Biochemical Characterization of Tannases from Paecilomyces variotii and Aspergillus niger

  • A biochemical characterization of the tannases from Paecilomyces variotii (produced at Unicamp), Aspergillus niger (purchased from Industrial Kerry Bio-Science) and A. niger cnpat 001 (purchased from Embrapa Agroindustrial Tropical-Brazil) was carried out. P variotii is a new strain obtained from the screening of 500 fungi that were tested for their production of tannase. The biochemical properties of this new tannase from P variotii were determined and compared with those of two other tannase preparations. The tannase produced from P. variotii showed optimum activity at pH 6.5. The functional temperature range of the tannases was from $20-70^{\circ}C$, with optima at $70^{\circ}C$ for P. variotii and at $60^{\circ}C$ for the commercially obtained tannase, whereas A. niger cnpat 001 showed optimum activity at $40^{\circ}C$. The effects of 1 mM preparations of cations and anions, inhibitors, surfactants, and chelators on the tannase activity from P. variotii were also studied.

  • The polygalacturonase (PG) production in rotten apples by Botryosphaeria dothidea was purified by using gel filtration and ion exchange column chromatography, and the biochemical characters of PG were investigated. The purified PG appeared as a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with approximate molecular weight of 49 kilodalton (kDa). The molecular weight was equal to the native molecular weight estimated by gel filtration. The Km and Vmax values of PG were 0.51 mg/ml and 90.9 $\mu$M/min/ml, respectively. Optimum pH was 4.0~5.0, and the PG activity was stable from pH 5.0~10.0. Optimum temperature of the enzyme activity was 4$0^{\circ}C$. The PG activity was relatively stable at 2$0^{\circ}C$, but it was reduced 45% at 4$0^{\circ}C$ and completely inactivated at 8$0^{\circ}C$. The PG activity was considerably inhibited by Cu2+, Zn2+, SDS and EDTA, whereas it was not effected by Ca2+, K+, Mg2+ or Na+ ions.

Effects of Replacement of Fish Meal by Soy Protein Isolate on the Growth, Digestive Enzyme Activity and Serum Biochemical Parameters for Juvenile Amur Sturgeon (Acipenser schrenckii)

  • An 8-wk experiment was conducted to evaluate the effect of replacing fish meal (FM) with soy protein isolate (SPI) on the growth, digestive enzyme activity and serum biochemical parameters of juvenile Amur sturgeon (Acipenser schrenckii). SPI was used to replace 0, 25, 50, 62.5, 75, 87.5, 100% of dietary FM and 100% replacement supplemented crystalline amino acid. Healthy sturgeon with an average initial weight of $26.38{\pm}0.24$ g were randomly assigned to 24 aquaria (8 treatments with triplicates each) at an initial stocking density of 11 fish per aquarium and cultured for 8 wks. The results showed that 75.00% or more substitution resulted in a poor weight gain rate, feed conversion ratio and survival rate compared to that of fish fed the control diet (p<0.05), whereas no significant differences were observed between diets of 25.00% to 62.50% substitution. Protease, lipase and amylase activity in foregut, mid-gut and hindgut were significantly (p<0.05) decreased by diets where SPI replacement levels were 62.50% or more. Levels of serum total protein (TP) and globulin decreased significantly from 21.03, 10.34 to 14.05, 5.63 g/L with the increasing dietary SPI (p<0.05), but alkaline phosphatase activity significantly increased (p<0.05). In addition, supplemental crystalline amino acid in the FM absence diet did not improve growth performance, intestine digestive enzyme activities and serum biochemical parameters. In conclusion, the results from this study showed adverse effects of inclusion of SPI in diets on growth performance, feed utilization and serum biochemical parameters in juvenile Amur sturgeon. Based on WGR and replacement ratio presented in this report, a 57.64% replacement level was recommended.

Effect of Hot Water Extract of Undaria pinnatifida on the Activities of Antioxidant and Nitrite Scavenging (미역 열수 추출물의 항산화 및 아질산 제거효과)

  • For the development of functional food and cosmetics using the hot water extract of Undaria pinnatifida, the concentrations of vitamin, amino acid and element and activities of antioxidant and nitrite scavenging were investigated. The results are shown as follows: Vitamin C and E concentrations were 0.301 and 0.11 mg/100 g, respectively. Mineral concentrations were an order of Ca > Mg > K > Fe. The concentrations of total amino acids were an order of Glu > Ala > Val > Leu > Gly > Pro. Total phenol concentration and DPPH radical scavenging activity were increased with the increase of the concentration of extract. Especially, when the extract concentrationwas increased from 1.0 to 10.0 mg/mL, the total phenol concentration was increased from 0.043 to 0.125 OD 725 nm. DPPH radical scavenging activity at 50 mg/mL was 70.1%. The antioxidant activity of extract was stable in range of 80 to $140^{\circ}C$ and pH 3-9. The nitrite scavenging activity was increased with the decreaseof pH and the increase of it's extract concentration. Especially, it was 83.4% at 50 mg/mL (pH 1.2). These results showed that the hot water extract of U. pinnatifida can be applied to functional food and cosmetics.

Comparison of Riboflavin Status between Traditional Farming Women and Commercial Farming Women in Korea

  • this study was undertaken to compare the riboflavin status of rural women with different physical activity intensity and to determine factors influencing biochemical fiboflavin status. The study was carried out over three different farming seasons : planting (June), harvest(October) and interim(February) in two rural regions of Korea. One was a traditional farming region, the other a commercial farming region with heavier work intensity. Twenty women in the traditional region and eighteen women in the commercial region were involved. The intensity of physical activity was determined by a daily activity record. Body composition was assessed by bioelectrical impedence. Dietary riboflavin intake was measured by the food frequency method. Riboflavin biochemical status was assessed by erythrocyte glutathione reductase activity coefficient (EGR AC) and ruinary riboflavin excretion. The results from the EGR AC and urinary riboflavin excretion during the period showed the overall riboflavin status of the commercial farming women was significantly worse than that of the traditional farming women(EGR AC p<0.0001, urinary riboflavin excretion p<0.05). The traditional farming group had about 40% with risk of riboflavin deficiency, whereas the commercial farming group had about 70%. Overall mean nutrient intake was not significantly different between the two groups, however, overall mean percent lean body mass representing long term physical activity was significantly higher in the commercial farming group ( <0.005). It appears that the biochemical riboflavin status of traditional farming women was significantly influenced by riboflavin intake and crude nitrogen balance while the biochemical riboflavin status of the commercial farming women was significantly influenced by riboflavin intake and percent of lean body mass over the three seasons. Effects of Different Dietary Oil and d-Limonene on Histopathological and Biochemical Changes in Experimental Hepatocarcinogenesis (식이지방의 종류 및 d-Limonene 투여가 간 발암과정에 미치는 영향)

  • The purpose of this study was to investigate the effcts of n-3, n-6 fatty arid and d-limonene on histopathological and biochemical changes in experimental rat hepatocarcinogenesis. To attain the above objectives, weanling Sprague-Dawley female rats were intraperitoneally injected twice with a dose of diethylnitrosamine(DEN, 50mg/kg body weight) and after 1 week 0.05% phenobarbital was provided with water. Sardine oil rich in n-3 fatty acids and corn oil rich in n-6 fatty acids were fed at 15% by weight and 5% d-limonene was added to the diet in each group. Ten weeks or 20 weeks after DEN treatment, rats were sacrifirced. The formation of glutathione S-transferase placental form positive(GST-P$\^$+/) foci was significantly decreased by the treatment of either sardine oil or d-limonene HMG-CoA reductase activity was not affected by dietary oils and d-limonene. Protein kinase C (PKC) activity was decreased by either sardine oil or d-limonene. Particularly d-limonene decreased the membrane PKC activity. Membrane Cholesterol/Phospholipid(Chol/PL) ratio was significantly decreased by d-limonene in sardine oil group. The data showed that GST-P$\^$+/ foci number was positively correlated with membrane PKC activity and serum cholesterol and negatively correlated with liver cholesterol level. These results suggest informations about the correlation between histopathological and biochemical changes such as cholesterol metabolism and PKC activity in experimental hepatocarcinogenesis and thereby can elucidate the possible mechanism related to the cancer inhibition.(Korean J Nutrition 33(1) : 23-32, 2000)

Serum Biochemical Values during Antler Growth in Sika Deer (Cervus nippon)

  • Serum biochemical values were measured in blood samples collected from 8 fasted stags from both jugular and femoral veins at 18-day intervals during antler growth. Samples were analyzed for blood substrate, enzyme activity values, minerals and electrolyte. There were no significant differences in total protein, albumin, urea, creatinine, triglyceride, glucose or cholesterol concentration between veins or sampling dates. However, total-bilirubin concentration in the jugular vein on the casting date was three times higher than on the other sampling dates (p<0.05). There were no significant differences in alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase with progressing antler growth. The highest level of alkaline phosphatase concentration was on day 55 after casting. There were no significant differences in inorganic phosphorus, sodium and chloride concentration between jugular and femoral samples. Calcium concentration was significantly higher in the femoral vein on the cutting date (55 day) than in the jugular vein (p<0.05). There were few changes in serum biochemical values. However, some electrolytes and minerals had differences during antler growth. It is suggested that despite such a big event as antler growth, blood biochemical values are not variable if feeding conditions are consistently maintained as was the case in this study.

  • There are many cyclic peptides with diverse biological activities, such as antibacterial activity, immunosuppressive activity, and anti-tumor activity, and so on. Encouraged by natural cyclic peptides with biological activity, efforts have been made to develop cyclic peptides with both genetic and synthetic methods. The genetic methods include phage display, intein-based cyclic peptides, and mRNA display. The synthetic methods involve individual synthesis, parallel synthesis, as well as split-and-pool synthesis. Recent development of cyclic peptide library based on split-and-pool synthesis allows on-bead screening, in-solution screening, and microarray screening of cyclic peptides for biological activity. Cyclic peptides will be useful as receptor agonist/antagonist, RNA binding molecule, enzyme inhibitor and so on, and more cyclic peptides will emerge as therapeutic agents and biochemical tools.

  • The use of biochemical markers of bone turnover may be particular interest in the investigation of bone disorders with osteoporosis. Serum osteocalcin(OC), total alkaline phosphatase and procollagen C, reflecting bone formation, and urinary pyridinium cross-links excretion, reflecting bone reabsorption have been measured in hyperthyroidism, postmenopause women, after testosterone supplementation, androgen, testosterone and estrogen deficiency, bone mineral density degree, age duration. Bone marks which is reflect to metabolic bone disorders are biochemical indices method to measure enzyme activity about bone formation, bone absorption and bone components in blood or urine. Bone metabolism biochemical marks are correlated with osteophorotic agents and also represent significantly different between bone mineral density and bone biochemical marks. Therefore if we develope and use bone metabolism marks which have higher sensitivity and specificity in bone formation and bone absorption, I think that these bone biochemical marks can have utility in the clinical application to predict osteoporosis risk group, bone loss, bone fracture and response degree to treatment of osteoporosis risk groups.
