Batch Time Course Behaviors of Growth and Berberine Production in Plant cell suspension Cultures of Thalictrum rugosum. (Thalictrum rugosum 식물세포배양에 있어서 시간에 따른 세포성장 및 Berberine 생산의 변화)

  • Batch growth of plant cell suspension cultures of Thalictrum rugosum was studied to clarify the kinetic behaviors. It was found that the product formation was growth associated. The specific growth rate was $0.20-0.25\;day\;^{-1}$/TEX> at the growth phase and the FW/DCW ratio was an interesting parameter which represented the status of the cells or the status of sugar concentration. The cell yield was 0.36 g cells/g sugar. The maximum berberine level was 139 mg/L of which 120 mg/L was intracellular. In terms of the specific content of berberine, the product was 1.10% of dry cell weight. At the growth phase, the relationship between the specific growth rate and sugar concentration was described well by Monod kinetics.

Mathematical Model for Adsorption of Berberine on Encapsulated Adsorbent (캡슬에 고정화된 흡착제에의 Berberine의 흡착에 관한 수학적 모델)

  • A mathematical model using local thermodynamic equilibrium isotherms for adsorption in encapsulated adsorbent is proposed in order to optimize the design parameters in situ bioproduct separation process. The model accurately follows the experimental data on the adsorption of berberine, secondary metabolite produced in Thaictrum rugosum plant cell culture. The adsorption rate on encapsulated adsorbent is compared with that on alginate-entrapped adsorbent. The result shows that the higher loading capacity in encapsulated adsorbent is mainly due to the increase in the maximum solid phase concentration. Based on the adsorption rate and loading capacity, the encapsulated adsorbent would be more useful than the entrapped adsorbent when used in situ bioproduct separation process. Design parameters in situ bioproduct separation process, such as the size of the capsule, membrane thickness, the ratio of capsule volume to bulk volume, the ratio of single capsule volume to total capsule volume and the adsorbent content in the capsule, are evaluated by using the model. The ratio of single capsule volume to total capsule volume is the most effective parameter for adsorption of berberine on encapsulated adsorbent.

Antibacterial Activity of Oriental Medicinal Herb Extracts against Skin Pathogens (한약재 추출물의 피부 염증 유발 세균에 대한 항균활성)

  • The antibacterial activity of methanol extracts from 70 kinds of oriental medicinal herbs on four strains of Staphylococcus aureus and S. epidermidis was investigated. The results showed that C. japonica, C. sappan, R. javanica, R. tanguticum, and S. miltiorrhiza had an antibacterial activity on all the strains used. Among these, C. japonica and R. javanica, which showed excellent antibacterial activity, were extracted with water, ethanol, methanol, and ethyl acetate for further study of antibacterial activities. The results showed that the boiled water extract of C. japonica had the best antibacterial activity. Assuming that the antibacterial activity of C. japonica originated from berberine, which has been reported many times, the berberine content of the boiled water extract of C. japonica was analyzed and the result was 13.88%. To compare the antibacterial activity of berberine with that of other antibiotics, berberine chloride and three other kinds of antibiotics were investigated, which showed that berberine chloride had an antibacterial activity on KCCM 35494 S. epidermidis only when the concentration was higher than 600 mg/l, and it did not show antibacterial activity in the other strains. Based on these results, it was concluded that the antibacterial activity of the boiled water extract of C. japonica on the strains used in this study originated from high concentration of berberine or substances other than berberine. Therefore, identification of the substance will be necessary.

Development and Validation of Analytical Method and Antioxidant Effect for Berberine and Palmatine in P.amurense (황백의 지표성분 berberine과 palmatine의 분석법 개발과 검증 및 항산화 효능 평가)

  • The aim of this study was to develop and validate a simultaneous analytical method for berberine and palmatine, which are representative substances of Phellodendron amurense, and to evaluate the antioxidant activity. We evaluated the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) of analytical methods for berberine and palmatine using high-performance liquid chromatography. Our result showed that the correlation coefficients of the calibration curve for berberine and palmatine exhibited 0.9999. The LODs for berberine and palmatine were 0.32 to 0.35 µg/mL and the LOQs were 0.97 to 1.06 µg/mL, respectively. The inter-day and intra-day precision values for berberine and palmatine were from 0.12 to 1.93 and 0.19 to 2.89%, respectively. The inter-day and intra-day accuracies were 98.43-101.45% and 92.39-100.60%, respectively. In addition, the simultaneous analytical method was validated for the detection of berberine and palmatine. Moreover, we conducted FRAP and NaNO2 scavenging activity assays to measure the antioxidant activities of berberine and palmatine, and both showed antioxidant activity. These results suggest that P.amurense could be a potential natural resource for antioxidant activity and that the efficacy can be confirmed by investigating the content of the berberine and palmatine.

  • This study was conducted to test antifungal activity and to separate bioactive compound using C. japonica extract which was shown the most effective control on the B. berengeriana, G. cingulata and P. expansum. $EC_{50}$ on the test pathogene of C. japonica extract was $400{\sim}500{\mu}g/mL$. Bioactive compound analyzed by TLC and HPLC method was identified as a kind of alkaloids, berberine, and the content was 81.14%.

  • A study was carried out to investigate the possibility of acclimation of Coptis japonica Makino in Korea. Coptis japonica could be grown within 4 years as a medicinal crude drug if it is cultivated in the area of mountain facing north(300m above sea level) with 70% light interception and appropriate soil texture. It is to be noted that the propagation of the plant is more important than the yield of berberine because the berberine contents do not change in different conditions. The results were as follows; 1. The germination rates of the seeds by covering with the pine tree leaves were the highest and averaged 63%. 2. The germination rates of dry seeds were not affected in spite of low temperature and $GA_3$ treatment. 3. The germination rates of the seeds stored in moistured sand were averaged 85%, but in dry state, they were not germinated. 4. The number of leaves and fresh weight of Coptis japonica from the area of mountain facing north were increased significantly. 5. The growth and fresh weight of Coptis japonica were reduced in clay. 6. The best growth was obtained under 70% light interception. It is possible to intercrop between evergreen trees. 7. The foliage dressing did not affect the growth. 8. The number of leaves and fresh weight were increased from the 3rd year, and the average weight of rhizomes was 5.7g in 4th year. 9. The berberine contents of the rhizomes of Coptis japonica under various treatment conditions ranged from 9 to 10%. Coptidis Rhizoma from this study and from Korean market showed much higher berberine contents than those from Japanese market.

A monitoring study of marker contents in the Hwangnyeonhaedok-tang Ex preparations on the market (시판 황련해독탕 Ex제제의 성분함량 모니터링 연구)

  • Objectives : Hwangnyeonhaedok-tang(HNHDT) is the one of well-known traditional herbal formula and has been widely used for the treatment of various diseases in Korea. For the quality improvement of commercial traditional herbal formula, we investigated the quality of prepared HNHDT. Methods: We measured the contents of baicalin, berberine and geniposide in the HNHDT Ex preparations on the market by HPLC and compared with the official notice of ingredients. Results: In 3 preparations of HNHDT covered with the national health insurance(NHI), the content of baicalin was various from 32.83mg to 185.05mg, the total berberine was $12.0{\sim}50.75mg$ and geniposide was $12.57{\sim}44.84mg$. Contents of marker ingredients in NHI Preparations were below the standard and showed a large variation. In 7 non NHI Preparations of HNHDT and the standard(STD) HNHDT manufactured in laboratory, the content of baicalin was $37.16{\sim}115.69mg$, the total berberine was $5.62{\sim}28.43mg$ and geniposide was $10.98{\sim}113.47mg$. But, because composition of non-NHI preparations is different from NHI preparations, every maker's contents were up to the notice for baicalin and berberine. There was newly enforced standard regulations of geniposide in non-NHI preparations, so contents of geniposide was up to that standard only in JI and STD. Conclusions: There was a large variation of contents in the HNHDT Ex preparations on the market and the quality differences were existed between NHI and non-NHI preparations of HNHDT. Further studies for evaluation of efficacy differences and standardization of manufacturing processes are necessary.

Comparison of Hwangbek Dyeability for Cotton, Silk, and Wool from Different Tree Species and Origin (황백의 품종 및 산지에 따른 면, 견, 양모직물에 대한 염색성 비교)

  • This study examined hwangbek dyeability for cotton, silk, and wool from different tree species and origin as well as their berberine and palmatine content. A total of 12 different hwangbek were examined, six purchased directly from different locations of China and six purchased from the Seoul Medicine Market. Hwangbek purchased from China were 3 Kwanhwangbek (Phellodendron amurense Rupr.) from Jilin, Liaoning, Heilongjiang and 3 Chunhwangbek (Phellodendron chinense Schneid.) from Sichuan, Yunnan, and Guangxi. Hwangbek purchased from the Seoul Medicine Market included 3 hwangbek that originated from Korea and 3 hwangbek imported from China. We extracted 2.5g of each hwangbek into 500mL of water for 60 min at $80^{\circ}C$. Dyeing was conducted using the IR dyeing machine for 30min at $55^{\circ}C$ using a 1:100 liquor ratio. Cotton was mordanted using tannic acid followed by potassium antimonyl tartrate trihydrate treatment before dyeing. Berberine and palmatine pigments in hwangbek were identified with the HPLC-DAD-MS instrument and the amount of pigment was quantified using an ion chromatogram. The results indicated that the K/S values of dyed fabrics were highly related to the amount of pigment in hwangbek used for dyeing.

  • In this study, Aftertreatment properties of PCM/Nylon sheath/core fabrics have been determined. Especially, the relationship between finishing property and content including of PCM ratio. Samples of PCM/Nylon fabrics were monitored, separately, with 2% o.w.f solutions of each of the berberine chloride, cetylpyridinium chloride(CPC), benzyldimethylhexadecyl ammonium chloride(BDHAC) and dodecyltrimetyl ammonium bromide(DTAB). Various temperatures and liquor ratio and pH conditions were also studied to optimize aftertreatment properties. Berberine chloride finished sample showed the good color fastness. Cetylpyridinium chloride(CPC) finished sample showed very effective antibacterial properties against Staphylococcus aureus and Klebsiella pneumoniae.

A Study for the isolation of the Berberine-type Alkaloid from Coptidis Rhizoma and for their Antitumor Activities

  • The purpose of this study is the separation of biologically active ingredients from Coptidis Rhizoma which has been widely used as one of oriental herbal medicine for body fever. In this study, berberine-type alkaloids were tested on their biological activities in the aspect of antibacterial, antitumor, anti-herpetic and anti-HIV activity. Contents of five major alkaloids for the various origin of Coptidis Rhizoma were assayed by HPLC. As the results, the content of berberine from Coptis chinensis and Coptis japonica were 6.78% and 7.09%, respectively. The contents of coptisine, jatrorrhizine, berberastine from Coptis chinensis were higher than those of Coptis japonica. The amount of palmatine from both species were almost the same. Surprisingly for antitumor experiment, all compounds have been shown remarkable activity, especially against SNU-l(human stomach cancer) cell line. Among the compounds purified through column chromatography, palmatine, coptisine, and jatrorrhizine inhibited the growth of K-562(human chronic myelogenous leukemia) cell line whereas jatrorrhizine has been shown the effective inhibition of A-549 (human lung) cell line at the same time.
