황련 중 berberine의 추출효율에 관한 연구 (A Study on the Extraction Efficiency of Berberine in Coptidis Rhizoma)

  • 본 연구에서는 황련의 지표물질인 berberine의 추출효율을 측정하기 위하여 추출온도, 추출용매, 추출시간 등의 조건 변화에 따른 최적의 추출조건을 찾았다. $60^{\circ}C$에서 2~3시간 환류하였을 때 추출효율이 가장 좋았으며, 추출용매로는 메탄올이나 아세트산을 사용하였을 때 각각 추출효율이 우수하였다. 이온쌍 추출법에서는 사용되는 sodium lauryl sulfate의 첨가 효과는 추출 효율을 오히려 감소시키는 결과를 나타내었다.

바이칼린 함유생약의 제제화 및 생체이용률 (제 1보): 황금 및 황련 공침물의 확인 및 물리화학적 성질 (Preparation and Bioavailability of Oriental Medicine Containing Baicalin (I) : Identification and Physicochemical Properties of Coprecipitated Product of Scutellariae Radix and Coptidis Rhizoma)

  • Precipitation was formed during the preparation of decoction from a mixture of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine were identified in coprecipitated product and these components were the active ingredients of two herbal medicine. The coprecipitated product was very slightly soluble in water and sparingly soluble in ethanol. The stoichiometric ratio of baicalin and berberine was found to be 1:1. The lipid-water partition coefficients of coprecipitated product were increased more than baicalin and berberine in chloroform, but were decreased in other organic solvents. The content of baicalin and berberine in coprecipitated product, determined by HPLC, were 23.08% and 26.75%, but the content of active ingredients in supernatant were 0.66% and 0.26%, respectively. The dissolution profile of baicalin of coprecipitated product was increased more than extract of Scutellariae Radix in artificial gastric juice, but was decreased in artificial intestinal juice. The dissolution rate of berberine of coprecipitated product was lower than extract of Coptidis Rhizoma in artificial gastric juice and intestinal juice commonly.

Quantitative and Qualitative Analysis of Alkaloids in Coptis chinensis (Coptidis Rhizoma) by LC-DAD and LC-ESI/MS

  • The quality control of natural products is principal key to guarantee the Good Manufacturing Practices (GMP) and Good Clinical Practices (GCP) for the functional food, pharmaceuticals and cosmeceuticals in the industry. In this study, we examined the quantitative analysis of berberine as marker substance of Coptidis Rhizoma by high performance liquid chromatography-photodiode array detector (HPLC-DAD). The HPLC method was validated and met all the requirements for the quality control analysis recommended by FDA and ICH. The berberine was separated on a Xterra $C_{18}$ column ($5{\mu}m$, $4.6{\times}250mm$) using mobile phase consisting of distilled water and acetonitrile with $KH_2PO_4$ (3.4 g) and $Na_2SO_4$ (1.7 g). Calibration curve of berberine has been estimated (y = 42293.47x-41589 with the correlation coefficient 0.9999). The amount of berberine was calculated as 4.25%. And berberastine, palmatine, columbamine, jatrorrhizine, epiberberine, berberine and coptisine in the Coptidis Rhizoma were identified by high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-ESI-MS) method.

  • 본 논문에서는 Thalictrum rugosum 식물세포의 회분식 배양에 있어서 동역학적 특성들을 연구하였다. 본 실험에서 alkaloid의 생성은 일반적인 2차대사 산물의 생성 특성과 달리 세포성장에 비례함이 확인되었다. 세포 성장기의 비증식속도는 $0.20~0.25\;day\;^{-1}이었으며 FW / DCW 비가 배양상태를 나타내는 중요한 인자임을 알수 있었다. Cell yield는 0.36g cells/g sugar이었으며 berberine은 139mg/L까지 생성되었고 그중 120mg/L는 세포내에 존재하였다. 이를 specific content로 계산하면 dry cell weight의 1.10%가 된다. 세포 성장기에서 비증식속도와 당농도간의 관계는 Monod kinetics로 잘 기술될 수 있었다.

  • In oriental medicine, Coptis japonica $M_{AKINO}$ (Ranunculaceae) has been known as bitter stomachics. This plant originally grows wild and has been cultivated in Japan but not in Korea. However, a few years ago, some workers have been trying to cultivate this plant in Korea and have made much progress. Therefore, this . study was made for the purpose of conforming possibility in cultivation of Coptis japonica $M_{AKINO}$ in Korea. The results were as follows: 1) Berberine contents of rhizomes are much more than of K.P.. Difference in contents between Eu Jeon Bu $(4.88{\pm}0.77%)$ and Bo Seong area $(3.62{\pm}0.50%)$ is significant (p<0.05). 2) Berberine contents of fibrous roots of Eu jeong Bu $(0.97{\pm}0.28%)$ area was similar to that of Gang Nam $(0.93{\pm}0.21%)$, Yang Pyung area $(0.91{\pm}0.14%)$ but only contents of Bo Seong area $(0.74{\pm}0.25%)$ is less than that of other area and it's difference is not significant. 3) It was considered that the cultivation of Coptis japonica $M_{AKINO}$ in Korea is possible considering the cultivation results and berberine contents, and also more moderate than south region.

Synergistic Antifungal Activity of Phellodendri Cortex and Magnoliae Cortex against Candida albicans

  • Many studies on plant extracts have been reported for the treatment of candidiasis caused by Candida albicans, a representative fungal infection. This study demonstrates the synergistic antifungal activity of the combination of Phellodendri Cortex and Magnoliae Cortex, previously reported to have antifungal efficacy. Considering the antifungal efficacy and the separation of the active constituents, berberine and magnolol, hot water extraction and carbon dioxide supercritical extraction were selected for Phellodendri Cortex and Magnoliae Cortex, respectively. A combination of 0.55 g/L hot water extract of Phellodendri Cortex and 0.59 g/L carbon dioxide supercritical extract of Magnoliae Cortex showed synergistic antifungal activity. The synergistic antifungal activity of 160 μM berberine and 100 μM magnolol, which are representative antifungal compounds of Phellodendri Cortex and Magnoliae Cortex, respectively, contributes to the synergistic antifungal effect of their extracts. The additive decrease in cellular ergosterol level and the increased antifungal efficacy by extracellular ergosterol suggest that disruption of the biological function of ergosterol in the cell membrane is not responsible for the synergistic antifungal activity of berberine and magnolol. Synergistic cellular release of chromosomal DNA upon mixing berberine and magnolol indicates that disruption of the cellular structure is responsible for the synergistic antifungal effect of berberine and magnolol.

  • Apical to basal transport of organic cations (OCs) such as tributylmethylammonium (TBuMA), triethyimethylammonium (TEMA). 1-methyl-4-phenylpyridinium (MPP), and berberine across Caco-2 cell monolayers was measured to elucidate the intestinal absorption of OCs. Basal to apical transport of MPP and berberine was larger than apical to basal transport and showed temperature dependency and concentration dependency. indicating that MPP and berberine are secreted into the inteslinal lumen.   • The present study was investigated the effect of berberine on the development of behavioral sensitization by morphine, methampethamine, and cocaine. Repeated administration of morphine (10 mg/kg), methampethamine (2 mg/kg), and cocaine (15 mg/kg) produced behavioral sensitization in mice. Pretreatment with berberine (2 mg/kg) did not inhibit methampethamine-and cocaine-induced behavioral sensitization. (omitted)

Antifungal Activity of Chaerophylline and Berberine Hydroxide Isolated from Corydalis Species

  • Chaerophylline and berberine hydroxide, both being alkaloids, isolated from Corydalis chaerophylla and C. longipes respectively were assayed against spore germination of some fungi, e.g. Alternaria solani, A. brassicola, A. brassicicola, Helminthosporium penniseti, Helminthosporium sp., Heterosporium sp., Curvularia penniseti, C. maculens and C. palliscens. While chaerophylline inhibited spore germination of most of the fungi at 1000 ppm, being also effective at 50, 100, 200 and 500 ppm, berberine hydroxide was significantly effective at much lower concentration, i.e., 400 ppm against several fungi. This compound was also effective against some fungi at 50, 100, 150, 200 ppm. There was 100% inhibition of spore germination in several fungi at highest concentration of both the compounds. Some of the fungi showed similar results even at lower concentrations.

천연염색에 관한 연구(6) - 황벽색소의 추출, 정제 및 특성- (A Study on Natural Dyeing(6) - Extract, Purification and Characters of Berberine -)

  • The most effective solvent for extract of dye from amur cork tree was methanol. Two protoberberine alkaloids, berberine and palmatine, were isolated from amur cork tree by Prep-TLC, and the developing solvent was Benzene : AcOEt ; PrOH ; MeOH ; EtNH$_2$(8: 4: 2: 0.5: 0.5). Dyes were stable at a high temperature and there wasn't any change of color at pH 3~9 and with added metal mordants. In adsorption spectra of aqueous solution of berberine and tannin mixture, two isosbestic points$(328^{nm},\;357^{nm})$ were found and the mole fraction of reaction of components was 1:1.
