A bacterial strain capable of producing a novel bioflocculant was isolated from a biofilm sample and identified as Bacillus megaterium G31. The highest biopolymer yield was achieved when the organism was cultivated in a medium containing acetate as the sole carbon source and ($NH_4)_2HPO_4$as the nitrogen source. In kaolin suspension, the flocculating activity was highest at 170 mg I$^{-1}$ and decreased at the higher bioflocculant concentrations. The crude bioflocculant produced by the organism was purified by ethanol precipitation and gel permeation chromatography. The FTIR spectrum of the purified bioflocculant revealed that the bioflocculant might be a heterogeneous polysaccharide composed of hexosamines and neutral sugars. The analysis of sugar components of the bioflocculant using high performance anion-exchange chromatography showed that the sugar constituents of the bioflocculant were glucosamine, fucose, galactosamine, galactose, glucose, mannose in approximate molar ratio of 4 : 1 : 6 : 3 : 8 : 19. Its flocculating activity was stimulated by various cations. The bioflocculant was thermo-stable and retained 64% of its original activity after heating at $100^{\circ}C$ for 50 min.
The flocculating properties of bioflocculant Biopol32 produced by Pseudomonas sp. GP32 were investigated for application in industrial wastewater treatment. The major flocculating substance of bioflocculant Biopol32 was identified as polysaccharide. Many anionic flocculants need a counter ion to induce higher flocculating activity. The flocculating activity of bioflocculant Biopol32 was markedly increased by the addition of cationic ions ($Ca^{2+}$, $Al^{3+}$). The flocculating activity of bioflocculant Biopol32 was the most effective when 7.0 mM $CaCl_2{\cdot}2H_2O$ as coflocculant was added. The flocculating activity on the effect of pH and the temperature of the bioflocculant Biopol32 was compared with anionic commercial flocculant (polyacrylamide) and bioflocculant (zooglan from Zoogloea ramigera). In kaolin suspension, the highest flocculating activity was obtained at the bioflocculant Biopol32 concentration of 1.5 mg/l. A high flocculating activity was observed in the pH range of 5.0 to 8.0. The flocculating activity of bioflocculant Biopol32 was sustained up to $60^{\circ}C$, but decreased rapidly at over $70^{\circ}C$. In the batch culture, the charge density of bioflocculant Biopol32 was compared with flocculating activity. The larger the anionic charge density and apparent viscosity of bioflocculant Biopol32, the higher the flocculating activity. Therefore, we confirmed that the flocculating activity and apparent viscosity of bioflocculant Biopol32 was closely related to the charge density of bioflocculant Biopol32.
The purpose of this study was to develop the new microbial bioflocculant available in a food and fer-mentation industal. This study was reported the results of the composition for optimum culture medium and elemental characteristic of crude purification bioflocculant following the previous report(I). The maximum production of the flocculant from Bacillus megaterium was observated in the culture medium containing 2% sucrose 0.3% NaNO3 0.01% tryptone 0.01% beef extract 0.05% MgSO4 ·7 H2O 0.005% CaCO3 Addition of the sucrose as carbon sources and inorganic salt such as MgSO4, CaCO3 significantly increased the production of flocculant more than nitrogen sources. In the result of color reaction of the crude purified bioflocculant it was investgated that anthrone was positive and benedict burette and nin-hydrin was negative. These result were indicated that the flocculant produced from Bacillus megaterium was a kind of exopolysaccharide.
In this study, we analyzed composition of the bioflocculant, which strain YG-02 produces. First, supernatant and suspension from centrifugation of culture fluid of the strain, were used in the flocculation experiment. As a result, the SVI(sludge volume index) added with the suspension, was 182 mL/g, same as the control group with no additive, and the SVI added with supernatant, was 164 mL/g. So, the result above showed that flocculation capacity of the bioflocculant, was dependent on the substance which strain YG-02 produces, not on factors such as the body of germs. As a result of the thermostability test on substances that cause flocculation, the flocculation effect was significantly reduced, compared to the result of the flocculation test, before applying heat to the culture fluid, and it was able to assume that the substance that causes flocculation, was damaged by heat. Additionally, to understand the component of the bioflocculant, analyzation of sugar composition and fatty acid, was conducted. As a result, sugar composition was the polysaccharide consisting of glucose: lactose with molar ratio of 90.75:9.25. Fatty acid content was detected, as 0.0012 g/100g, showing that it contained glycolipid in the bioflocculant. Such results show that the bioflocculant which strain YG-02 produces, is the new bioflocculant, different from bioflocculantstudiedto date.
Bacillus sp. A56 was studied, because of its high flocculating activity. The flocculating substance produced by this strain was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography (GPC). The FT-IR spectrum of the purified bioflocculant, designated as BF-56, showed typical characteristics of polysaccharides. The non-sugar substituents, and sugar components of BF-56 containing glucose, fucose, glucuronic acid, and galactose in an approximate molar ratio of 2.76:1.10:1:0.12, suggested that it was a novel bioflocculant with an estimated molecular mass of over $7{\times}10^3$ kDa. Rheological analysis of BF-56 revealed that it was a pseudoplastic that had higher apparent viscosity rate at dilute concentrations than those of zooglan. The solution of bioflocculant BF-56 exhibited non-Newtonian characteristics and it was compatible to high concentrations of salts such as KCl, NaCl, $CaCl_2,\;or\;FeCl_3.$ The present results suggested strong possibility of bioflocculant BF-56 to be fully applicable to industries such as wastewater treatment.
The effects of CellCaSi and bioflocculant on the control of algal bloom were investigated in enclosures in a small eutrophic pond. The bioflocculant produced by a bacterial strain S-2 was finally selected to remove Microcystis aeruginosa which was a dominant species of algal bloom in the pond. Enclosure experiment showed that phosphorus concentration decreased dramatically from $131\mu{g}\ell^{-1}$ (Control) to $1-14\mu{g}\ell^{-1}$ in three CellCaSi-enriched enclosures. Chlorophyll $-\alpha$ concentration also decreased from $215\mu{g}\ell^{-1}$ (Control) to $59\mu{g}\ell^{-1}$ by the addition of CellCaSi $(1g\ell^{-1}$, bioflocculant $(2ml\ell^{-1}$, calcium chloride $(1g\ell^{-1}$ and ferric chloride $(2mg\;Fe\ell^{-1})$ in Enclosure 4. From the results of the mouse acute toxicity test of the S-2 bioflocculant and the goldfish survival test in enclosures, it seems that both the S-2 bioflocculant and the CellCaSi do not show any severe toxicity in water system. Consequently, it was concluded that the bioflocculant and the CellCaSi could be used to control algal bloom in eutrophic waters by removing phosphorus and chlorophyll$-\alpha$.

To develop bioflocculant for wastewater treatment, about 60 type culture strains and 450 strains isolated from natural sources were examined for screening their ability to flocculate the swine wastewater. Among them, YWO-5 showed the highest activity for NTU removal efficiency and was identified as Bacillus megaterium according to the cultural, morphological and physiological properties. The maximum production of the flocculant was achieved in culture medium containing 2% glucose, 0.05% soytone, 0.01% $CaCl_2$, 0.05% $KH_2PO_4$, and 0.05% yeast extract with initial pH 6.5 when cultured with rotary shaker controlled at 20$^{\circ}C$ and 150 rpm. With jar fermentor, the maximum production was reached to NTU removal efficiency of 93% after 3 days under the optimal conditions. The bioflocculant produced by Bacillus megaterium YWO-5 was effective on various suspended solids and organic wastewaters.
Among microorganisms isolated from soil, YJ-66 strain was the best producer of flocculant and was examined for flocculating ability in the active carbon and CaCl2. Addition of the carbon and inorganic salts significantly increased the production of flocculant. Compositions of optimized culture medium for bioflocculant production by Achromobacter sp. YJ-66 were 2% sucrose, 0.067% peptone, 0.1% MgSO4 and 0.1% KH2PO4 in initial pH 7.5 during at 3$0^{\circ}C$ for 72hrs.

The ubiquinone and G+C contents of the bioflocculant-producing fungus, a new Aspergillus strain, were detennined using high-perfonnance liquid chromatography. The internal transcribed spacers 1 and 2 (ITS1 and ITS2), and the 5.8S ribosomal DNA (rDNA) of the strain were amplified and sequenced. The strain contained ubiquinone-l0($H_2$)as a major quinone and the G+C content was 49 mol%. A phylogenetic analysis of the ITS regions indicated that the strain belonged to the genus Aspergillus according to its previously classified morphological characteristics. Based on a sequence homology search, the strain was most closely related to Petromyces muricatus (anamorph, A. muricatus; accession number, AJ005674). The sequence of a new Aspergillus strain in ITS1 and ITS2, and 5.8S rDNA showed 97% homology to P. muricatus. Therefore, the strain is believed to be a new bioflocculant-producing Aspergillus strain.
The culture conditions for Bacillus sp. DP-152 in the flask were investigated for the production of polysaccharide locculant, DP-152. The optimum pH and temperature for the locculant production were 8.0 and $30^{\circ}C$, respectively. The avorable substrates for flocculant production were soluble tarch and ammonium nitrate. The medium composition was optimized as follows: 30 g soluble starch, 0.75 g $NH_4NO_3,\; 2.0g\; K_2\;HPO_4,\; 0.1\; g KH_2PO_4,\; 0.2g\; MgSO_4.\; 7H_2O,\; and\; 0.2g\; MnSO_4~5H_2O$ in 11 of distilled water. Under this optimized condition, flocculating activity has been improved 4-fold compared with that of the basal medium. In the culture flask, the highest flocculating activity was obtained after 70 h of cultivation and the amount of bioflocculant DP-152 yielded was 12.4 g/$\ell$. The solution of bioflocculant DP-152 showed non-Newtonian characteristics. Bioflocculant DP-152 exhibited apparently higher viscosity at all concentrations compared to that of zooglan (from Zoogloea ramigera), and it was stable over a wide range of temperatures and pHs.
