There are many of anaerobic culture methods and equipments for isolation and cultivation of anaerabic bacteria, but most of these methods are used without pre-reduced media. Gas-jet method is a recommend. able method for the culture of anaerobes, resently developed. Bacteriological studies were experimented of liver abscess of cattle by the use of gas. jet method. The results were summarised as follows; 1. Gas-jet method for anaerobic culture are expedient for the making of pre-reduced media, maintaining of oxygen free condition in the culture tube, picking of bacteria from colony and colony counting etc. 2. A 121 strains of facultative anaerobic and anaerobic bacteria were isolated from liver abscess of 27 head of cattle, and the isolated anaerobic bacteria were as follows. Peptostreptococcus spp. 7 strains Acid aminococcus fermentans 1 Veillonella spp. 1 Bacterioides spp. 6 Bifidobacterium spp. 4 Arachinia propionica 2 Lactobacillus spp. 4 Propionibacterium acnes 1 3. Liver abscess were infected with many of bacteria, about $10^3-10^9$ numbers per gram of abcessed tissue. Almost of abscess were mixed infection of various bacterial species rather than simple species.
Schizandrae Fruits has been used as a traditional Oriental medicine for treatment of many stress-induced diseases. In the present study, we investigated inhibitory activity of enzymatic hydrolysate of Schizandrae fructus (SC-EX) in growth of tested intestinal microorganism and activity of bowel inflammation related enzyme. SC-EX was added to the proteose peptone-yeast extract-fildes (PYF) media to investigation the effect on the growth of type culture of intestinal microorganism. The growth of lactic acid bacteria such as Bifidobacterium species and Lactobacillus species was accelerated by more than 3% concentration of SC-EX. But, growth of harmfulness bacteria such as E.coli, Clostridium sp. Staphylococcus sp. Streptococcus sp. was inhibited by more than 3% concentration of SC-EX. Also, SC-EX was exhibited a concentration-dependent inhibitory activity of the bowel inflammation related enzymes. The SC-EX was showed 76% and 92% inhibitory activity of 5-lipoxygenase and cyclooygenase at 5% additional concentration respectively. Our results indicated that SC-EX may possess improvement effect on the intestinal flora and Anti-inflammatory effect on the bowel.
Microbial dysbiosis in the gut is associated with human diseases, and variations in mucus alter gut microbiota. Therefore, we explored the effects of mucin on the gut microbiota using a community of 19 synthetic gut microbial species. Cultivation of these species in modified Gifu anaerobic medium (GAM) supplemented with mucin before synthetic community assembly facilitated substantial growth of the Bacteroides, Akkermansia, and Clostridium genera. The results of 16S rRNA microbial relative abundance profiling revealed more of the beneficial microbes Collinsella, Bifidobacterium, Ruminococcus, and Lactobacillus. This increased acetate levels in the community cultivated with, rather than without (control), mucin. We identified differences in predicted cell function and metabolism between microbes cultivated in GAM with and without mucin. Mucin not only changed the composition of the gut microbial community, but also modulated metabolic functions, indicating that it could help to modulate microbial changes associated with human diseases.
26종 콩과식물 종실의 메탄올 조추출물의 Bifidobacterium adolescentis, B. longun, B. bifidum, Lactobacillus casei에 대한 생육촉진 및 생육억제 활성을 탄소원 배지와 비탄소원 배지를 이용하여 in vitro 혐기조건하에서 각각 분광학적 및 여지확산법으로 검정한 결과 생육반응은 공시세균과 콩의 종류에 따라 달리 나타났다. 콩과식물 종실중, 대두(밝은 청색 종실)와 땅콩 (짙은 갈색 종실) 추출물은 유산균의 생육을 촉진하였는데, 이러한 결과로 부터 이들 콩과식물 종실에 탄소원과 비탄소원의 비피더스 인자의 존재를 알 수 있었다. 공시 유산균 중에서 L. casei에서 가장 현저한 생육측진 반응을 나타내었다. 또한, 모든 콩과식물 종실시료는 상기 유산균의 생육을 저해하지 않았다.
In this study, lactic acid bacteria were isolated from 21 top-selling probiotic products on Korean market and their antimicrobial resistance were analyzed. A total 152 strains were claimed to be contained in these products and 70 isolates belonging to three genera (Bifidobacterium, Lactobacillus, and Lactococcus) were obtained from these products. RAPD-PCR showed diversity among isolates of the same species except for two isolates of Lacticaibacillus rhamnosus from two different products. The agar dilution method and the broth dilution method produced different MICs for several antimicrobials. With the agar dilution method, five isolates (three isolates of Bifidobacterium animalis subsp. lactis, one isolate of B. breve, one isolate of B. longum) were susceptible to all nine antimicrobials and 15 isolates were multi-drug resistant. With the broth microdilution method, only two isolates (one isolate of B. breve and one isolate of B. longum) were susceptible while 16 isolates were multi-drug resistant. In this study, only two AMR genes were detected: 1) lnu(A) in one isolate of clindamycin-susceptible and lincomycin-resistant Limosilactobacillus reuteri; and 2) tet(W) in one tetracycline-susceptible isolate of B. longum B1-1 and two tetracycline-susceptible isolates and three tetracycline resistant isolates of B. animalis subsp. lactis. Transfer of these two genes via conjugation with a filter mating technique was not observed. These results suggest a need to monitor antimicrobial resistance in newly registered probiotics as well as probiotics with a long history of use.
Recent developments in the application of molecular biology to food grade lactic acid bacteria (LAB) have shown that it could be feasible to engineer metabolic pathways to either enhance specific metabolic fluxes or to divert metabolites for the production of different or new end products. This engineering requires detailed knowledge of enzymes involved in metabolism and regulation within the targeted organism but little works have been done in this area. During biochemical and molecular characterisation of lactic bacterial enzymes, some of probiotic Lactobacillus and Bifidobacterium species were found to be very useful for food, nutraceutical and pharmaceutical industries. The enzymes are usually intracellular and the yields are very low to be useful for industrial applications. Among many enzymes and proteins of lactic bacteria studied, some of our gene cloning achievements have contributed to overproduction of lactic bacterial enzymes such as peptidases, esterases, lactases, bile salt hydrolases and linoleate isomerases for foods and nutraceuticals.
This study was carried out to survey the effect of Jujube extract on the growth of 3 strains of lactic acid bacteria(LBA) starter cultures in the MRS broth by the addition of 0, 0.1, 0.5, 1 and 2% extract The pH, titratable acidity and OD of LAB were investigated in order to get fundamental knowledge for the development a new product. The effects of Jujube extract on the growth of LAB were variable depending upon the LAB species and concentrations of Jujube extract significantly (p<0.05). In the results, Jujube extract enhanced the acid production and propagation by the 3 LAB strains with increasing concentration of Jujube extracts until 2.0% added was to the MRS broth medium (p<0.05). Addition of Jujube extract markedly stimulated the acid production and propagation of Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium longum.
Probiotics are traditionally defined as viable microorganisms that have a beneficial effect in the prevention and treatment of pathologic conditions when they are ingested. Although there is a relatively large volume of literature that supports the use of probiotics to prevent or treat intestinal disorders, the scientific basis behind probiotic use has only recently been established, and clinical studies on this topic are just beginning to get published. Currently, the best studied probiotics are lactic acid bacteria, particularly Lactobacillus and Bifidobacterium species. Other organisms used as probiotics in humans include Escherichia coli, Streptococcus sp., Enterococcus sp., Bacteroides sp., Bacillus sp., Propionibacterium sp., and various fungi, and some probiotic preparations contain more than one bacterial strain. Probiotic use for the prevention and treatment of antibiotic-associated diarrhea caused by Clostridium difficile induced intestinal disease as well as for other gastrointestinal disorders has been discussed in this review.
Twelve strains of Lactobacillus acidophilus isolated from feces of human or animal sources were tested for probiotic properties such as cholesterol assimilation, bile and acid tolerances, and CLA production. Although the cultures showed some variation with respect to each test, the 12 strains could be classified into 3 groups based on their ability to assimilate cholesterol. The cholesterol assimilation showed positive correlation with bile tolerance and negative correlation with acid tolerance. The cholesterol assimilation of L. acidophilus strains may not be related to the deconjugation activity, but may in fact be attributed to its bile tolerance. CLA production by lactic acid bacteria (LAB) exhibited a wide variation that ranged from 2.69 to 7.64 mg/g fat. CLA production of Bifidobacterium longum ATCC 15707 was the highest among the LAB tested, but there was no evidence for differences in CLA production between genus and species.
프로바이오틱 박테리아는 면역력 활성 조절, 콜레스테롤 수치 억제, 유당내성 강화, 항종양 활성 등의 다양한 생리활성 기능으로 건강 증진 효과가 있는 것으로 보고되고 있다. 프로바이오틱 박테리아는 일단 섭취하게 되면 위장관을 통과하는 동안 산도가 낮거나 단백질분해 효소가 많은 열악한 환경에서 생존해야 하며 프로바이오틱 효과를 발휘하기 위해 증식해야 한다. 이중 코팅 기술은 펩타이드와 다당류의 이중코팅으로 섭취된 프로바이오틱 박테리아를 열악한 조건으로부터 보호하기 위해 개발되었다. 본 연구에서는 이중코팅 된 4종의 비피도박테리움 혼합물의 생존 안정성을 평가하기 위해 코팅이 되지 않은 비피도박테리움 혼합물과 담즙, 산 저항성 및 열 안정성을 비교 평가하였다. 이중 코팅 된 균주와 코팅이 되지 않은 균주를 산과 담즙 조건 및 $40^{\circ}C$에 노출 시킨 후 한천배지에 배양하여 생존생육 세포수를 측정하였으며, BacLigtht 키트를 이용하여 염색 한 후 유세포 분석기를 이용하여 생균과 사균의 세포수를 평가하였다. 이중코팅 된 균주 혼합물의 경우 코팅이 되지 않은 균주 혼합물 보다 산, 담즙 내성이 더 높았으며, 열 안전성 또한 코팅 되지 않은 균주 혼합물보다 높은 것으로 나타났다. 이 같은 결과들로 이중코팅 기술은 프로바이오틱 박테리아의 안정성 및 섭취 후 위장관 트랙을 통과하는 동안 균주의 생존률을 향상시킬 수 있음을 확인하였다.
