that RSC extract has no cytotoxicity in HaCaT cells. Furthermore, the mRNA expression levels of tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), microphthalmia-associated transcription factor (MITF), and matrix metalloproteinase (MMPs) genes treated with RSC extract in B16F10 and HaCaT cells decreased. Moreover, a wound-healing assay was performed to identify the cell regeneration effect of RSC extracts. Also, a skin turnover effect was confirmed by creating a three-dimensional cell culture with HaCaT and human fibroblasts. Altogether, the results suggested that Red Sea Cucumber may possess a high ability to induce whitening and anti-wrinkle effects as a cosmeceutical ingredient.
