Analysis of Crude Fat and Fatty Acid in Collections of Ricinus communis L. (아주까리(비마자) 수집종의 조지방 및 지방산 분석)

  • Forty Ricinus communis collections were obtained from RDA National Agrobiodiversity Center for knowing the possibility of the use as a bio-diesel possibility crop. These are analysis results about crude fat and fatty acid. Gas chromatogram of seed collections analysis showed 6 peaks and retention time of ricinoleic acid was about 17.1 minute. Average oil content of collections were ranged from 44.6 to 49.4% and the difference was between maximum 52.5% and minimum 41.4%. Fatty acid composition was almost unsaturated fatty acid of 97.6% and saturated fatty acid showed low content of 2.4%. Ricinoleic acid was 87.3% and the content of oleic acid and linoleic acid in fatty acid was 4.6%, and 5.2%, respectively. The content of palmitic acid and stearic acid was about 1% and the difference was insignificant. The content of linolenic acid was extremely low as 0.6%.

Biochemical Classification of Coliforms Isolated from Drinking Water (식수에서 분리한 대장균군의 생화학적 성상에 의한 균종별 분포)

  • A total of 136 coliform bacteria isolated from spring water (112 strains) and ground water (24 strains), submitted to Seoul Health and Environmental Research Institute from June to July in 1997, were characterized biochemically and microbiologically. Colonical characteristics of each isolate were also noted, including color and texture on EMB agar. Among the 136 isolates, 50.7% were greenish metallic sheen color, 44.2% were pink and 5.1% were violet. The sixty four percent were smooth, 34.6% were mucoid and 0.7%. were rough. Twenty three bacterial species were identified by IMViC and API 20E test. Among the 136 coliform bacteria known to species, 39 isolates (28.6%) were Escherichia spp., 32 isolates (23.5%) were Klebsiella ssp., 30 isolates (22.1%) were Enterobacter spp., 19 isolates (14.0%) were Serratia spp., 6 isolates (4.4%) were Citrobacter spp., 4 isolates (3.0%) were Kluyvera spp. and 7 isolates (5.1%) were other bacterial species. Strains, which were gas-positive in lactose broth but gas-negative in Kligler Iron Agar were Ent. intermedium, Ser. liquefaciens, Ser. marcescenes and Salmonella arizoae. Strains, which were H2S production were also Kleb. pneumoniae, Kleb. oxytoca, Kleb. ornithinolytica, Ent. sahazahii, Ent. cloacae, Ser. liquefaciens, Ser. fica ria, Cit. freundii and Sal. arizoae. In the present study, most of coliform isolated from spring and ground water were E. coli, Klebsiella spp. and Enterobacter spp. Since coliform with pink colony in EMB agar was isolated as frequent as coliform with greenish metallic sheen colony, coliform with pink colony should be considered as important colony. Our results suggested that new coliform strains may be emerging on the basis of biochemical and microbiological testes.

Analysis on Ventilation Efficiency of Standard Duck House using Computational Fluid Dynamics (전산유체역학을 이용한 표준 오리사 설계안에 대한 환기효율성 분석)

  • In Korea, 69.4 % of duck farms had utilized conventional plastic greenhouses. In this facilities, there are difficulties in controlling indoor environments for raising duck. High rearing density in duct farms also made the environmental control difficult resulting in getting more stressed making their immune system weaker. Therefore, a facility is needed to having structurally enough solidity and high efficiency on the environmental control. So, new design plans of duck house have recently been conducted by National Institute of Animal Science in Korea. As a study in advance to establish standard, computational fluid dynamics (CFD) was used to estimate the aerodynamic problems according to the designs by means of overall and regional ventilation efficiencies quantitatively and qualitatively. Tracer gas decay (TGD) method was used to calculate ventilation rate according to the structural characteristics of duck houses including installation of indoor circulation fan. The results showed that natural ventilation rate was averagely 164 % higher than typically designed ventilation rate, 1 AER ($min^{-1}$). Meanwhile, mechanically ventilated duck houses made 81.2 % of summer ventilation rate requirement. Therefore, it is urgent to develop a new duck house considering more structural safety as well as higher efficiency of environmental control.

Improved Vapor Recognition in Electronic Nose (E-Nose) System by Using the Time-Profile of Sensor Array Response (센서 응답의 Time-Profile 을 이용한 전자 후각 (E-Nose) 시스템의 Vapor 인식 성능 향상)

  • The electronic nose (E-nose) recently finds its applications in medical diagnosis, specifically on detection of diabetes, pulmonary or gastrointestinal problem, or infections by examining odors in the breath or tissues with its odor characterizing ability. The odor recognition performance of E-nose can be improved by manipulating the sensor array responses of vapors in time-profile forms. The different chemical interactions between the sensor materials and the volatile organic compounds (VOC's) leave unique marks in the signal profiles giving more information than collection of the conventional piecemal features, i.e., maximum sensitivity, signal slopes, rising time. In this study, to use them in vapor recognition task conveniently, a novel time-profile method was proposed, which is adopted from digital image pattern matching. The degrees of matching between 8 different vapors were evaluated by using the proposed method. The test vapors are measured by the silicon-based gas sensor array with 16 CB-polymer composites installed in membrane structure. The results by the proposed method showed clear discrimination of vapor species than by the conventional method.

Screening of GLA (γ-Linolenic Acid) from Fungi by Gas Chromatography and Mass Spectroscopy (Gas Chromatograph-Mass Spectrometer를 이용한 γ-Linolenic Acid(GLA) 생성 균주탐색 및 확인)

  • In order to select ${\gamma}-Linolenic$ acid (GLA)-producing fungi, a total of forty-four strains of 4 genera such as Phytophthora, Pythium, Mucor and Rhizopus were obtained from Koran Agricultural Culture Collection (KACC) and then analysed by using GC-FID and GC-MS. GLA was detected on 39 fungal strains, and the highest rate of GLA was found as 24.8% of total fatty acids on Mucor hiemalis f. sp. hiemalis KACC 40264. Total GLA content of Zygomycota was comparatively high - Mucor (14.2%) and Rhizopus (14.3%), whereas that of Oomycetes was low - Phytophthora (3.3%) and Pythium (3.0%). Moreover, total fatty acids of the Zygomycota fungi such as Mucor (15.4 mg/100 ml) and Rhizopus (7.1 mg/100 ml) were higher compared with the Oomycetes such as Phytophthora (2.6 mg/100 ml) and Pythium (4.5 mg/100 ml). Thus, two genera such as Mucor and Rhizopus have higher potential as an useful microbial resource. The total fatty acid content varies even within the strains of the same genus e.g. Mucor. M. blumbeus KACC 40935 showed the highest values on productivity (18.2%) of GLA and total fatty acid contents (50.8 mg/100 ml liquid medium).

  • High thermal efficiency and the ability to use various types of fuel are a few of the many advantages of diesel engines. However, a major disadvantage is that their exhaust emissions are more harmful to humans and the environment than that of conventional engine. Consequently, the provisions of the international emissions standards for diesel engine equipped passenger cars, commercial vehicles, and ships have become more stringent. These standards include the EU Euro 6, the IMO MEPC Tier 3, and the US EPA Tier 4. Ryu et al. published a study that applied fuel additives to two-stroke diesel engines. In this study, a four-stroke diesel engine using diesel oil for a generator is utilized as the test subject, and an experiment is performed to verify whether fuel additive can be used to improve performance and exhaust emissions. In addition, this experimental study presents research results for the application of fuel additives in both two-stroke and four-stroke diesel engines. The experimental results were compared and analyzed by placing an oil-soluble calcium-based organometallic compound in diesel oil. The results confirmed that the addition of fuel additive improved the performance (fuel consumption rate, exhaust gas temperature) and exhaust emissions (NOx, CO) of the diesel engine.

  • Soil erosion is often extreme in Korea due to high rainfall intensities and steep slopes, and climate change has also increased the risk of erosion. Despite its significane, erosion-induced soil organic carbon (SOC) emission and water resource loss are not well understood, along with the lack of an integrated surface soil erosion protection policy. Therefore, to design adequate protection policies, land users, scientists, engineers and decision makers need proper information about surface soil and watershed properties related to greenhouse gas emission potential and water conservation capability, respectively. Assuming the total soil erosion of $346Tg\;yr^{-1}$, soil organic matter (SOM) content of 2% (58% of SOM is SOC), and mineralization rate of 20% of the displaced carbon, erosion-induced carbon emission could reach $800Gg\;C\;yr^{-1}$. Also the available water capacity of the soil was estimated to be 15.8 billion tons, which was 14 times higher than the yearly water supply demand in Seoul, Korea. Therefore, in order to prevent of soil erosion, this study proposes a three-stage plan for surface soil erosion prevention: 1) classification of soil erosion risk and scoring of surface soil quality, 2) selection of priority areas for conservation and best management practices (BMP), and 3) application of BMP and post management.

  • Biodiesel is estimated to be the best recycling energy source as an alternative fuel for transportation vehicles which represents the biggest share of greenhouse effect gas exhausts. Thus, in order to widely expand use of biodiesel and to enhancement its reliability, studies on quality improvement of biodiesel is needed. In this study, we have produced biodiesel(BD100, BD20) through esterification reaction using raw material of waste frying oil and analyzed compatibility with 24 items of quality criteria. As waste frying oil has high contents of unsaturated fatty acid such as Oleic acid, Linoleic acid and Linolenic acid, it is confirmed that there is no problem in using the same as a raw material of biodiesel. The result of analyzing the quality criteria items of biodiesel showed that it satisfied all the quality criteria except the oxidation stability of BD100, which was 2 hours, fatty acid methyl ester of BD20, which was 18.6w% and the filter plugging point, which was $-5^{\circ}C$. We believe that it will contribute to improved utilization of waste resources as alternative energy if studies on technology to improve quality of some items are provided.

  • Recently, as the interest in the accelerated global warming and the food shortage problem is increased, the concerns for microalgae as photosynthetic microorganisms are also increased. Specially, photosynthetic microalgae, Spriulina platensis have been an attractive source for $CO_2$ gas fixations and for a vast array of valuable nutritious compounds. In this paper, to culture the microalgal Spirulina platensis NIES 39 in a batch culture with high mass, optimal conditions for the culture temperature, initial pH, light intensity and concentration of carbon and nitrogen, were tested. At the most favorable culture condition, $35^{\circ}C$, initial pH 9.5, 4500 lux and carbon and nitrogen concentration of 16.8 g/L $NaHCO_3$ and 2.5 g/L $NaNO_3$, the excellent yields of 2.10 g/L biomass and 29.53 mg/L chlorophyll were obtained.

Quantitative Analysis of Coumarins from Angelica gigas Using $^1H$-NMR

  • $^1H$-Nuclear magnetic resonance (NMR) spectrometry was applied to the quantitative analysis of coumarins in the roots of Angelica gigas without any chromatographic purification. The experiment was performed by the analysis of each singlet germinal methyl, which was well separated in the range of 1.0-2.0 ppm in the $^1H$-NMR spectrum. The quantity of the compounds was calculated by the ratio of the intensity of each compound to the known amount of internal standard (dimethyl terephthalate). These results were compared with the conventional gas chromatography (GC) method. The contents of decursin and decursinol angelate in A. gigas were determined $1.98{\pm}0.07$, $1.13{\pm}0.08%$ in quantitative $^1H$-NMR method and $2.06{\pm}0.24$, $1.17{\pm}0.24%$ in GC method, respectively. The advantages of quantitative $^1H$-NMR analysis are that can be analyzed to identify and quantify, and no reference compounds required for calibration curves. Besides, it allows rapid and simple quantification for coumarins with an analysis time for only 10 min without any preprocessing.
