Binding Model of Amentoflavone to Peroxisome Proliferator-Activated Receptor γ

  • Human peroxisome proliferator-activated receptor gamma ($hPPAR{\gamma}$) has been implicated in numerous pathologies, including obesity, diabetes, and cancer. In this study, we verified that amentoflavone is an agonist of $hPPAR{\gamma}$ and probed the molecular basis of its action. It was demonstrated that amentoflavone bound $hPPAR{\gamma}$ with high (picomolar) affinity and increased the binding between $hPPAR{\gamma}$ and steroid receptor coactivator-1 (SRC-1) by approximately 4-fold. Based on a docking study, for the first time, we propose a model of amentoflavone and $hPPAR{\gamma}$ binding in which amentoflavone forms three hydrogen bonds with the side chains of His323, Tyr327, and Arg280 in $hPPAR{\gamma}$ and participates in two hydrophobic interactions.

Continuous Bio-hydrogen Production from Food Waste and Waste Activated Sludge (음식물 쓰레기와 폐활성 슬러지를 이용한 생물학적 수소생산 및 수소생산 미생물 군집분석)

  • Batch experiments were performed to investigate the effects of volumetric mixing ratio(v/v) of two substrates, food wastes(FW) and waste activated sludge(WAS). In batch experiments, optimum mixing ratio for hydrogen production was found at $10{\sim}20$ v/v % addition of WAS. CSTR(Continuous Stirred tank reactor) was operated to investigate the hydrogen productivity and the microbial community under various HRTs and volumetric mixing ratio(v/v) of two substrates. The maximum yield of specific hydrogen production, 140 mL/g VSS, was found at HRT of 2 day and the volumetric mixing ratio of 20:80(WAS:FW). The spatial distribution of hydrogen producing bacteria was observed in anaerobic fermentative reactor using fluorescent in situ hybridization(FISH) method.

Sole and Combined Usage of Ultra-sonication and Hydrogen Peroxide as Mitigation Techniques of Bio-fouling

  • Mussels are stubborn organism attached to solid substrate by byssus threads and caused operational problems in utility of power generating stations. Sole and combined usage of ultrasonic (28 kHz- and 42 kHz- frequencies) and hydrogen peroxide ($H_2O_2$) has studied for control of blue mussel larvae and adult stage in seawater condition. A theoretical wo rking model using disinfection (Chick and Watson type) approaches is presented based on helpful results of experiments. This study also demonstrate that the combined treatment (ultra-sonication with $H_2O_2$) is overall highly efficient than individual treatment would, but on the basis of exposure time, the ultra-sonication was the most efficient among them. Therefore the development of sole and combined technique might be effective practical mitigation strategy against mussel attachment for water handling facilities.

A Complete, Reductive Depolymerization of Concentrated Sulfuric Acid Hydrolysis Lignin into a High Calorific Bio-oil using Supercritical Ethanol

  • It is imperative to develop an effective pathway to depolymerize lignin into liquid fuel that can be used as a bioheavy oil. Lignin can be converted into liquid products either by a solvent-free thermal cracking in the absence air, or thermo-chemical degradation in the presence of suitable solvents and chemicals. Here we show that the solvent-assisted liquefaction has produced promising results in the presence of metal-based catalysts. The supercritical ethanol is an efficient liquefaction solvent, which not only provides better solubility to lignin, but also scavenges the intermediate species. The concentrated sulfuric acid hydrolysis lignin (CSAHL) was completely liquefied in the presence of solid catalysts (Ni, Pd and Ru) with no char formation. The effective deoxy-liquefaction nature associated with scEtOH with aid hydrodeoxygenation catalysts, resulted in significant reduction in oxygen-to-carbon (O/C) molar ratio up to 61%. The decrease in oxygen content and increase in carbon and hydrogen contents increased the calorific value bio-oil, with higher heating value (HHV) of $34.6MJ{\cdot}Kg^{-1}$. The overall process is energetically efficient with 129.8% energy recovery (ER) and 70.8% energy efficiency (EE). The GC-TOF/MS analysis of bio-oil shows that the bio-oil mainly consists of monomeric species such as phenols, esters, furans, alcohols, and traces of aliphatic hydrocarbons. The bio-oil produced has better flow properties, low molecular weight, and high aromaticity.

Bio-functionalized Gold Nanoparticles for Surface-Plasmon- Absorption-Based Protein Detection

  • Bio-functionalized gold nanoparticles (AuNPs), which bio-specifically interact with biotin-(strept)avidin, were investigated in this study. AuNPs were functionalized with a synthetically-provided biotin-linked thiol (BLT), which was synthesized by amidation of the active ester of biotin with 2-mercaptoethylamine. The BLT-attached AuNP was bio-specific for streptavidin, making it potentially useful for biosensor applications. To test the bio-specific interactions, the colors, absorption spectra and TEM images were investigated for proteins such as streptavidin, cytochrome C, myoglobin and hemoglobin. The colors and absorption spectra changed when streptavidin was added to the BLT-attached AuNP solution. However, the color and spectra did not change when the other proteins were added to the same solution. These results show that the AuNPs provided a colloidal solution with excellent stability and highly selective absorption characteristics for streptavidin as a target molecule. Proteins were also screened in order to identify a general strategy for the use of optical biosensing proteins based on AuNPs. In addition, TEM images confirmed that streptavidin led the BLT-attached AuNPs to aggregate or precipitate.

UV-induced Crosslinking of Poly(vinyl acetate) Films Containing Benzophenone (벤조페논을 함유한 폴리비닐아세테이트 필름의 자외선 조사에 의한 가교)

  • Poly(vinyl acetate) films containing benzophenone were photocrosslinked by continuous UV irradiation.UV irradiation of PVAc film containing 5% benzophenone induced bulk crosslinking of the polymer indicated by 84.1% of gel fraction after ethyl acetate extraction. The crosslinking was attributed to the recombination of tertiary polymer radicals generated upon UV irradiation, which was enhanced by the hydrogen abstraction of benzophenone. Also the UV irradiation resulted in scission of ester linkage and photooxidation of PVAc surface, which was verified by ATR and zeta potential analysis, implying that the PVAc surface became more polar and hydrophilic. The zeta potential proportionally increased from +4.5mV to -26.8 mV with increasing UV irradiation. Also the surface energy of the PVAc film increased with higher UV irradiation upto 56.5 $mJ/m^2$ by the enhanced Lewis acid/base component with larger contribution of Lewis acid parameter. Accordingly the crosslinked PVAc showed higher thermal stability with increasing UV energy.

Conversion of organic residue from solid-state anaerobic digestion of livestock waste to produce the solid fuel through hydrothermal carbonization

  • The solid-state anaerobic digestion (SS-AD) has promoted the development and application for biogas production from biomass which operate a high solid content feedstock, as higher than 15% of total solids. However, the digested byproduct of SS-AD can be used as a fertilizer or as solid fuel, but it has serious problems: high moisture content and poor dewaterability. The organic residue from SS-AD has to be improved to address these problems and to make it a useful alternative energy source. Hydrothermal carbonization was investigated for conversion of the organic residue from the SS-AD of livestock waste to solid fuels. The effects of hydrothermal carbonization were evaluated by varying the reaction temperatures within the range of $180-240^{\circ}C$. Hydrothermal carbonization increased the calorific value through the reduction of the hydrogen and oxygen contents of the solid fuel, in addition to its drying performance. Therefore, after the hydrothermal carbonization, the H/C and O/C atomic ratios decreased through the chemical conversion. Thermogravimatric analysis provided the changed combustion characteristics due to the improvement of the fuel properties. As a result, the hydrothermal carbonization process can be said to be an advantageous technology in terms of improving the properties of organic waste as a solid-recovered fuel product.

  • Core-shell hydrogel nanocomposite was fabricated by encapsulating a silica-gold nanoshell (SGNS) with poly(N-isopropylacrylamide-co-acrylic acid) (PNIPAM-co-AAc) copolymer. The oleylamine-functionalized SONS was used as a nanotemplate for the shell-layer growth of hydrogel copolymer. APS (ammonium persulfate) was used as a polymerization initiator to produce a hydrogel-encapsulated SGNS (H-SGNS). The amounts of NIPAM (N-isopropylacrylamide) monomers were optimized to reproduce the hydrogel-encapsulated SGNS. The shell-layer thickness was increased with the increase of polymerization time and no further increase in the shell-layer thickness was clearly observed over 16 h. H-SGNS exhibited the systematic changes of particle size corresponding to the variation of pH and temperature, which was originated from hydrogen-bonding interaction between PNIPAM amide groups and water, as well as electrostatic forces attributed by the ionization of carboxylic groups in acrylic acid.
