The Effect of Glucose and Glucose Transporter on Regulation of Lactation in Dairy Cow

  • Glucose is universal and essential fuel of energy metabolism and in the synthesis pathways of all mammalian cells. Glucose is the one of the major precursors of lactose synthesis using glycolysis result in producing milk fat and protein. During the milk fat synthesis, lipoprotein lipase (LPL) and CD36 are required for glucose uptake. Various morecules such as acyl-CoA synthetase 1 (ACSL1) activity of acetyl-CoA synthetase 2 (ACSS2), ACACA, FASN AGPAT6, GPAM, LPIN1 are closely related with milk fat synthesis. Additionally, glucose plays a major role for synthesizing lactose. Activations of lactose synthesize enzymes such as membranebound enzyme, beta-1,4-galactosyl transferase (B4GALT), glucose-6-phosphate dehydrogenase (G6PD) are changed by concentration of glucose in blood resulting change of amount of lactose production. Glucose transporters are a wide group of membrane proteins that facilitate the transport of glucose over a plasma membrane. There are 2 types of glucose transporters which consisted facilitative glucose transporters (GLUT); and sodium-dependent transport, mediated by the Na+/glucose cotransporters (SGLT). Among them, GLUT1, GLUT8, GLUT12, SGLT1, SGLT2 are main glucose transporters which involved in mammary gland development and milk synthesis. However, more studies are required for revealing clear mechanism and function of other unknown genes and transporters. Therefore, understanding of the mechanisms of glucose usage and its regulation in mammary gland is very essential for enhancing the glucose utilization in the mammary gland and improving dairy productivity and efficiency.

  • An experiment was conducted to examine the effects of ceramic sheet on concentration of students studies. To demonstrate the improvement in the concentration of study, we obtained functional magnetic resonance imaging (fMRI), which has superior time resolution and measures brain noninvasively by using intrinsic contrast agent. As a result of Brainwave measurement, we could verify the blood flow's activate in the nearby frontal lobe related to memory process and noticeable ratio change in absolute alpha wave and beta wave after the analysis of Brainwave measurement. fMRI ascertains the physiological function of the brain and is being used to prevent the trouble medically that can be caused before and after the operation. For the visibility of cranial nerve network, many researches will be carried out to develope the product which is related to brain like concentration of study.

DEVELOPMENT OF ANIMAL CELL EXPRESSION SYSTEM WHICH CONFERS POSITION-INDEPENDENT AND ENHANCED FOREIGN GENE EXPRESSION

  • In order to develop the novel gene expression system, we introduced new control elements which could influence the foreign gene expression in animal cells. When the foreign genes are introduced into the genome of higher eukaryotic cells, the expressions from these integrated genes are often low and can vary greatly depending on the positions of the integration sites due to the complex nature of the chromatin structures (1). First we screened the various DNA sequence elements which can function as an insulator of gene expression from these position effects and can cooperate with the SV40 enhancer/promoter. Among the several DNA elements from the various sources, we identified the particular DNA element which confers the increased frequency of the positive colonies, assayed by the reporter gene from stable selections indicating significantly reduced position effects. This element also showed the several fold-increased expression level as well as the copy-number dependent expression with host cell specificity. Second we modified the transcription termination element where we introduced the specific terminator in combination with SV40 polyA signal. This modified terminator showed the increased efficiency and the level of the gene expression. By combining these two elements, we made the animal cell expression system and tested successfully for the recombinant protein productions of TGF ${\beta}$-soluble receptor, Antithrombin III, and single chain Pro-Urokinase. Vitamin D Sufficiency: How Should it be Defined and what are its Functional Indicators?

  • It has been more than three decades since the first assay assessing circulating 25 (OH)D in human subjects was performed That publication as well as several that followed it defined 'normal' nutritional vitamin D status in human populations. Recently, the wisdom by which 'normal' circulating 25 (OH)D levels in human subjects were assigned in the past has come under question. It appears that sampling human subjects, who appear to be free from disease, and assessing 'normal' circulating 25 (OH)D levels by plotting a Gaussian distribution is grossly inaccurate. There are many reasons why this method is inaccurate, including race, lifestyle habits, sunscreen usage, age, latitude, and inappropriately low dietary recommendations for vitamin D. For instance, a 400 IU/day. AI for vitamin D is insignificant when one considers that a 10-15 minute whole body exposure to peak summer sun will generate and release up to 20,000 IU vitamin $D_3$ into the circulation. Recent studies, which orally administered up to 10,000 IU/day vitamin $D_3$ to human subjects for several months, have successfully elevated circulating 25 (OH)D levels to those observed in individuals from sun-rich environments. Further, we are now able to accurately assess sufficient circulating 25 (OH)D levels utilizing specific biomarkers instead of guessing what an adequate level is. These biomarkers include intact parathyroid hormone (PTH), calcium absorption, bone mineral density (BMD), insulin resistance and pancreatic beta cell function. Using the data from these biomarkers, vitamin D deficiency should be defined as circulating levels of 25 (OH)D$\leq$30 ng/mL. In certain cases, such as pregnancy and lactation, significantly higher circulating 25 (OH)D levels would almost certainly be beneficial to both the mother and recipient fetus/infant.

Usefullness of Mask Pack Sheets Including Marine Materials: Skin Regeneration Effect (해양소재를 이용한 마스크팩의 유용성 : 피부재생효과)

  • Minerals from marine materials such as deep ocean water and Dead Sea water have been used since ancient times. We made a mask pack sheet including deep ocean water and salt from the Dead Sea and evaluated the function of the mask pack sheet through animal study. Three full-thickness skin defects were made on the backs of Sprague-Dawley rats. The wounds were left untreated in group Con, and mask pack sheets including deep ocean water or deep ocean water and Dead Sea water were used as treatment for 20 min on the skin of animals in groups DP and DDP, respectively. We analyzed the gross, histological and biochemical findings. Groups DDP and DP showed decreases in wound size, as compared to group Con at 7 days after wound infliction. The histological findings revealed that wound healing had progressed further in groups DP and DDP than in group Con, with more rapid collagen deposition and regression of neutrophils. Also, the expression of vascular endothelial growth factor and transforming growth factor ${\beta}1$ were increased in groups DDP and DP compared with those in group Con at 3 days after wound infliction. Mask sheet packs including deep ocean water and Dead Sea salt affected wound healing by reducing the inflammatory phase and stimulated wound contracture by facilitating the deposition of collagen.

  • The chemistry of 5-methyl-4-imidazolecarboxylic acid ester synthesis involves three distinct reaction steps. Of these the rate of formation of diketone was found to be a function of oxime and HCl concentration and temperature by the batch experiment. The decomposition of diketone was found to be a slow process whereas temperature was the only variable to affect it significantly. An empirical rate expression for the net formation of diketone fits the experminetal data satisfactorily. The principal objectives of this study are to study the kinetics of the diketone formation reaction and to develop the empirical rate expression.

Antidiabetic Synergetic Effects of Plant Extract-Mixtures in Streptozotocin-Diabetes Rats (STZ으로 유발된 당뇨쥐에 대한 식물추출 혼합물의 항당뇨 상승효과)

  • This study investigates the effects of Psidium guajava L. leaf (Pg), Lagerstroemia speciosa L. leaf (Ls) and mixture A (Pg, Ls, Morus indica L. leaf extract, Pinus densiflora needles extract, Acanthopanax senticosus M. root extract) on streptozotocin (STZ)-diabetes rats. For four weeks, STZ-diabetes rats were fed crystallized extracts of Pg, Ls, and mixture A. Compared to the diabetic control group, extracts of Pg, Ls, and mixture A decreased glucose levels in rats by 20%, 14% and 24% respectively. These extracts also decreased the level of total cholesterol, triglyceride and free fatty acid, compared to the diabetic control group, while effectively increasing levels of insulin and high-density lipoprotein-cholesterol. These results showed that mixture A had greater antihyperglycemic, antihyperlipidemic, and insulin-increasing effects than the Pg and Ls extracts. Mixture A also showed better restoration of damaged beta cell function compared to Pg and Ls extracts. Therefore, it was proved that mixture A provides a beneficial synergistic effect when compared with Pg and Ls extracts used individually.

SD and EEG Evaluation of the Visual Cognition to the Natural and Urban Landscape (SD 및 EEG 기법을 통한 자연 및 도시경관의 시지각적 인지분석)

  • The color and structure of urban constructions is a factor of urban landscape and shows their characteristics. Hence the modern buildings deal with their materials and external appearance as an important factor, making up the urban image. But it was nearby impossible to evaluate the value of visual landscape with objective measuring method. Most of all, it depends on the subjective estimation of a few talented or high educated experts with a sense of beauty. Such kinds of estimation can in some cases include arbitrary interpretations. In relation to this kind of problems, it is tried here in this study to analyse the human response of brain wave pattern (EEG) with use of SD method, while the tested persons watch the urban landscape scenery constructed in a visual reality. The tested persons were 20 adult male and female with no color blindness and intact cognitive function. Light source with color filter was used for color environment in a dark soundproof chamber. The signal of EEG is analysed digitally and grouped into the ${\alpha}$ and ${\beta}$ waves. The result showed that relative power of ${\alpha}$ wave ratio increased in the natural landscape scenery with blue and green color. From these results it was possible to evaluate the human response, which is affected by urban and natural color and structure stimulation and it might be useful as an indicator of visual cognition amenity toward the design of urban construction environment.

Prescription Patterns and Factors Related to the Number of Medications in Chronic Obstructive Pulmonary Disease in Non-elderly Adults (비고령 성인환자의 만성폐쇄성폐질환 약물사용 현황과 영향인자)

  • Background: This study is to investigate the prescription patterns and factors related to the number of medications treating chronic obstructive pulmonary disease (COPD) in patients under 65 years old according to GOLD guidelines. Methods: We retrospectively analyzed the medical records of patients aged 40-64 years with a diagnosis of COPD from January to March 2016. Patients were classified by combined assessment of COPD (grades A, B, C, D) using spirometry, exacerbation history, mMRC, and/or CAT results. We analyzed prescribed medications, treatment options and factors related to the numbers of COPD medications. Results: The total number of prescriptions were 251. About 35.5% of patients were classified as GOLD A, 34.2% as GOLD B, 17.1% as GOLD C and 13.2% as GOLD D. Inhaled bronchodilator was prescribed for 86.9% of patients and the most frequent COPD medication was long-acting muscarinic antagonist (LAMA) followed by inhaled corticosteroids/long acting beta agonist (ICS/LABA). The majority of low risk patients (GOLD A/B) were prescribed a monotherapy with LAMA or LABA. For high risk patients (GOLD C/D), combination treatment with ICS+LAMA+LABA was mostly prescribed. The 21.2% of patients in GOLD D received systemic corticosteroid. The average number of medications per prescription was 3.7, and this number increased with increasing COPD grade, COPD duration and lung function reduction ($FEV_1$, $FEV_1/FVC$). Conclusion: Generally high adherence to GOLD guideline recommendations was reported. Given the progressive nature of the disease, results suggest that closer attention to respiratory symptoms for early detection, diagnosis, and appropriate treatment of COPD is warranted.

Brainwave Activities of the Cognitive Individual Differences in Computerized Arithmetic Addition by Implicit Association Test (컴퓨터 덧셈학습의 인지적 개인차에 대한 암묵적 연합검사를 적용한 뇌파 분석)

  • This research analyzed the brainwave activities and brain hemispherity to find out any implications to design the connections between the activities of the brain function and the computerized arithmetic addition in two difficulty levels: easy: 1-5 vs. hard: 6-9. Thus, in developing the brain based math learning for the computer education by implicit association test(IAT) indicated the significant results for the exclusive brain location and the brain hemispherity on the theta, alpha, low alpha, beta brainwaves by QEEG analysis. The results of this study physiologically supported the theoretical background for the computerized math learning skills as well as the math learning material development. It shows the difficulty levels of math information education and the brain activities on cognitive process of the learner continued on the possible investigation of the brain science.
