  • Microstructural analysis of a (${\alpha}+{\beta}$) Ti alloy was investigated to consider phase transformation in each step of the thermo-mechanical process using by SEM and TEM EDS. The TAF (Ti-6Al-4Fe) alloy was thermo-mechanically treated with solid solution at $880^{\circ}C$, rolling at $880^{\circ}C$ and annealing at $800^{\circ}C$. In the STQ state, the TAF microstructure was composed of a normal hcp ${\alpha}$ and metastable ${\beta}$ phase. In a rolled state, it was composed of fine B2 precipitates in an ${\alpha}$ phase, which had high Fe segregation and a coherent relationship with the ${\beta}$ matrix. Finally, in the annealing state, the fine B2 precipitates had disappeared in the ${\alpha}$ phase and had gone to the boundary of the ${\alpha}$ and ${\beta}$ phase. On the other hand, in a lower rolling temperature of $704^{\circ}C$, the B2 precipitates were more coarse in both the ${\alpha}$ and the boundary of ${\alpha}$ and ${\beta}$ phase. We concluded that microstructural change affects the mechanical properties of formability including rolling defects and cracks.

  • The purpose of this study was to determine the dose effects of taurine supplementation and effects of taurine depletion in diabetic rats. Sprague-Dawley male rats were fed the purified diets and supplemented with 1, 2 or 3% taurine in drinking water for 7 weeks(E1, E2 and E3, respectively). To induce taurine depletion, rats were treated with 5% $\beta$-alanine in drinkeng water (EA). Diabetes was induced by streptozotocin injection(50mg/kg B.W) after 3 weeks. Taurine-depleted diabetic rats(EA)had significantly lower body weight compared to those of nondiabetic(CO), nontaurine-supplemented diabetic(E0) and taurine-supplemented diabetic rats(E1, E2 and E3). E0 had significantly higher food intake compared to that of CO and EA. E1, E2 and E3, however, had significantly lower food intake compared to that of E0, and E3 had significantly lower food intake compared to that of E1 and E2. E0 had significantly higher water intake compare to that of CO and EA. EI had significantly lower water intake compared to that of E0. Blood glucose concentration of E0 was significantly increased compared to that of CO and EA. E2 and E3 had significantly lower blood glucose concentration compared to E0 and E1. Also there was a dose effect in blood glucose concentration between E2 and E3. The total cholesterol and LDL-cholesterol concentrations of EA were significantly increased compared to those of other groups. Therefore, it may be suggested that taurine supplementation is necessary for diabetes in order to prevent diabetic complications such as cardiac vascular diseases.

  • The growth-inhibiting effects of Pinus densiflpora leaf-derived materials on nine human intestinal bacteria were investigated using the impregnated paper disk method, and their activities were compared with those of 13 commercially available terpenes. The biologically active constituent of the extract of P densiflora leaf was characterized as the monoterpene (1R)-(+)-$\alpha$-pinene by various spectroscopic analyses. Responses varied according to bacterial strain, chemicals, and dose. At 10 mg/disk, limonene and (1R)-(+)-$\alpha$-pinene strongly inhibited the growth of Clostridium perfringens, Staphylococcus aureus, and Escherichia coli, without adverse effects on the growth of five lactic acid-bacteria (Bifidobacterium adolescentis, B. bifidum, B. longum, Lactobacillus acidophilus, and L. casei). Little or no inhibition against seven bacteria was observed with anethole, borneol, camphor, caryophyllene, 1,8-cineole, estragole, linalool, and $\alpha$-terpineol. Structure-activity relationship revealed that (1R)-(+)-$\alpha$-pinene had more growth-inhibiting activity against C. perfringens than (1R)-(+)-$\beta$-, (1S-(-)-$\alpha$-, and (1S-(-)-$\beta$-pinenes. Furthermore, the growth-inhibition against L. casei was much more pronounced in (1R)-(+)-$\beta$- and (In-(-)-$\beta$-pinenes than (1R)-(+)-$\alpha$- and (1S)-(-)-$\alpha$-pinenes. These results indicate that the (+)-$\alpha$ form seems to be required against C. perfringens and $\beta$ form against L. casei for growth-inhibiting activity. Morphologically, most strains of C. perfringens were damaged and disappeared at 5 and 2 mg/disk of (1R)-(+)-$\alpha$-pinene. Morphological study revealed that (1R)-(+)-$\alpha$-pinene had more growth-inhibiting activity against C. perfringens than (1R)-(+)-$\beta$-, (1S)-(-)-$\alpha$-, and (1S)-(-)-$\beta$-pinenes. As naturally occurring growth-inhibiting agents, the Pinus leaf-derived materials described above could be useful preventive agents against diseases caused by harmful intestinal bacteria such as clostridia.

  • The purpose of this study was to determine the effect of ${\beta}$-mannanase supplementation on digestible energy (DE) and metabolizable energy (ME) contents of copra expellers (CE) and palm kernel expellers (PKE) fed to pigs. Six barrows with an initial body weight of 38.0 kg (standard deviation = 1.5) were randomly allotted to a $6{\times}6$ Latin square design with 6 dietary treatments and 6 periods. Six experimental diets were prepared in a $3{\times}2$ factorial treatment arrangement with 3 diets of a corn-soybean meal-based diet, a CE 30% diet, and a PKE 30% diet and with 2 concentrations of supplemental ${\beta}$-mannanase at 0 or 2,400 U/kg. All diets had the same proportion of corn:soybean meal ratio at 2.88:1. The marker-to-marker procedure was used for fecal and urine collection with 4-d adaptation and 5-d collection periods. No interactive effects were observed between diet and ${\beta}$-mannanase on energy digestibility and DE and ME contents of experimental diets. However, diets containing CE or PKE had less (p<0.05) DE and ME contents compared with the corn-soybean meal-based diet. The DE and ME contents in CE and PKE were not affected by supplemental ${\beta}$-mannanase. Taken together, we failed to find the effect of ${\beta}$-mannanase supplementation on energy utilization in CE and PKE fed to pigs.

  • The molecular weight of the purified $\beta$-galactosidase of Penicillium sp. was estimated to be 130000 by both Sephadex G-200 gel filtration and SDS-polyacrylamide del electrophoresis. The SDS-electrophoresis gave two protein bands corresponding to the two molecular weights of 130000 and 70000. These results indicated that the enzyme consisted of two probably identical subunits which had a molecular weight of 70000. The optimum pH of the enzyme activity was 4.7 and maximum activity appeared at 5$0^{\circ}C$. The stable pH range for the enzyme was from 4.5 to 7.0. The purified $\beta$-galactosidase had no metal ion requirement for its activity or stability. The enzyme activity was inhibited by C $u^{++}$(1mM)and galactose (100mM). The hydrolysis of lactose in 5% lactose solution, pasteurized milk and 10% skim milk solution were 69.5%, 88.7% and 72.6% after 4 hr incubation at 5$0^{\circ}C$, when 10 units of $\beta$-glucosidase were used per $m\ell$ of the substrate solutions.s.

  • The hot-water extract of fruiting bodies in Fomitella fraxinea had potent anti-complementary activities. After fractionation of water-soluble polysaccharides by DEAE-Sephadex A-25 column chromatography, major anti-complementary activity was concentrated into the FF-AP1 among three polysaccharides (FF-NP, FF-AP1, FF-AP2). FF-AP1 was fractionated into $FF-AP1{\alpha}$ and $FF-AP1{\beta}$ obtained from the adsorbed fraction and unadsorbed fraction by affinity chromatography using a ConA-sepharose 4B column, respectively. $FF-AP1{\beta}$, which exihibited the highest anti-complementary activities had an IR absorption peak of $890cm^{-1}$, and a M.W. of about 15,000 (gel filtration). Anti-complementary activity of FF-AP1 decreased greatly by pronase treatment and periodate oxidation. $FF-AP1{\beta}$ responsible for potent anti-complemenary activities of Fomitella fraxinea was an acidic protein-containing heteroglycan consisted of 48% glucose, 13% mannose, and 12% galactose as major component sugars, 9.6% protein, 6% uronic acids.

  • An agar-degrading bacterium was isolated from red seaweed (Gelidium amansii) on a natural seawater agar plate, and identified as Saccharophagus sp. AG21. The ${\beta}$-agarase gene from Saccharophagus sp. AG21 (agy1) was screened by long and accurate (LA)-PCR. The predicted sequence has a 1,908 bp open reading frame encoding 636 amino acids (aa), and includes a glycosyl hydrolase family 16 (GH16) ${\beta}$-agarase module and two carbohydrate binding modules of family 6 (CBM6). The deduced aa sequence showed 93.7% and 84.9% similarity to ${\beta}$-agarase of Saccharophagus degradans and Microbulbifer agarilyticus, respectively. The mature agy1 was cloned and overexpressed as a His-tagged recombinant ${\beta}$-agarase (rAgy1) in Escherichia coli, and had a predicted molecular mass of 69 kDa and an isoelectric point of 4.5. rAgy1 showed optimum activity at $55^{\circ}C$ and pH 7.6, and had a specific activity of 85 U/mg. The rAgy1 activity was enhanced by $FeSO_4$ (40%), KCl (34%), and NaCl (34%), compared with the control. The newly identified rAgy1 is a ${\beta}$-agarase, which acts to degrade agarose to neoagarotetraose (NA4) and neoagarohexaose (NA6) and may be useful for applications in the cosmetics, food, bioethanol, and reagent industries.

  • Purpose: To evaluate the influence of experiences of witnessing tobacco advertising and preferences of tobacco companies' social responsibility (CSR) on current and future smoking intentions in adolescents. Methods: Data were collected from 700 adolescents living in Korea representing 7 metropolitan cities and 8 provinces in the method of proportional random sampling. The Gallup's data collection system was adopted using 1:1 face to face interviews. The witness of tobacco advertising was determined by 'yes', or 'no' responses to three survey items and to four survey items for preferences of tobacco CSR. For statistical analysis, ${\chi}^2$ test, Fisher's exact test and logistic regression were used. Results: 98.4% of the adolescents had experiences of witnessing tobacco advertising. POP (OR=103.44, 95% CI: 8.22~1301.45) and magazine advertisement (OR=6.07, 95% CI: 1.34~22.58) had a significant effect on their current smoking. Also, POP (${\beta}$=.24, p<.001) and movie advertisement (${\beta}$=.42, p<.001) had a significant effect on their future smoking intentions. As for the preferences of tobacco CSR, cultural-art support activities (${\beta}$=.15, p=.025) and environmental purifying campaigns (${\beta}$=-.15, p=.034) had a significant effect on their future smoking intentions. Conclusion: Witness of tobacco advertising and positive responses to tobacco CSR are associated with current and future smoking intentions in adolescents.

  • The grain and agronomic characteristics of Korean barley cultivars were investigated with respect to ethanol yield. Test weight, grain yield, and starch yield showed noticeable variation among the cultivars. Grain yields were higher in covered barley and non-waxy barley. Starch yield was higher in non-waxy barley than waxy barley. Protein, ${\beta}$-glucan, and starch content of tested cultivars ranged in 10.0-12.9%, 4.4-7.5% and 49.7-65.3%, respectively. Naked barley cultivar had higher starch content than covered barley cultivar. However, covered barley had high starch yield because it has higher grain yield than naked barley. Covered barley cultivar had higher husk content, ranging 7.6-14.0%, than that of naked barley cultivar, ranging 5.3-8.0%. Starch content was positively correlated with amylose content, test weight, ethanol yield and negatively correlated with protein, husk, ${\beta}$-glucan content. Ethanol yield per ton was positively correlated with starch content, but negatively correlated with husk content. Ethanol yield per hectare was positively correlated with starch yield, grain yield, grain weight and negatively correlated with protein, test weight. From this research, the important characteristics of barley cultivar as a bioethanol producing material were starch content and grain yield. Optimum barley genotype was non-waxy naked barley that had low protein, ${\beta}$-glucan, husk content, and high starch content and grain yield.
