Purification and Characterization of Cyclodextrin Glucanotransferase from Bacillus sp. El (Bacillus sp. E1이 생성하는 Cyclodextrin Glucanotransferase의 정제 및 특성)

  • Bacillus sp. was isolated from soil for its strong activity of cyclodextrin glucanotransferase (CGTase, EC 2.4.1.19). The enzyme was purified by gel filtration and anion exchange column chromatography using FPLC. The purified enzyme exhibited its maximum CGTase activity in the pH range of 6~8 and the temperature range of 50~$70^{\circ}C$. The molecular weight was estimated as 114,000 by SDS-PAGE. The isoelectric point of the enzyme was 4.3. The CGTase of Bacillus sp. E l produced $\beta$-cyclodextrin mainly and did not produce a-cyclodextrin. The product ratio of $\beta$-cyclodextrin to $\gamma$-cyclodextrin was 7:l.

Rhynchosia volubilis Lour. and Beta vulgaris Modulate Extracts Regulate UV-Induced Retinal Pigment Epithelial Cell and Eye Damage in Mice (약콩, 비트 추출물의 자외선에 의한 망막 상피세포와 마우스의 눈 손상 조절 효능)

  • Ultraviolet (UV)-induced damage plays a major role in ocular diseases, such as cataracts and retinal degeneration. UV irradiation can generate free radicals including reactive oxygen species (ROS), which are known to cause lipid peroxidation of cellular membranes. It has also been shown that UV can damage DNA directly and induce apoptosis. Rhynchosia volubilis Loureiro (the small black bean or yak-kong, RV) and Beta bulgaris (beet, BB) are used as health supplements. In this study, we explored the protective effects of RV and BB against UVA-induced damage in human pigment epithelial (ARPE-19) cells and in mice. RV and BB mixture and their effective constituents (cyanidin, delphidin, petunidin glycosides) improved cell viability and suppressed intracelluar ROS generation. Phosphorylation of p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), Erk1/2 was analyzed by immunoblotting. RV and BB mixture inhibited UVA-induced phosphorylation of p38 MAPK, JNK, Erk1/2 in APRE-19 cells. RV and BB treatment also showed protective effects on ocular damage in UVA-irradiated mice by increasing the levels of endogenous antioxidants such as superoxide dismutase and glutathione. RV and BB have the potential to be used in a range of ocular diseases and conditions, based on in vitro and in vivo study.

In Vitro Growth-inhibiting Effects of Leaf Extracts from Pinus Species on Human Intestinal Bacteria

  • Methanol extracts of leaves from 15 Pinus species belonging to the family Pinaceae were tested for their in vitro growth-inhibiting activities against 10 bacteria commonly found in the gastrointestinal tracts of human, using impregnated paper disk methods. The inhibitory activities varied with both bacterial strain and Pinus species used. At a concentration of 10 mg/disk, a clear growth inhibition was produced from the extracts of Pinus armandii, P. banksiana, P. bungeana, P. densiflora, P. rigida, and P. thunbergii against Clostridium perfringens, whereas all Pinus samples revealed weak or little growth-inhibiting activity against Escherichia coli, Bacteroides fragilis, and Staphylococcus aureus. At 5 mg/disk, the extracts of P. banksiana and P. thunbergii exhibited potent growth inhibition toward C. perfringens. All the extracts except the one from P. densiflora did not adversely affect growth of Bifidobacterium adolescentis, B. longum, B. bifidum, B. breve, B. animalis, and Lactobacillus casei. The growth-inhibiting activity was more pronounced in C. perfringens, as compared to the lactic acid-producing bacteria. These results may be an indication of at least one of the pharmacological activities of these Pinus species.

Synthesis and Electrochemical Characterization of Reduced Graphene Oxide-Manganese Oxide Nanocomposites

  • Nanocomposites of reduced graphene oxide and manganese (II,III) oxide can be synthesized by the freeze-drying process of the mixed colloidal suspension of graphene oxide and manganese oxide, and the subsequent heat-treatment. The calcined reduced graphene oxide-manganese (II,III) oxide nanocomposites are X-ray amorphous, suggesting the formation of homogeneous and disordered mixture without any phase separation. The reduction of graphene oxide to reduced graphene oxide upon the heat-treatment is evidenced by Fourier-transformed infrared spectroscopy. Field emission-scanning electronic microscopy and energy dispersive spectrometry clearly demonstrate the formation of porous structure by the house-of-cards type stacking of reduced graphene oxide nanosheets and the homogeneous distribution of manganese ions in the nanocomposites. According to Mn K-edge X-ray absorption spectroscopy, manganese ions in the calcined nanocomposites are stabilized in octahedral symmetry with mixed Mn oxidation state of Mn(II)/Mn(III). The present reduced graphene oxide-manganese oxide nanocomposites show characteristic pseudocapacitance behavior superior to the pristine manganese oxide, suggesting their applicability as electrode material for supercapacitors.

Application of spacers for increasing OCC solid content in wet pressing process (I) (스페이서 사용에 의한 OCC 압착공정의 고형분 증대 (I))

  • The increase of OCC solid content after wet pressing will save drying energy greatly. We applied spacers, which used to increase draining rate and bulk in fiber furnishes, to increase the OCC solid contents. MDF fibers (fibers for making medium density fiberboard) and diatomaceous earth were used as spacers, and added 10% by weight to the OCC fiber furnish. Application of high wet pressing pressure to the mixed furnish of spacers and refined OCC did not deteriorate bulk and drainage rate, but their solid contents were increased to 0.5-1.5% without loss of compressive strength when compared to those of unrefined OCC, which is the furnish normally used for mill commercial practice. It is believed that the spacers caused the rate of solid content increase faster in the mixed furnish with OCC at high wet pressing pressure area than the unrefined OCC furnish did. Little amount of starch addition (0.5%) to the spacers helped to keep the strength properties.

Synthesis and Antiwear Properties of Ammonium Dithiocarbamate-based Ionic Liquid (I) (암모니움 디티오카바메이트계 이온성 액체의 합성 및 내마모성능 (I))

  • The friction-reducing properties of lubricants containing ionic liquids based on ammonium dithiocarbamate are studied. The ionic liquids are produced through the following two steps: the synthesis of sodium alkyl dithiocarbamates via the substitution reaction of dialkylamine and carbon disulfide and their subsequent conversion into ammonium dithiocarbamate-based ionic liquids through an ion-exchange reaction with a quaternary alkyl ammonium halide salt. The structures of the ionic liquids are characterized by NMR spectroscopy and Fourier transform infrared spectroscopy. The isolated yields of the ionic liquids, which are viscous and pale yellow, are approximately 92%. The Brookfield viscosities and pour points of the ionic liquids are determined. Further, their wear resistances are measured through the four-ball wear test and the Schwingung Reibung Ver-schleiss (oscillation, friction, wear) test. The wear scar diameter of the lubricants containing 1 wt of the quaternary alkyl ammonium dithiocarbamate-based ionic liquids (0.475-0.631 mm) is significantly lower than that of the base oil (0.825 mm), proving that the ammonium dithiocarbamate-based ionic liquids have good friction-reducing characteristics. However, these friction-reducing characteristics fade significantly after long-term storage, owing to the degradation of the ionic liquids.

Effects of enzymatically hydrolyzed fish by-products in diet of juvenile rainbow trout (Oncorhynchus mykiss)

  • Five experimental diets were formulated to evaluate the effects of dietary enzymatically hydrolyzed tuna by-product on growth, non-specific immune responses, and hematology of juvenile rainbow trout (Oncorhynchus mykiss). A basal diet with 50% of fishmeal was used as control (CON) and four other diets replaced 12.5% ($TBB_{12.5}$), 25% ($TBB_{25}$), 37.5% ($TBB_{37.5}$), and 50% ($TBB_{50}$) of fish meal in the CON diet. Juvenile rainbow trout ($4.87{\pm}0.05g$) were randomly distributed into 15 tanks (50 L) and fed 3-4% of wet body weight two times a day. At the end of 7 weeks of feeding trial, weight gain, specific growth rate, feed efficiency, and protein efficiency ratio of fish fed CON diet were significantly higher than those of fish fed $TB_{50}$ diet (P < 0.05). But there were no significant differences among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets (P > 0.05). There were no significant differences in GPT levels among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets. Also, there were no significant differences in lysozyme, superoxide dismutase, glucose, and total protein levels in all experimental diet (P > 0.05). The broken-line analysis indicated that the minimum dietary level of enzymatically hydrolyzed tuna by-product to replace fishmeal could be 29.7% in rainbow trout. These results indicated that the optimum level of dietary enzymatically hydrolyzed tuna by-product could replace greater than 29.7% but less than 37.5% of fishmeal in juvenile rainbow trout diet.

Effect of Cheonggukjang Pills Product Containing Blueberry and Aronia in Mouse Inflammatory Bowel Disease (블루베리와 아로니아를 함유한 청국장 환 제품의 마우스 염증성 장질환 개선 효과)

  • The purpose of this study was to investigate the effect of Cheonggukjang pills with aronia and blueberries on dextran sulfate sodium (DSS)-induced colitis in mice. There have been several reports that Cheonggukjang is effective for intestinal health, but the efficacy of Cheonggukjang containing fruits has not yet been reported. In this study, we showed the effect of cheonggukjang pills with blueberries and aronia (CPBA) on DSS-induced colitis in BALB/c mice. CPBA was obtained from Soonchang Moonokae foods and orally administered once a day for 2 weeks before DSS treatment. Colitis was induced in mice by feeding 5% (w/v) DSS drinking water for 7 days. The results showed that CPBA treatment significantly alleviated DSS-induced disease activity index associated with a decrease in colon length. CPBA improved DSS-induced histological changes and intestinal epithelial barrier integrity in mice colon. In addition, CPBA administration significantly reduced the levels of DSS-mediated interferon-γ and interleukin-6 in serum and tumor necrosis factor-α in colon tissue. Moreover, the gene expression of COX-2 and iNOS, which are factors involved in inflammatory signaling, was significantly reduced by CPBA treatment. These results suggest that CPBA have a protective effect against DSS-induced mice colitis and may be a candidate for colitis treatment.

Analysis of Growth and Flowering of Thymus quinquecostatus Using Smart Farming System (스마트 재배시스템을 활용한 백리향 생장 및 개화 분석)

  • 백리향(Thymus quinquecostatus)은 꿀풀과의 낙엽반관목으로 국내에서 자생하는 허브 식물 중 하나이다. 백리향은 특유의 좋은 향기 및 항산화, 항염증, 항균, 미백 등의 효능을 가지는 각종 폴리페놀 성분을 함유하고 있어 의약품이나 기능성 식품, 화장품의 천연 소재 원료로 활용되고 있다. 국내에서 백리향은 재배 환경에 맞추어 주로 고산지대의 노지에서 재배되고 있다. 노지 재배는 지역, 시기, 기후 등의 외부환경에 영향을 받아서 백리향의 유효성분, 품질 및 생산성을 안정적으로 유지하기 어렵다는 한계를 가지고 있다. 따라서 본 연구에서는 스마트팜 시스템을 활용하여 백리향의 유효 성분 등을 안정적으로 얻기 위한 생장조건을 탐색하기 위해 4종의 Light Emitting Diode(LED) 광원과 4종의 토양 조성에 따라서 백리향의 생육조건을 수행하였다. LED는 white, purple, RGB1, RGB2를 사용하였으며, 토양은 상토:펄라이트 비율(상토, 5:1, 3:1, 1:1)로 조성하여 백리향 묘목을 이식한 뒤 생장과 개화시기를 분석하였다. 재배환경은 백리향 재배지의 기상 데이터를 참고하여 동일하게 설정하였으며, 총 8주 동안 생육상태를 관찰하였다. 연구 결과 백리향 재배 4주차에 일부 개체에서 봉오리가 올라오며 개화를 시작하였으며, 8주차에는 대부분의 조건에서 개화를 관찰할 수 있었다. 백리향의 지상부 면적을 비교한 결과 가장 우수한 생장을 보이는 조건은 토양은 3(상토):1(펄라이트) 비율로 분석되었다. 따라서 이번 연구 결과를 바탕으로 백리향 재배에 스마트팜 농업 기술을 활용한다면 기존 노지 재배 한계를 보완하여 안정적이고 지속적인 백리향을 생산할 수 있을것으로 기대된다.
