Nonketotic hyperglycinemia (NKH) is a rare inborn error of amino acid metabolism. A defect in the glycine cleavage enzyme system results in highly elevated concentrations of glycine in the plasma, urine, cerebrospinal fluid, and brain, resulting in glycine-induced encephalopathy and neuropathy. The prevalence of NKH in Korea is very low, and no reports of surviving patients are available, given the scarcity and poor prognosis of this disease. In the current study, we present a patient with NKH diagnosed on the basis of clinical features, biochemical profiles, and genetic analysis. Magnetic resonance spectroscopy (MRS) allowed the measurement of absolute glycine concentrations in different parts of the brain that showed a significantly increased glycine peak, consolidating the diagnosis of NKH. In additional, serial MRS follow-up showed changes in the glycine/creatinine ratios in different parts of the brain. In conclusion, MRS is an effective, noninvasive diagnostic tool for NKH that can be used to distinguish this disease from other glycine metabolism disorders. It may also be useful for monitoring NKH treatment.

Yeasts that are present in marine environments have evolved to survive hostile environments that are characterized by high exogenous salt content, high concentrations of inhibitory compounds, and low soluble carbon and nitrogen levels. Therefore, yeasts isolated from marine environments could have interesting characteristics for industrial applications. However, the application of marine yeast in research or industry is currently very limited owing to the lack of a suitable isolation method. Current methods for isolation suffer from fungal interference and/or low number of yeast isolates. In this paper, an efficient and non-laborious isolation method has been developed and successfully isolated large numbers of yeasts without bacterial or fungal growth. The new method includes a three-cycle enrichment step followed by an isolation step and a confirmation step. Using this method, 116 marine yeast strains were isolated from 14 marine samples collected in the UK, Egypt, and the USA. These strains were further evaluated for the utilization of fermentable sugars (glucose, xylose, mannitol, and galactose) using a phenotypic microarray assay. Seventeen strains with higher sugar utilization capacity than the reference terrestrial yeast Saccharomyces cerevisiae NCYC 2592 were selected for identification by sequencing of the ITS and D1/D2 domains. These strains belonged to six species: S. cerevisiae, Candida tropicalis, Candida viswanathii, Wickerhamomyces anomalus, Candida glabrata, and Pichia kudriavzevii. The ability of these strains for improved sugar utilization using seawater-based media was confirmed and, therefore, they could potentially be utilized in fermentations using marine biomass in seawater media, particularly for the production of bioethanol and other biochemical products.
Antibodies raised against the purified p-subunit of $\beta$-conglycinin were used in immunohistochemical studies to monitor the pattern of $\beta$-conglycinin mobilization in the cotyledons during soybean [Glycine max (L.) Merr.] seed germination. Western blot analysis revealed that the break down of the $\beta$-subunit of $\beta$-conglycinin commenced as early as 2 days after seed imbibition (DAI). Concurrent with the degradation of the $\beta$-subunit of $\beta$-conglycinin, accumulation of 48, 28, and 26 kD proteolytic intermediates was observed from 2 to 6 DAI. Western blot analysis also revealed that the acidic subunit of glycinin was mobilized earlier than the basic subunit. The basic glycinin subunit was subjected to proteolysis within 2 DAI resulting in the appearance of an intermediate product approximately 2 kD smaller than the native basic glycinin subunit. In contrast to the major seed storage proteins, lipoxygenase was subjected to limited proteolysis and was detected even after 8 DAI. The first sign of $\beta$-conglycinin breakdown was observed near the vascular strands and proceeded from the vascular strands towards the epidermis. Protein A-gold localization studies using thin sections of soybean cotyledons and antibodies raised against the $\beta$-subunit of $\beta$-conglycinin revealed intense labeling over protein bodies. A pronounced decrease in the protein A-gold labeling intensity over protein bodies was observed at later stages of seed germination. The protein bodies, which were converted into a large central vacuole by 8 DAI, contained very little 7S protein as evidenced by sparse protein A-gold labeling in the vacuoles.

Cerebrotendinous xanthomatosis (CTX) is a rare genetic disease caused by a deficiency of enzymes for the synthesis of bile acid, resulting in the accumulation of cholestanol with reduced chenodeoxycholic acid (CDCA) production and causing various symptoms such as chronic diarrhea in infancy, juvenile cataracts in childhood, tendon xanthomas in adolescence and young adulthood, and progressive neurologic dysfunction in adulthood. Because oral CDCA replacement therapy can effectively prevent disease progression, early diagnosis and treatment are critical in CTX. This study reports the case of CTX in a 10-year-old male who presented with Achilles tendon xanthoma and mild intellectual disability. Biochemical testing showed normal cholesterol and sitosterol levels but elevated cholestanol levels. Genetic testing showed compound heterozygous variants of CYP27A1, c.379C>T (p.Arg127Trp), and c.1214G>A (p.Arg405Gln), which confirmed the diagnosis of CTX. The patient had neither cataracts nor other focal neurologic deficits and showed no abnormalities on brain imaging. The patient received oral CDCA replacement therapy without any adverse effects; thereafter, the cholestanol level decreased and no disease progression was noted. The diagnostic possibility of CTX should be considered in patients with tendon xanthoma and normolipidemic conditions to prevent neurological deterioration.
Hyperornithinemia-hyperammonemia-homocitrullinuria 증후군(HHH 증후군)은 경한 학습장애에서 심각한 뇌증에 이르기까지 다양한 신경학적 증상을 보일 수 있는 신경대사질환이다. HHH 증후군은 임상 증상이나 대사 검사 결과가 뚜렷하지 않아 진단이 늦어지는 경우가 흔한데, 이러한 경우 유전자 검사로 확진하는 것이 중요하다. 저자들은 강직성 하반신 마비를 가진 소아와 무증상의 여동생에서 HHH 증후군을 진단하고 유전자 검사를 통해 SLC25A15 유전자의 c.535C>T (p.R179*)와 c.116C>A (p.T39K) 변이를 확인하였다. p.R179*는 일본과 중동의 HHH 환자들에서 흔히 발견되는 변이로 한국인에서도 발견됨을 확인하였고, p.T39K는 HHH 증후군을 유발하는 새로운 변이임을 최초로 보고하는 바이다.
벼 자색 종자과피(Purple pericarp, Prp) 형질은 주요 생리활성물질인 안토시아닌 C3G 생성에 관여하며 흑미를 결정하는 주요 유전형질이다. Prp 유전형질을 가진 흑미와 종자과피에 색이 없는 벼를 교배할 경우 그 후대는 검정색, 갈색, 백색이 각각 9:3:4로 분리된다. 1921년 Nagai에 의하여 제시된 바 벼 종자 색의 9:3:4 유전분리비로 인하여 벼 Prp 형질은 유전자의 열성상위(recessive epistasis) 현상으로 해석되었다. 그러나 흑미를 결정하는 Prp 형질은 두 개의 상보적 유전자들의 상호관계(complementary gene interaction)에 의한 것이기도 하다. 본 연구에서는 이러한 논란이 발생되는 이유를 설명하기 위하여 두 유전자의 조성이 완전한 이형접합인 Pbpb Pppp 유전자형을 가진 $F_1$ 잡종을 만들었다. 이들의 자손은 진한자색(검정), 중간자색, 갈색, 백색 종자이며 각각 3:6:3:4로 분리되었다. 즉, 검정색, 갈색, 백색의 종자가 각각 9:3:4의 비율로 분리된다. 그러나 생화학적인 분석결과 이들은 안토시아닌 중 cyanidin 3-O-glucoside (C3G)가 축적된 검정색 종자와 C3G가 없는 갈색 또는 백색 종자인 두 개의 집단을 분리되며 정확히 9:7의 분리 비를 갖는다. 이 경우 벼 Prp 형질을 갖는 검정쌀 또는 흑미는 전형적인 상보적 유전자의 상호관계에 의한 유전현상이다. 즉, 흑미의 자색 종피 형질 발현에는 Pb 유전자와 Pp 유전자에서 각각 한 개 이상의 우성대립인자의 발현이 필요하다. 그러나 Pb 유전자만 우성대립인자가 존재하는 Pb_ pppp 유전자형의 벼는 C3G를 생성하지 못하고 갈색 종자과피(Brown pericarp, Brp) 형질을 갖게 된다. 즉 갈색쌀는 우성 Pb 유전자의 우성대립인자에 의하여 결정된다. 그러므로 종피색을 결정하는 Prp 형질의 유전양상은 열성상위 현상으로 보이나 흑미의 결정요소인 안토시아닌 C3G의 함유 여부에 관한 유전분석을 시행하면 9:7의 비율로서 전형적인 두 개의 유전자가 모두 관여하는 상보적 유전현상이다. 유전적 정의는 유전자의 최종산물에 의한 물리적 또는 화학적 결정이다. 그러므로 결론하여 검정 쌀의 주요 생리활성물질인 안토시아닌 C3G 생성에 관한 유전현상은 Pb와 Pp 유전자의 상보적 유전자의 상호에 의한 것이다.
뮤코지방증 2형과 3형은 GlcNAc-1-phosphotransferase 효소의 기능 이상으로 인해 발생하는 상염색체 열성 유전질환이다. GlcNAc-1-phosphotransferase 효소의 기능 이상은 리소좀 효소의 세포 내 이동에 관여하는mannose-6-phosphate와 리소좀 효소간의 결합을 저해하여 리소좀 효소들이 세포 안으로 들어오지 못하게 된다. 리소좀 효소의 부족으로 인해 리소좀 기능 이상 및 뮤코지질의 축적으로 인해 다양한 임상을 보이게 된다. 본 연구는 새롭게 진단된 뮤코지방증 환자의 임상 및 생화학, 분자유전학적 양상을 기술하고 본원에서 치료받은 11명의 뮤코지방증 2, 3형 환자들의 임상 양상을 비교하여 분석하였다. 본 저자들은 17개월에 특징적 얼굴 모양 및 관절 구축을 주소로 내원한 환자에서 효소 검사를 통해 리조솜 효소의 혈장 내 증가를 확인하였으며 GNPTAB 유전자의 직접염기 서열분석을 통해 복합이형접합자 돌연변이 (c.3428 dupA [pAsn1143Lysfs*3], c.673C>T [p.Gln225*])를 발견하였다. 새로 진단된 환자를 포함하여 총 11명의 환자(뮤코지방증 2형: 7명, 3형: 4명)들을 대상으로 임상 양상을 분석하였고 진단 시 나이는 뮤코지방증 2형은 2세 1개월, 3형은 6세 9개월이었으며 진단 시 신장 표준편차지수는 각각 -3.2 (${\pm}1.5$), -1.3 (${\pm}1.1$)였다. 심초음파 결과에서 뮤코지방증 3형 환자들은 심장 판막의 이상만을 보였으나 2형 환자들에서는 비후성 심근증(n=3), 좌심비대(n=1)이 동반되었다. 간비비대(n=4)는 뮤코지방증 2형 환자에서만 확인되었다. 뮤코지방증 2형의 경우, 3명의 환자가 호흡 곤란으로 기관 절개 및 호흡기 보조를 받았으며 2 명의 환자가 호흡기 문제로 사망하였다. Pamidronate 치료는 4명의 뮤코지방증 3형 환자와 4명의 2형 환자에서 시행하고 있으며 유효성 및 안정성에 대한 추가적인 평가가 필요하다.
I 세포 질환(뮤코지방증 2형; MIM 252500)과 pseudo-Hurler polydystrophy (뮤코지방증 3형; MIM 252600)는 세포내 비정상적인 리소좀 관련 운송으로 인해 발병한다. 특징적인 소견으로는 섬유아 세포의 세포질에 다수의 봉입체, 뮤코다당뇨의 부재, 혈청 내 리소좀 효소 활성도의 증가, GlcNAc-phosphotransferase 활성도의 감소를 보인다. 이 연구에서는 GlcNAc-phosphotransferase 알파/베타 아형에 대한 knockout 마우스의 표현형과 생화학적 특징을 조사하였다. 또한, 태반으로부터 추출한 리소좀 농축 분획을 knockout 마우스에 투여하였을 때 체중 증가에 대한 효과를 볼 수 있는지에 대해 알아보고자 하였다. knockout 마우스는 뮤코지방증 2형 환자에서 그렇듯이 정상적인 체중 증가를 보이지 않았고 낮은 골밀도를 보였다. 게다가 knockout 마우스의 피부 섬유 아세포의 배양액에서는 리소좀 효소 활성도가 증가한 반면, 세포 내에서는 리소좀 효소 활성도가 감소되어 있는 것을 확인할 수 있었으며 이러한 특징은 뮤코지방증 2형 환자에서 볼 수 있는 특징과 일치한다. knockout 마우스의 꼬리 정맥내로 태반에서 추출한 리소좀 농축 분획을 투여한 결과, 체중이 증가하는 것을 확인할 수 있었고, 반면 생리식염수를 투여한 knockout 마우스의 경우는 체중이 증가하지 않았다. 결론적으로, knockout 마우스의 표현형과 생화학적 특징이 뮤코지방증 2형 환자와 유사하다는 것을 확인하였으며, 리소좀 농축 분획의 치료적 가능성을 증명하였다. 더 큰 범위의 동물 실험을 진행할 필요가 있으나, 이 연구는 질병에 대한 동물 모델을 개발하고 리조솜 농축 분획의 치료적 가능성을 제시하는 것을 통해 현재까지 치료가 불가능한 뮤코지방증 2형의 새로운 치료 방법의 가능성을 열었다고 볼 수 있다.
제주말의 혈통보존을 위한 기초자료를 마련할 목적으로 국내에서 사육중인 몽고말 19두를 대상으로 적혈구항원형 및 혈액단백질형의 표현형 분포와 유전자 빈도를 조사한 결과는 다음과 같다. 적혈구항원형의 표현형은 $A^af$(42.1%), $C^a$(89.5%), $K^-$((84.2%), $U^a$(63.2%), $P^a$(42.1%) $P^-$(42.1%), $Q^c$(31.6%), $Q^-$(31.6%)에서 높은 빈도를 나타냈으며 적혈구항원형 D 시스템의 유전자형은 $D^cgm/dghm$ (15.8%), $D^dghm/dghm$(15.8%), $D^ad/dghm$(10.5%), $D^ade/dghm$ (10.5%)가 높은 빈도 순으로 관찰되었다. 혈액단백질형 7개의 좌위에서는 $AL^{AB}$(52.6%), $AIB^K$( (89.5%), $ES^I$(63.2%), $GC^F$(78.9%), $HB^{BI}$(68.4%),$PGD^F$(84.2%), $TF^{F1R}I$(21.1%), $TF^{F2R}I$(21.1%)로 높은 빈도를 보였으며, $HB^{A2B2}I$(5.3%), $PGD^{DS}I$(5.3%)가 특이하게 관찰되었다. 적혈구항원형의 유전자 빈도를 조사한 결과 $A^a$(0.4211), $C^a$(0.8947), $K^-$(0.8421), $U^a$(0.6316), $P^a$ (0.4474), $Q^c$(0.4474), $D^{dghm}$(0.4211)로 가장 높은 빈도를 보였으며, 혈액단백질형 7개 좌위에 대해서 유전자 빈도를 조사한 결과 $AL^B$(0.6579), $AIB^K$(0.9211), $ES^I$(0.7895), $GC^F$(0.8947), $HB^BI$ (0.7895), $PGD^F$(0.8421), $TF^R$(0.3421)가 높은 빈도로 관찰되었다.
Soyfoods have potential roles in the prevention and treatment of chronic diseases, most notably cancer, osteoporosis, and heart disease. There is evidence that carcinogenesis are supressed by isolated soybean derived products in vivo such as a protease inhibitor, phytic acid, saponins and isoflavones. It is believed that supplementation of human diets with soybean products markedly reduces human cancer mortality rates. Especially, recent papers recognize the potential benefit of soybean isoflavone components for reducing the risk of various cancers. Isoflavones exhibit a multitude of medicinal effects that influence cell growth and regulation, which may have potential value in the prevention and treatment of cancer. In addition to potential biological effects, soybean isoflavones have the important physiological functions such as the induction of Bradyrizobium japonicum nod genes and the responses of soybean tissues to infection by Phytophthora megasperma as well as biochemical activities such as antifungal and antibacterial actions. Genistin, daidzin, glycitin and their aglycone (genistein, daidzein, glycitein) are the principal isoflavones found in soybean. Malonyl and acetyl forms have also been detected but they are thermally unstable and are usually transformed during the processing in glucoside form. Most soy products, with the exception of soy sauce, alcohol-extracted soy protein concentrate, and soy protein isolate, have total isoflavone concentrations similar to those in the whole soybean. Soybean-containing diets inhibit mammary tumorigenesis in animal models of breast cancer, therefore, it is possible that dietary isoflavones are an important factor accounting for the lower incidence and mortality from breast cancer. Of the total soybean seed isoflavones, $80\~90\%$ were located in cotyledons, with the remainder in the hypocotyls. The hypocotyls had a higher concentrations of isoflavones on a weight basis compared with cotyledons. Isoflavone contents were influenced by genetics, crop years, and growth locations. The effect of crop year had a greater impact on the isoflavone contents than that of location. The climate condition might be the attribution factor to variation in isoflavone contents. Also, while the isoflavone content of cotyledons exhibited large variations in response to high temperature during seed development, hypocotyls showed high concentration in isoflavone content. So, it is concluded that one of the factors affecting isoflavone content in soybean seeds is temperature during seed development. High temperature, especially in maturity stage, causes lower isoflavone content in soybean seed. It is also suggested that there may exist a different mechanism to maintain isoflavone contents between cotyledon and seed hypocotyls. In a conclusion, soy foods may be able to have a significant beneficial impact on public health.

저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
